We compared the immunologic characteristics of mycoplasma pneumoniae-triggered Kawasaki disease (MP-KD) with Kawasaki disease (KD) not associated with mycoplasma pneumoniae (MP), with mycoplasma pneumoniae-triggered Henoch-Schönlein purpura (MP-HSP), and with healthy controls.Complement levels, cellular and humoral immunity were assessed in KD, in MP-KD, in MP-HSP, and in healthy children.Of 622 children with KD, 74 had MP-KD. Complement C3 and CD4/CD8 ratio were significantly increased in MP-KD compared to KD. C3, C4, and the ratio of CD4/CD8 in the MP-KD group were higher than those in the MP-HSP group. Aims Phenazines, such as phenazine-1-carboxylic acid (PCA), phenazine-1-carboxamide (PCN), 2-hydroxyphenazine-1-carboxylic acid (2-OH-PCA), 2-hydroxyphenazine (2-OH-PHZ), are a class of secondary metabolites secreted by plant-beneficial Pseudomonas. Ps. chlororaphis GP72 utilizes glycerol to synthesize PCA, 2-OH-PCA and 2-OH-PHZ, exhibiting broad-spectrum antifungal activity. Previous studies showed that the addition of dithiothreitol (DTT) could increase the phenazines production in Ps. chlororaphis GP72AN. However, the mechanism of high yield of phenazine by adding DTT is still unclear. Methods and Results In this study, untargeted and targeted metabolomic analysis were adopted to determine the content of metabolites. The results showed that the addition of DTT to GP72AN affected the content of metabolites of central carbon metabolism, shikimate pathway and phenazine competitive pathway. Transcriptome analysis was conducted to investigate the changed cellular process, and the result indicated that the addition of DTT affected the expression of genes involved in phenazine biosynthetic cluster and genes involved in phenazine competitive pathway, driving more carbon flux into phenazine biosynthetic pathway. Furthermore, genes involved in antioxidative stress, phosphate transport system and mexGHI-opmD efflux pump were also affected by adding DTT. Conclusion This study demonstrated that the addition of DTT altered the expression of genes related to phenazine biosynthesis, resulting in the change of metabolites involved in central carbon metabolism, shikimate pathway and phenazine competitive pathway. Significance and Impact of the Study This work expands the understanding of high yield of phenazine by the addition of DTT and provides several targets for increasing phenazine production.

Objective Cytomegalovirus (CMV) virulence may depend on genetic variability in several regions of the genome. This study aimed to assess specific CMV genotypes' association with the severity of symptomatic congenital CMV disease at birth. Methods CMV glycoprotein B (gB), glycoprotein N (gN), glycoprotein H (gH), and UL144 strains were identified by nested polymerase chain reaction, restriction fragment length polymorphism, and heteroduplex mobility assay single-stranded conformation polymorphism in 50 infants infected congenitally and 25 asymptomatic infants. Results gN1 (p = 0.010) and UL144-B (p = 0.034) genotypes were associated, by logistic regression, with reduced risk of developing symptomatic congenital CMV infection. gN1 (p = 0.020) and gN3 (p = 0.022) genotypes were associated with reduced risk of severe symptomatic disease. Conversely, gB1 (p = 0.018) was the most virulent genotype and was associated with severe symptoms. Conclusion An association among gB1, gN1, gN3, and UL144-B genotypes of CMV and severity of congenital CMV disease might exist. gB, gN, and UL144 genotypes could be important virological markers of infant infection.

To induce the development of tertiary lymphoid organs (TLO) in a mouse model of melanoma and to evaluate TLO's functions in antitumor immunity.Lymphotoxin-beta receptor (LTβR) was overexpressed in NIH3T3 cells through the lentivirus system and the overexpression efficiency of LTβR in LTβR-NIH3T3 cells was examined. Western blot and qPCR were used to examine the non-canonical nuclear factor (NF)-κB signaling pathway in NIH3T3 cells overexpressing LTβR. B16-OVA melanoma mouse model was constructed to explore the induction of TLO and anti-tumor functions of TLO in LTβR-NIH3T3 cells.LTβR was overexpressed in NIH3T3 cells through the lentivirus system, and flow cytometry showed that the proportion of GFPLTβR-NIH3T3 cells promoted anti-tumor immunity by inducing TLO development, which may provide new perspectives for tumor immunotherapy.

BACKGROUND Apart from the controversy of high glycemic index and glycemic load associated with precancerous cervical lesions, few studies have examined the impact of fasting blood glucose levels on HPV multiple infections. In a population of HPV-positive women with high-grade cervical lesions, we conducted the current study to appraisal the relationship between blood glucose levels and multiple HPV infections. METHODS The present study was a cross-sectional correlative study. A total of 629 participants with a pathologically confirmed HSIL with HPV infection were included from a hospital in China between 2018-1-1 and 2019-12-31. The target independent variable and the dependent variable were glucose level at baseline and multiplicity of HPV respectively. The ratio (OR) and 95% confidence intervals (95% CIs) for the risk of multiple infections were estimated using logistic regression analysis. RESULTS The average age of 560 participants was 44.63 ± 10.61 years old, Result of fully-adjusted binary logistic regression showed glucose was negatively associated with risk of event of multiple infection after adjusting confounders (Odds ratio (OR)=0.84, 95%CI(0.57-1.22). Non-linear relationship was detected between glucose and multiplicity of HPV, with point at 5.4. After adjusting for the full range of variables, the effect sizes and confidence intervals for the left and right sides of the inflection points were 0.379 (0.196, 0.732) and 5.083 (1.592, 16.229), respectively. CONCLUSION In this cross-sectional study, both hypoglycemia and hyperglycemia predicted an increased risk of multiple infections, demonstrating a U-shaped association between blood glucose levels and HPV multiple infections.

Introduction: Henoch-Schönlein purpura (HSP) is an immune-mediated vasculitis, and the formation of immune complexes may be triggered by exposure to Epstein-Barr virus (EBV) infection. Methods: We performed a five-year case-control study to evaluate the epidemiology and clinical characteristics of HSP associated with EBV infection. Results: The incidence of EBV-triggered HSP was 4.2%, while EBV infection in children with HSP was 0.9%; The EBV-triggered HSP cases had a significantly higher frequency of abdominal pain than the MP-triggered HSP group (χ2 = 8.024, p = 0.005); Significant differences were observed in the duration of abdominal pain (Z = -1.935, p = 0.027) between the two groups; C3 (t = 9.709, p < 0.001), IgA (t = 20.39, p < 0.001) and IgG (t = 6.407, p < 0.001) were significantly increased in the EBV infection group than those in the healthy control group. Notably, significantly higher proportion of CD19 (t = 6.773, p < 0.001) and lower proportion of CD56 (t = 11.13, p < 0.001) was found in EBV infection group compared with healthy control group. The IgA level was higher than that of the non-infectious group (t = 2.162, p = 0.032), but their CD4/CD8 ratio (t = 10.070, p < 0.001) and CD56 proportion (t = 2.096, p = 0.037) were significantly lower. Conclusions: Both cellular and humoral immunity were involved in the pathogenesis of EBV-triggered HSP, leading to increased production of inflammatory mediators and immunoglobulins. Those events may cause or promote the development of systemic vessel vasculitis.

Mupirocin, a polyketide antibiotic produced by Pseudomonas fluorescens, is used as a topical antimicrobial treatment to cure various skin infections. Quorum sensing system plays an important role in regulation of mupirocin biosynthesis in P. fluorescens NCIMB 10586. In Pseudomonas, the RpeA/RpeB two-component signal transduction (TCST) system regulates quorum sensing system. However, the influences of the RpeA/RpeB TCST system on mupirocin production or other cell activities have not been studied. In this work, the homologous genes of rpeA and rpeB in P. fluorescens NCIMB 10586 were identified and inactivated in the chromosome, respectively. The deletion of rpeA reduced the mupirocin production from 160 in the wild-type to 21.3 mg/L along with slightly decreased cell growth, while no significant effected on mupirocin production in the rpeB mutant. Next, it was found that the RpeA/RpeB TCST system regulated the biosynthesis of mupirocin by modulating the quorum sensing system. Furthermore, untargeted metabolomics analysis was employed to detect the influences of RpeA on other cell activities modulated by quorum sensing system. Combined with quantitative real-time PCR, the results demonstrated that RpeA also regulated other cell activities including central carbon, amino acids, fatty acids, and purine metabolism. Overall, this study expands the current understanding of the RpeA/RpeB TCST system and provides several targets for increasing yields of mupirocin. KEY POINTS: • In P. fluorescens, the RpeA/RpeB TCST system regulates the biosynthesis of mupirocin. • RpeA modulates the cell activities through effecting the central carbon metabolism.

The precise measurements of energy spectra and anisotropy could help us uncover the local cosmic-ray accelerators. Our recent works have shown that spectral hardening above $200$ GeV in the energy spectra and transition of large-scale anisotropy at $\sim 100$ TeV are of local source origin. Less than $100$ TeV, both spectral hardening and anisotropy explicitly indicate the dominant contribution from nearby sources. In this work, we further investigate the parameter space of sources allowed by the observational energy spectra and anisotropy amplitude. To obtain the best-fit source parameters, a numerical package to compute the parameter posterior distributions based on Bayesian inference, which is applied to perform an elaborate scan of parameter space. We find that by combining the energy spectra and anisotropy data, the permissible range of location and age of local source is considerably reduced. The observation of very-high-energy (VHE, $\rm >10~GeV$) $\gamma$-ray emission from $\rm \gamma$-ray bursts (GRBs), especially in the prompt phase, will provide critical information for understanding many aspects of their nature including the physical environment, the relativistic bulk motion, the mechanisms of particle acceleration of GRBs and for studying Lorentz invariance violation, etc. For the afterglow phase, the highest energy photons detected to date by the imaging atmospheric Cherenkov telescopes extend to the TeV regime. However, for the prompt phase, years of efforts in searching for the VHE emission has yielded no statistically significant detections. A wide field-of-view (FOV) and large effective area above tens of GeV are essential for detecting the VHE emissions from GRBs in the prompt phase. The High Altitude Detection of Astronomical Radiation (HADAR) experiment has such merits. In this paper, we report the estimates of its expected annual GRB detection rate, which are obtained by combining the performance of the HADAR instrument with the theoretical calculations based on a phenomenological model to generate the pseudo-GRB population. The expected detectable gamma-ray signal from GRBs above the background is then obtained to give the detection rate. In the spectral model, an extra component is assigned to every GRB event in addition to the Band function. The results indicate that if the energy of the cutoff due to internal absorption is higher than 50 GeV, the detection rate for GRBs for the HADAR experiment is approximately two or three GRBs per year, which varies slightly depending upon the characteristics of the extra component., Comment: accepted by ApJ

Recent studies have revealed that long non-coding RNAs (lncRNAs) serve important roles in carcinogenesis and that this type of gene may be used as biomarkers in cancer. A high level of lncRNA HOXA distal transcript antisense RNA (HOTTIP) is associated with unfavorable prognosis for patients with ovarian cancer (OC), but the mechanism of HOTTIP involved in OC development remains to be elucidated. The present study aimed to investigate the mechanism of HOTTIP in metastasis-associated OC cell behaviors. HOTTIP levels in ovarian cells were quantified by reverse transcription-quantitative PCR, cell proliferation was analyzed by colony formation assay, and apoptosis was assessed by flow cytometry. Cell migratory and invasive abilities were evaluated by wound healing and Transwell assays, respectively. The expression levels of mitogen-activated protein kinase kinase (MEK)/ERK pathway-associated proteins were detected by western blotting. The results demonstrated that knockdown of HOTTIP in OC cells significantly reduced the phosphorylation levels of MEK and ERK, inhibited the proliferation and invasion of OC cells and promoted their apoptosis. Furthermore, the effects of HOTTIP on cell migration and invasion were partly associated with the epithelial-mesenchymal transition (EMT) process. Proliferation, invasion and EMT of OC cells were enhanced following overexpression of HOTTIP; however, these effects were reversed by the MEK/ERK pathway inhibitor U0126. In conclusion, HOTTIP was demonstrated to promote the proliferation, migration and invasion of OC cells by activating the MEK/ERK pathway. Therefore, HOTTIP may serve as a potential therapeutic target for OC.

As one of the pilot experiments of LHAASO, a hybrid array (HA) covering an area of ${20000 m^{2}}$ was successfully constructed by the end of ${2016}$ at the international Cosmic Ray Observatory at YangBajing (YBJ) in Tibet of China. This array consists of 115 scintillation detectors and 16 underground muon detectors of ${900 m^{2}}$. Using muons information, most of hadronic air showers are rejected at several dozens of TeV energy regime. With the data collected from ${2017}$ to ${2018}$, this work presents preliminary results on diffuse $\gamma$-ray emission from galactic plane. As a result, there has no significant excess of TeV $\gamma$-ray in the galactic plane with the YBJ-HA observation. Therefore, we put a 90% CL upper limit. We foresee that the under-construction LHAASO will have great potential in observing $\gamma$-rays in several dozens of even up to hundreds of TeV energy range.

Conventional cosmic-ray propagation models usually assume an isotropic diffusion coefficient to account for the random deflection of cosmic rays by the turbulent interstellar magnetic field. Such a picture is very successful in explaining a lot of observational phenomena related to the propagation of galactic cosmic rays, such as broken power-law energy spectra, secondary-to-primary ratios, etc. However, the isotropic diffusion presupposition is facing severe challenges from observations. In particular, recent observations on the large-scale anisotropy of TeV cosmic rays show that the dipole direction differs from the prediction of the conventional model. One possible reason is that the large-scale regular magnetic field, which leads to an anisotropic diffusion of cosmic rays, has not been included in the model provided by the public numerical packages. In this work, we propose a pseudo source method to solve the 3-dimensional anisotropic transport equation. Based on a realistic configuration of Galactic magnetic field, both proton energy spectrum and B/C ratio are reproduced with proper transport parameters.

The Water Cherenkov Detector Array (WCDA) is one of the major components of the Large High Altitude Air Shower Observatory (LHAASO). The installation of the first pool of WCDA is already finished and normally operated. Except for the initially designed 8-inch PMTs (big PMTs) of WCDA installed, each of the 900 cells of the first pool was installed another 1.5-inch PMT (small PMTs) to extend the dynamic range, which is called the WCDA dynamic range extension system (WCDA++). The 900 1.5-inch small PMTs work together with WCDA big PMTs and WFCTA and KM2A to measure the energy spectra of cosmic rays with energy from TeVs to 10 PeV. This paper presents the data analysis and simulation of the system, such as the single rate study, the match of single-channel triggered hits of big and small PMTs, the match of cosmic ray events, the core reconstruction, and full Geant4 simulation and fast simulation based on Geant4.

Spectra of cosmic rays (CRs), and particularly their features, contain a lot of important information about the astroparticle physics and the fundamental physics. Realizing that the 4 PeV knee of CR nuclei and the 1 TeV spectral cutoff of the electron share almost the same Lorentz factor, we propose that CRs experience a threshold interaction with a new light particle $X$ abundant in the Galaxy. The interaction $\mathrm{CR}+X\ensuremath{\rightarrow}\mathrm{CR}+{X}^{\ensuremath{'}}$ can take place when the effective energy is sufficient to convert it into another heavier unknown particle ${X}^{\ensuremath{'}}$ (as a representative to all possible threshold inelastic interactions). Under this scenario, we can reproduce the spectral break for both the nuclei and the electron and predict a zigzag spectrum for them. Given that there are great uncertainties in experiments, our model accommodates a wide mass range of the $X$ from the ultralow value to around 1 eV.

The key factors involved in the train derailment on bridges or lines have been discussed in this paper. Also the statistics of derailment accidents with an upward trend occurred in recent years, all over the world, are listed. Analysis of the reasons for train derailment, geometric criterion and mechanical mechanism of train derailment are discussed. Then the authors make an analysis of difference of train derailment between bridges and lines from five aspects. The conclusions have been drawn from the comparative analysis of train derailment involved in lateral stiffness, derailment time history, flow field, motion equation and resonance. There is no characteristic of time history for a certain distance on line derailment. And the mechanical characteristics of vehicle-bridge system are more complicated than the vehicle-line system under wind loads.

Research in mesenchymal stem cells (MSCs) is mainly focused on applications for treatments of brain and spinal cord injury as well as mechanisms underlying effects of MSCs. However, due to numerous limitations, there is little information on selection of appropriate sources of MSCs for transplantation in clinical applications. Therefore, in this study we compared various properties of human umbilical cord-derived MSCs (HUCMSCs) with human placenta-derived MSCs (HPDMSCs), including cell proliferation, apoptosis, cellular morphology, ultrastructure, and their ability to secrete various growth factors (i.e. vascular endothelial growth factor, insulin-like growth factors-1, and hepatocyte growth factor), which will allow us to select appropriate MSC sources for cellular therapy. Cell culture, flow cytometry, transmission electron microscope (TEM) and atomic force microscope (AFM) were used for assessment of HUCMSCs and HPDMSCs. Results showed that the two types of cells appeared slightly different when they were observed under AFM. HUCMSCs appeared more fibroblast-like, whereas HPDMSCs appeared as large flat cells. To analyze the second plastic hinge of high bridge pier formed in the earthquake and study the pier seismic performance, this paper takes the Kunming Slope Railway Bridge as an example. The simulation model was built by the elastic-plastic finite element ANSYS software. The buckling load of piers was calculated by buckling analysis capabilities of ANSYS software, a small enough initial offset given to the model, then the buckling load of piers put into the model. The deformation of non-linear model under the loads was calculated, the process was recorded that the reinforced concrete bridge piers initially yielding on the cross-section, continuously rotating around the neutral axis, and finally forming the eventually plastic hinges in certain parts of pier. The location of the plastic hinges can be inferred by final data of analysis software. By analyzing the calculated results, the seismic performance of the second plastic hinge in high bridge is studied.

Background Ventilator-associated pneumonia (VAP) is an important nosocomial infection at neonatal intensive care units (NICU), frequently caused by Pseudomonas aeruginosa . A 6-month prospective study from January 2009 through June 2009 was performed to investigate the respective contribution of endogenous and exogenous transmission of P aeruginosa in the respiratory colonization or/and infection in the mechanically ventilated patients at a NICU to identify routes of lung infection with P aeruginosa and to assess risk factors for colonization or respiratory infection with P aeruginosa . Methods Samples from oropharyngeal swab, tracheobronchial aspirates, gastric aspirate, and rectal swab were obtained in each patient after intubation and then twice a week. Surveillance cultures for the presence of P aeruginosa from environmental surfaces of the ICU were taken once every 5 days during the study period. Pulsed-field gel electrophoresis was used to characterize the clonal relatedness of the strains by Spe I-digested genomic DNA. Results Eighteen patients (78.3%) had colonization of the upper respiratory tract. Sixteen (69.6%) patients with colonization of the respiratory tract were infected from other patients or environmental surfaces, which was considered exogenous, and, among strains causing pulmonary infection, there were 4 (50%) patients with exogenous infection. Eight of these developed VAP after a mean of 9 ± 3.4 days. The incidence of P aeruginosa VAP on the unit was 6.2%. The respiratory tract was the earliest site of colonization in all patients of VAP. Low birth weight, duration of mechanical ventilation, previous ampicillin group use, and previous second-generation cephalosporins use were independently associated with patient-related acquisition of P aeruginosa . Conclusion Our results confirm that the upper respiratory tract acts as an important reservoir of P aeruginosa colonization and infection in the mechanically ventilated patients and emphasize the importance of exogenous acquisition of P aeruginosa . A combination of early identification and eradication of airways colonization by P aeruginosa plus infection control measures may be the basis to prevent pulmonary infection.

The aim of this study is to investigate the denitrification efficiency enhancement and to evaluate the denitrification potential at the condition of lower temperature. Three identical biofilter were operated in parallel at batch tests. Results showed that the denitrification potential can be substantially increased by addition of ethanol and hydrolysate of sludge as external carbon sources compared with raw sewage. The mean denitrification rates of ethanol and hydrolysate reached up to 0.058 and 0.08 mgN/(g MLVSS•h), respectively, while that of raw sewage was only 0.041 mgN/(g MLVSS•h). The denitrification potential increased respectively from 0.76 to 1.54 and 5.3 mg NO3-N/L owing to ethanol and hydrolysate addition. By comparison, hydrolysate of sludge was found to be the better carbon source for denitrification considering strengthening denitrification performance and price. This study provides evidence for external carbon sources choice of sewage plant and surplus sludge reutilization.

This paper presents the novel implementation of a primary standard for low-g shock acceleration calibration based on rigid body collision using laser interferometry at National Institute of Metrology (NIM), China. The combination of an electromagnetic exciter and a pneumatic exciter as mechanical power supply of the shock excitation system are built up to achieve a wider acceleration range. Three types of material for shock pulse generators between airborne anvil and hammer are investigated and compared in the aspects of pulse shapes and acceleration levels. A heterodyne He-Ne laser interferometer is employed for precise measurement of shock acceleration with less electronic and mechanical influences from both the standard device itself and its surroundings. For signal acquisition and processing, virtual instrument technology is used to build up data acquisition PXI hardware from National Instrument and calibration software developed by LabVIEW. Some calibration results of a standard accelerometer measuring chain are shown accompany with the uncertainty evaluation budget. The expanded calibration uncertainty of shock sensitivity of the accelerometer measuring chain is 0.8%, k =2, with the peak range of half-sine squared acceleration shape from 20m/s2 to 10000 m/s2 and pulse duration from 0.5 ms to 10 ms. This primary shock standard can meet the traceability requirements of shock acceleration from various applications of industries from automobile to civil engineering and is used for piloting ongoing international shock comparison APMP.AUV.V-P1.

The CACTUS experiment recently observed a gamma ray excess above 50 GeV from the direction of the Draco dwarf spheroidal galaxy. Considering that Draco is dark matter dominated the gamma rays may be generated through dark matter annihilation in the Draco halo. In the framework of the minimal supersymmetric extension of the standard model we explore the parameter space to account for the gamma ray signals at CACTUS. We find that the neutralino mass is constrained to be approximately in the range between 100 GeV ~ 400 GeV and a sharp central cuspy of the dark halo profile in Draco is necessary to explain the CACTUS results. We then discuss further constraints on the supersymmetric parameter space by observations at the ground based ARGO detector. It is found that the parameter space can be strongly constrained by ARGO if no excess from Draco is observed above 100 GeV., 15 pages, 4 figures

With the improvement of living standard of people in China, the degree of people’s desire towards spiritual life is becoming higher and higher, and the dance hall has become one of the preferred place for recreation. Although most of the dance halls have good hard wares, the audio effect is not satisfying which is related with the ignorance of acoustic environment. If the distribution of sound field is irregular in the hall, and the reverberation time is over-long or over-short which will affect the musical sense of beauty. So this paper makes a detailed description of the methods of creating the sound field of dance hall.

Ovarian injury because of chemotherapy can decrease the levels of sexual hormones and potentia generandi of patients, thereby greatly reducing quality of life. The goal of this study was to investigate which transplantation method for human umbilical cord mesenchymal stem cells (HUMSCs) can recover ovarian function that has been damaged by chemotherapy. A rat model of ovarian injury was established using an intraperitoneal injection of cyclophosphamide. Membrane-labelled HUMSCs were subsequently injected directly into ovary tissue or tail vein. The distribution of fluorescently labelled HUMSCs, estrous cycle, sexual hormone levels, and potentia generandi of treated and control rats were then examined. HUMSCs injected into the ovary only distributed to the ovary and uterus, while HUMSCs injected via tail vein were detected in the ovary, uterus, kidney, liver and lung. The estrous cycle, levels of sex hormones and potentia generandi of the treated rats were also recovered to a certain degree. Moreover, in some transplanted rats, fertility was restored and their offspring developed normally. While ovary injection could recover ovarian function faster, both methods produced similar results in the later stages of observation. Therefore, our results suggest that transplantation of HUMSCs by tail vein injection represents a minimally invasive and effective treatment method for ovarian injury.
