The main issues that limit the large-scale application of TiO2 in photocatalysis is its low utilization of solar energy and high recombination rate of photogenerated charges. To overcome these limitations, a type II WO3·H2O/TiO2 heterojunction has been prepared via a one-step mild hydrothermal method that uses Na2WO4·2H2O and P25 as the starting materials and proceeds at 150°C without requiring any post-heat treatment for crystallization, special solvents or additives. The XRD and HRTEM results show that the prepared powders were composed of orthorhombic WO3·H2O and TiO2, and demonstrate the existence of the WO3·H2O/TiO2 heterojunctions. The photocatalytic activity of the as-prepared WO3·H2O/TiO2 heterojunction powders was investigated by using the degradation of phenol. The WO3·H2O/TiO2 heterojunction powders exhibited the best photocatalytic performance when the nominal W/Ti ratio was 6.57%, and showed better photocatalytic performance than commercial P25 and orthorhombic WO3·H2O.

The tumour microenvironment contributes to chemotherapy resistance in gliomas, and glioma-associated mesenchymal stromal/stem cells (gaMSCs) are important stromal cell components that play multiple roles in tumour progression. However, whether gaMSCs affect chemotherapy resistance to the first-line agent temozolomide (TMZ) remains unclear. Herein, we explored the effect and mechanism of gaMSCs on resistance to TMZ in glioma cells.Human glioma cells (cell line U87MG and primary glioblastoma cell line GBM-1) were cultured in conditioned media of gaMSCs and further treated with TMZ. The proliferation, apoptosis and migration of glioma cells were detected by Cell Counting Kit-8 (CCK-8), flow cytometry and wound-healing assays. The expression of FOXS1 in glioma cells was analysed by gene microarray, PCR and Western blotting. Then, FOXS1 expression in glioma cells was up- and downregulated by lentivirus transfection, and markers of the epithelial-mesenchymal transformation (EMT) process were detected. Tumour-bearing nude mice were established with different glioma cells and treated with TMZ to measure tumour size, survival time and Ki-67 expression. Finally, the expression of IL-6 in gaMSC subpopulations and its effects on FOXS1 expression in glioma cells were also investigated.Conditioned media of gaMSCs promoted the proliferation, migration and chemotherapy resistance of glioma cells. The increased expression of FOXS1 and activation of the EMT process in glioma cells under gaMSC-conditioned media were detected. The relationship of FOXS1, EMT and chemotherapy resistance in glioma cells was demonstrated through the regulation of FOXS1 expression in vitro and in vivo. Moreover, FOXS1 expression in glioma cells was increased by secretion of IL-6 mainly from the CD90CD90

Total resection of meningiomas involving the major dural sinuses (MIMDS) is still challenging for neurosurgeons. Gamma knife radiosurgery (GKRS) was shown to have a high probability of tumor control. The current study evaluated the clinical outcomes of patients who underwent subtotal resection alone or in combination with postoperative GKRS for the treatment of WHO grade I MIMDS.From January 2006 to December 2016, 204 patients with MIMDS underwent Simpson IV subtotal resection in Wuhan Union Hospital. In 151 patients, no additional treatment was performed, while the tumor remnant was treated with GKRS in 53 patients. All patients were monitored with regular MR follow-ups. We retrospectively reviewed the clinical data, radiological characteristics, and outcomes of these 204 patients. Progression-free survival (PFS) was determined by Kaplan-Meier analysis. Related factors were determined by univariate Cox regression analyses.The mean follow-up period was 75.5 months. The tumor recurrence/progression rates were 13.9% in the microsurgery group and 3.8% in the combined therapy group (p = 0.045). The 5- and 10- year progression-free survival (PFS) rates were 92.3 and 80.7%, respectively, in the microsurgery group and 100.0 and 88.5% in the combined therapy group. Treatment approach was found to be an independent prognostic factor for tumor recurrence/progression in the univariable analyses (p=0.04).Compared with microsurgery alone, targeted Simpson grade IV resection combined with early gamma knife treatment resulted in longer progression-free survival without increased complications for WHO grade I MIMDS.

The drug resistance in cancer treatment with DOX is mainly related to the overexpression of drug efflux proteins, residing in the plasma and nuclear membranes. Delivering DOX into the mitochondria, lacking drug efflux proteins, is an interesting method to overcome DOX resistance. To solve the problem of positively charged triphenylphosphonium (TPP) for mitochondrial targeting in vivo, a charge reversal strategy was developed. Methods: An acidity triggered cleavable polyanion PEI-DMMA (PD) was coated on the surface of positively charged lipid-polymer hybrid nanoparticle (DOX-PLGA/CPT) to form DOX-PLGA/CPT/PD via electrostatic interaction. The mitochondrial localization and anticancer efficacy of DOX-PLGA/CPT/PD was evaluated both in vitro and in vivo. Results: The surface negative charge of DOX-PLGA/CPT/PD prevents from rapid clearance in the blood and improved the accumulation in tumor tissue through the enhanced permeability and retention (EPR) effect. The hydrolysis of amide bonds in PD in weakly acidic tumor tissue leads to the conversion of DOX-PLGA/CPT/PD to DOX-PLGA/CPT. The positive charge of DOX-PLGA/CPT enhances the interaction with tumor cells, promotes the uptake and improves DOX contents in tumor cells. Once endocytosed by tumor cells, the exposed TPP in nanomedicine results in effective mitochondrial localization of DOX-PLGA/CPT. Afterward, DOX can release from the nanomedicine in the mitochondria, target mtDNA, induce tumor cells apoptosis and overcome DOX resistance of MCF-7/ADR breast cancer. Conclusion: Tumor acidity triggered charge reversal of TPP-containing nanomedicine and activation of mitochondrial targeting is a simple and effective strategy for the delivery of DOX into the mitochondria of cancer cells and overcoming DOX resistance of MCF-7/ADR tumor both in vitro and in vivo, providing new insight in the design of nanomedicines for cancer chemotherapy.

Singlet oxygen is regarded as the primary cytotoxic agent in cancer photodynamic therapy (PDT). Despite the advances in optical methods to image singlet oxygen, it remains a challenge for in vivo application due to the limited tissue penetration depth of light. Up to date, no singlet oxygen-specific magnetic resonance imaging (MRI) probe has been reported. Herein, a T2 -weighted MRI probe is reported to visually detect singlet oxygen generated in PDT in vitro and in vivo. The MRI probe Ce6/Fe3 O4 -M is constructed by co-encapsulation of photosensitizer Ce6 and Fe3 O4 nanoparticles in mPEG2000 -TK-C16 micelles. Thioketal (TK) linker in the probe is highly sensitive to singlet oxygen, but lowly sensitive to other reactive oxygen species (ROS) existing in physiological and pathological environments. Singlet oxygen, generated with light irradiation, triggers the cleavage of TK, which leads to loss of surface polyethylene glycol, increment of the hydrophobicity, and aggregation of Fe3 O4 nanoparticles. Subsequently, negatively enhanced T2 -weighted MRI signal is obtained for visual detection of singlet oxygen in the solution, cancer cells, and in vivo. This oxidation responsive MRI probe is expected to hold great promise in evaluating the ability of photosensitizers to generate singlet oxygen and in predicting the therapeutic efficacies of PDT in vivo.

Nanotechnology provides a powerful tool to overcome many disadvantages of small-molecule photosensitizers for photodynamic cancer therapy, such as hydrophobicity, rapid blood clearance, low accumulation in tumor tissue and low cell penetration, etc. The occurrence of quench in photosensitizer-loaded nanoparticle greatly downregulates the ability to generate singlet oxygen with light irradiation. Stimuli-responsive nanocarriers can improve the efficacy of PDT to a certain extent. However, insufficient release of photosensitizer from either endogenous or exogenous stimuli responsive nanocarriers in the short period of light irradiation restricts full usage of the photosensitizer delivered into cancer cells. We here report a dual-step light irradiation strategy to enhance the efficacy of cancer PDT. Ce6 as a photosensitizer is loaded in singlet oxygen-sensitive micelles (Ce6-M) via self-assembly of amphiphilic polymer mPEG2000-TK-C16. After co-incubation of Ce6-M with cancer cells or i.v. injection of Ce6-M, cancer cells or tumor tissues are irradiated with light for a short time to trigger Ce6 release, and 2 h later, re-irradiated for relatively long time. The sufficient release of Ce6 in the period between twice light irradiation significantly improves the generation of singlet oxygen, leading to more efficient cancer therapeutic effects of dual-step irradiation than that of single-step irradiation for the same total irradiation time.

This current study intends to investigate the effect of microRNA-128 (miR-128) on cisplatin (DDP) resistance in glioma SHG-44 cells. SHG-44/DDP cells were transfected with miR-128 antisense oligonucleotide (ASO) and assigned into blank, resistance, NC, anti-miR-128, miR-128 mimic, si-JAG1, and anti-miR-128 + si-JAG1 groups. qRT-PCR and Western blotting were employed for determining expression of miR-128, JAG1, Bax and Bcl-2. MTT assay, Giemsa staining, and flow cytometry were applied to detect DDP resistance, cellular morphology, and cell cycle, respectively. JAG1 is targeted and negatively regulated by miR-128. In in vitro experiments, compared with the blank group, the rest groups exhibited declined miR-28 and Bax expression, lowered cell inhibition rate and apoptosis rate, but elevated JAG1 and Bcl-2 expression with cells arrested in the S phase. Compared with the resistance group, the anti-miR-128 group showed decreasedBax expression along with a lowered cell inhibition rate and apoptosis rate, but increased JAG1 and Bcl-2 expression with reduced cells arrested in the S phase; while the miR-128 mimic group showed an opposite trend; the si-JAG1 group showed decreased Bcl-2 expression and reduced cells in the S phase. In in vivo experiments, compared with the resistance group, the tumor growth rate, tumor volume, and weight as well as JAG1 expression accelerated in the anti-miR-128 group; whereas the miR-128 mimic and si-JAG1 groups exhibited an opposite trend. Our findings demonstrated that miR-128 ASO transfection might down-regulate the expression of miR-128 in SHG-44/DDP and up-regulate the DDP resistance in SHG-44/DDP cells, providing a potential treatment target for glioma.

Glioma is one of the most common types of tumor of the central nervous system. Due to the aggressiveness and invasiveness of high-level gliomas, the survival time of patients with these tumors is short, at ~15 months, even after combined treatment with surgery, radiotherapy and/or chemotherapy. Recently, a number of studies have demonstrated that long non-coding RNA (lncRNAs) serve crucial roles in the multistep development of human gliomas. Gliomas acquire numerous biological abilities during multistep development that collectively constitute the hallmarks of glioma. Thus, in this review, the roles of lncRNAs associated with glioma hallmarks and the current and future prospects for their development are summarized.

Gamma knife surgery (GKS) is now used as a treatment option for glossopharyngeal neuralgia (GPN). Most authors have selected the distal part of the nerve as the gamma knife target. Here we report on 3 patients with medically intractable GPN who were treated with GKS. All 3 patients had a single shot with a 4-mm collimator which was used to deliver 80 Gy to the 100% isodose line. The GKS targets were the medial cisternal segments of the glossopharyngeal nerve. Patients were investigated prospectively, treated, and then assessed periodically with respect to pain relief and neurological function. Three patients felt pain reduced at 2, 7, and 11 days, respectively. None of them have suffered recurrent pain since becoming pain free. The follow-up after radiosurgery was 25 months, 22 months, and 20 months, respectively. This preliminary experience provides encouraging evidence that choosing the medial cisternal segment of the glossopharyngeal nerve as the target is also an option for the treatment of GPN with stereotactic radiosurgery and is consistent with previous reports. Additional follow-ups and a larger number of patients are needed to demonstrate the long-term safety and effectiveness for this indication.

Although it is recognized that thrombin plays a key role in airway remodeling during chronic asthma. In a previous study, we have proved that thrombin promotes airway remodeling via PAR-1 in OVA-allergic rats, but little is known about intracellular signaling pathway involved in the event.In this study, we intend to explore the impact of pERK1/2 signaling pathway on the process of thrombin-induced airway remodeling in OVA-allergic rats.A rat model of chronic asthma was set up by systemic sensitization and repeated challenge to OVA. The doses of thrombin, recombinant hirudin, PAR-1 inhibitor ER-112780-06, and pERK1/2 inhibitor PD98059 varied for different groups. The expression of pERK1/2 was analyzed by western blot and RT-PCR. Secretion of TGF-β1 and IL-6 was detected by ELISA.The expression of pERK1/2 was higher in the airway of asthmatic rats than those of normal rats, and was significantly increased by thrombin treatment but decreased by thrombin-inhibitor treatment. Airway remodeling was enhanced by thrombin but weakened by pERK1/2 inhibitor. Expression of growth factors and IL-6 in asthmatic rats was significantly increased by thrombin treatment and decreased by thrombin-inhibitor treatment and pERK1/2 inhibitor treatment.These results suggest that ERK1/2 signaling pathway may play an important role in the process of thrombin-promoting airway remodeling in OVA-allergic rats, and pERK1/2 inhibitor effectively inhibits the process.

Coiling tension control system is the main control means to ensure the quality of the rolling process. According to the characteristics of coiling tension control characteristics and the requirements of actual engineering, establish the mathematical model of the double closed loop control system and use the traditional PID to tuning the model parameters. It is proved by Matlab simulation experiments that traditional PID can effectively improve the response process of coiling tension control system and greatly enhance the dynamic and steady-state performance.

In this study, 7050 aluminum alloy strip was produced using the twin-roll casting technique. Microstructures and properties of the cast-rolling 7050 aluminum alloy were investigated by means of optical microscope (OM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and Vickers hardness test. The results indicate that microstructures of cast-rolling 7050 alloy strip are inhomogeneous on the cross section. From the edge to the center, the size of grain decreases gradually and becomes more homogeneous, showing a transition from dendrite grains to equiaxed grains. There are undissolved second phase particles in grains and at grain boundaries. And the dendrite grains and second phase particles can be reduced by solution treatment, and the hardness of the cast-rolling strip can be improved by aging treatment.

Aims: We conducted a case-control study in a Chinese population to clarify the association between polymorphisms in ERCC1 and XPD and susceptibility and survival of glioma. Methods: A total of 393 cases and 410 controls were selected from March 2007 to December 2011. Genotyping of ERCC1 and XPD was conducted by TaqMan assays using the ABI Prism 7911HT Sequence Detection System. All analyses were performed using the STATA statistical package. Results: Polymorphisms in ERCC1 118C/T, ERCC1 8092C/A and XPD Asp312Asn showed no statistically significant difference between glioma cases and controls. However, individuals with the XPD 751Gln/Gln genotype had an increased risk of developing glioma compared with those with the Lys/Lys genotype (adjusted OR=1.64, 95% CI: 1.06-2.89). The ERCC1 118T/T genotype was associated with significantly higher median survival than the ERCC1 C/C genotype (HR=0.67, 95%CI=0.35-0.96). In addition, individuals with XPD 751Gln/Gln had a lower median survival time than XPD Lys/Lys carriers (HR=0.54, 95%CI=0.370.93). Conclusion: In conclusion, we observed that the XPD 751Gln/Gln genotype is associated with glioma susceptibility, and ERCC1 118 T/T and XPD 751Gln/Gln genotypes confer a significantly better prognosis.

In hot-dip galvanizing process, air jet wiping control is so crucial to decide the coating thickness and uniformity of the zinc layer on the steel strip. The mathematical models developed predict the zinc coating thickness as a function of pressure and shear stress. The required pressure and shear stress profile on the strip surface were calculated using regression analysis, and carried out using numerical simulation as FLUENT, a finite element analysis software. The influences of the outlet pressure, the nozzle to strip distance, the slot opening, the edge baffle plate, as well as the tilting angle of air knife were discussed. Combining with these results and regression analysis on the practical data, four first-order polynomial multi-parameter models were established for different targeted coating thicknesses with better regression coefficients. The validated model was used to carry out sensitivity analysis to determine the favorable controlling regime for the air jet wiping process.

Background: Microvascular decompression (MVD) and rhizotomy are all selected for treating vagoglossopharyngeal neuralgia (VGPN). Nonetheless, controversies still exist about their curative effect on VGPN. Here we evaluate the effectiveness of MVD together with rhizotomy of the glossopharyngeal nerve for the treatment of VGPN. Methods: This study was carried out on 21 patients who were diagnosed with VGPN between the years 2005 and 2010. Patients underwent MVD and glossopharyngeal rhizotomy through a retromastoid keyhole approach. Surgical technique, operation results and complications were our particular concern. Results: Eighteen (85.7%) of 21 patients experienced immediate and complete relief of pain after surgery. In the remaining 3 patients (14.3%), the pain faded away within the following week. No patient complained of dysphonia or dysphagia. All 21 patients reported no change in their outcome at follow-up. Conclusions: Intracranial vagoglossopharyngeal nerve MVD with glossopharyngeal rhizotomy is an effective and safe procedure to treat VGPN.

The Shoushan Stone is one of the famous craft carving stones in China and transparency is one of the key indicators to evaluate the value of Shoushan Stone. The better the transparency is, the higher the value is. In this study, the mineral compositions and morphologies were investigated by XRD and SEM, respectively. Combining with the characterization of Shoushan Stone samples, the relationships between mineral composition, morphology and transparency were studied. The results show that the transparency of Shoushan Stone is determined by the mineral categories and microstructures. The sample with high transparency is fewer mineral composition numbers, similar mineral compositions and bigger crystal particles

The Shoushan Stone is one of the famous craft carving stones in China and the transparency is one of the decisive factors to appraise its value. The better the transparency is, the higher the value is. In this paper, the mineral compositions and morphology of some Shoushan Stone samples were investigated by XRD and SEM methods. Combining with the characterization of Shoushan Stone, the relationships between the morphology and the transparency of Shoushan Stone were studied. The results show that the transparency of Shoushan Stone samples was determined by their microstructures, such as the mineral particle sizes and its arrangement, the gaps and porosity. The Shoushan Stone samples with high transparency are those with good arrangement, bigger crystal particle and less porosity and gaps.

Stress intensity factors of thin AZ31B magnesium alloy sheet under biaxial tension loading were analyzed by modified Dugdale model. K-values with crack angle of 90° obviously show that there is no influence of the loading condition in Mode-I. In the 45° case, KI values are obtained within 10% errors when they are calculated by modified Dugdale model under biaxial loading. It is concluded that the modified Dugdale model is one of effective ways to evaluate stress intensity factor of AZ31 magnesium alloy sheet appropriately.

A new magnesium alloy plate added elements Zn, Sn and In was manufactured by twin-roll continuous casting method to improve the precipitation of AZ91 alloy. The effects of elements addition and casting method on microstructure and mechanical properties of the Mg-Zn-In-Sn alloys were investigated by optical microscopy (OM), transmission electron microscopy (TEM), electron probe micro-analysis (EPMA), X-ray diffractometry (XRD) and energy dispersion spectrograph (EDS). The results show that the Mg-Zn-In-Sn alloy has higher tensile strength and better corrosion protection than the AZ91 alloy. The outstanding precipitation strengthening effect of the alloy is attributed to the small grain size and the hard precipitates between the grain boundaries.

With five varieties alfalfa, seleted the highest cell medium from callus . and detected the biomass and protein content in cell grows from different nitrogen source and sucrose concentration. Rusult is the highest protein in 12th -18th days of grows. There are different protein content in different nitrogen source (from Fig.2) and in different sucrose concentration range medium(from Fig.3).Conclusion is obtained the protein from alfalfal cells that is possibally.

Stress intensity factor K is analyzed by measuring distribution of sum of the principal stresses around slit tip by an infrared stress measuring device. Under the biaxial tensile stress, both K-values of the isotropic materials with slit angle 0° and 45° show there is no influence of the slit angle at Mode I. The tolerance of the measured K-values of the test specimens which are compared with measured K-values by photoelastic method are around 10-20and these increase as applied cyclic load increases. These tolerance can be reduced up to about 8% by modifying measurements. The effectiveness of the thermoelastic stress measurement method to KI value analyzing under biaxial tensile stress was confirmed.

Estrogen has been implicated as a risk factor for meningioma. Bisphenol A (BPA), a widely used synthetic xenoestrogen, has already been reported to be associated with several estrogen-sensitive tumors. An exploratory association study of 243 meningioma cases and 258 frequency-matched healthy controls was conducted, using subjects from a hospital-based study to demonstrate the association of urine BPA concentration and the risk of meningioma. The specimens and data of patients were collected at Union Hospital, Wuhan, China, from 2009 to 2010. A positive association between increasing levels of urinary BPA and meningioma was observed, independent of confounding factors such as gender, age, race, body mass index, HRT use, BMI, and family history of cancer. Compared to quartile 1 (referent), the multivariate-adjusted odds ratio of meningioma associated with quartile 4 was 1.45 (95 % CI, 1.02–1.98) (P trend = 0.03). In this case–control study from China, a clear association between urinary BPA concentrations and diagnosis of meningioma was detected.

To evaluate the microsurgical treatment for sylvian fissure arteriovenous malformations (AVMs).The clinical data of 94 sylvian fissure AVMs treated microsurgically was retrospectively collected. They were classified as medial, lateral, deep, anterior and simple AVMs according to the anatomic location of nidus in sylvian fissure.Total lesion removal was achieved in all patients and it was confirmed by postoperative digital subtraction angiography. Transient neurological deficits, including slight hemiparesis (n = 5, 5.3%) and aphasia (n = 4, 4.3%), were documented. Another 2 patients (2.1%) presented with a postoperative epidural and an intracranial hematoma respectively. However, an excellent outcome was observed in all patients as indicated by the Karnofsky performance scale90.The microsurgical treatment for sylvian fissure AVMs has been greatly restrained by the involvement of multiple brain functional regions and its anatomic complexity. A more aggressive approach is recommended.

The hypoxic-ischemic encephalopathy caused by asphyxia in peripartum is a serious disease in newborn infants, with a high disability and mortality rate. Lack of regenerative ability in central nervous system after injury is considered as the fundamental cause. However, in recent years many studies have revealed that there are myelin-associated neurite growth inhibitory factors that exert inhibiting effect through the Nogo receptor (NgR). This study aimed to investigate the expression level of NgR and the possible neuroprotective effect of NEP1-40 in newborn rats with hypoxic ischemic brain damage (HIBD).Eighty healthy Wistar rats aged 7 days were randomly divided into 4 groups; 8 in control group, 24 in HIBD model group, 24 in GM-1 group and 24 in NEP1-40 group. The rats of the control group and HIBD group were injected with normal saline (0.25 ml/kg) intraperitoneally, while those in NEP1-40 group and GM-1 group with NEP1-40 12.5 microg/d, GM-1 10 mg/(kg.d) for continuous 3 days of 72-hour group or 7 days of 168-hour group, respectively. In situ hybridization was adopted for detecting the expression of NgR in the brain of the rats at the time point of 24 hours, 72 hours and 7 days. Meanwhile histopathological changes of neurons and axon were detected by transmission electron microscopy (TEM). The SPSS statistical software package for Windows, version 10.0, was used to run Chi-square tests and least significance difference (LSD-t) on the data presented, and P value of less than 0.05 was regarded as statistically significant.The expression level of Nogo-A receptor in the control group was higher than that of the other groups at different time point (t value was 5.48, 6.11, 6.96, 8.24, 5.99 and 5.34, respectively, and all P values were less than 0.05). There were no significant differences in Nogo-A receptor level among the HIBD group, the GM-1 group and the NEP1-40 at 24 hours (t was 1.48, 2.76 and 1.29, respectively, and all P0.05), while the expression of Nogo-A receptor of NEP1-40 at 72 hours and 7 days was lower than that of the HIBD group and the GM-1 group at the same time point, respectively (all P0.05). Repair of neurons in damaged brain to some extent was found after GM-1 treatment and satisfactory repair of neurons and axon regeneration was obtained with NEP1-40 administration as shown by TEM.Hypoxic ischemic brain damage can down-regulate the expression of Nogo-A receptor in the central nervous system. NEP1-40 contributes to the regeneration of axon and repair of brain damage, thus exerts neuroprotective effect.

To investigate the effect of wt p53 gene transfection on the sensitivity of hyperthermia in C6 glioma.The recombinant eukaryotic expressive vector pCMV-p53 plasmids were extracted, identified and transmitted to C6 glioma cells by stable transfection. The biological characteristics were compared among C6/p53 (+) cells, thermo-treated C6/p53 (+) cells, thermo-treated C6 cells, thermotolerance C6 cells and control C6 cells. Transplanted glioma was treated by laser interstitial thermotherapy (LITT) in tumor-bearing nude mice.The wt p53 gene segments were extracted and correctly identified by restriction enzyme (Hind and EcoR). The optimal transfection concentration of plasmid to liposome was 1:6, neo gene expressed stably in C6 cells transfected with positive and blank plasmid. Thermotolerance C6 cell growth was normal in vitro, growth of C6/p53 (+) cells was inhibited. Growth of hyperthermia treated C6 cells was inhibited significantly 12 h after thermal treatment, cell proliferation activity of C6/p53 (+) cells was suppressed significantly 6 h after hyperthermia treatment. Apoptosis was observed in C6/p53 (+), thermo-treat C6 and thermo-treat C6/p53 (+) cells. The apoptosis ratio of hyperthermia treated C6/p53(+) cells was increased by 30 folds compared with that of control groups. Study in nude mouse model demonstrated lower positive inoculation rate in groups of C6/p53 (+) cells (57 %), hyperthermia treated C6 cells (75 %) and hyperthermia treated C6/p53(+) cells (20 %). C6/p53(+) LITT group showed the highest antitumor effect, compared with C6 LITT group(P0.05). However, thermotolerance C6 LITT group had the lowest effect.Transfection of wt p53 gene can increase the sensitivity of hyperthermia in glioma cells.

To isolate cancer stem cells glioblastoma cells and detect the expression of anti-apoptotic and multi-drug resistance-associated protein (MRP) genes thereof.CD133 positive cells were isolated from human glioblastoma multiforme cells of the line TJ905 by immunomagnetic beads technique and nestin, beta-tubulin and GFAP expression were examined by Immunofluorescence staining. RT-PCR was used to detect the expression of livin, livinalpha, Livinbeta, Survivin, MRP1, and MRP3.Only 0.21% of the TJ905 cells maintained in serum was CD133(+) and showed characteristics of cancer stem cells, positive in nestin. These cells maintained a sphere-like growth status in serum-free medium in vitro, and could self-renew, proliferate, conditionally differentiate into tubulin-beta(+) and GFAP(+) cells, and produce neurons as well as glial cells. The mRNA expression levels of livin, livinalpha, survivin, MRP1, and MRP3 of the TJ905 tumor stem cells were significantly lower than those f the of TJ905 cells.Cancer stem cells can be isolated from TJ905 glioblastoma multiforme cells. However, the generating rate of the tumor stem cells is lower than that of the TJ905 cells, and the expression levels of anti-apoptotic and MRP genes are lower than those of the progenitor cells. Showing that cancer stem cells are not the solo factor to maintain tumor growth and resist apoptosis and to pump the anti-tumor drugs out of cells.

Reticulon proteins, which are localized in the endoplasmic reticulum, have recently been shown to be involved in hormone secretion, in particular RTN1 and RTN3. The aim of the present study was to examine the effects of Nogo-A gene expression knockdown by RNA interference (RNAi) on dopamine release in PC12 cells.A small hairpin RNA (shRNA) eukaryotic expression vector, targeting the Nogo-A gene, was constructed and transfected into cultured PC12 cells by lipofecamine2000. Inhibition of Nogo-A gene expression was detected by semi-quantitative reverse transcription PCR and Western blot analysis. Following transfection, dopamine release was detected by high performance liquid chromatography.The pGenesil-1-Nogo-A-2 plasmid was identified by gene sequencing. After transfection of the recombinant vector in PC12 cells, Nogo-A gene expression was significantly inhibited (p0.01). Compared with the empty vector control group, dopamine release significantly decreased within 48 hours after transfection.Results from this study suggest that Nogo-A might be involved in the mechanism of DA release in PC12 cells.

To evaluate the applied value of magnetoencephalography (MEG) localization in microsurgery of brain tumors in the region of motor cortex.From January 2003 to April 2005, 36 patients with 19 meningomas, 14 gliomas, 2 metastases and 1 cavernous angioma underwent MEG and MRI before operation. These individualized functional brain maps were integrated into a neuronavigation system. Preoperative mapping of somatosensory and/or motor cortex was performed, and sites were compared, and all tumors were resected microsurgerically.A space-occupying lesion could change the region of motor cortex in all the patients. Thirty-four cases were resected totally. The patient's myodynamia after operation improved in 19 cases, unchanged in 15 cases, descended in 2 cases.MEG is an effective localization method of function, providing the relationship between the region of motor cortex and the tumor. And it is valuable for accurate planning in the treatment of brain tumors in the region of motor cortex.
