Abstract Prostate cancer (PC) is an epithelial malignancy occurring in the prostate. PC ranks second in incidence among all male malignancies globally by the latest statistics from the World Health Organization. Notably, China has seen a more rapid increase in PC incidence compared to developed European and American nations. By 2022, the newly reported cases and deaths due to PC in China increased to 134,200 and 47,500, respectively. Thus, early diagnosis and standardized treatment for prostate cancer in China remain far-reaching objectives. Burgeoning research on advanced PC and castration-resistant prostate cancer in recent years have paved the way for a new era of integrated treatment methods including novel endocrine drugs, chemotherapy, targeted therapy, and immunotherapy. Future therapies involve precision treatment guided by genetic testing and individualized integrated treatment as part of a multidisciplinary integrated diagnosis and treatment model for PC. The Genitourinary Oncology Committee of the China Anti-Cancer Association (CACA-GU) has invited multidisciplinary experts across fields including surgery, oncology, pathology, radiology, herbal medicine, physiatry, and psychology to collaboratively write, discuss, and revise guidelines on managing PC. The CACA Guidelines for Holistic Integrative Management of Prostate Cancer includes epidemiology, screening and diagnosis, treatment for localized PC, diagnosis and treatment of PC recurrence after radical prostatectomy, management of metastatic PC, traditional Chinese medicine diagnosis and treatment of PC, and rehabilitation from PC. This guideline aims to standardize the clinical diagnosis and treatment management of PC in China. It is more aligned with China’s clinical practice, highlights Chinese characteristics, and bears significant clinical importance.

Abstract To reveal the molecular function of elongation family of very long chain fatty acids(ELO) protein in Cyrtotrachelus buqueti, we have identified 15 ELO proteins from C.buqueti genome. 15 CbuELO proteins were located on four chromosomes. Their isoelectric points ranged from 9.22 to 9.68, and they were alkaline. These CbuELO proteins were stable and hydrophobic. CbuELO proteins had transmembrane movement, and had multiple phosphorylation sites. The secondary structure of CbuELO proteins was mainly α-helix. A total of 10 conserved motifs were identified in CbuELO protein family. Phylogenetic analysis showed that molecular evolutionary relationships of ELO protein family between C. buqueti and Tribolium castaneum was the closest. Developmental transcriptome analysis indicated that CbuELO10, CbuELO13 and CbuELO02 genes were key enzyme genes that determine the synthesis of very long chain fatty acids in pupae and eggs, CbuELO6 and CbuELO7 were that in the male, and CbuELO8 and CbuELO11 were that in the larva. Transcriptome analysis under different temperature conditions indicated that CbuELO1, CbuELO5, CbuELO12 and CbuELO14 participated in regulating temperature stress responses. Transcriptome analysis at different feeding times showed CbuELO12 gene expression level in all feeding time periods was significant downregulation. The qRT-PCR experiment verified expression level changes of CbuELO gene family under different temperature and feeding time conditions. Protein-protein interaction analysis showed that 9 CbuELO proteins were related to each other, CbuELO1, CbuELO4 and CbuELO12 had more than one interaction relationship. These results lay a theoretical foundation for further studying its molecular function during growth and development of C. buqueti.

The analysis of foundation's bearing capacity is generally conducted under the assumption of plane strain. However, the damage of rectangular foundation usually presents obvious three-dimensional (3D) effect. This article considers this 3D effect, which, for the first time, provides a theoretical framework for assessing the bearing capacity of rectangular foundations placed on rock slopes under seismic condition. In order to apply the kinematic method of limit analysis, a new 3D kinematically admissible collapse mechanism is first constructed. Owing to the point-to-point discretized technique, this 3D mechanism can avoid complex surface integration and effectively reduce spatial coordinate iterative calculations while maintaining high accuracy. The pseudo-static method is adopted to simulate the effects of earthquakes. Generalized multi‑tangential technique is employed to derive the Mohr-Coulomb constants from Hoek-Brown criterion. The comparison between the present results and results of previous literatures proves the validity of the theoretical framework in this paper. Shape factor and reduction factor are introduced in the parametric study, showing that the 3D effect is more obvious when the rectangular foundation aspect ratio is reduced. As the rectangular foundation location moves away from the slope, its bearing capacity gradually converges towards that of the horizontal foundation. The critical collapse mechanism is also explored, demonstrating that the collapse extent expands with increasing internal friction angle of the rock.

The three-dimensional (3D) stability problem of triple-step slopes is important due to their unique profiles under natural rainfall conditions. In this paper, an innovative approach was proposed for rapidly evaluating the stability of triple-step slopes, which considers the effects of diverse hydraulic hysteresis paths caused by rainfall and drought alternation. Considering diverse hydraulic hysteresis paths, a 3D upper-bound analysis (UBA) mechanical model was developed. On this basis, a new analytical solution for the stability number (NS) of 3D slopes under the influence of hydraulic hysteresis was derived through the work and energy equation. The comparisons showed that the new analytical solution and proposed method are correct. Then, ranges of the controlling parameters of triple-step slopes were designed for practical use in slope engineering. Comprehensive discussions were presented, considering four distinct hydraulic hysteresis cases within the context of two typical scenarios involving silty and sandy soils. The results could provide a reference for the design and construction of triple-step slopes where rainy and drought conditions frequently alternate.

Abstract Purpose To evaluate the predictors for short and long term urinary continence (UC) recovery after laparoscopic radical prostatectomy (LRP) from clinical and oncological variables. Methods We retrospectively collected data from 142 prostate cancer patients who underwent LRP between September 2014 and June 2021 at a tumor specialist diagnosis and treatment center in China. The rate of post-prostatectomy incontinence (PPI) was evaluated from immediate and at 3, 6 and 12 mo after LRP, and UC was defined as the use of no or one safety pad. Sixteen clinical and oncological variables were analyzed by univariate and multivariate regression analysis to determine whether they were associated with short (3 mo) or long term (12 mo) UC recovery after LRP. Results After eliminating patients who were lost to follow-up, 129 patients were eventually included. The mean ± SD age was 68 ± 6.3 years. The UC rates of immediate, 3, 6 and 12 mo after the operation were 27.9%, 54.3%, 75.2% and 88.4%, respectively. Abstract Ischemic stroke is the leading cause of adult disability. Ischemia leads to progressive neuronal death and synapse loss. The engulfment of stressed synapses by microglia further contributes to the disruption of the surviving neuronal network and related brain function. Unfortunately, there is currently no effective target for suppressing the microglia-mediated synapse engulfment. Stimulator of interferon genes (STING) is an important participant in innate immune response. In the brain, microglia are the primary cell type that mediate immune response after brain insult. The intimate relationship between STING and microglia-mediated neuroinflammation has been gradually established. However, whether STING affects other functions of microglia remains elusive. In this study, we found that STING regulated microglial phagocytosis of synapses after photothrombotic stroke. The treatment of STING inhibitor H151 significantly improved the behavioral performance of injured mice in grid-walking test, cylinder test, and adhesive removal test after stroke. Moreover, the puncta number of engulfed SYP or PSD95 in microglia was reduced after consecutive H151 administration. Further analysis showed that the mRNA levels of several complement components and phagocytotic receptors were decreased after STING inhibition. Transcriptional factor STAT1 is known for regulating most of the decreased molecules. After STING inhibition, the nucleus translocation of phosphorylated STAT1 was also suppressed in microglia. Our data uncovered the novel regulatory effects of STING in microglial phagocytosis after stroke, and further emphasized STING as a potential drug-able target for post-stroke functional recovery.

Abstract Ozone (O3) pollution is a severe air quality issue in China, posing a threat to human health and ecosystems. The climate change will affect O3 levels by directly changing physical and chemical processes of O3 and indirectly changing natural emissions of O3 precursors. In this study, near‐surface O3 concentrations in China in 2030 and 2060 are predicted using the process‐based interpretable Extreme Gradient Boosting (XGBoost) model integrated with multi‐source data. The results show that the climate‐driven O3 levels over eastern China are projected to decrease by more than 0.4 ppb in 2060 under the carbon neutral scenario (SSP1‐1.9) compared with the high emission scenario (SSP5‐8.5). Among this reduction, 80% is attributed to the changes in physical and chemical processes of O3 related to a cooler climate, while the remaining 20% is attributed to the reduced biogenic isoprene emissions.

Abstract The climate commitment to achieving carbon neutrality before 2060 in China has been announced recently. In the context of pursuing carbon neutrality, sharing similar sources as greenhouse gases, aerosol particle and precursor emissions are projected to substantially decrease in China, which can potentially have a great impact on climate. Here, we investigate the effects of future aerosol reductions, because of achieving carbon neutrality, on solar and wind energy in China by using an earth system model. We show that significant reductions in aerosol emissions, particularly in eastern China, lead to increases in the surface downwelling shortwave radiation, surface air temperature and wind speed, which can further enhance the potential of solar and wind energy production. The findings underline that the pursuit of carbon neutrality can yield co‐benefits of not only mitigating climate change and air pollution but also fortifying the stability of renewable energy sources.

Levulinic acid, a hydrolysis product of lignocellulose, can be metabolized into important compounds in the field of medicine and pesticides by engineered strains of Saccharomyces cerevisiae. Levulinic acid, as an intermediate product widely found in the conversion process of lignocellulosic biomass, has multiple applications. However, its toxicity to Saccharomyces cerevisiae reduces its conversion efficiency, so screening Saccharomyces cerevisiae genes that can tolerate levulinic acid becomes the key. By creating a whole-genome knockout library and bioinformatics analysis, this study used the phenotypic characteristics of cells as the basis for screening and found the HMX1 gene that is highly sensitive to levulinic acid in the oxidative stress pathway. After knocking out HMX1 and treating with levulinic acid, the omics data of the strain revealed that multiple affected pathways, especially the expression of 14 genes related to the cell wall and membrane system, were significantly downregulated. The levels of acetyl-CoA and riboflavin decreased by 1.02-fold and 1.44-fold, respectively, while the content of pantothenic acid increased. These findings indicate that the cell wall-membrane system, as well as the metabolism of acetyl-CoA and riboflavin, are important in improving the resistance of Saccharomyces cerevisiae to levulinic acid. They provide theoretical support for enhancing the tolerance of microorganisms to levulinic acid, which is significant for optimizing the conversion process of lignocellulosic biomass to levulinic acid.

Abstract Passiflora is a plant genus known for its extremely distinctive and colorful flowers and a wide range of genome size variation. However, how genome characteristics are related to flower traits among Passiflora species remains poorly understood. Here, we assembled a chromosome-scale genome of P. foetida, which belongs to the same subgenus as the commercial passionfruit P. edulis. The genome of P. foetida is smaller (424.16 Mb) and contains fewer copies of long terminal repeat retrotransposons (LTR-RTs). The disparity in LTR-RTs is one of the main contributors to the differences in genome sizes between these two species and possibly in floral traits. Additionally, we observed variation in insertion times and copy numbers of LTR-RTs across different transposable element (TE) lineages. Then, by integrating transcriptomic data from 33 samples (eight floral organs and flower buds at three developmental stages) with phylogenomic and metabolomic data, we conducted an in-depth analysis of the expression, phylogeny, and copy number of MIKC-type MADS-box genes and identified essential biosynthetic genes responsible for flower color and scent from glandular bracts and other floral organs. Our study pinpoints LRT-RTs as an important player in genome size variation in Passiflora species and provides insights into future genetic improvement. Graphical Abstract

Abstract To mitigate climate warming, many countries have committed to achieve carbon neutrality in the mid-21st century. Here, we assess the global impacts of changing greenhouse gases (GHGs), aerosols, and tropospheric ozone (O3) following a carbon neutrality pathway on climate and extreme weather events individually using the Community Earth System Model version 1 (CESM1). The results suggest that the future aerosol reductions significantly contribute to climate warming and increase the frequency and intensity of extreme weathers toward carbon neutrality and aerosol impacts far outweigh those of GHGs and tropospheric O3. It reverses the knowledge that the changing GHGs dominate the future climate changes as predicted in the middle of the road pathway. Therefore, substantial reductions in GHGs and tropospheric O3 are necessary to reach the 1.5 °C warming target and mitigate the harmful effects of concomitant aerosol reductions on climate and extreme weather events under carbon neutrality in the future.

In the field of tunnels, the stability of tunnel faces is generally considered in dry, saturated and homogeneous soils. However, the actual condition of some soils has been found to be inhomogeneous, with unsaturated seepage. In this paper, an analytical method is applied to estimate the safety factor when the supporting force at the tunnel face is zero under steady unsaturated seepage and inhomogeneous conditions. This method combines kinematic limit analysis techniques with strength reduction techniques; an efficient stress formulation utilizing suction stress is employed to determine the apparent cohesive force to obtain the solution of the steady unsaturated seepage problem, and indicators of soil inhomogeneity are attributed to the effect on cohesion. A 3D log-spiral collapse mechanism is used to find the zero supporting pressure and determine the safety factor through an iterative method. This paper analyzes the effect of variations in the unsaturated parameters, inhomogeneity parameters and tunnel dimensional parameters on the stability of the tunnel face.

The construction industry plays a pivotal role in China’s achievement of its “dual carbon” goals. This study conducts a decomposition analysis of the carbon emissions from the construction industry (CECI) at both national and provincial levels for the period 2010–2020 and employs the ARIMA model to predict the short-term peak trends at the provincial level. The findings are as follows. (1) Inner Mongolia, Shandong, Sichuan, and Chongqing exhibit an N-shaped trend in CECI, while the northeast region shows an inverted U-shaped trend. (2) Labor productivity and energy intensity are identified as the largest and smallest drivers of national CECI growth, respectively, with the driving force of the study’s identified factors fluctuating between 1% and 60%. (3) Energy intensity significantly contributes to the growth of CECI in Tianjin and Zhejiang, while it aids in reducing CECI in western provinces. The “rebound effect” of building energy efficiency is particularly pronounced in provinces with strong resource endowments, such as Ningxia. (4) Between 2021 and 2025, CECI is predicted to decrease in the northern and economically developed provinces, while it is expected to increase in central and western provinces, with Heilongjiang, Shandong, Guangdong, Shanghai, and Shaanxi potentially reaching their peaks within the forecast period. The paper concludes with several recommendations.

This research introduces an innovative construction method based on magnesia expansive agent concrete for the seamless construction of hundred-meter scale super-length raft structures, corroborated by the on-site test. The basic principle of this construction method is to use the pre compression stress generated by magnesia expansive agent to offset temperature and shrinkage stress. A temperature-strain monitoring system was employed to gather data, affirming the technique’s applicability and safety. Through the examination of temperature and strain dispersion trends in super-length raft structure, recommendations for the configuration of temperature-strain sensors have been put forth. Through the scrutiny of the temporal evolution pattern of temperature, the specific temporal and spatial coordinates that warrant particular vigilance during the surveillance of the raft’s inner-surface temperature difference were identified. Upon evaluating the correlation between strain dispersion and strain-temperature differential in the raft’s thickness dimension, a novel temperature control index (the bottom-surface temperature difference) was introduced. The threshold for this metric was established at 30°C, derived from empirical test outcomes conducted on-site. Furthermore, the critical regions for monitoring the bottom-surface temperature difference were specified.

Engineering Saccharomyces cerevisiae for biodegradation and transformation of industrial toxic substances such as catechol (CA) has received widespread attention, but the low tolerance of S. cerevisiae to CA has limited its development. The exploration and modification of genes or pathways related to CA tolerance in S. cerevisiae is an effective way to further improve the utilization efficiency of CA. This study identified 36 genes associated with CA tolerance in S. cerevisiae through genome-wide identification and bioinformatics analysis and the ERG6 knockout strain (ERG6Δ) is the most sensitive to CA. Based on the omics analysis of ERG6Δ under CA stress, it was found that ERG6 knockout affects pathways such as intrinsic component of membrane and pentose phosphate pathway. In addition, the study revealed that 29 genes related to the cell wall-membrane system were up-regulated by more than twice, NADPH and NADP+ were increased by 2.48 and 4.41 times respectively, and spermidine and spermine were increased by 2.85 and 2.14 times, respectively, in ERG6Δ. Overall, the response of cell wall-membrane system, the accumulation of spermidine and NADPH, as well as the increased levels of metabolites in pentose phosphate pathway are important findings in improving the CA resistance. This study provides a theoretical basis for improving the tolerance of strains to CA and reducing the damage caused by CA to the ecological environment and human health.

The density peaks clustering (DPC) algorithm plays an important role in data mining by quickly identifying cluster centers using decision graphs to identify arbitrary clusters. However, the decision graph introduces uncertainty in determining the cluster centers, which can result in an incorrect number of clusters. In addition, the cut-off distance parameter relies on prior knowledge, which poses a limitation. To address these issues, we propose an improved automatic density peaks clustering (ADPC) algorithm. First, a novel clustering validity index called density-distance clustering (DDC) is introduced. The DDC index draws inspiration from the density and distance characteristics of cluster centers, which is applicable to DPC and aligns with the general definition of clustering. Based on the DDC index, the ADPC algorithm automatically selects the suitable cut-off distance and acquires the optimal number of clusters without additional parameters. Numerical experimental results validate that the introduced ADPC algorithm successfully automatically determines the optimal number of clusters and cut-off distance, significantly outperforming DPC, AP and DBSCAN algorithms.

Abstract In recent years, to improve air quality, significant efforts have been made to reduce regional aerosols including China, Europe and North America, which have potential impacts on climate. In this study, fast and slow climate responses in China to changes in domestic and foreign anthropogenic emissions of aerosols and precursors from 2013 to 2019 are investigated using the Community Earth System Model version 1 (CESM1). Aerosol emissions changes, especially reductions in China, North America and Europe, during 2013–2019 resulted in surface air temperature increases across the Northern Hemisphere. Global aerosol changes induce a regional warming of 0.2 °C in China, equally contributed by domestic and foreign emissions changes. China’s domestic emission reductions lead to a regional average temperature rise of 0.1°C, primarily driven by rapid atmospheric adjustments over eastern China. Foreign aerosol changes, particularly reductions in North America and Europe, also contributed to a 0.1°C warming in China through slow oceanic processes. This warming in China induced by foreign aerosol changes is due to a teleconnection between the aerosol-induced anomalous regional warming in Eastern U.S.-North Atlantic Ocean-Europe and the downstream East Asian climate through anomalous wave trains propagation. The comparable influence of domestic and foreign aerosol changes on climate in China underscores the importance of international collaboration in climate mitigation endeavors.

Acid soils occupy approximately 50% of potentially arable lands. Improving crop productivity in acid soils, therefore, will be crucial for ensuring food security and agricultural sustainability. High soil acidity often coexists with phosphorus (P) deficiency and aluminum (Al) toxicity, a combination that severely impedes crop growth and yield across wide areas. As roots explore soil for the nutrients and water required for plant growth and development, they also sense and respond to below-ground stresses. Within the terrestrial context of widespread P deficiency and Al toxicity pressures, plants, particularly roots, have evolved a variety of mechanisms for adapting to these stresses. As legumes, soybean (Glycine max) plants may acquire nitrogen (N) through symbiotic nitrogen fixation (SNF), an adaptation that can be useful for mitigating excessive N fertilizer use, either directly as leguminous crop participants in rotation and intercropping systems, or secondarily as green manure cover crops. In this review, we investigate legumes, especially soybean, for recent advances in our understanding of root-based mechanisms linked with root architecture modification, exudation and symbiosis, together with associated genetic and molecular strategies in adaptation to individual and/or interacting P and Al conditions in acid soils. We propose that breeding legume cultivars with superior nutrient efficiency and/or Al tolerance traits through genetic selection might become a potentially powerful strategy for producing crop varieties capable of maintaining or improving yields in more stressful soil conditions subjected to increasingly challenging environmental conditions.

Interprovincial disparities in carbon emissions from the construction industry (CECI) are an important challenge for future emissions reductions. Based on the CECI data of 30 provinces in China from 2010 to 2020, the interprovincial equity and decoupling of CECI were examined in this study. The conclusions are as follows: (1): The main CECI growth regions were the eastern Bohai Rim region and the Pearl River Delta region. Heilongjiang basically achieved a CECI carbon peak in 2016. (2) The three northeastern provinces and western provinces were the main high–high and low–low aggregation areas, respectively. The greatest degree of inequality was found in the western provinces. The inequality between the eastern and western areas was the largest, while the inequality between the central and western areas was the smallest. The inequality in CECI in the north–south region decreased year by year. (3) The decoupling status of Shandong and some western provinces has significantly worsened, while the decoupling status of Sichuan, Yunnan, and some eastern developed provinces has been improving. These conclusions will contribute to the improvement of regional emission reduction strategies.

The drive for continuous innovation in large-scale infrastructure necessitates advancements in techniques, addressing the challenges of constructing super-length concrete structures. This study investigated the emerging shift from traditional united expanding agent (UEA) to magnesia expansive agent (MEA) in conjunction with expansive strengthening bands (ESBs), marking a pivotal transition in ensuring monolithic integrity. Despite a decade of exploration, MEA–ESB implementation in real-world projects remains underdocumented, with scholarly focus primarily confined to material characterization. This research integrated empirical on-site tests of MEA–ESB with high-fidelity numerical simulations in ABAQUS. The finite element model (FEM) validation against actual test data underscored the precision of our modeling, capturing the complex thermomechanical behavior of the system. We introduced a sophisticated parametric analysis framework, elucidating the influence of critical parameters like the ESB-to-raft-width ratio and MEA concrete expansion rates. This granular understanding facilitated the fine-tuning of design parameters, advancing the practical application of MEA methodologies. A groundbreaking contribution entailed the formulation of predictive models for early-stage cracking, anchored in the guidelines of the ACI Committee 207 and refined through extensive parametric exploration. These formulae empower engineers to anticipate and mitigate cracking risks during the design phase, thereby enhancing project safety and efficiency. Notably, this study identified limitations in current prediction models, highlighting the need for future research to incorporate comprehensive lifecycle considerations, including hydration heat effects and time-dependent mechanical property evolution.

Currently, the study of analytical solutions for the seismic stability of slopes under anchorage conditions is one of the hottest subjects in engineering. In this paper, an analytical solution for the seismic safety of the three-dimensional (3D) two-stage rock slope reinforced with prestressed anchor cables governed by the nonlinear Hoek–Brown criterion was deduced, in which the analyses of seismicity were performed by the latest modified pseudo-dynamic method. This method supplements the consideration of the damping effect of the rock medium on seismic waves, which is more in line with the real seismic situation. A mathematical geometric model was developed for calculating the external forces work and internal energy dissipation acting on 3D rotating rock masses reinforced with prestressed anchor cables, in which the seismic work rate was calculated using a new layer-by-layer superposition summation method. The analytical solution of the safety factor could be collated as an explicit function of several variables, and then, the optimal value was obtained by the Particle Swarm Optimization (PSO) algorithm. To corroborate the accuracy of new analytical solutions, the results were contrasted with those of the pertinent literature. The results of the two comparisons were very close. Ultimately, the sensitivity analyses and coupling effects of seismic pseudo-dynamic factors and prestressing anchorage factors were carried out. It was found that even small seismic intensities had a large effect on the stability of rock slopes with developed joints. Increasing the number of steps and prestressing anchors can effectively improve the stability of rock slopes under seismic effects. The conclusions have significant implications for the anchorage design of the 3D two-stage rock slope as seismic events occur.

Abstract. Background:. LY01005 (Goserelin acetate sustained-release microsphere injection) is a modified gonadotropin-releasing hormone (GnRH) agonist injected monthly. This phase III trial study aimed to evaluated the efficacy and safety of LY01005 in Chinese patients with prostate cancer. Methods:. We conducted a randomized controlled, open-label, non-inferiority trial across 49 sites in China. This study included 290 patients with prostate cancer who received either LY01005 or goserelin implants every 28 days for three injections. The primary efficacy endpoints were the percentage of patients with testosterone suppression ≤50 ng/dL at day 29 and the cumulative probability of testosterone ≤50 ng/dL from day 29 to 85. Non-inferiority was prespecified at a margin of -10%. Secondary endpoints included significant castration (≤20 ng/dL), testosterone surge within 72 h following repeated dosing, and changes in luteinizing hormone, follicle-stimulating hormone, and prostate specific antigen levels. Results:. On day 29, in the LY01005 and goserelin implant groups, testosterone concentrations fell below medical-castration levels in 99.3% (142/143) and 100% (140/140) of patients, respectively, with a difference of -0.7% (95% confidence interval [CI], -3.9% to 2.0%) between the two groups. The cumulative probabilities of maintaining castration from days 29 to 85 were 99.3% and 97.8%, respectively, with a between-group difference of 1.5% (95% CI, -1.3% to 4.4%). Both results met the criterion for non-inferiority. Secondary endpoints were similar between groups. Both treatments were well-tolerated. LY01005 was associated with fewer injection-site reactions than the goserelin implant (0% vs. 1.4% [2/145]). Conclusion:. LY01005 is as effective as goserelin implants in reducing testosterone to castration levels, with a similar safety profile. Trial registration:. ClinicalTrials.gov, NCT04563936.

Aim. To explore the potential factors that influence the presentation and recovery of postoperative chylous ascites (CA) in gynecological malignancies. Methods. We reported two cases of postoperative CA following gynecological surgery and reviewed the clinical features of 140 patients from 16 relevant papers. Patients’ clinicopathological characteristics, surgical approach, and management were summarized. The onset and resolution times of postoperative CA in different groups were analyzed separately. Results. The two patients in our report had recovery after conservative treatments. According to the literature review, the median time of onset of postoperative CA was 5 days (range, 0–75 days) after surgery. The median resolution time was 9 days (range, 2–90 days). Among patients, 87.14% of them had lymphadenectomy during gynecological surgeries, while 92.86% of the patients had resolution after conservative treatments. Conclusions. Lymphadenectomy during surgery may be relevant to the postoperative CA. Conservative management could be the initial choice for postoperative CA treatment.

With the advancement of information technology and artificial intelligence, translation technologies have seen rapid development in language services and increasing integration in higher education. However, research on factors affecting students' acceptance of these technologies remains limited. This study intends to formulate and test an extended Technology Acceptance Model (TAM) incorporating computer self-efficacy and perceived enjoyment to investigate students' adoption of translation technologies. A questionnaire survey was conducted among 370 college students in China experienced with using translation technologies. The results from the structural equation modeling demonstrated a positive prediction on perceived ease of use and enjoyment from computer self-efficacy. Perceived enjoyment increased perceived ease of use and attitudes. Perceived ease of use positively influenced perceived usefulness and attitudes. Finally, attitudes positively predicted greater behavioral intentions to use translation technologies. However, computer self-efficacy was identified to have no significant effect on perceived usefulness. The study makes significant theoretical contributions by expanding TAM and offering practical guidance to improve students' acceptance of translation technologies in tertiary education.

Abstract Particulate nitrate is a major component of fine particulate matter (PM2.5). Its formation may be varyingly sensitive to emissions of ammonia (NH3), nitrogen oxides (NOx ≡ NO + NO2), and volatile organic compounds (VOCs), depending on local conditions. Diagnosing these sensitivities is critical for successful air quality management. Here, we show that satellite measurements of tropospheric NH3 and NO2 columns can be used as a quick indicator of the dominant sensitivity regime through the NH3/NO2 column ratio together with the NO2 column. We demonstrate the effectiveness of this indicator with the GEOS‐Chem chemical transport model and define thresholds to separate the different sensitivity regimes. Applying the method to wintertime IASI and OMI observations in East Asia reveals that surface nitrate is dominantly VOC‐sensitive in the southern North China Plain (NCP), NOx‐sensitive in most of the East China Plain, and NH3‐sensitive in the northern NCP, southern China, and Korea.

The shear strength of unsaturated soils exhibits significant nonlinearity, while previous studies often simplified it with linear strength models. The objective of this paper is to investigate the distinctions in the stability of three-dimensional (3D) tunnel faces when using linear and nonlinear strength models. A new 3D rotational failure mechanism and an extended form of the Mohr–Coulomb (M-C) failure criterion were integrated into the kinematically limited analysis (KLA) framework to describe the failure characteristics of tunnel faces. Subsequently, the factor of safety (FS) of the 3D tunnel faces was calculated using the strength reduction method (SRM). In the discussion section, the impacts of nonlinear shear strength, matric suction in the unsaturated soils, and the 3D geometric parameters of the tunnel on the stability of the tunnel face were analyzed. The outcomes indicate that, in unsaturated soil conditions, diverse nonlinear strength calculation models and soil types exert disparate influences on the FS of 3D tunnel faces. The main novelty of this study lies in establishing an effective method for assessing the stability of tunnel faces in unsaturated soils.

The vertical mass exchange of ozone (O3) plays an important role in determining surface O3 air quality, the understanding of which, however, is greatly limited by the lack of continuous measurements in the vertical direction. Here, we characterize O3 variations at a high-altitude monitoring site at the top of Shanghai Tower (SHT) and discuss the potential impacts of the vertical exchange of air pollutants on O3 air quality within the urban planetary boundary layer (PBL) based on continuous measurements during 2017–2018. During the daytime, two distinct patterns of vertical O3 gradient are detected. In summer, the daytime O3 formation at SHT is observed to be more limited by nitrogen oxides (NOx) than the surface, which, together with the efficient vertical mixings, results in higher O3 levels in the upper mixing layer. In cold months, the opposite vertical gradient is observed, which is associated with weak vertical exchange and NOx-saturated O3 formation. A nighttime O3 reservoir layer and consistent morning O3 entrainments are detected all year round. These results provide direct evidence of the vertical mixings within the urban PBL, underscoring the pressing need for improving vertical resolution in near-surface layers of air quality models.

Abstract Atmospheric black carbon (BC) aerosols have been long‐lasting uncertain components in environmental and climate studies. Global climate models (GCMs) potentially overestimate BC absorption efficiency due to a lack of consideration of complex BC microphysical and mixing properties. We extract multiple BC properties from observations and develop an aerosol optical module known as Advanced Black Carbon (ABC) in the framework of the Modal Aerosol Model version 4 (MAM4). The ABC module is implemented in the Community Atmosphere Model version 6 (CAM6) and evaluated by in situ and remote sensing observations. CAM6‐ABC addresses the shortcomings of CAM6‐MAM4 in terms of BC microphysical and mixing properties, particularly their size, mixing state and optical simulations. Sensitivity simulations show that the global BC absorption aerosol optical depth at 550 nm simulated by CAM6‐ABC is reduced by ∼29% compared with that in CAM6‐MAM4. The BC absorption enhancement simulated by CAM6‐ABC is reduced from ∼2.6 of the default MAM4 to ∼1.4, which is closer to the observed values (mostly less than 1.5). With improved BC absorption estimation, the biases of aerosol single‐scattering coalbedo simulations are reduced by 18%–69% compared with global Aerosol Robotic Network observations. Moreover, the globally averaged BC direct radiative effect is reduced from 0.37 to 0.28 W/m2 at the top of the atmosphere. Our new scheme alleviates the overestimation of BC absorption in GCMs by constraining BC microphysical and mixing properties when assessing aerosol radiative and climate effects, and it can be easily implemented in most modal‐based aerosol modules of climate models.

Abstract A global chemical transport model GEOS‐Chem was applied to examine the concentrations of PM2.5 and O3 in China, related health burdens and the relative importance of changes in air pollutants and population aging over 2015–2060 under carbon neutral pathway (Shared Socioeconomic Pathway limiting end‐of‐century radiative forcing to 1.9 W m−2, SSP1‐1.9). Results showed synergistic reductions of 100.0%, 72.0%, and 24.4% in net CO2 emission, concentration of annual mean PM2.5, and the 90th maximum daily average 8‐hr (MDA8) O3 over China, respectively, during 2015–2060. Pollutants‐related deaths owing to changes in pollutants and population were simulated to increase by 38.1% over 2015–2060 because of population aging. Over 2015–2060, sensitivity studies showed that the decline in pollutants alone would mitigate deaths by 5,172.7 thousand, while the population aging alone would increase deaths by 2,633.3 thousand. Carbon neutrality will avoid 3,520.0 thousand deaths in 2060 relative to a higher‐carbon pathway (SSP5‐8.5).

Abstract Background High‐risk human papillomavirus (hrHPV) infection is a key factor that alters cervicovaginal microbiota patterns and causes cervical intraepithelial neoplasias (CINs) or even cervical cancer. Although local excisional treatment can clear hrHPV infection and restore the cervicovaginal microbiota, it is unclear which cervicovaginal microbiota represents recovery. Our objective was to describe the cervicovaginal microbiota before and after treatments and to assess the association between the microbiota and HPV persistence. Results A cohort of 91 participants was classified into four groups (healthy control women and HPV16‐infected women with CIN I, CIN II/III, and squamous cell carcinoma [SCC]). Endocervical swabs were collected 3 months prior to treatment and at 3 months post‐treatment for bacterial 16S rRNA gene pyrosequencing and for HPV DNA testing. There was an increase in the number of Lactobacillus bacterial species present after the clinical treatments, and the community state type (CST) profiles were shifted from dysbiotic CSTs II and IV to Lactobacillus‐dominated CSTs I and III. Specifically, the composition of Geobacter and Prevotella before treatment and Lactobacillus secaliphilus after treatment might have been related to CIN I, the composition of Burkholderia before treatment and Lactobacillus iners after treatment might have been related to CIN II/III, and the composition of Atopobium and Aerococcus before treatment and Bacilli after treatment might have been related to SCC. Further functional predictions revealed that the composition differences were linked to infectious disease‐ and cancer‐related genes. Conclusion Our study provides an illustration of the changes in CSTs and the cervicovaginal microbiota before and after HPV16 clearance in each disease state.

Abstract Background Although evidence has revealed that miR-200a-3p is involved in the malignant progression of various tumors, the regulatory mechanism of miR-200a-3p in the development of cervical cancer (CC) cells with different HPV statuses remains unknown. The present study was to investigate the differential effects of either miR-200a-3p or YAP on tumorous cells’ fate in vitro in HPV-negative and HPV-positive cervical cancer cell models, and to explore if the changes in proliferation, migration, and invasion of the CC cells with different HPV statuses could be attributed to the differential interactions between miR-200a-3p and YAP. Methods The colony formation assays, EDU assays and Transwell assays were performed for CC cell proliferation, migration and invasion capacities analysis. The prediction of downstream targets of miR-200a-3p was performed by bioinformatical databases. The dual-luciferase reporter assays were used to validate the binding sites of miR-200a-3p and YAP. The qRT-PCR assays were performed to quantify the mRNA expression of miR-200a-3p and YAP, and the protein levels of YAP were examined by Western blot analysis. Results The results demonstrated that miR-200a-3p overexpression suppressed proliferation, migration, and invasion of the HPV-negative C33A cells but promoted the growth and metastasis of HPV-positive CC cells, while YAP promoted the cell growth and metastasis not only in HPV-negative but also in the HPV-positive CC cells. The suppressive role of miR-200a-3p in C33A cells appeared to be mediated partially by direct interaction with YAP, and YAP might participate in miR-200a-3p-mediated cellular changes in CC cells differing from not only the presence or absence of HPV but even also the subtypes of HPV of CC cells. Meanwhile, we preliminarily revealed that the expression level of miR-200a-3p was significantly decreased in HPV-negative, but not in HPV16-positive cervical neoplasm mucus samples. Conclusion miR-200a-3p-mediated functional changes of YAP exhibited regulatory effects on cells’ fate differentially in HPV-negative and HPV-positive cervical cancer cells.

Abstract Background The traditional prostate cancer (PCa) model is established by injecting cell suspension and is associated with a low tumor formation rate. Cell sheet technology is one of the advancements in tissue engineering for 3D cell-based therapy. In this study, we established ectopic and orthotopic PCa models by cell sheet technology, and then compared the efficiency of tumor formation with cell suspension injection. Methods DU145 cells were seeded on 35 mm temperature-sensitive dishes to form PCa cell sheets, while the cell suspension with the same cell density was prepared. After transplanting into the nude mice, the tumor volumes were measured every 3 days and the tumor growth curves were conducted. At the time points of 2 weeks and 4 weeks after the transplantation, magnetic resonance imaging (MRI) was used to evaluate the transplanting site and distant metastasis. Finally, the mice were sacrificed, and the related tissues were harvested for the further histological evaluation. Results The orthotopic tumor formation rate of the cell sheet injection group was obviously better than that in cell suspension injection group (100% vs 67%). Compared with cell suspension injection, the tumors of DU145 cell sheet fragments injection had the higher density of micro-vessels, more collagen deposition, and lower apoptosis rate. There was no evidence of metastasis in forelimb, lung and liver was found by MRI and histological tests. Conclusion We successfully cultured the DU145 cell sheet and can be used to establish ectopic and orthotopic PCa tumor-bearing models, which provide an application potential for preclinical drug development, drug-resistance mechanisms and patient individualized therapy.

The microphysical attributes of black carbon (BC) can determine its absorption and hygroscopic properties. However, long-term information is difficult to obtain from the field. In this study, the BC properties including mass concentration, the coating volume ratio (VR) relative to the refractory BC (rBC), the rBC diameter and the fraction of cloud condensation nuclei (CCN), are derived from a number of field experiments using a random forest model. This model effectively derives the long-term BC microphysical properties in the Beijing region from 2013 to 2020 using continuous measurements of particulate matter, gas, BC mass concentration and meteorological parameters. The results reveal notably higher BC coatings (mean VR = 7.2) and a greater fraction of CCN-like BC (51%) in the winter compared to other seasons. Following the implementation of national air pollution control measures in 2017, BC mass exhibited a substantial reduction of 60% (29%) in the winter (summer), and VR decreased by 45% (24%). Apart from the influence of meteorological variations, these can be attributed to the declined primary emissions and the gas precursors which are associated with secondary formation of BC coatings. The reduction of both BC mass loading and coatings leads to its solar absorption decreasing by 50%, and the fraction of CCN-like BC (likely in clouds) decreasing by 23%. Environmental regulation will therefore continue to reduce both direct and indirect radiative impacts of BC in this region.

Third Front construction was a massive defensive industrialization and economic campaign in response to the threat of war from both the Soviet Union and the United States in the 1960s in the remote and isolated hinterlands of China, which remained a top secret for many years before 1980s. Academic community has been making growing efforts in exploring its origin, history, development and influence ever since. As the largest industrial relocation in modern Chinese history, it is crucial for understanding the complexity of China today. Based on the bibliometric analysis of the retrieved and pruned 370 bibliographic references from Google Scholar, this article identified the major references, leading authors, contributing journals, productive countries or territories and cooperating institutions, as well as hot topics and emerging trends of the research on Third Front construction. By adopting VOSviewer as the visualizing tool, the paper found that Third Front construction continues to be a hot research topic with growing trend from the perspectives of historical background, macro-policy, industrial development, third-front cities and modern development of the Third Front construction in China. It also revealed the potential research trends in urbanization, industrial heritage and Cold War related to Third Front construction. Meanwhile, the paper made a brief comparison between the research on China’s third front construction in English and Chinese languages. The findings could be of theoretical values to the academic community for the revelation of the knowledge landscape of Third Front construction and the entry point to the modern history of China.

In the summer of 2020, China experienced record rainfall and flooding. Here, the authors investigate the connection between the rainfall and the abrupt decrease in emissions due to Covid-19 and find that the emission decrease may have contributed to the rainfall.

Background: Pyroptosis, gasdermin-mediated programmed cell death, is readily induced in macrophages by activation of the canonical inflammasome (caspase-1) or by intracellular lipopolysaccharide (LPS)-mediated non-canonical inflammasome (caspase-11) activation. However, whether pyroptosis is induced similarly in hepatocytes is still largely controversial but highly relevant to liver pathologies such as alcoholic/nonalcoholic liver disease, drug-induced liver injury, ischemia-reperfusion and liver transplant injury, or organ damage secondary to sepsis. Methods and Results: In this study we found that hepatocytes activate and cleave gasdermin-D (GSDMD) at low levels after treatment with LPS. Overexpression of caspase-1 or caspase-11 p10/p20 activated domains was able to induce typical GSDMD-dependent pyroptosis in hepatocytes both in vitro and in vivo. However, morphologic features of pyroptosis in macrophages (eg, pyroptotic bodies, cell flattening, loss of cell structure) did not occur in pyroptotic hepatocytes, with cell structure remaining relatively intact despite the cell membrane being breached. Our results suggest that hepatocytes activate pyroptosis pathways and cleave GSDMD, but this does not result in cell rupture and confer the same pyroptotic morphologic changes as previously reported in macrophages. This is true even with caspase-1 or caspase-11 artificial overexpression way above levels seen endogenously even after priming or in pathologic conditions. Conclusions: Our novel findings characterize hepatocyte morphology in pyroptosis and suggest alternative use for canonical/non-canonical inflammasome activation/signaling and subsequent GSDMD cleavage because there is no rapid cell death as in macrophages. Improved understanding and recognition of the role of these pathways in hepatocytes may result in novel therapeutics for a range of liver diseases.

Stratospheric intrusion (SI) is an important source of tropospheric ozone (O3). Here, we used the online coupled Weather Research and Forecasting-Chemistry (WRF-Chem) model to simulate a typical SI event that occurred over eastern China on 15–19 July 2016 and investigate the impacts of SI on near-surface O3 pollution. The results show that the large-scale circulation of SI was characterized by a deep trough over central China and South Asia high and Western Pacific Subtropical high located to the east and west of the deep trough, respectively. With the evolution of the deep trough, the strong downdrafts behind the trough lead to O3-rich air injected into the lower troposphere across eastern China. By using a tracer tagging method in WRF-Chem, we quantified the SI contributed up to 6.5 ppb to the surface O3 concentration. According to the integrated process rate analyses, which were employed to quantify the contribution of different physical/chemical processes to O3, the advection process dominated variations in troposphere O3 with positive contribution ranging from 0.1 to 13.8 ppb h−1. As altitude decreases, the contribution of advection diminishes as the intensity of the SI gradually weakens. Although the vertical advection contributed limited O3 that directly reached the ground, we revealed that the SI has significant impact on near-surface O3 over a large territory of eastern China through regional transport by horizontal advection process. Below 691 m, the positive contribution of horizontal advection to O3 ranges from 1.6 to 13.4 ppb h−1. This study highlights the natural effect of SI on summertime O3 pollution in eastern China and will help the development of a future O3 pollution alert system.

The Clean Air Plan has been active in China since 2013 to mitigate severe PM2.5 pollution. In this study, we applied the air quality model WRF-Chem to simulate PM2.5 in the Yangtze River Delta (YRD) region of China in 2017, with the aim of assessing the air quality improvement and its associated health burden in the final year of the Clean Air Plan. To better describe the fate of various PM2.5 compositions, we updated the chemical mechanisms in the model beforehand, including heterogeneous sulfate reactions, aqueous secondary organic aerosol (SOA) uptake, and volatility basis set (VBS) based SOA production. Both the observation and simulation results agreed that the stringent clear air action effectively reduced the PM2.5 pollution levels by ∼ 30 %. The primary PM2.5 (−6 ∼ − 16 % yr−1) showed a more significant decreasing trend than the secondary PM2.5 (−2 ∼ − 8 % yr−1), which was mainly caused by the directivity of the clear air actions and the worsening ozone pollution in the recent years. The inconsistent decreasing trends of PM2.5 components subsequently led to an increasing proportion of secondary PM2.5. Nitrate particles, higher in the central and western YRD region, have replaced sulfate and have become the largest component of secondary inorganic aerosols year-round, except in summer, when strong ammonium nitrate evaporation occurs. In addition, SOA remains an important component (21 ∼ 22 %) especially in summer, most of which is produced from the oxidation and ageing of semi/intermediate volatile organic compounds (S/IVOC). Furthermore, we quantified the associated health impacts and found that the Clean Air Plan has largely reduced premature mortality due to PM2.5 exposure in the YRD region from 399.1 thousand to 295.7 thousand. Our study highlights the benefits of the Clean Air Plan and suggests that subsequent PM2.5 improvement should be geared more towards controlling secondary pollutants.

Geogrid reinforced soil slope is a new type of slope supporting structure, which is of great significance for improving the slope stability, saving construction land and preserving the ecological environment. In order to provide the optimal design solution of the reinforced soil slope for high fill slopes, a case study is conducted on the No.6 slope at the northwest corner of the airport runway. Three design schemes with different slope ratios are carried out firstly based on the geological conditions of the slope and the field situation of the high fill slope. Secondly, the simplified Bishop method, Spencer wedge method and Morgenstern price method are used to calculate the stability factors of the reinforced slopes under the natural, rainstorm and earthquake conditions. Finally, the deformation characteristics of the slope and tensile force in reinforcements are investigated based on the finite element method under the natural conditions. The results show that all the proposed design schemes can meet the requirements of slope stability under different working conditions. A zigzag distribution of the tensile force in reinforcements in the multi-stage reinforced soil slope is observed along the slope height, and the maximum axial force of reinforcement increases abruptly at the toe of each slope. Compared with the design scheme of the gentle reinforced soil slope with the slope ratio of 1∶1.5, the reinforced retaining wall with the slope ratio of 1∶0.25 shows obvious advantages in reducing the slope height, consumption of geosynthetics, filling and excavation amount as well as slope protection area. The key issues including the slope stability, construction cost and duration are comprehensively considered, and the design scheme of the reinforced earth wall is reasonable.

Biomass burning emissions have been shown to influence clouds in the Atlantic, but its influence in other regions is not well known. Here, the authors show that biomass burning aerosols increase the low-cloud cover over subtropical southeastern Asia by a similar magnitude than over the Atlantic.

Introduction: As the world is consistently driven by the infusion of new-generation information technology and the knowledge economy, college students are placed under mounting pressure in developing occupation-related competencies. Their employability has been receiving growing concerns from stakeholders such as higher education institutions, governments, employers, parents, and even student groups themselves as it plays a decisive role in occupational success, social stability, and economic prosperity. Under the theoretical guidance of social cognitive theory, this study set out to investigate the cognitive and psychological mechanisms through which innovation capability, social adaptability, and self-efficacy influence the employability of college students. It also attempts to analyze the mediating role of self-efficacy in the relations between innovation capability, social adaptability, and employability which has been rarely studied in academia.Methods: A quantitative approach was employed in this study. Data was collected from 726 undergraduates from 9 higher education institutions in the mainland of China by questionnaire survey method. The research model showed a good fit (χ2/df=4.46, RMSEA=0.069, SRMR=0.049, GFI=0.934, CFI=0.965, NFI=0.955, TLI=0.955). Structural equation modeling (SEM) was applied to this study for data analysis.Results: The findings showed that innovation capability, social adaptability, and self-efficacy significantly and positively correlates with undergraduates’ employability. University students with stronger innovation capability, social adaptability, and self-efficacy tend to be more employable in the job market. Model 4 of SPSS PROCESS Macro revealed that self-efficacy played a mediating role in the correlation between innovation capability, social adaptability, and employability.Discussion: Undergraduates with higher levels of innovation capability and social adaptability are more confident in their abilities to take specific actions and achieve expected goals, which in turn intensifies their employability. The study suggests the possibility of improving undergraduates’ employability through positive interference of innovation capability, social adaptability, and self-efficacy in the era of information technology and knowledge-based economy.

Projecting future air pollution and related health burdens remains challenging because of the complex interactions among future emissions, population, and climate change. In this study, we estimated the premature deaths attributed to ambient fine particulate matter (PM2.5) and ozone (O3) from 2015 to 2100 under four socioeconomic climate scenarios based on an age-stratified assessment method. We found that PM2.5 will decrease in all shared socioeconomic pathway (SSP) scenarios and O3 will decrease in the SSP1-2.6 and SSP2-4.5 scenarios, contributing to a decrease in premature mortality together with the declining total population in China. However, the benefits of a decline in population size and PM2.5 and O3 concentrations over time will be largely offset by population aging, and premature death caused by PM2.5 and O3 will continue to rise till 2060–2080. This impact was greater for the O3-related deaths than those for PM2.5. Our study highlights the importance of future prevention strategies that must jointly improve air quality and susceptibility to aging.

Peritoneal metastases are associated with a low response rate to immune checkpoint blockade (ICB) therapy. The numbers of peritoneal resident macrophages (PRMs) are reversely correlated with the response rate to ICB therapy. We have previously shown that TLR9 in fibroblastic reticular cells (FRCs) plays a critical role in regulating peritoneal immune cell recruitment. However, the role of TLR9 in FRCs in regulating PRMs is unclear. Here, we demonstrated that the class A TLR9 agonist, ODN1585, markedly enhanced the efficacy of anti–PD-1 therapy in mouse models of colorectal peritoneal metastases. ODN1585 injected i.p. reduced the numbers of Tim4+ PRMs and enhanced CD8+ T cell antitumor immunity. Mechanistically, treatment of ODN1585 suppressed the expression of genes required for retinoid metabolism in FRCs, and this was associated with reduced expression of the PRM lineage–defining transcription factor GATA6. Selective deletion of TLR9 in FRCs diminished the benefit of ODN1585 in anti–PD-1 therapy in reducing peritoneal metastases. The crosstalk between PRMs and FRCs may be utilized to develop new strategies to improve the efficacy of ICB therapy for peritoneal metastases.

Pruning is an important strategy for increasing tea production. However, the effects of pruning on tea quality are not well understood. In this study, tea leaves were collected from Wuyi Mountain for both ionomic and metabolomic analyses. A total of 1962 and 1188 fresh tea leaves were respectively collected from pruned and unpruned tea plants sampled across 350 tea plantations. Ionomic profiles of fresh tea leaves varied significantly between pruned and unpruned sources. For tea plants, pruning was tied to decreases in the concentrations of mobile elements, such as nitrogen (N), phosphorus (P), potassium (K) and magnesium (Mg), and dramatic increases in the concentrations of the immobile ions calcium (Ca), aluminum (Al), manganese (Mn), boron (B) and cobalt (Co). Clustering and heatmap analysis showed that pruning also affected tea leaf metabolism. Among 85 metabolites that were significantly impacted by pruning, 30 were identified through random forest analysis as characteristic differential metabolites with a prediction rate of 86.21%. Redundancy analysis showed that pruning effects on mineral nutrient concentrations accounted for 25.54% of the variation in characteristic metabolites between treatments, with the highest contributions of 6.64% and 3.69% coming from Ca and Mg, respectively. In correlation network analysis, Ca and Mg both exhibited close, though opposing correlations with six key metabolites, including key quality indicators 1,3-dicaffeoylquinic acid and 2-O-caffeoyl arbutin. In summary, large scale sampling over hundreds of tea plantations demonstrated that pruning affects tea quality, mainly through influences on leaf mineral composition, with Ca and Mg playing large roles. These results may provide a solid scientific basis for improved management of high-quality tea plantations.

Symbiotic nitrogen fixation (SNF) provides sufficient nitrogen (N) to meet most legume nutrition demands. In return, host plants feed symbionts carbohydrates produced in shoots. However, the molecular dialogue between shoots and symbionts remains largely mysterious. Here, we report the map-based cloning and characterization of a natural variation in GmNN1, the ortholog of Arabidopsis thaliana FLOWERING LOCUS T (FT2a) that simultaneously triggers nodulation in soybean and modulates leaf N nutrition. A 43-bp insertion in the promoter region of GmNN1/FT2a significantly decreased its transcription level and yielded N deficiency phenotypes. Manipulating GmNN1/GmFT2a significantly enhanced soybean nodulation, plant growth, and N nutrition. The near-isogenic lines (NILs) carrying low mRNA abundance alleles of GmNN1/FT2a, along with stable transgenic soybeans with CRISPR/Cas9 knockouts of GmNN1/FT2a, had yellower leaves, lower N concentrations, and fewer nodules than wild-type control plants. Grafting together with split-root experiments demonstrated that only shoot GmNN1/FT2a was responsible for regulating nodulation and thereby N nutrition through shoot-to-root translocation, and this process depends on rhizobial infection. After translocating into roots, shoot-derived GmNN1/FT2a was found to interact with GmNFYA-C (nuclear factor-Y subunit A-C) to activate symbiotic signaling through the previously reported GmNFYA-C-ENOD40 module. In short, the description of the critical soybean nodulation regulatory pathway outlined herein sheds novel insights into the shoot-to-root signaling required for communications between host plants and root nodulating symbionts.

Most of the published literature regarding bearing capacity are often focused on linear and associative soils. Concerning the intrinsic strength nonlinearity in dilatancy soils, this study investigates the problem of the seismic bearing capacity in the framework of the kinematic theorem of limit analysis. The conventional linear Mohr–Coulomb criterion is substituted with a nonlinear power law criterion to depict the nonlinearity of the soil strength. The non-associative feature of soil materials is considered by defining a nonlinear dilatancy coefficient. A generalized tangential technique is accordingly introduced to linearize the strength envelope for making the nonlinear criterion tractable in the analysis. A non-symmetrical translational failure mechanism that is comprised of several rigid wedges is used to characterize the failure of the foundation at the limit state. Moreover, the seismic action is considered by the classic pseudo-static method. Based upon the energy equilibrium theory of the upper-bound limit analysis, new analytical solutions are derived from the work-balanced equation with nonlinearity and dilatancy. This rigorous upper-bound solution is formulated as a multivariate optimization problem and is readily addressed by sequential quadratic programming (SQP). To verify the reliability of the new expressions, the present results are compared with already posted solutions and the original pseudo-dynamic solutions. The comparative results show a good agreement with previous works, and the correctness and rationality of the new analytical solutions are validated. The detailed parametric study reveals that, in the non-associative flow soils, the ultimate bearing capacity is significantly decreased with a reduction in the dilatancy coefficient. Particularly in the linear condition, namely m = 1, the larger the internal friction angle is, the more obvious the influence of the non-associative feature on the bearing capacity is.

Soybean (Glycine max (L.) Merr.) is an important nutritional crop with high seed protein content. Production of high protein concentrations relies on sufficient nutrient supplies, especially of nitrogen (N) and phosphorus (P). Although the genetic basis for seed quality traits has been well studied, little information exists on any genetic connections between seed quality and nutrient supplies in soybean. Here, a recombinant inbred line (RIL) population of 179 progeny was generated using HC6 and JD17 as parents contrasting in seed quality and N and P translocation efficiencies. Seed protein and N and P translocation efficiencies were higher in HC6 than in JD17. Meanwhile, positive correlations were observed between seed protein content and translocation efficiency of N and P in RILs, implying that high N and P translocation efficiencies might facilitate seed protein accumulation. A genetic map was constructed using 5250 SNP markers covering a genetic distance of 3154.83 cM. A total of 6 loci for quality and 13 loci for N and P translocation efficiency were detected. Among them, two fragments on chromosome 6 and chromosome 20 contained multiple significant markers for both quality and N and P translocation efficiencies, with the respective observed LOD values ranging from 2.98 to 5.61, and 3.01 to 11.91, while the respective PVE values ranged from 8.2% to 13.9%, and 8.3% to 28.0%. Interestingly, one significant locus on chromosome 20 appears to be the product of a transposable element (TE) InDel in Glyma.20G085100, with progeny lacking the TE also exhibiting higher N and P translocation efficiencies, along with higher seed protein contents. Taken together, these results provide genetic evidence that increasing N and P translocation efficiencies may lead to increasing protein contents in soybean seeds. Furthermore, a TE InDel may be used as a genetic marker for breeding elite soybean cultivars with high protein content and N and P translocation efficiencies.

Abstract Background Shoot architecture is fundamentally crucial to crop growth and productivity. As a key component of shoot architecture, plant height is known to be controlled by both genetic and environmental factors, though specific details remain scarce. Results In this study, 308 representative soybean lines from a core collection and 168 F9 soybean progeny were planted at distinct field sites. The results demonstrated the presence of significant genotype × environment interaction (G × E) effects on traits associated with plant height in a natural soybean population. In total, 19 loci containing 51 QTLs (quantitative trait locus) for plant height were identified across four environments, with 23, 13 and 15 being QTLs for SH (shoot height), SNN (stem node number) and AIL (average internode length), respectively. Significant LOD ranging from 2.50 to 16.46 explained 2.80–26.10% of phenotypic variation. Intriguingly, only two loci, Loc11 and Loc19–1, containing 20 QTLs, were simultaneously detected across all environments. Results from Pearson correlation analysis and PCA (principal component analysis) revealed that each of the five agro-meteorological factors and four soil properties significantly affected soybean plant height traits, and that the corresponding QTLs had additive effects. Among significant environmental factors, AD (average day-length), AMaT (average maximum temperature), pH, and AN (available nitrogen) had the largest impacts on soybean plant height. Therefore, in spite of uncontrollable agro-meteorological factors, soybean shoot architecture might be remolded through combined efforts to produce superior soybean genetic materials while also optimizing soil properties. Conclusions Overall, the comprehensive set of relationships outlined herein among environment factors, soybean genotypes and QTLs in effects on plant height opens new avenues to explore in work aiming to increase soybean yield through improvements in shoot architecture.

Surface ozone concentrations in the Yangtze River Delta (YRD) region in China have shown a significant increase with the dramatic reduction of anthropogenic nitrogen oxide (NOx) emissions since 2013. As the nonlinearity between ozone and its precursors (i.e., NOx and volatile organic compounds (VOCs)) varies greatly in time and space, we quantify the monthly changes of surface ozone with the co-control of NOx and VOCs anthropogenic emissions in the YRD region from May to October 2017 by using the GEOS-Chem model. Model evaluations show that the GEOS-Chem model exhibits good performance to simulate ozone concentrations in the region. For May–September 2017, most areas in the YRD region are under a transitional regime, but the regions with high anthropogenic emissions including Shanghai and southern Jiangsu are under a VOCs-limited regime. In October, basically, the entire YRD region is under a VOCs-limited regime. Generally, reducing VOCs emissions only is the most effective method for ozone control in the YRD. Nanjing is under a VOCs-limited regime in May, June, September, and October, and under a transition regime from July to August. The VOCs/NOx emission reduction ratio of 1:1 is effective for ozone mitigation in Nanjing (Shanghai) in May, June, and September (for May–September); the corresponding ratio is 2:1 in October. Hangzhou belongs to a transitional regime from May to September and is under a VOCs-limited regime in October. Reducing NOx emissions only would control ozone in Hangzhou from May to September, while the VOCs/NOx emission reduction ratio of 1:1 is favorable to reduce ozone concentrations in October. During high pollution days on July 22–27, 2017, the three cities belong to a transitional regime and reducing NOx emissions only is generally the most effective way to control high ozone pollution. GEOS-Chem tagged ozone simulation shows that ozone problem in the region is caused by the joint effect of local generation and regional and long-distance transport. Local generation (19.0–50.7%) is generally the largest contributor to monthly mean ozone concentrations in Jiangsu and Shanghai, Zhejiang, and central eastern China; the contribution of ozone from regions outside the YRD is larger in spring and autumn (42–76.0%) than in summer (23.3–51.8%). Since the annual VOCs (NOx) anthropogenic emissions in the region have shown a decline by 8% (11%) from 2017 to 2020 and would continue to reduce by 10% (10%) by 2025 according to the Chinese government requirement, the growth of ozone would be stopped in the YRD for May–September but likely to continue in October. Our study thus would provide a scientific base for guiding the effective emission reduction strategies to control ozone pollution in the YRD region.
