Abstract

The effects of muscarinic ligands on acid secretion were examined by estimating the accumulation of [ 14 C]aminopyrine in gastric glands isolated from guinea pigs. The accumulation of [ 14 C]aminopyrine in the presence of 0.1 mM histamine was potentiated by 1 μM carbachol but suppressed by 1 mM. These two effects of carbachol were abolished by atropine, pirenzepine and AF-DX 116. Assuming that the binding of carbachol to one site (Site 1) increases [ 14 C]aminopyrine accumulation but its binding to the other site (Site 2) reduces [ 14 C]aminopyrine accumulation, we analysed the dose-response curves for the carbachol effects in the absence and presence of different concentrations of atropine, pirenzepine and AF-DX 116. The dissociation constants determined for these ligands at Sites 1 and 2 were as follows: carbachol, 0.28 and 7.1 μM; atropine, 0.28 and 0.54 nM; pirenzepine, 45 and 560 nM; and AF-DX 116, 380 and 4400 nM, respectively. The binding of [ 3 H]N-methylscopolamine to the gastric glands indicated the presence of two populations of binding sites with different affinities for the above ligands, other than atropine. The apparent dissociation constants, which were estimated by analysing the displacement curves for [ 3 H]N-methylscopolamine binding, were as follows: carbachol, 0.18 μM (10%) and 31 μM (90%); atropine, 1.24 nM; pirenzepine, 15 nM (16%) and 220 nM (84%); and AF-DX 116, 370 nM (10%) and 2970 nM (90%). These results suggest that there are two kinds of muscarinic acetylcholine receptors in the guinea pig gastric gland, one potentiating and the other inhibiting the acid secretion induced by histamine.

Abstract

The relation between finger blood flow level and the periodicity in slower finger volume fluctuations was studied in humans by using mercury plethysmography. The periodicity was analysed by means of autocorrelographic and harmonic analysis. Slower periodic waves, not originating in the heart beat or the respiratory cycle, were found throughout the range of finger blood flow above 10 ml/min 100 ml tissue, but not below this blood flow level. The period of the shortest volume wave, except the pulse and respiratory waves, prolonged with increased finger blood flow. [ABSTRACT FROM AUTHOR]

Abstract

A family with a complete deficiency of lactate dehydrogenase M-subunit was investigated. The propositus was an 18-year-old male who complained of exertional pigmenturia and easy fatigue. Marked discrepancy was observed in the ratio between creatine kinase and lactate dehydrogenase (CK/LDH). Electrophoretic analysis of serum LDH isoenzymes of the propositus demonstrated only one activity band of LDH H4. A complete lack of the LDH M-subunit was similarly demonstrated in erythrocytes, leukocytes and in the intermediate vastus muscle. LDH levels in the muscle specimen were markedly decreased in the patient, whereas CK and aspartate aminotransferase were almost the same as in a control subject. LDH isoenzymes of erythrocytes were analyzed in 5 siblings and in the parents. This demonstrated a complete lack of LDH M-subunit in 3 siblings. The ratio between H-subunit and M-subunit (H/M) in erythrocyte LDH suggested a partial absence of the M-subunit in two siblings and in the parents. An abortive increase of blood lactate and a marked increase in blood pyruvate were observed immediately after ischemic work of the forearm, accompanied by an increase in serum creatine kinase and myoglobinuria. The present case represents a newly described form of genetically determined myopathy.

38. Cyclic amp in gastric juice does not reflect histamine H2receptor activity in heidenhain pouch dog

Abstract

It is strongly believed that cAMP mediates histamine H2receptor activity, but does not mediate gastrin and acetylcholine stimulation of gastric acid secretion. Therefore, cAMP production could be a marker of H2receptor activity. Whether endogenous histamine mediates gastrin and/or acetylcholine stimulation, at least partialy, remains to be elucidated. If cAMP in the gastric juice reflects H2receptor activity, we can investigate whether endogenous histamine mediates gastrin and/or acetylcholine stimulation in vivo. In this study, we investigated whether cAMP in the gastric juice reflected histamine H2receptor activity in the Heidenhain pouch dog in vivo using different kinds of inhibitors of gastric secretion. Our hypothesis was as follows: Upon betazole stimulation, cimetidine, an H2receptor antagonist, should decrease cAMP output into the gastric juice, but omeprazole, an H+, K+-ATPase blocker, should not, because it blocks at a site more peripheral than the H2receptor and the production of cAMP. Sixty minutes after betazole administration, 4.0 µmol/kg of cimetidine and 0.18 µmol/kg omeprazole were administered intravenously and they inhibited gastric juice volume to a similar degree, that is, 49.6% and 52.1%, respectively. However, omeprazole caused a greater decrease in cAMP output than cimetidine. Inhibition with 4 /µmol/kg/h of cimetidine or 0.2 µmol/kg of omeprazole from the beginning of betazole stimulation also caused similar decreases in gastric juice volume, 66.6% and 60.6%, respectively. Both inhibitors decreased cAMP output into the gastric juice in a similar fashion in the first two 30 minute periods. These results do not agree with our hypothesis. It seems that cAMP in the gastric juice does not reflect histamine H2receptor activity following betazole stimulation, at least as a major component.

39. The role of carbonic anhydrase in gastric mucosal protection with special reference to H+back diffusion and concomitant metabolic acidosis induced by acetazolamide

It is suggested that carbonic anhydrase is implicated not only in gastric acid secretion, but in mucosal protection. It was reported that acetazolamide induced gastric mucosal lesions. But acetazolamide also caused concomitant metabolic acidosis by inhibiting H+secretion from renal tubules.

Abstract

Analog chromatography with glomerular basement membrane isolated from normal and nephrotic rabbits was performed to see if there was a molecular sieving effect by the basement membrane. In a normal glomerular basement membrane column, 125I-labeled human serum globulin and 131I-labeled human serum albumin were eluted at the same position, probably at the void volume of the column. However, 125I-labeled insulin and tritiated water appeared with a delay and gave separated peaks. In nephrotic glomerular basement membrane, 125I-labeled human globulin, 131I-labeled serum albumin, 125I-labeled insulin and tritiated water gave four distinctly separated peaks in that order. By analogy with Sephadex, the sieving effect of glomerular basement membrane was demonstrated.

Abstract

The temperature compensation for the mercury-inrubber strain gauge by a copper wire was examined. It was found that the influence of a temperature change on the resistance of the mercury-in-rubber strain gauge can be adequately compensated for by a fine copper wire. A transient imbalance of the gauge circuit cannot be avoided after he exposure to abrupt temperature change. With he copper compensation wire described, the effect of a slow, irregular temperature change on the resistance of the gauge can be eliminated. The addition of the rubber tube as insulation markedly improves the timecourse compensation with abrupt temperature changes. (Author)

The role of precooling of peripheral arterial blood in temperature regulation has been studied in cold-adapted and warm-adapted rabbits. It was found that precooling of arterial blood during its flow from the interior to the surface (ear) of the body in a cold environment is more marked in the cold-adapted rabbits.
