Abstract

We describe the in-orbit performance of the soft X-ray imaging system consisting of the Soft X-ray Telescope and the Soft X-ray Imager aboard Hitomi. Verification and calibration of imaging and spectroscopic performance are carried out making the best use of the limited data of less than three weeks. Basic performance including a large field of view of 38'x38' is verified with the first light image of the Perseus cluster of galaxies. Amongst the small number of observed targets, the on-minus-off pulse image for the out-of-time events of the Crab pulsar enables us to measure a half power diameter of the telescope as about 1.3'. The average energy resolution measured with the onboard calibration source events at 5.89 keV is 179 pm 3 eV in full width at half maximum. Light leak and cross talk issues affected the effective exposure time and the effective area, respectively, because all the observations were performed before optimizing an observation schedule and parameters for the dark level calculation. Screening the data affected by these two issues, we measure the background level to be 5.6x10^{-6} counts s^{-1} arcmin^{-2} cm^{-2} in the energy band of 5-12 keV, which is seven times lower than that of the Suzaku XIS-BI., Comment: Accepted for publication in PASJ. 17 pages, 18 figures

Abstract

Since the pioneering experiment, E89-009 studying hypernuclear spectroscopy using the $(e,e^{\prime}K^+)$ reaction was completed, two additional experiments, E01-011 and E05-115, were performed at Jefferson Lab. These later experiments used a modified experimental design, the "tilt method", to dramatically suppress the large electromagnetic background, and allowed for a substantial increase in luminosity. Additionally, a new kaon spectrometer, HKS (E01-011), a new electron spectrometer, HES, and a new splitting magnet were added to produce precision, high-resolution hypernuclear spectroscopy. These two experiments, E01-011 and E05-115, resulted in two new data sets, producing sub-MeV energy resolution in the spectra of ${}^{7}_{\Lambda}\text{He}$, ${}^{12}_{\Lambda}\text{B}$ and ${}^{28}_{\Lambda}\text{Al}$ and ${}^{7}_{\Lambda}\text{He}$, ${}^{10}_{\Lambda}\text{Be}$, ${}^{12}_{\Lambda}\text{B}$ and ${}^{52}_{\Lambda}\text{V}$. All three experiments obtained a ${}^{12}_{\Lambda}\text{B}$, spectrum, which is the most characteristic $p$-shell hypernucleus and is commonly used for calibration. Independent analyses of these different experiments demonstrate excellent consistency and provide the clearest level structure to date of this hypernucleus as produced by the $(e,e^{\prime}K^+)$ reaction. This paper presents details of these experiments, and the extraction and analysis of the observed ${}^{12}_{\Lambda}\text{B}$ spectrum.

Abstract

We consider a hybrid Monte Carlo algorithm which is applicable to lattice theories defined on Lefschetz thimbles. In the algorithm, any point (field configuration) on a thimble is parametrized uniquely by the flow-direction and the flow-time defined at a certain asymptotic region close to the critical point, and it is generated by solving the gradient flow equation downward. The associated complete set of tangent vectors is also generated in the same manner. Molecular dynamics is then formulated as a constrained dynamical system, where the equations of motion with Lagrange multipliers are solved by the second-order constraint-preserving symmetric integrator. The algorithm is tested in the lambda phi^4 model at finite density, by choosing the thimbles associated with the classical vacua for subcritical and supercritical values of chemical potential. For the lattice size L=4, we find that the residual sign factors average to not less than 0.99 and are safely included by reweighting and that the results of the number density are consistent with those obtained by the complex Langevin simulations., Comment: 25 pages, 10 figures, references updated, comments added, typos corrected

Abstract

An experiment with a newly developed high-resolution kaon spectrometer (HKS) and a scattered electron spectrometer with a novel configuration was performed in Hall C at Jefferson Lab (JLab). The ground state of a neutron-rich hypernucleus, He 7 Lambda, was observed for the first time with the (e,e'K+) reaction with an energy resolution of ~0.6 MeV. This resolution is the best reported to date for hypernuclear reaction spectroscopy. The he 7 Lambda binding energy supplies the last missing information of the A=7, T=1 hypernuclear iso-triplet, providing a new input for the charge symmetry breaking (CSB) effect of \Lambda N potential., Comment: 5 pages, 4 figures, submitted to PRL

Abstract

Red tides of Chattonella spp. have caused continuous damage to Japanese aquaculture, however, the life cycle of this organism remains incompletely understood. To further investigate this matter, we assessed genotypes at 14 microsatellite markers in three varieties of Chattonella marina, viz., C. marina var. antiqua, C. marina var. marina, and C. marina var. ovata, to establish whether Chattonella undergoes asexual diploidization or sexual reproduction. After genotyping 287 strains of C. marina, all but one of these strains was shown to be heterozygous for at least some loci, and thus, in the diploid state, suggesting that Chattonella strains undergo sexual reproduction. In addition, we performed single-cell amplification on 'small cells' that are derived from vegetative cells under dark and low-nutrient conditions. The results indicated the existence of two types of small cells. The ' Small cell Type 1' was found to be heterozygous, genotypically equivalent to the vegetative cells, and is therefore diploid. These small cells may change to resting cells (cysts) directly. The ' Small cell Type 2' was homozygous at all analyzed loci, suggesting that these small cells are haploid and may be derived by meiosis. As fusion between small cells has previously been observed, the ' Small cell Type 2' may be the gamete of Chattonella. Abstract

BACKGROUND: Approved on-demand treatments for hereditary angioedema attacks need to be administered parenterally, a route of administration that is associated with delays in treatment Or withholding of therapy. METHODS: In this phase 3, double-blind, three-way crossover trial, we randomly assigned par- ticipants at least 12 years of age with type 1 or type 2 hereditary angioedema to take up to two oral doses of sebetralstat (300 mg or 600 mg) or placebo for an angioedema attack. The primary end point, assessed in a time-to-event analysis, was the beginning of symptom relief, defined as a rating of "a little better" on the Patient Global Impression of Change scale (ratings range from "much worse" to much better") at two or more consecutive time points within 12 hours after the first administration of the trial agent. Key secondary end points, assessed in time-to-event analysis, were a reduction in attack severity (an improved rating on the Patient Global Impression of Severity [PGI-S] scale, with ratings ranging from none" to "very severe") at two or more consecutive time points within 12 hours and complete attack resolution (a rating of "none" on the PGI-S scale) within 24 hours. RESULTS: A total of 136 participants were assigned to one of six trial sequences, with 110 treating 264 attacks. The time to the beginning of symptom relief with the 300-mg dose and the 600-mg dose was faster than with placebo (P<0.001 and P=0.001 for the two comparisons, respectively), with median times of 1.61 hours (interquartile range, 0.78 to 7.04), 1.79 hours (1.02 to 3.79), and 6.72 hours (1.34 to >12), respectively. The time to reduction in the attack severity with the 300-mg dose and the 600-mg dose was faster than with placebo (P=0.004 and P=0.003), with median times of9.27 hours (interquartile range, 1.53 to >12), Z75 hours (2.19 to >12), and more than 12 hours (6.23 to >12). The time to complete resolution was faster with the 300-mg and 600-mg doses than with placebo (P=0.002 and P<0.001). The percentage of attacks with complete resolution within 24 hours was 42.5°/o with the 300-mg dose, 49.5% with the 600-mg dose, and 274% with placebo. Sebetralstat and placebo had similar safety profiles; no serious adverse events related to the trial agents were reported. CONCLUSIONS: Oral sebetralstat provided faster times to the beginning of symptom relief, reduction in attack severity, and complete attack resolution than placebo. [ABSTRACT FROM AUTHOR]

Abstract

A marine microbe (strain SR21) from the coral reef area of the Yap Islands was isolated by a screening test for polyunsaturated fatty acids and was found to accumulate lipid that contained 22:5n-6 docosapentaenoic acid (DPA) as well as 22:6n-3 docosahexaenoic acid (DHA). Strain SR21 was identified as genus Schizochytrium in Labyrinthulomycota, owing to its ultrastructural character and life cycle, which is composed of vegetative cell, zoosporangium, and zoospore stages. After cultural optimization, both in flask and fermenter, the highest DHA and DPA productivities of 2.0 and 0.44 g/L per day, respectively, were obtained in a medium of 60 g/L glucose and corn steep liquor/ammonium sulfate in a half salt concentration of seawater in fermenter culture at 28°C and pH 4. This productivity was almost twice that obtained with flask culture, indicating its high resistance to mechanical stirring. The lipid extracted from the cell was about 50% of the dry cell weight and was composed of 93% triacylglycerol (TG). DHA content of the lipid was 34% of total fatty acids. The TG profile was simple, and the content of the most dominant TG, 1,3-dipalmitoyl-2-DHA-TG, was 27%. TG that contained DHA and n-6-DPA amounted to 57 and 17%, respectively, of total TG molecules. Abstract

A novel series of substituted N-benzyl-N-phenyl-trifluoro-3-amino-2-propanols are described that reversibly inhibit cholesteryl ester transfer protein (CETP). Starting with screening lead 22, various structural features were explored with respect to inhibition of the CETP-mediated transfer of [3H]cholesterol from high-density cholesterol donor particles to low-density cholesterol acceptor particles. The free hydroxyl group of the propanol was required for high potency, since acylation or alkylation reduced activity. High inhibitory potency was also associated with 3-ether moieties in the aniline ring, and the highest potencies were exhibited by 3-phenoxyaniline analogues. Activity was substantially reduced by oxidation or substitution in the methylene of the benzylic group, implying that the benzyl ring orientation was important for activity. In the benzylic group, substitution at the 3-position was preferred over either the 2- or the 4-positions. Highest potencies were observed with inhibitors in which the 3-benzylic substituent had the potential to adopt an out of plane orientation with respect to the phenyl ring. The best 3-benzylic substituents were OCF2CF2H (42, IC50 0.14 μM in buffer, 5.6 μM in human serum), cyclopentyl (39), 3-iso-propoxy (27), SCF3 (67), and C(CF3)2OH (36). Separation of 42 into its enantiomers unexpectedly showed that the minor R(+) enantiomer 1a was 40-fold more potent (IC50 0.02 μM in buffer, 0.6 μM in human serum) than the major S(−) enantiomer 1b, demonstrating that the R-chirality at the propanol 2-position is key to high potency in this series. The R(+) enantiomer 1a represents the first reported acyclic CETP inhibitor with submicromolar potency in plasma. A chiral synthesis of 1a is reported.
