3. Transition to tolling third-party gas at Karratha Gas Plant: an attractive investment which unlocks infrastructure utilisation

Abstract

In December 2020, the North West Shelf (NWS) Project participants executed fully-termed third-party Gas Processing Agreements with Woodside Burrup Pty Ltd (Woodside Burrup), for gas from the Pluto fields, and with subsidiaries of Mitsui & Co Ltd and Beach Energy Limited (Mitsui and Beach), for the Waitsia Gas Project Stage 2. These agreements underpinned final investment decisions in the infrastructure projects, business processes, and digital tools, necessary to process third-party gas from the Pluto-Karratha Gas Plant (KGP) Interconnector, and the Burrup Extension Pipeline (BEP). Tolling operations commenced at the NWS Project in March 2022 with the processing of gas delivered from Pluto through the Interconnector. The first Liquefied Natural Gas (LNG) cargo produced from gas through the Interconnector was loaded in April 2022. KGP is also expected to commence processing Waitsia gas via the BEP from 2023. Woodside CEO Meg O’Neill stated: ‘The start-up of the Pluto-KGP Interconnector provides access to spare capacity at Karratha Gas Plant to process gas owned by other resource owners, both onshore and offshore Western Australia. The commencement of Pluto gas flowing through the Interconnector is the first example of this… The commercial agreements underpinning third-party gas processing at the North West Shelf reflect the commitment of Woodside and the North West Shelf Project to maximising value from the significant infrastructure on the Burrup Peninsula’. The development of the necessary infrastructure, business/operational processes, and digital tools is a forward looking investment which unlocks an efficient market in third-party gas processing market at KGP.

Topoisomerase 2 (Top2) is known to resolve DNA topological stress through double strand breaks (DSBs), yet Top2 inhibition has been reported to result in a significant amount of single-strand breaks (SSBs). Here the authors develop CC-seq—a method that allows direct mapping of both Top2-linked SSBs and DSBs—and reveal a significant impact of primary DNA sequence on Top2 directed cleavage.

Abstract

Tinnitus is often triggered by cochlear damage and has been linked with aberrant patterns of neuronal activity. Acoustic Coordinated Reset (CR®) Neuromodulation is a sound therapy hypothesised to reduce tinnitus symptoms by desynchronising pathological brain activity using a portable acoustic device (the T30 neurostimulator). We report results of a pivotal trial to test the efficacy of this intervention. This two-centre, double-blind randomised controlled trial with long-term open-label extension, was undertaken between February 2012 and February 2014 in the UK. Participants were 100 adults with tinnitus as a primary complaint, recruited through hearing clinics and media advertisements. Intervention was the device programmed either with the proprietary sound sequence or placebo algorithm, fit by one of five trained audiologists. Minimisation software provided group allocation (1:1 randomisation), with groups matched for age, gender, hearing loss and tinnitus severity. Allocation was masked from participants and assessors during the trial. The primary measure of efficacy was change in tinnitus symptom severity between groups, measured using the Tinnitus Handicap Questionnaire at 12 weeks. Secondary outcomes were other measures of tinnitus symptom severity, health-related quality of life, and perceptual characteristics (pitch, loudness, bandwidth) at 12 weeks, and Tinnitus Handicap Questionnaire at 36 weeks (open-label extension). A statistician blinded to the allocation conducted an intention-to-treat analysis that employed linear regressions on minimisation variables, trial centre and intervention group, with multiple imputations for missing data. The study was registered on clinicaltrials.gov (NCT01541969). We screened 391 individuals and assigned interventions to 100 eligible participants. The primary outcome was not statistically significant between groups (mean group = −0.45, 95% CI −5.25 to 4.35; p = 0.85), nor were any of the secondary outcomes. Four adverse events occurred during the trial. Analysis of tinnitus symptom severity data collected across the 24-week open-label extension showed no statistically significant within-group changes after 12, 24, or 36 weeks treatment with the proprietary sound sequence. While individual participants may benefit from sound therapy, Acoustic CR® Neuromodulation did not lead to group-mean reductions on tinnitus symptom severity or other measures compared to placebo, or over time.

Abstract

Background Laboratory diagnosis of Lyme disease (LD) relies on a two-tier protocol. We have observed disproportionate equivocal serologies in children requiring reflex western blot (WB) using manufacturer-provided ranges based on adult studies. We aimed to determine appropriate ranges for our pediatric population. Methods We performed a one-year retrospective institutional review of all 2755 children with LD testing with the Vidas® Lyme IgM II/IgG II immunoassays with reflex to WB for equivocal/positive serologies. Results were assessed by frequency distributions, optimization via percent agreement analysis, and clinical adjudication. Results The proposed ranges for IgM (negative ≤0.20, equivocal ≥0.21 to 0.50) allowed for a decrease in the IgM equivocal rate (7% to 2%) and IgG positive rate (15% to 13%). There was a decrease in the positive percent agreement between tiers (95% to 83% and 98% to 95%) with increase in the negative (32% to 63% and 70% to 81%) and overall (65% to 73% and 85% to 88%) percent agreements for IgM and IgG, respectively. Of 15 IgM serologies reclassified as negative with a positive WB and not positive for IgG, 8 were clinically negative, 5 were clinically positive, and two had insufficient history. Of the 10 IgG serologies reclassified as negative with a positive WB 3 were clinically positive, 6 were clinically negative and one had insufficient history. Conclusions Our modified ranges are more suitable for our pediatric population while reducing overdiagnosis, unnecessary treatment, diagnostic uncertainty, and turnaround time.

Abstract

The distribution of moraines in the Transantarctic Mountains affords direct constraint of past ice-marginal positions of the East Antarctic Ice Sheet (EAIS). Here, we describe glacial geologic observations and cosmogenic-nuclide exposure ages from Roberts Massif, an ice-free area in the central Transantarctic Mountains. We measured cosmogenic 3He, 10Be, 21Ne, and 26Al in 168 dolerite and sandstone boulders collected from 24 distinct deposits. Our data show that a cold-based EAIS was present, in a configuration similar to today, for many periods over the last ∼14.5 Myr, including the mid-Miocene, late Pliocene, and early to Middle Pleistocene. Moraine ages at Roberts Massif increase with distance from, and elevation above, the modern ice margin, which is consistent with a persistent EAIS extent during glacial maxima and slow, isostatic uplift of the massif itself in response to trough incision by outlet glaciers. We also employ the exceptionally high cosmogenic-nuclide concentrations in several boulders, along with multi-isotope measurements in sandstone boulders, to infer extremely low erosion rates (≪5 cm Myr−1) over the period covered by our record. Although our data are not a direct measure of ice volume, the Roberts Massif glacial record indicates that the EAIS was present and similar to its current configuration during at least some periods when the global temperature was believed to be warmer and/or atmospheric CO2 concentrations were likely higher than today.

Abstract

RATIONALE AND OBJECTIVE: The safety of intensive blood pressure (BP) targets is controversial for persons with chronic kidney disease (CKD). We studied the effects of hypertension treatment on cerebral perfusion and structure in those with and without CKD. STUDY DESIGN: Neuroimaging substudy of a randomized trial. SETTING & PARTICIPANTS: A subset of participants in the Systolic Blood Pressure Intervention Trial who underwent brain MRI studies. Presence of baseline CKD was assessed by estimated glomerular filtration rate (eGFR) and urine albumin to creatinine ratio (UACR). INTERVENTION: Participants were randomly assigned to intensive (systolic BP 30 mg/g (N=151), the effects of intensive versus standard BP treatment on change in global cerebral blood flow, WMLs and total brain volume were 1.91 ml/100g/min (95% CI −3.01, 6.82), 0.003 cm(3) (asinh transformed, 95% CI −0.13, 0.13), and −7.0 cm(3) (95% CI −13.3, −0.3), respectively. The overall treatment effects on cerebral blood flow and total brain volume were not modified by baseline eGFR or UACR; however the effect on WMLs was attenuated in participants with albuminuria (interaction p-value 0.04). LIMITATIONS: Measurement variability due to multi-site design. CONCLUSIONS: Among hypertensive adults with primarily early kidney disease, intensive versus standard blood pressure treatment did not appear to have a detrimental effect on brain perfusion or structure. The findings support the safety of intensive blood pressure treatment targets on brain health in persons with early kidney disease. Abstract

DNA topoisomerases are required to resolve DNA topological stress. Despite this essential role, abortive topoisomerase activity generates aberrant protein-linked DNA breaks, jeopardising genome stability. Here, to understand the genomic distribution and mechanisms underpinning topoisomerase-induced DNA breaks, we map Top2 DNA cleavage with strand-specific nucleotide resolution across the S. cerevisiae and human genomes—and use the meiotic Spo11 protein to validate the broad applicability of this method to explore the role of diverse topoisomerase family members. Our data characterises Mre11-dependent repair in yeast and defines two strikingly different fractions of Top2 activity in humans: tightly localised CTCF-proximal, and broadly distributed transcription-proximal, the latter correlated with gene length and expression. Moreover, single nucleotide accuracy reveals the influence primary DNA sequence has upon Top2 cleavage—distinguishing sites likely to form canonical DNA double-strand breaks (DSBs) from those predisposed to form strand-biased DNA single-strand breaks (SSBs) induced by etoposide (VP16) in vivo., Topoisomerase 2 (Top2) is known to resolve DNA topological stress through double strand breaks (DSBs), yet Top2 inhibition has been reported to result in a significant amount of single-strand breaks (SSBs). Here the authors develop CC-seq—a method that allows direct mapping of both Top2-linked SSBs and DSBs—and reveal a significant impact of primary DNA sequence on Top2 directed cleavage.

Abstract

The distribution of moraines in the Transantarctic Mountains affords direct constraint of past ice-marginal positions of the East Antarctic Ice Sheet (EAIS). Here, we describe glacial-geologic observations and cosmogenic-nuclide exposure ages from Roberts Massif, an ice-free area in the central Transantarctic Mountains. We measured cosmogenic 3He, 10Be, 21Ne, and 26Al in 180 dolerite and sandstone boulders collected from 24 distinct deposits. Our data show that a cold-based EAIS was present, in a configuration similar to today, for many periods over the last ~ 14.5 Myr, including the mid-Miocene, Late Pliocene, and early-to-mid Pleistocene. Moraine ages at Roberts Massif increase with distance from, and elevation above the modern ice margin, which is consistent with a persistent EAIS extent during glacial maxima, and slow, isostatic uplift of the massif itself in response to trough incision by outlet glaciers.
