Abstract

Global biodiversity gradients are generally expected to reflect greater species replacement closer to the equator. However, empirical validation of global biodiversity gradients largely relies on vertebrates, plants, and other less diverse taxa. Here we assess the temporal and spatial dynamics of global arthropod biodiversity dynamics using a beta-diversity framework. Sampling includes 129 sampling sites whereby malaise traps are deployed to monitor temporal changes in arthropod communities. Overall, we encountered more than 150,000 unique barcode index numbers (BINs) (i.e. species proxies). We assess between site differences in community diversity using beta-diversity and the partitioned components of species replacement and richness difference. Global total beta-diversity (dissimilarity) increases with decreasing latitude, greater spatial distance and greater temporal distance. Species replacement and richness difference patterns vary across biogeographic regions. Our findings support long-standing, general expectations of global biodiversity patterns. However, we also show that the underlying processes driving patterns may be regionally linked.

Abstract

Assessing genetic differentiation among natural populations can aid understanding of dispersal patterns and connectivity among habitats. Several molecular markers have become increasingly popular in determining population genetic structure for this purpose. Here, we compared the resolution of mitochondrial cytochrome c oxidase subunit I (COI) and nuclear single nucleotide polymorphism (SNP) markers for detecting population structure among stream insects at small spatial scales. Individuals of three endemic taxa--Coloburiscus humeralis (Ephemeroptera), Zelandobius confusus (Plecoptera), and Hydropsyche fimbriata (Trichoptera)--were collected from forested streams that flow across open pasture in the North Island of New Zealand. Both COI and SNP data indicated limited population structure across the study area, and small differences observed among these species were likely related to their putative dispersal abilities. For example, fine-scale genetic differentiation between and among neighbouring stream populations for H. fimbriata suggests that gene flow, and hence dispersal, may be more limited for this species relative to the others. Based on the generally similar results provided by both types of markers, we suggest that either COI or SNP markers can provide suitable initial estimates of fine-scale population genetic differentiation in stream insects. Dans ce travail, les auteurs ont compare la resolution de la structure des populations, parmi des insectes de ruisseaux a une echelle spatiale reduite, obtenue a l'aide de la sous-unite I de la cytochrome c oxydase (COI) mitochondriale et de marqueurs mononucleotidiques (SNP) nucleaires. Abstract

Novel methods for sampling and characterizing biodiversity hold great promise for re-evaluating patterns of life across the planet. The sampling of airborne spores with a cyclone sampler, and the sequencing of their DNA, have been suggested as an efficient and well-calibrated tool for surveying fungal diversity across various environments. Here we present data originating from the Global Spore Sampling Project, comprising 2,768 samples collected during two years at 47 outdoor locations across the world. Each sample represents fungal DNA extracted from 24 m 3 of air. We applied a conservative bioinformatics pipeline that filtered out sequences that did not show strong evidence of representing a fungal species. The pipeline yielded 27,954 species-level operational taxonomic units (OTUs). Each OTU is accompanied by a probabilistic taxonomic classification, validated through comparison with expert evaluations. To examine the potential of the data for ecological analyses, we partitioned the variation in species distributions into spatial and seasonal components, showing a strong effect of the annual mean temperature on community composition.

Abstract

DNA barcoding was intended as a means to provide species-level identifications through associating DNA sequences from unknown specimens to those from curated reference specimens. Although barcodes were not designed for phylogenetics, they can be beneficial to the completion of the Tree of Life. The barcode database for Trichoptera is relatively comprehensive, with data from every family, approximately two-thirds of the genera, and one-third of the described species. Most Trichoptera, as with most of life's species, have never been subjected to any formal phylogenetic analysis. Here, we present a phylogeny with over 16 000 unique haplotypes as a working hypothesis that can be updated as our estimates improve. We suggest a strategy of implementing constrained tree searches, which allow larger datasets to dictate the backbone phylogeny, while the barcode data fill out the tips of the tree. We also discuss how this phylogeny could be used to focus taxonomic attention on ambiguous species boundaries and hidden biodiversity. We suggest that systematists continue to differentiate between 'Barcode Index Numbers' (BINs) and 'species' that have been formally described. Each has utility, but they are not synonyms. We highlight examples of integrative taxonomy, using both barcodes and morphology for species description.This article is part of the themed issue 'From DNA barcodes to biomes'.

Abstract

Polar Collembola have adapted over millions of years to an environment that is changing faster than any other on earth. Globally, Collembola (springtails) are among the most abundant and widely-distributed arthropods and are key components of species-poor polar ecosystems. Understanding the resilience of polar collembola to climate change is therefore an urgent research priority. Here we explore the links between genetic diversity and physiology in shaping the resilience of polar Collembola to climate change. I have reviewed the resilience of polar Collembola considering genetic diversity, behavioural avoidance and physiological tolerances along with an examination of the potential impacts of biotic interactions. I also explored potential recovery dynamics with reference to temperate taxa and colonisation patterns of new habitat exposed by glacial retreat. This review illustrated that polar Collembola exhibit a suite of traits that have enabled their survival in extreme conditions and may serve as pre-adaptations to changing conditions. However, if resistance capacities are insufficient, complete community level recovery following disturbances is exceedingly slow, especially among Collembola that inhabit deeper microhabitats within the soil column (deeper-dwelling). Overall, it appears that deeper-dwelling species that fail to resist climate changes may not recover in ecologically realistic timescales, especially given the projected pace of climate changes. The largest spatial scale study and analysis of the genetic diversity of Collembola from the central Canadian High Arctic location of Cambridge Bay (Ikaluktuktiak) was undertaken to refine species identifications, examine levels of population diversity, and explore the role of geological processes and glacial dynamics in shaping the current Arctic collembolan fauna. 23. Biogeography and genetic diversity of terrestrial mites in the Ross Sea region, Antarctica

Abstract

Free-living terrestrial mites (Acari) have persisted through numerous glacial cycles in Antarctica. Very little is known, however, of their genetic diversity and distribution, particularly within the Ross Sea region. To redress this gap, we sampled mites throughout the Ross Sea region, East Antarctica, including Victoria Land and the Queen Maud Mountains (QMM), covering a latitudinal range of 72–85 °S, as well as Lauft Island near Mt. Siple (73 °S) in West Antarctica and Macquarie Island (54oS) in the sub-Antarctic. We assessed genetic diversity using mitochondrial cytochrome c oxidase subunit I gene sequences (COI-5P DNA barcode region), and also morphologically identified voucher specimens. We obtained 130 sequences representing four genera: Nanorchestes (n = 30 sequences), Stereotydeus (n = 46), Coccorhagidia (n = 18) and Eupodes (n = 36). Tree-based analyses (maximum likelihood) revealed 13 genetic clusters, representing as many as 23 putative species indicated by barcode index numbers (BINs) from the Barcode of Life Datasystems (BOLD) database. We found evidence for geographically-isolated cryptic species, e.g., within Stereotydeus belli and S. punctatus, as well as unique genetic groups occurring in sympatry (e.g., Nanorchestes spp. in QMM). Collectively, these data confirm high genetic divergence as a consequence of geographic isolation over evolutionary timescales. From a conservation perspective, additional targeted sampling of understudied areas in the Ross Sea region should be prioritised, as further diversity is likely to be found in these short-range endemic mites.

Abstract

Increasing anthropogenic habitat fragmentation in riverine ecosystems, in particular the loss of natural riparian vegetation, has profound consequences for dispersal and connectivity of aquatic insects. Aquatic insects depend on dispersal to colonise new sites and expand their ranges, but colonisation is only effective if followed by successful reproduction of the colonising individual within the receiving population. Populations that are connected by dispersal are likely to possess and maintain higher levels of genetic diversity through gene flow, which enhances their long-term viability. Conversely, altered and/or restricted movement of individuals in fragmented landscapes may disrupt the connectivity of populations across habitats, reducing gene flow and decreasing genetic diversity of remnant populations. Thus, the ability to successfully disperse through fragmented and disturbed patches can be a key determinant of the long-term viability of populations. Genetic markers provide an indirect approach to estimate dispersal potential and infer a species’ gene flow, genetic diversity, and population connectivity. A variety of different genetic markers have been used over the past decades, each with advantages and disadvantages. Mitochondrial DNA (mtDNA) is commonly used in aquatic insect research and provides insight into historic dispersal, connectivity and isolation, while nuclear markers identified through next-generation sequencing approaches can reflect changes on a more contemporary time scale. The application of such markers in landscape genetics research further enables quantification of the effects of landscape and environmental features on gene flow, and identification of potential barriers to dispersal. This thesis is a collection of individual research papers that together provide new knowledge of functional connectivity and dispersal patterns of stream insect populations in altered landscapes. Nevertheless, Antarctic soils harbor unique bacterial and archaeal (prokaryotic) communities able to cope with extremely cold and dry conditions. These communities are not homogeneous, and the taxonomic composition and functional capabilities (genomic attributes) of these communities across environmental gradients remain largely undetermined. We analyzed the prokaryotic communities in soil samples collected from across the Shackleton Glacier region of Antarctica by coupling quantitative PCR, marker gene amplicon sequencing, and shotgun metagenomic sequencing. We found that elevation was the dominant factor explaining differences in the structures of the soil prokaryotic communities, with the drier and saltier soils found at higher elevations harboring less diverse communities and unique assemblages of cooccurring taxa. The higher-elevation soil communities also had lower maximum potential growth rates (as inferred from metagenome-based estimates of codon usage bias) and an overrepresentation of genes associated with trace gas metabolism. 47. Relationship between meteoric 10Be and NO3− concentrations in soils along Shackleton Glacier, Antarctica

Abstract

Outlet glaciers that flow through the Transantarctic Mountains (TAM) experienced changes in ice thickness greater than other coastal regions of Antarctica during glacial maxima. As a result, ice-free areas that are currently exposed may have been covered by ice at various points during the Cenozoic, complicating our understanding of ecological succession in TAM soils. Our knowledge of glacial extent on small spatial scales is limited for the TAM, and studies of soil exposure duration and disturbance, in particular, are rare. We collected surface soil samples and, in some places, depth profiles every 5 cm to refusal (up to 30 cm) from 11 ice-free areas along Shackleton Glacier, a major outlet glacier of the East Antarctic Ice Sheet. We explored the relationship between meteoric 10Be and NO3- in these soils as a tool for understanding landscape disturbance and wetting history and as exposure proxies. Concentrations of meteoric 10Be spanned more than an order of magnitude across the region (2.9×108 to 73×108 atoms g−1) and are among the highest measured in polar regions. The concentrations of NO3- were similarly variable and ranged from ∼1 µg g−1 to 15 mg g−1. In examining differences and similarities in the concentrations of 10Be and NO3- with depth, we suggest that much of the southern portion of the Shackleton Glacier region has likely developed under a hyper-arid climate regime with minimal disturbance. Finally, we inferred exposure time using 10Be concentrations. This analysis indicates that the soils we analyzed likely range from recent exposure (following the Last Glacial Maximum) to possibly >6 Myr. We suggest that further testing and interrogation of meteoric 10Be and NO3- concentrations and relationships in soils can provide important information regarding landscape development, soil evolution processes, and inferred exposure durations of surfaces in the TAM.
