A key challenge in the synthesis of diterpenoid alkaloids lies in identifying strategies that rapidly construct their multiply bridged polycyclic skeletons. Existing approaches to these structurally intricate secondary metabolites are discussed in the context of a "bond-network analysis" of molecular frameworks, which was originally devised by Corey some 40 years ago. The retrosynthesis plans that emerge from a topological analysis of the highly bridged frameworks of the diterpenoid alkaloids are discussed in the context of eight recent syntheses of hetidine and hetisine natural products and their derivatives. This Minireview highlights the extent to which network analyses of the type described here sufficed for designing synthesis plans, as well as areas where they had to be amalgamated with functional group oriented synthetic planning considerations., (© 2019 The Authors. Two carbenoids combine to generate an olefin by a mechanism involving formation of an ate complex, 1,2-metalate rearrangement, and β-elimination. As each stage of this eliminative coupling is stereospecific, the overall stereochemical outcome can be understood and, in principle fully controlled, providing that the absolute stereochemical configurations of the reacting carbenoid species are defined. In contrast to traditional alkene syntheses, the eliminative cross-coupling of carbenoids offers a connective approach to olefins capable of precisely targeting a given isomer regardless of the nature of the features distinguishing the isomers. The formation of olefins by the eliminative dimerization and eliminative cross-coupling of carbenoids is reviewed with a range of illustrative examples, including the reactions of α-lithiated haloalkanes, epoxides, and carbamates. This review covers free radical additions, which are initiated by the formal addition of a hydrogen atom to a C[double bond, length as m-dash]C double bond. These reactions originated in the realms of inorganic chemistry, polymer chemistry, and organic chemistry, whereby barriers between these disciplines impeded the rapid implementation of the findings.

For almost 200 years, the synthesis of natural products has been practiced. In this time span, not only the target structures have become increasingly more complex, the objectives of natural product synthesis have also changed. Likewise, the standards and criteria for the conduction of natural product synthesis have changed. In this tutorial review the economies of synthesis are analysed from both detailed and macroscopic perspectives, using case-studies from complex molecule synthesis. Atom, step, and redox economy are more than philosophical constructs, but rather guidelines, which enable the synthetic chemist to design and execute an efficient synthesis. Students entering the field of synthesis might find this tutorial helpful for understanding the subtle differences between these economic principles and also see real-world situations where such principles are put into practice.

In July 2005, 107 rainbow trout in age 1+ from a salmonid farm in Southern Germany situated in the southern tributary area of the Danube river were examined. The aim of this study was to determine the gyrodactylid species found on rainbow trout and to identify their location on the host's body. In total, 291 specimens from genus Gyrodactylus were collected. The most abundantly occurring species was Gyrodactylus truttae (181 specimens), whilst the others were less abundant. For the first time in Germany, Gyrodactylus teuchis and Gyrodactylus derjavinoides on rainbow trout were found. Most parasites occurred on the pectoral and ventral fins. Few specimens were found on the anal or caudal fins, in the oral cavity or on the gills. The only uninfected place was the nasal cavity.

"Economy" is referred to as the thrifty and efficient use of material resources, as the principle of "minimum effort to reach a goal." More illuminating is: "the aim to portion one's forces in order to use as little as possible of them to reach a goal." Such statements certainly apply when the goal is to synthesize a complex target molecule. Redox economy then implies the use of as few redox steps as possible in the synthetic conquest of a target compound. While any sort of economy will help to streamline the effort of total synthesis, redox economy addresses a particularly weak area in present-day total synthesis. It is not enough to point out the present deficiencies, rather the purpose of this Review is to serve as a teaching tool for all practitioners of the field by giving and illustrating guidelines to increase redox economy in multistep organic synthesis.

[structure: see text] An efficient total synthesis of the annonaceous acetogenin jimenezin was achieved. The key steps used were a highly stereoselective intramolecular allylboration to establish the tetrahydropyran ring and an intramolecular Williamson reaction to close the tetrahydrofuran ring.

A diagnostic procedure, based on a polymerase chain reaction method (PCR) was developed to detect infection of crayfish with the Oomycete Aphanomyces astaci. A set of oligonucleotide primers was designed to specifically amplify A. astaci DNA in the ITS region surrounding the 5.8S rDNA gene. The PCR amplifies a 115bp amplicon. The specificity of the primers was demonstrated by testing on 27 A. astaci strains and against 20 non-A. astaci Oomycetes and 5 fungal species. Most of the non-A. astaci Oomycete or fungal species included in the study are either known parasites of freshwater crayfish cuticle or can be found in their natural environment. Specificity was also tested against crayfish tissue and some known parasites and bacteria infecting crayfish. A protocol for the extraction of A. astaci DNA from infected crayfish tissue was developed. The optimised method allows the detection of two genome equivalents of purified A. astaci genomic DNA. The method was tested on noble crayfish (Astacus astacus), artificially infected with A. astaci. Detection of A. astaci was possible at the very first time of sampling, which was 2 days after the beginning of spore exposure.

The involvement of single electron transfer, i.e. of free radicals in the reactions of organomagnesium reagents could be detected with the aid of a chiral secondary Grignard reagent, in which the magnesium-bearing carbon atom is the sole stereogenic centre. So far, however, such reagents have not been accessible, because the standard preparation of Grignard reagents proceeds via free radicals. We review and summarize here our efforts to generate such a Grignard reagent 36 by asymmetric synthesis starting from an enantiomerically pure alpha-chloroalkyl-sulfoxide 30b using a sulfoxide/magnesium exchange reaction to give 33 followed by a carbenoid homologation reaction. Grignard reagent 36 turned out to be an ideal probe to learn about the extent to which SET is involved in reactions of organomagnesium reagents.

The addition of alpha-haloalkyl Grignard reagents to benzaldehyde occurs with simple diastereoselectivity substantially higher than that of the corresponding lithium reagents. Reaction in the presence of dimethyl-aluminum chloride suppresses subsequent Oppenauer oxidation of the resulting Mg-alkoxides by excess benzaldehyde.

Present knowledge on the capability of fish to feel pain and of suffering is presented. According to these data it has to be supposed, that fish are able to feel fish. Suffering and damage can be proven by biochemistry and morphology. Under these aspects, angling ponds and cesarean section in sturgeon have to be regarded as a violation of the German animal welfare act.

The Grignard reagent 2, in which the magnesium-bearing carbon atom is the sole stereogenic centre has been coupled with vinyl bromide under Pd(0) or Ni(0)-catalysis to give compound 3 with full retention of configuration. Coupling using Fe(acac)3 or Co(acac)2 as catalyst was accompanied by considerable racemisation. These findings are discussed with respect to a dichotomy between concerted polar and stepwise SET transmetallation pathways.

After more than a quarter of a century of development, the methodology of stereoselective synthesis appears to be fully matured. In line with this, the potential that meso compounds offer in stereoselective synthesis is clearly recognized. The use of meso compounds in synthesis is, however, in no way commensurate with this potential, because, ironically, the synthesis of meso compounds in the first place is a problem of stereoselective synthesis. Present-day methodology does not provide many useful solutions to this problem. This Review therefore addresses the strategies available for the synthesis of more elaborate meso compounds whose stereogenic centers have a distance >1,4 between them. meso Compounds with more than four stereogenic centers are also considered. The criteria used in choosing from several strategies in the synthesis of such compounds are discussed.

[reaction: see text] The conversion of the iodine ate-complex 5 to the Grignard reagent 6 follows a rate law first order in 5 and in magnesium ions. In addition to this normal pathway of the halogen-metal exchange process, the conversion of 5 to 6 is catalyzed by a number of diverse "electrophiles". This points to a SET-initiated radical chain process as a second pathway for the conversion of the iodine ate-complex 5 to the Grignard reagent 6.

A systematic inventory of actinosporeans and oligochaetes conducted over 3 years at a trout fish farm in Bavaria, Germany, allowed the identification of 12 actinosporeans from five collective groups: four Triactinomyxon (Triactinomyxon nov. types 1-4), two Raabeia (Raabeia nov. types 1, 2), two Echinactinomyxon ( E. radiatum, Echinactinomyxon nov. type 1), two Aurantiactinomyxon ( A. pavinsis, Aurantiactinomyxon nov. type 1) and two Neoactinomyxum (Neoactinomyxum nov. types 1, 2). Nine forms are novel but can be placed within existing collective groups. All 12 forms were detected in the laboratory in aquarium water associated with farm sediment. However, only four of these could be linked with an oligochaete host. Three families of oligochaetes were identified from the sediment: Tubificidae, represented by eight species, Lumbriculidae with one species and Naididae with two species. Only tubificid oligochaetes were found to host actinosporeans.

A set of three-armed urea-containing anion receptors was prepared. The receptors all have the same binding topology but differ in the level of conformational preorganization with respect to the arrangement of the side-arms relative to the platform and within the side arms themselves. This is mirrored in a specific increase (x 2.5) in the binding constant for chloride and in a 12-fold increase in the chloride/nitrate-selectivity.

Transmetalation of a chiral secondary Grignard reagent to Cu(I) or Mn(II) occurs with partial racemization in a manner that suggests the sequence of two transmetalation cycles, one with racemization (caused by ET ?) and the other one with (predominant) retention of configuration.

The anion binding of tripodal hosts 2-4 has been studied. Increasing levels of conformational preorganisation of the side arms of the hosts led to increased (Cl-) unaltered (Br-) or decreased (NO3-) binding; it was thus possible to change guest selectivities by about an order of magnitude through conformational preorganisation of the flexible host.

The development of actinosporeans in their oligochaete host proceeding pansporocyst formation is relatively well documented, however, phases preceding it are not as well known. The initial stages in the development of two actinosporeans, Raabeia type 1 of Oumouna et al. [Parasitol. Res. 2002] and Aurantiactinomyxon pavinsis (Ormières, 1968) Marquès [Languedoc, Universite des Sciences et Techniques, Dissertation, 1984] from schizogony to gametogony and sporogony are described. Both actinosporeans begin their development as multinucleate stages near the basal lamina of the oligochaete intestine. Proximal to these stages and between the host epithelium cells are uninucleate cells whose nuclei divide to produce binucleate cells. These divide mitotically to produce cells with four nuclei which then undergo plasmotomy to yield a tetracellular stage and the first phase in pansporocyst formation. From the uninucleate stage to the tetranucleate stage, the cell membrane of the parasite is associated closely via finger-like projections with the intestinal epithelial and glandular cells of the host., (2002 Elsevier Science (USA))

Efforts are described to design simple, fully flexible but conformationally preorganised omega-hydroxy-nonanoic acids that could serve as the conformation controlling unit in analogues of the potent protein-kinase C activator aplysiatoxin. Such analogues are macrodilactones incorporating the designed omega-hydroxy-nonanoic acid and 3,4-dihydroxy-pentanoic acid, which contains the pharmacophoric groups. The design process (replacement of CH(2) groups by an oxygen atom, annelation of a six-membered ring and placement of alkyl substituents) of the omega-hydroxy-nonanoic acids was monitored by force-field calculations. In the end of this process simple analogues of aplysiatoxin are proposed in which the proper disposition of the pharmacophoric groups is secured by a conformationally flexible but preorganised template structure as part of the macrodilactone ring., (Copyright 2002 The Japan Chemical Journal Forum and Wiley Periodicals, Inc.)

The question on the capability of fish to feel pain and of suffering are still subject of discussion nowadays. In the article presented, the information available in the literature to date is summarised. Based on this knowledge, the conclusion is drawn that fish are capable of feeling pain and that they are able to suffer in the sense of the word as used in the German animal welfare law.

In the last few years two factors have helped to significantly advance our understanding of the Myxozoa. First, the phenomenal increase in fin fish aquaculture in the 1990s has lead to the increased importance of these parasites; in turn this has lead to intensified research efforts, which have increased knowledge of the development, diagnosis. and pathogenesis of myxozoans. The hallmark discovery in the 1980s that the life cycle of Myxobolus cerebralis requires development of an actinosporean stage in the oligochaete. Tubifex tubifex, led to the elucidation of the life cycles of several other myxozoans. Also, the life cycle and taxonomy of the enigmatic PKX myxozoan has been resolved: it is the alternate stage of the unusual myxozoan, Tetracapsula bryosalmonae, from bryozoans. The 18S rDNA gene of many species has been sequenced, and here we add 22 new sequences to the data set. Phylogenetic analyses using all these sequences indicate that: 1) the Myxozoa are closely related to Cnidaria (also supported by morphological data); 2) marine taxa at the genus level branch separately from genera that usually infect freshwater fishes; 3) taxa cluster more by development and tissue location than by spore morphology; 4) the tetracapsulids branched off early in myxozoan evolution, perhaps reflected by their having bryozoan, rather than annelid hosts; 5) the morphology of actinosporeans offers little information for determining their myxosporean counterparts (assuming that they exist); and 6) the marine actinosporeans from Australia appear to form a clade within the platysporinid myxosporeans. Ribosomal DNA sequences have also enabled development of diagnostic tests for myxozoans. PCR and in situ hybridisation tests based on rDNA sequences have been developed for Myxobolus cerebralis, Ceratomyxa shasta, Kudoa spp., and Tetracapsula bryosalmonae (PKX). Lectin-based and antibody tests have also been developed for certain myxozoans, such as PKX and C. shasta. We also review important diseases caused by myxozoans, which are emerging or re-emerging. Epizootics of whirling disease in wild rainbow trout (Oncorhynchus mykiss) have recently been reported throughout the Rocky Mountain states of the USA. With a dramatic increase in aquaculture of fishes using marine netpens, several marine myxozoans have been recognized or elevated in status as pathological agents. Kudoa thyrsites infections have caused severe post-harvest myoliquefaction in pen-reared Atlantic salmon (Salmo salar), and Ceratomyxa spp., Sphaerospora spp., and Myxidium leei cause disease in pen-reared sea bass (Dicentrarchus labrax) and sea bream species (family Sparidae) in Mediterranean countries.

[see reaction]. The stereochemistry of electrophilic amination has been probed using the chiral Grignard reagent 5, in which the magnesium-bearing carbon atom is the sole stereogenic center. Amination with azidomethyl phenyl sulfide 1 and with O-sulfonyloxime 2 were found to proceed with full retention of configuration.

alpha-Chloro- and alpha-bromoalkyl Grignard reagents 11 and 30 with > 97% ee (enantiomeric excess) were generated by a sulfoxide/magnesium exchange reaction from the enantiomerically and diastereomerically pure sulfoxides 25 and 27. The resulting alpha-haloalkyl Grignard reagents are configurationally stable at -78 degrees C. Racemization sets in at or above -60 degrees C, especially when the solution contains bromide ions. In the absence of halide ions, the configurational stability extends up to -20 degrees C, when chemical decomposition commences.

At the brink of the 21st century, chemistry is increasingly concerned with the function that molecules fulfil as drugs, receptors, or-as ensemble of molecules-as materials. The capability of compounds to fulfil such functions cannot sufficiently be described by using only the terms composition and configuration. A decisive role is played in addition by the conformation of the molecules, which serves as the link between molecular composition and molecular function. Expressions such as "active conformation" or "competent conformation" allude to this aspect. Chemists have to develop an understanding how a flexible molecule adopts the conformation (a distinct shape) which is optimal for the function in question and how this process can be controlled. On the outset of such considerations, we may ask how nature succeeded in the process of evolution to endow flexible molecules with a preference to adopt the conformation which is optimal for the function it has to serve. In this review, I report on how we have reached a crude level of understanding of conformation design in nature with reference to the class of polyketide natural products, how we developed these insights into a conformation design of open-chain compounds, and which applications are already in sight.

[formula: see text] A series of C2- or sigma-symmetric oligo-1,3-dioxanylmethanes, 2-5, have been prepared using a bidirectional approach. In bidirectional syntheses of meso compounds, only substrate-based asymmetric induction could be applied. 1,3-Asymmetric induction in Mukaiyama-aldol additions, 1,5-asymmetric induction in enol-borinate aldol reactions, and 1,3-anti-selective reduction of aldols turned out to be reliable tools in the preparation of compounds 3-5.

The cytology of a new microsporean parasite Microsporidium epithelialis sp. n. from the intestinal epithelial cells of the freshwater oligochaete Tubifex sp. (Tubificidae) is described. The microsporean occurred together with an actinosporean of the genus Triactinomyxon, which was found between the epithelial cells. The merogonic and sporogonic stages (mature spores included) of the microsporean parasite are monokaryotic. An individual sporophorous vesicle surrounds each spore. The fixed and stained spore has an average dimension of 1.9-2.5 x 0.9-1.2 microm. The spores are oval with a characteristic surface layer, showing ornamentation-like projections, which are in close contact to the exospore. A short polar filament forming three to four coils traverses the polaroplast with two lamellar layers. The ultrastructure and other characteristic features of this microsporean parasite are distinct from those of the microsporean species described so far from oligochaetes.

Not only for ligand exchange at sulfoxides can the sulfoxide/magnesium exchange reaction be used, but it also provides a possibility to generate Grignard reagents in way that avoids metallic magnesium and thus radical processes.
