This paper uses two triple-hurdle models to analyse the determinants of Ethiopian rural households decisions in (1) milk production, butter marketing and volume of butter sales; and (2) milk production, purchased input use and intensity of purchased input use. Results are based on data collected from 5000 households and 497 rural communities in the highlands of Ethiopia. Availability of feed stands out as an important factor influencing household decision to engage in milk production, indicating the dire need to develop feed resources to promote dairy production in rural Ethiopia. Milk production in rural Ethiopia seems to have an interesting and complex gender dimension. While female-headed households are less likely to be engaged in milk production, perhaps because of resource limitations, they are more likely to manage their dairy farms intensively. Marketing costs matter in dairy production and marketing in rural Ethiopia, suggesting for the need to develop market infrastructures for both dairy outputs and inputs. Our results further show that participation in butter markets as sellers or the amount of butter sales do not respond to price signals, suggesting the need to understand the behavioural aspect of dairy marketing decision in rural Ethiopia. Cationic lipids are widely used for gene delivery, and inclusion of dioleoylphosphatidylethanolamine (DOPE) as a helper lipid in cationic lipid-DNA formulations often promotes transfection efficacy. To investigate the significance of DOPE's preference to adopt a hexagonal phase in the mechanism of transfection, the properties and transfection efficiencies of SAINT-2/DOPE lipoplexes were compared to those of lipoplexes containing lamellar-phase-forming dipalmitoylphosphatidylethanolamine (DPPE). After interaction with anionic vesicles, to simulate lipoplex-endosomal membrane interaction, SAINT-2/DOPE. lipoplexes show a perfect hexagonal phase, whereas SAINT-2/DPPE lipoplexes form a mixed lamellar-hexagonal phase. The transition to the hexagonal phase is crucial for dissociation of DNA or oligonucleotides (ODN) from the lipoplexes. However, while the efficiencies of nucleic acid release from either complex were similar, SAINT-2/DOPE lipoplexes displayed a two- to threefold higher transfection efficiency or nuclear ODN delivery. Interestingly, rupture of endosomes following a cellular incubation with ODN-containing SAINT-2/DPPE complexes dramatically improved nuclear ODN delivery to a level that was similar to that observed for SAINT-2/DOPE complexes. Our data demonstrate that although hexagonal phase formation in lipoplexes is a prerequisite for nucleic acid release from the complex, it appears highly critical for accomplishing efficient translocation of nucleic acids across the endosomal membrane into the cytosol for transport to the nucleus.

In the central nervous system, the extracellular matrix (ECM) compound laminin-2, present on developing axons, is essential in regulating oligodendrocyte (OLG) maturation. For example, laminin-2 is involved in mediating interactions between integrins and growth factors, initially localizing in separate membrane microdomains. The galactosphingolipid sulfatide is an important constituent of these microdomains and may serve as a receptor for laminin-2. Here, we investigated whether sulfatide interferes with ECM-integrin interactions and, in this manner, modulates OLG maturation. Our data reveal that disruption of laminin-2-sulfatide interactions impeded OLG differentiation and myelin-like membrane formation. On laminin-2, but not on (re)myelination-inhibiting fibronectin, sulfatide laterally associated with integrin alpha 6 in membrane microdomains. Sulfatide was partly excluded from membrane microdomains on fibronectin, thereby likely precluding laminin-2-mediated myelination. Anti-sulfatide antibodies disrupted integrin alpha 6-PDGF alpha R interactions on laminin-2 and induced demyelination in myelinated spheroid cultures, but intriguingly stimulated myelin-like membrane formation on fibronectin. Taken together, these findings highlight the importance of laminin-sulfatide interactions in the formation of functional membrane microdomains essential for myelination. Thus, laminin-sulfatide interactions might control the asynchronous localized differentiation of OLGs, thereby allowing myelination to be triggered by axonal demand. Given the accumulation of fibronectin in multiple sclerosis lesions, the findings also provide a molecular rationale for the potential of anti-sulfatide antibodies to trigger quiescent endogenous OLG progenitor cells in axon remyelination. Both legal and illegal livestock marketing systems are operating at different magnitudes in the Amhara Region’s Ethio-Sudan cross-border cattle trade. Small farmer exporters and traders are the major actors in the illegal cattle marketing system while medium- to largescale licensed exporters are dominantly operating in the legal system. Most cattle sales are related to farm households’ cash needs and commercial orientation. However, cattle sales are also induced due to fear of theft and insecurity. The paper analyses volume of trade, transaction costs, trade barriers or constraints and how to address them.

Recent advances in studies involving the structure and dynamics of membranes have shown that fluorescently-tagged lipid probes have become versatile and, occasionally, indispensable tools in this area of research. These probes are applied in investigations as diverse as those dealing with biophysical aspects of membranes, such as lateral mobility, lipid phase transitions and phase separations (domain formation), nonbilayer formation (hexagonal phases) and lipid translocation, but also in studies of the cell biology of membranes, including membrane flow and lipid trafficking. In this brief overview, some aspects of the properties and the application of fluorescent lipid (-like) probes will be summarized. The primary focus will be on the use of nitro-benzoxadiazole-derivatized lipids (NBD-lipids).

A detailed physicochemical study is presented on a new class of cationic amphiphiles, Sunfish amphiphiles, recently designed, synthesized, and tested for gene delivery. These materials have two hydrophobic tails, connected to the cationic pyridinium headgroup at the 1- and 4-positions. Two extreme morphologies can be visualized, i.e. one by back-folding involving association of both tails at one side of the pyridinium ring and one by independent unfolding of the tails, the two molecular geometries leading to considerable differences in the aggregate morphology. The behavior of six members of the Sunfish family in mixtures with DOPE, applying different conditions relevant for transfection, has been studied by a combination of techniques (DLS, DSC, NMR, SAXS, Cryo-TEM, fluorescence, etc.). The effects of structural parameters such as the presence of unsaturation in the tails and length of the alkyl chains on the properties of the aggregates have been assessed. A correlation of these structural data with cellular transfection efficiencies reveals that the highest transfection efficiency is obtained with those amphiphiles that are easily hydrated, form fluid aggregates, and undergo a transition to the inverted hexagonal phase in the presence of plasmid DNA (p-DNA) at physiological ionic strength.

Multivesicular bodies contain membrane vesicles which either undergo lysosomal digestion or are released in the extracellular environment as exosomes. Evidence is accumulating that supports a physiological role for exosomes in, for example, antigen presentation or removal of transferrin receptor during reticulocyte development. Here, inspired by observations on exosomal release from reticulocytes, we discuss the potential involvement of the so-called ESCRT mechanism in the entrapment of both lysosomal and exosomal cargo within the intralumenal vesicles of multivesicular bodies. We propose that this mechanism operates at different sites in the endocytic itinerary in different cells, thereby providing a tool for directional sorting. We also explore the possibility that the efficiency of sorting of molecules into exosomes increases when the recycling kinetics of molecules decreases, exosomal sorting being favored by intermolecular interactions occurring within lipid domains, or with protein webs, that slow lateral mobility. These considerations are mirrored in the context of current knowledge on the mechanism of protein sorting for degradation in lysosomes, and the hijacking of such mechanisms by some retroviruses for particle budding.

A conceptually new class of cationic amphiphiles, Sunfish amphiphiles, designed for the delivery of genes into cells is introduced. Sunfish amphiphiles have two hydrophobic tails, connected at the 4- and the N-position to the cationic pyridinium headgroup. Two extreme morphologies visualised by backfolding and combining of both tails at one site (matching situation) or unfolding of the tails at distinct interaction sites at biological membranes will lead to considerable differences in morphological behaviour. The underlying rationale allows controlled release by using this morphological alteration of the Sunfish/helper-lipid/DNA complex (lipoplex). The often excellent transfection efficiencies are probably related to these morphological changes. In addition, the Sunfish amphiphiles possess low toxicities, resulting in high cell survival after internalisation. The underlying rationale, design, synthesis and in vitro transfection potential are discussed in detail. Moreover, some physico-chemical characteristics of the Sunfish amphiphiles have been studied. Insulin-like growth factor 1 (IGF-1) is a growth and survival factor for oligodendrocyte lineage cells and promotes myelination. We demonstrate that IGF-binding protein 6 (IGFBP-6) is expressed and localized to the Golgi complex in rat oligodendrocyte precursor (O2A) cells. IGFBP-6 mRNA showed a developmentally regulated expression pattern, displaying a transient decrease during early development, and enhanced levels upon cell maturation. IGFBP-6 mRNA expression could be reduced by addition of basic fibroblast growth factor and progesterone while estrogen increased IGFBP-6 mRNA. IGF-1, platelet-derived growth factor, and insulin had no effect. When added exogenously, IGFBP-6 reduced O2A cell survival in the absence of IGF-1 and inhibited IGF-1-stimulated survival in a partially IGF-1-dependent and partially IGF-1-independent fashion. In addition, IGFBP-6 reduced the IGF-stimulated expression of two myelin proteins, CNPase and MAG. Taken together, the data show that IGFBP-6 is a new negative effector of oligodendrocyte survival and differentiation. Copyright 2003 Wiley-Liss, Inc.

Under hypoglycemic conditions, concomitant hyperinsulinism causes an apparent modification of hemoglobin (Hb) which is manifested by its a aggregation (Niketic et al.. Clin. Chim. Acia 197 (1991) 47). In the present work the causes and mechanisms underlying this Hb modification were Studied. Hemoglobin isolated from normal erythrocytes incubated with insulin was analyzed by applying P-31-spectrometry and lipid extraction and analysis. To study the dynamics of the plasma membrane during hperinsulinisra a fluorescent lipid-analog was applied. In the presence of insulin phosphatidylserine (PS). phosphatidylethanolamine (PE) and cholesterol were found to bind to Hb. Lipid binding resulted in Hb aggregation, a condition that can be reproduced when phospholipids arc incubated with Hb in vitro. Using a fluorescent lipid-analog, it was also shown that exposing crythrocytes to supraphysiological concentrations of insulin in vitro resulted in the internalization of lipids. The results presented in this work, may have relevance to cases of diabetes mellitus and hypoglycemia. Uranijim radovima je pokazano da hiperinsulinizam u uslovima hipoglikemije izaziva modifikaciju molekula hemoglobina koja se manifestuje njegovim agregiranjem (Niketić et al., Clin. Chim. Acta 197 (1991) 47). U ovom radu ispitivana je ova modifikacija molekula hemoglobina, kao i mehanizam njenog nastajanja. Primenom 31P-spektrometrije i analizom lipidnog ekstrakta utvrđeno je da u normalnim eritrocitima inkubiranim sa insulinom dolazi do vezivanja fosfatidil-serina, fosfatidil-etanolmina i holesterola za molekul hemoglobina. Vezivanje fosfolipida za hemoglobin dovodi do njegovog agregiranja što je potvrđeno eksperimentima u kojima je hemoglobin inkubiran sa fosfolipidima in vitro. Primenom fluorescentnog lipidnog analoga pokazano je da pri izlaganju eritrocita suprafiziološkim koncentracijama insulina dolazi do internalizacije membranskih lipida. Dobijeni rezultati mogu biti od značaja za pacijente obolele od šećerne bolesti i hipoglikemije.

Plasma membranes of epithelial cells consist of two domains, an apical and a basolateral domain, the surfaces of which differ in composition. The separation of these domains by a tight junction and the fact that specific transport pathways exist for intracellular communication between these domains and distinct intracellular compartments relevant to cell polarity development, have triggered extensive research on issues that focus on how the polarity is generated and maintained. Apart from proper assembly of tight junctions, their potential functioning as landmark for the transport machinery, cell-cell adhesion is obviously instrumental in barrier formation. In recent years, distinct endocytic compartments, defined as subapical compartment or common endosome, were shown to play a prominent role in regulating membrane trafficking to and from polarized membrane domains. Sorting devices remain to be determined but likely include distinct rab proteins, and evidence is accumulating to indicate that signaling events may direct intracellular membrane transport, intimately involved in the biogenesis and maintenance of polarized membrane domains and hence the development of cell polarity.

In plasma membranes, lipids distribute asymmetrically across the bilayer, a process that requires proteins. Recent work identified novel lipid translocators in yeast, and their activity was functionally correlated to endocytosis, thus boosting investigations on identity, mechanism, and function of lipid translocases.

The data reveal that IGFBP-1 and -2 reduced O2A cell survival in the absence and presence of exogenously added IGF-1. The effects of IGFBP-1 on cell survival in the presence of exogenously added IGF-1 were IGF-1-dependent, whereas IGFBP-2 displayed both IGF-1-dependent and IGF-1-independent effects. Furthermore, IGFBP-1 and -2 inhibited O2A cell differentiation in the presence of IGF-1 as reflected by decreased expression levels of two myelin proteins, CNPase (2',3'-cyclic nucleotide 3'phosphohydrolase) and MAG (myelin associated glycoprotein). Analysis of medium samples revealed that O2A cells do not secrete proteases that degrade these IGFBPs. Taken together the data show that IGFBP-1 and -2 are negative effectors of oligodendrocyte survival and differentiation. Accordingly, the role of IGFBPs should be explicitly taken into account when investigating IGF-1 effects on oligodendrocytes, especially in the context of therapeutic purposes. Oligodendrocytes, the myelinating cells of the central nervous system, are capable of transporting vast quantities of proteins and of lipids, In particular galactosphingolipids, to the myelin sheath. The sheath is continuous with the plasma membrane of the oligodendrocyte, but the composition of both membrane domains differs substantially. Given its high glycosphingolipid and cholesterol content the myelin sheath bears similarity to the lipid composition of the apical domain of a polarized cell. The question thus arises whether myelin components, like typical apical membrane proteins are transported by an apical-like trafficking mechanism to the sheath, involving a 'raft'-mediated mechanism. Indeed, the evidence indicates the presence of cognate apical and basolateral pathways in oligodendrocytes. However, all major myelin proteins do not participate in this pathway, and remarkably apical-like trafficking seems to be restricted to the oligodendrocyte cell body. In this review, we summarize the evidence on the existence of different trafficking pathways in the oligodendrocyte, and discuss possible mechanisms separating the oligodendrocyte's membrane domains.
