Abstract

The Classical Armenian definite article system is unusual for Indo-European, but paralleled in prehistoric common Kartvelian. It may represent an intermediate stage in grammaticalization between demonstratives and definite articles. The Kartvelian articles progressed further down the path of grammaticalization, becoming markers of argument status, and eventually being incorporated into the case system. It has been suggested that the modern Armenian definite article has also progressed further than the stage of definiteness marking, becoming a marker of specificity, but in fact, its non-definite uses are better described as associated with nominalisation, and in particular with core argument status, than with specificity.

Abstract

Summary: Favourable outcomes with CPX‐351 versus conventional 7 + 3 were demonstrated in the pivotal phase III trial in adults aged 60–75 years with newly diagnosed, highrisk/secondary acute myeloid leukaemia (AML). As a complement to the clinical trial and to address important data gaps, the CPX‐351 Real‐World Effectiveness and SafeTy (CREST‐UK; NCT05169307) study evaluated the use of CPX‐351 in routine clinical practice in the UK, in 147 patients with newly diagnosed therapy‐related AML or AML with myelodysplasia‐related changes. Best response of complete remission or complete remission with incomplete platelet or neutrophil recovery was achieved by 53% of evaluable patients. Kaplan–Meier median overall survival (OS) was 12.8 months (95% confidence interval 9.2–15.3). Fifty (34%) patients proceeded to haematopoietic cell transplantation (HCT); median OS landmarked from the HCT date was not reached. There were no new safety concerns with CPX‐351 identified in CREST‐UK. Patients treated with CPX‐351 in the outpatient setting spent an average of 24.4, 16.7, 28.2, and 27.7 fewer days on the ward compared with inpatients during first induction, second induction, first consolidation, and second consolidation, respectively. The results from CREST‐UK provide valuable insights into the effectiveness, safety, and outpatient delivery of CPX‐351 in routine clinical practice in the UK. [ABSTRACT FROM AUTHOR]

Abstract

Therapeutic vaccination can harness the body's cellular immune system to target and destroy cancerous cells. Several treatment options are available to eliminate pre-cancerous and cancerous lesions caused by human papillomaviruses (HPV), but may not result in a long-term cure. Therapeutic vaccination may offer an effective, durable, and minimally intrusive alternative. We developed mucosally delivered, recombinant, non-replicating human adenovirus type 5 (rAd5)-vectored vaccines that encode HPV16′s oncogenic proteins E6 and E7 alongside a molecular dsRNA adjuvant. The induction of antigen-specific T cells and the therapeutic efficacy of rAd5 were evaluated in a mouse model of HPV tumorigenesis where E6E7-transformed cells, TC-1, were implanted subcutaneously in C57BL/6 mice. After tumor growth, mice were treated intranasally with rAd5 vaccines expressing the wildtype form of E6E7 (rAd5-16/E6E7Wt) in combination with an anti-PD-1 antibody or isotype control. Animals treated with rAd5-16/E6E7Wt with and without anti-PD-1 had significant reductions in tumor volume and increased survival compared to controls. Further, animals treated with rAd5-16/E6E7Wt had increased CD4+ and CD8+ tumor-infiltrating lymphocytes (TILs) and produced a cytotoxic tumor microenvironment. In a second study, the immunogenicity of a non-transformative form of E6E7 (rAd5-16/E6E7Mu) and a vaccine encoding predicted T cell epitopes of E6E7 (rAd5-16/E6E7epi) were evaluated. These vaccines elicited significant reductions in TC-1 tumor volume and increased survival of animals. Antigen-specific CD8+ T effector memory cells were observed in the animals treated with E6E7-encoding rAd5, but not in the rAd5-empty group. The work described here demonstrates that this mucosal vaccination can be used therapeutically to elicit specific cellular immunity and further identifies a clinical candidate with great potential for the treatment and prevention of human cervical cancer. Abstract

Patients with FLT3-mutated AML have a high relapse rate and suboptimal outcomes. Many have co-mutations suitable for measurable residual disease (MRD) monitoring by RT-qPCR and those destined to relapse can be identified by high or rising levels of MRD, called molecular failure.  This provides a window for pre-emptive intervention, but there is little evidence to guide treatment. The use of FLT3inhibitors (FLT3i) appears attractive but their use has not yet been evaluated.  We identified 56 patients treated with FLT3i at molecular failure.  The FLT3mutation was an ITD in 52, TKD in 7 and both in 3. Over half of patients had previously received midostaurin. Molecular failure occurred at a median 9.2 months from diagnosis and was treated with gilteritinib (n= 38), quizartinib (n= 7) or sorafenib (n= 11). 60% achieved a molecular response, with 45% reaching MRD negativity. Haematological toxicity was low, and 22 patients were bridged directly to allogeneic transplant with another 6 to donor lymphocyte infusion. 2-year overall survival was 80% (95%CI 69–93) and molecular event-free survival 56% (95%CI 44–72). High-sensitivity next-generation sequencing for FLT3-ITD at molecular failure identified patients more likely to benefit. FLT3i monotherapy for molecular failure is a promising strategy which merits evaluation in prospective studies.

Abstract

This thesis presents a study of the syntax and typology of relative clauses in colloquial Armenian. It proposes a syntactic analysis and classification of the relativization strategies available in Armenian within the framework of existing syntactic theoretical and typological proposals concerning relative clauses, and to identify the decisive factors associated with the distribution of these different strategies. As each of the available strategies is paralleled in other languages of the area, it is possible that language contact will have an impact on the choice of strategy. There is also evidence that the role of the relativized element in the relative clause is relevant for the choice of strategy; in particular, evidence presented in previous studies of Armenian suggests that the distribution of participial RCs may violate the Relativization Accessibility Hierarchy if this is envisaged as operating directly in terms of syntactic grammatical relations.The study is mainly based on data from sound recordings of native speaker consultants from various areas of Armenia, both spontaneous speech and responses to stimuli designed to elicit relative clauses with particular properties that have been proposed to affect the choice of relativization strategy. The relative clauses are entered into a database with filters for relevant features, which contains approximately 2000 examples. The results show that accessibility to relativization is determined by semantic (affectedness) and pragmatic (topicality) role prominence, and by frequent role-reference association patterns, rather than directly by syntactic grammatical relations. This provides a coherent explanation of the apparent Accessibility Hierarchy violations found in Armenian, as well as other phenomena that have proved problematic for syntactic structure-based interpretations of accessibility to relativization.; Cette thèse étudie la syntaxe et la typologie des propositions relatives en arménien parlé. Elle propose une analyse syntaxique et une classification des stratégies de relativisation disponibles en arménien, dans le cadre des approches théoriques et typologiques existantes de la relativisation, ainsi qu’à identifier les facteurs décisifs pour le choix des différentes stratégies. Chaque stratégie ayant des équivalents dans d'autres langues de la région, le contact linguistique peut influencer leur choix. Il est également probable que le rôle de l'élément relativisé dans la proposition relative soit pertinent pour le choix de la stratégie. En particulier, les données présentées dans de précédentes études sur l'arménien suggèrent que la distribution des relatives participiales peut constituer une violation de la hiérarchie d'accessibilité de la relativisation si elle est envisagée comme opérant directement en termes de fonctions grammaticales syntaxiques. L’étude se concentre sur les parlers de diverses régions d’Arménie. Les propositions relatives ont été saisies dans une base de données d’environ 2000 exemples avec des filtres portant sur les paramètres pertinents.Les résultats montrent que l’accessibilité à la relativisation est déterminée par la saillance liée aux rôles sémantiques (affectivité) et pragmatiques (thematicité) et par de fréquents modèles d’association rôle-référence plutôt que directement par des fonctions grammaticales syntaxiques. Ceci fournit une explication cohérente des violations apparentes de la hiérarchie d'accessibilité trouvées en arménien, ainsi que d'autres phénomènes qui se sont révélés problématiques pour les interprétations de l'accessibilité à la relativisation basées directement sur la structure syntaxique.

Abstract

The stratigraphic record of lahars in the Whangaehu River reveals that in the past 180,000 years this route has been one of the main conduits for lahars from Mount Ruapehu, the highest active andesitic stratovolcano in the Central North Island of New Zealand. Both debris flows and hyperconcentrated flows have engulfed surfaces up to 160 km distance from the Volcano. Eight episodes of laharic activity are recognized by the distinctive lithology and similar age of their deposits. The newly defined upper Pleistocene Whangaehu Formation provides evidence for the earliest lahar event in the Valley, c. 180,000- 140,000 years ago. There is only meagre evidence for laharic activity following this event until the Ohakean and Holocene, although two new informally named deposits - the Mangatipona pumice sand (c. 37,000 years B.P.) and Apitian lahars (c. 32,000-25,500 years B.P) - are recognized, of minor extent. The formerly defined late Quaternary Te Heuheu (c. 25,500- 14,700 years B.P.), Tangatu (c. 14,700-5,370 years B.P.), Manutahi (c. 5 ,370-3,4600 years B.P.), Mangaio (c. 4,600 years B.P.) and Onetapu (< c. 1,850 years B.P.) Formations are here described and interpreted. Triggering mechanisms for lahar deposits are distinguished based on lithological criteria. (a) Bouldery deposits in the Whangaehu Formation are interpreted to have been emplaced by a single highly competent debris flow triggered by a southerly-directed flank collapse at Mount Ruapehu. This debris flow was competent enough to transport boulders up to 2 m in diameter over 140 km from the Volcano. Bouldery deposits are also recognized in the Onetapu Formation, but are restricted to higher gradient surfaces on the Mount Ruapehu ring plain. The Onetapu Formation deposits are interpreted to have been emplaced by lahars resulting from catastrophic drainage of Crater Lake, which occupies the active crater on Mount Ruapehu.
