Abstract

The Large Hadron Collider will soon undergo an upgrade to increase its luminosity by a factor of ~10 [1]. A crucial part of this upgrade will be replacement of the NbTi focusing magnets with Nb3Sn magnets that achieve a ~50% increase in the field strength. This will be the first ever large-scale implementation of Nb3Sn magnets in a particle accelerator. The High-Luminosity LHC Upgrade, HL-LHC is a CERN project with a world-wide collaboration. It is under construction and utilizes Nb3Sn Magnets (named MQXF) as key ingredients to increase tenfold the integrated luminosity delivered to the CMS and ATLAS experiments in the next decade. The HL-LHC AUP is the US effort to contribute approximately 50% of the low-beta focusing magnets and crab cavities for the HL-LHC. This paper will present the program to fabricate the Nb3Sn superconducting magnets. Abstract

Abstract While there is a considerable body of data regarding the sources of minerals employed in Bronze Age Egypt, the supply chains to Sudanese Lower Nubia are virtually unknown. This paper presents results of lead isotope analysis of 11 samples of kohl from C-group, Pan-grave and New Kingdom funerary contexts across the Debeira and Ashkeit areas in Sudanese Lower Nubia during the Bronze Age. The samples could be divided in two groups based on their lead isotope ratios. A comparison between the kohl samples from Sudanese Lower Nubia and galena ores from various mining sites on the Red Sea coast of Egypt indicated that some of the galena used in kohl mixtures in Sudanese Lower Nubia came from the Pharaonic mining site of Gebel el-Zeit. The second group of Nubian samples seems to have originated from an alternative galena source yet to be determined. This is the first time that kohl samples from Sudanese Lower Nubia are integrated into known northeast African networks of supply by using Lead isotope analysis.

Abstract

By the end of October 2022, the US HL-LHC Accelerator Upgrade Project (AUP) had completed fabrication of ten MQXFA magnets and tested eight of them. The MQXFA magnets are the low beta quadrupole magnets to be used in the Q1 and Q3 Inner Triplet elements of the High Luminosity LHC. This AUP effort is shared by BNL, Fermilab, and LBNL, with strand verification tests at NHMFL. An important step of the AUP QA plan is the testing of MQXFA magnets in a vertical cryostat at BNL. The acceptance criteria that could be tested at BNL were all met by the first four production magnets (MQXFA03-MQXFA06). Subsequently, two magnets (MQXFA07 and MQXFA08) did not meet some criteria and were disassembled. Lessons learned during the disassembly of MQXFA07 caused a revision to the assembly specifications that were used for MQXFA10 and subsequent magnets. In this paper, we present a summary of: 1) the fabrication and test data of all the MQXFA magnets; 2) the analysis of MQXFA07/A08 test results with characterization of the limiting mechanism; 3) the outcome of the investigation, including the lessons learned during MQXFA07 disassembly; and 4) the finite element analysis correlating observations with test performance.

Abstract

Although the importance of genome organization for transcriptional regulation of cell-fate decisions and function is clear, the changes in chromatin architecture and how these impact effector and memory CD8+ T cell differentiation remain unknown. Using Hi-C, we studied how genome configuration is integrated with CD8+ T cell differentiation during infection and investigated the role of CTCF, a key chromatin remodeler, in modulating CD8+ T cell fates through CTCF knockdown approaches and perturbation of specific CTCF-binding sites. We observed subset-specific changes in chromatin organization and CTCF binding and revealed that weak-affinity CTCF binding promotes terminal differentiation of CD8+ T cells through the regulation of transcriptional programs. Further, patients with de novo CTCF mutations had reduced expression of the terminal-effector genes in peripheral blood lymphocytes. Therefore, in addition to establishing genome architecture, CTCF regulates effector CD8+ T cell heterogeneity through altering interactions that regulate the transcription factor landscape and transcriptome.

Abstract

As we age, structural changes contribute to progressive decline in organ function, which in the heart act through poorly characterized mechanisms. Taking advantage of the short lifespan and conserved cardiac proteome of the fruit fly, we found that cardiomyocytes exhibit progressive loss of Lamin C (mammalian Lamin A/C homologue) with age, coincident with decreasing nuclear size and increasing nuclear stiffness. Premature genetic reduction of Lamin C phenocopies aging's effects on the nucleus, and subsequently decreases heart contractility and sarcomere organization. Surprisingly, Lamin C reduction downregulates myogenic transcription factors and cytoskeletal regulators, possibly via reduced chromatin accessibility. Subsequently, we find a role for cardiac transcription factors in regulating adult heart contractility and show that maintenance of Lamin C, and cardiac transcription factor expression, prevents age-dependent cardiac decline. Our findings are conserved in aged non-human primates and mice, demonstrating that age-dependent nuclear remodeling is a major mechanism contributing to cardiac dysfunction.

13. Toceranib phosphate in the management of canine insulinoma: A retrospective multicentre study of 30 cases (2009-2019).

Abstract

BACKGROUND: Insulinomas are the most common tumour of the endocrine pancreas in dogs. These malignant tumours have a high metastatic rate and limited chemotherapeutic options. The multi-receptor tyrosine kinase inhibitor sunitinib malate has benefit in the treatment of metastatic insulinoma in people. Toceranib phosphate, an analogous veterinary agent, may provide benefit for dogs. METHODS: A retrospective study describing the extent and duration of clinical outcomes and adverse events (AEs) in dogs diagnosed with insulinoma and receiving toceranib. RESULTS: Records for 30 dogs diagnosed with insulinoma and having received toceranib were identified from a medical record search of five university and eight referral hospitals. The median progression-free interval and overall survival time were 561 days (95% confidence interval (CI): [246, 727 days]) and 656 days (95% CI: [310, 1045 days]), respectively. Of the dogs for which the canine Response evaluation criteria for solid tumours tool could be applied, the majority (66.7%) showed either a complete response, partial response or stable disease. Time to clinical progression was associated with prior intervention and type of veterinary practice. Larger dogs were at increased risk for disease progression and death. No novel AEs were reported. CONCLUSIONS: Most dogs diagnosed with insulinoma and receiving toceranib appeared to have a clinical benefit. Randomised, prospective studies are needed to better elucidate and objectively quantify the potential effect and survival benefit of toceranib therapy for management of insulinoma in dogs.

Abstract

An observation of Charged Lepton Flavor Violation (CLFV) would be unambiguous evidence for physics beyond the Standard Model. The Mu2e and COMET experiments, under construction, are designed to push the sensitivity to CLFV in the mu to e conversion process to unprecedented levels. Whether conversion is observed or not, there is a strong case to be made for further improving sensitivity, or for examining the process on additional target materials. Mu2e-II is a proposed upgrade to Mu2e, with at least an additional order of magnitude in sensitivity to the conversion rate over Mu2e. The approach and challenges for this proposal are summarized. Abstract

The MQXFA Quadrupole magnets will be installed in High Luminosity LHC to form the Q1 and Q3 inner triplet optical elements in front of the interaction points 1 (ATLAS) and 5 (CMS). A pair of MQXFA units is assembled in a stainless steel helium vessel, including the end domes, to make the Q1 Cold Mass or the Q3 Cold Mass. The US HL LHC Accelerator Upgrade Project* is responsible for the design, manufacturing and test of the Q1/Q3 Cold Masses and the complete MQXFA magnets. CERN provides the cryostat components and is responsible for integration and installation in HL LHC. The MQXFA quadrupoles have 150 mm aperture, 4.2 m magnetic length, nominal gradient of 132.2 T/m, and coil peak field of 11.3 T. They use Nb_3Sn conductor and a support structure made of segmented aluminum shells pre-loaded by using bladders and keys. Abstract

The muon-to-electron conversion (Mu2e) experiment at Fermilab will be used to search for the charged lepton flavor-violating conversion of muons to electrons in the field of an atomic nucleus. The Mu2e experiment is currently in the construction stage. The scope of this paper is the cryogenic distribution system and superconducting power leads for four superconducting solenoid magnets: Production Solenoid (PS), an Upstream and Downstream Transport Solenoids (TSu and TSd) and Detector Solenoid (DS). The design of the cryogenic distribution system and the fabrication of several sub-systems was reported previously. This paper reports on additional fabrication and installation progress that has been performed over the past two years. Lessons learned during fabrication and testing of the cryogenic distribution system components are described. In particular, the challenges and solutions implemented for aluminum welding are reported. A description of the process used to qualify the welding procedure and welders for welding the aluminium stabilized NbTi superconducting power leads is provided. Additionally, the progress made with regards to installing the power leads into the cryogenic Feedboxes is covered.

Abstract

A quench detection system was developed for protecting and monitoring the superconducting solenoids for the Muon-to-Electron Conversion Experiment (Mu2e) at Fermilab. The quench system was designed for a high level of dependability and long-term continuous operation. It is based on three tiers: Tier-I, FPGA-based Digital Quench Detection (DQD); Tier-II, Analog Quench Detection (AQD); and Tier-3, the quench controls and data management system. The Tier-I and Tier-II are completely independent and fully redundant systems. The Tier-3 system is based on National Instruments (NI) C-RIO and provides the user interface for quench controls and data management. It is independent from Tiers I & II. The DQD provides both quench detection and quench characterization (monitoring) capability. Both DQD and AQD have built-in high voltage isolation and user programmable gains and attenuations. The DQD and AQD also includes user configured current dependent thresholding and validation times. A 1st article of the three-tier system was fully implemented on the new Fermilab magnet test stand for the HL-LHC Accelerator Up-grade Project (AUP). It successfully provided quench protection and monitoring (QPM) for a cold superconducting bus test in November 2020. The Mu2e quench detection design has since been implemented for production testing of the AUP magnets. A detailed description of the system along with results from the AUP superconducting bus test will be presented.

Abstract

A study of initial-state gluon radiation (ISR) in hadron collisions is presented using Drell-Yan (DY) events produced in proton-antiproton collisions by the Tevatron collider at a center-of-mass energy of 1.96 TeV. This paper adopts a novel approach which uses the mean value of the Z/$\gamma^*$ transverse momentum $$ in DY events as a powerful observable to characterize the effect of ISR. In a data sample corresponding to an integrated luminosity of 9.4 fb$^{-1}$ collected with the CDF Run II detector, $$ is measured as a function of the Z/$\gamma^*$ invariant mass. It is found that these two observables have a dependence, $ = -8 + 2.2 \ln m_{DY}^2$ [GeV/c], where $m_{DY}$ is the value of the Z/$\gamma^*$ mass measured in units of GeV/$c^2$. This linear dependence is observed for the first time in this analysis. It may be exploited to model the effect of ISR and constrain its impact in other processes., Comment: 14 pages, 14 figures

Abstract

Since the initial isolation of human embryonic stem cells and subsequent discovery of reprogramming methods for somatic cells, thousands of protocols have been developed to create each of the hundreds of cell types found in-vivo with significant focus on disease-prone systems, e.g., cardiovascular. Robust protocols exist for many of these cell types, except for cardiac fibroblasts (CF). Very recently, several competing methods have been developed to generate these cells through a developmentally conserved epicardial pathway. Such methods generate epicardial cells, but here we report that prolonged exposure to growth factors such as bFGF induces fibroblast spindle-like morphology and similar chromatin architecture to primary CFs. Media conditions for growth and assays are provided, as well as suggestions for seeding densities and timepoints for protein harvest of extracellular matrix. We demonstrate marker expression and matrix competency of resultant cells as shown next to primary human cardiac fibroblasts. These methods provide additional guidance to the original protocol and result in an increasingly stable phenotype.

Abstract

At the Fermilab Tevatron proton-antiproton ($p\bar{p}$) collider, high-mass electron-neutrino ($e\nu$) pairs are produced predominantly in the process $p \bar{p} \rightarrow W(\rightarrow e\nu) + X$. The asymmetry of the electron and positron yield as a function of their pseudorapidity constrain the slope of the ratio of the $u$- to $d$-quark parton distributions versus the fraction of the proton momentum carried by the quarks. This paper reports on the measurement of the electron-charge asymmetry using the full data set recorded by the Collider Detector at Fermilab in 2001--2011 and corresponding to 9.1~fb$^{-1}$ of integrated luminosity. The measurement significantly improves the precision of the Tevatron constraints on the parton-distribution functions of the proton. Abstract

This paper introduces the processing element architecture of the second generation SpiNNaker chip, implemented in 22nm FDSOI. On circuit level, the chip features adaptive body biasing for near-threshold operation, and dynamic voltage-and-frequency scaling driven by spiking activity. On system level, processing is centered around an ARM M4 core, similar to the processor-centric architecture of the first generation SpiNNaker. To speed operation of subtasks, we have added accelerators for numerical operations of both spiking (SNN) and rate based (deep) neural networks (DNN). PEs communicate via a dedicated, custom-designed network-on-chip. We present three benchmarks showing operation of the whole processor element on SNN, DNN and hybrid SNN/DNN networks.

Abstract

Thermal inspections of a structure typically utilize a flash or quartz lamp heat source located on the same side of an infrared camera. The heat source provides light energy for heating while the infrared camera measures the surface transient temperature response. The inspection can be difficult for low emissivity surfaces for several reasons. First, the high intensity light can reflect off the surface and cause “burn-in” to the camera’s detector. The “burn-in” can take time for the sensors to recover and potentially damage the detector. Secondly, the heat source after pulsing, has a transient cool down component. The cool down component can be reflected and therefore superimposed over the structure’s thermal response which can cause an error (false defect indications) in the inspection. Lastly, the heat source is spectrally broad and therefore while heating, infrared components of the heat source can produce non-uniformity in the measured temperature field. Typically for the inspection of reflective surfaces, paint or other emissivity enhancing coatings are applied before inspection. In this paper, a pulsed light emitting diodes (PLED) heat source is used. The PLED heat source is spectrally narrow, contained within the visible band, and therefore not detectable by the infrared camera. The PLED heat source is configured to reduce any transient cool down components that could produce false defect indications. The PLED thermal inspections are compared to commercially available flash thermography inspections on unpainted aluminum samples with simulated corrosion and additively manufactured Ti-6AL-4V metal specimens.

To profile the activity of gene regulatory elements in diverse cell types and tissues in the human body, we applied single-cell chromatin accessibility assays to 30 adult human tissue types from multiple donors. We integrated these datasets with previous single-cell chromatin accessibility data from 15 fetal tissue types to reveal the status of open chromatin for ∼1.2 million candidate cis-regulatory elements (cCREs) in 222 distinct cell types comprised of >1.3 million nuclei. We used these chromatin accessibility maps to delineate cell-type-specificity of fetal and adult human cCREs and to systematically interpret the noncoding variants associated with complex human traits and diseases. This rich resource provides a foundation for the analysis of gene regulatory programs in human cell types across tissues, life stages, and organ systems.

Abstract

Misregulated gene expression in human hearts can result in cardiovascular diseases that are leading causes of mortality worldwide. However, the limited information on the genomic location of candidate cis-regulatory elements (cCREs) such as enhancers and promoters in distinct cardiac cell types has restricted the understanding of these diseases. Here, we defined >287,000 cCREs in the four chambers of the human heart at single-cell resolution, which revealed cCREs and candidate transcription factors associated with cardiac cell types in a region-dependent manner and during heart failure. We further found cardiovascular disease-associated genetic variants enriched within these cCREs including 38 candidate causal atrial fibrillation variants localized to cardiomyocyte cCREs. Additional functional studies revealed that two of these variants affect a cCRE controlling KCNH2/HERG expression and action potential repolarization. Overall, this atlas of human cardiac cCREs provides the foundation for illuminating cell type-specific gene regulation in human hearts during health and disease.

Abstract

The CCCTC-binding factor (CTCF) works together with the cohesin complex to drive the formation of chromatin loops and topologically associating domains, but its role in gene regulation has not been fully defined. Here, we investigated the effects of acute CTCF loss on chromatin architecture and transcriptional programs in mouse embryonic stem cells undergoing differentiation to neural precursor cells. We identified CTCF-dependent enhancer-promoter contacts genome-wide and found that they disproportionately affect genes that are bound by CTCF at the promoter and are dependent on long-distance enhancers. Disruption of promoter-proximal CTCF binding reduced both long-range enhancer-promoter contacts and transcription, which were restored by artificial tethering of CTCF to the promoter. Promoter-proximal CTCF binding is correlated with the transcription of over 2,000 genes across a diverse set of adult tissues. Taken together, the results of our study show that CTCF binding to promoters may promote long-distance enhancer-dependent transcription at specific genes in diverse cell types.

38. Lowered Expectations and Power: A Multi-Site Case Study of K-12 Educational Technology Decision-Makers

Abstract

This dissertation seeks to better understand the individual educational technology decision-maker and how they interact with other members of the educational technology decision making process in a public k-12 school setting. To do this, I asked the questions: who are the people who make educational technology decision in a K-12 school district? What qualifications regarding technology do the people who make educational technology decisions hold? How does the technology decision making process unfold within a given district? What personal experiences impact educational technology decisions? This study also introduces the Essential Aspects of Educational Technology Decision-Making Framework to understand how different aspects of technology and leadership influence the types of educational technology problems a district will face, what individual decision-makers will draw in from their background when they participate in the educational technology decision making process, and the power dynamics of the process itself. This mixed method study used descriptive statistical analysis to provide context to the interview data collected from participants from four Pennsylvania school districts. This study found that most of the participating educational technology decision-makers had no formal technology training and relied heavily on their educational experience as their primary qualification for making decisions about technology. The lack of formal training and over-reliance on educational experience, along with other factors, perpetuated a series of unconscious, yet pervasive lowered expectations toward educational technology. This study also found that Technology Directors have an outsized amount of practical power in a school district as a result of the nature of the position and other members of the decision-making process deferring their judgment to that of the Technology Director. These findings show the need to further understand how the lack of formal technology qualifications are influencing how decisions about educational technology are made, the nature and magnitude of the influence Technology Directors wield, and how the community's perceptions of educational technology can be better understood and, if need be, improved. Abstract

We report on a search for a spin-zero non-standard-model particle in proton-antiproton collisions collected by the Collider Detector at Fermilab at a center-of-mass-energy of 1.96 TeV. This particle, the $\phi$ boson, is expected to decay into a bottom-antibottom quark pair and to be produced in association with at least one bottom quark. The data sample consists of events with three jets identified as initiated by bottom quarks and corresponds to $5.4~\text{fb}^{-1}$ of integrated luminosity. In each event, the invariant mass of the two most energetic jets is studied by looking for deviations from the multijet background, which is modeled using data. No evidence is found for such particle. Exclusion upper limits ranging from 20 to 2 pb are set for the product of production cross sections times branching fraction for hypothetical $\phi$ boson with mass between 100 and 300 GeV/$c^2$. Abstract

Disorders of consciousness are neurological conditions characterized by impaired arousal and awareness of self and environment. Behavioural responses are absent or are present but fluctuate. Disorders of consciousness are commonly encountered as a consequence of both acute and chronic brain injuries, yet reliable epidemiological estimates would require inclusive, operational definitions of the concept, as well as wider knowledge dissemination among involved professionals. Whereas several manifestations have been described, including coma, vegetative state/unresponsive wakefulness syndrome and minimally conscious state, a comprehensive neurobiological definition for disorders of consciousness is still lacking. The scientific literature is primarily observational, and studies-specific aetiologies lead to disorders of consciousness. Despite advances in these disease-related forms, there remains uncertainty about whether disorders of consciousness are a disease-agnostic unitary entity with a common mechanism, prognosis or treatment response paradigm. Our knowledge of disorders of consciousness has also been hampered by heterogeneity of study designs, variables, and outcomes, leading to results that are not comparable for evidence synthesis. The different backgrounds of professionals caring for patients with disorders of consciousness and the different goals at different stages of care could partly explain this variability. The Prospective Studies working group of the Neurocritical Care Society Curing Coma Campaign was established to create a platform for observational studies and future clinical trials on disorders of consciousness and coma across the continuum of care. In this narrative review, the author panel presents limitations of prior observational clinical research and outlines practical considerations for future investigations.
