The role of parental literacy skills in cross-linguistic transfer, the phenomenon of the sharing of skills and competence from one language to another, is under-researched. The present study tested 147 Hong Kong Chinese children and their parents (76 boys; mean age = 7.16). Parental literacy skills significantly explained children's reading and spelling in English after controlling for socioeconomic status, highlighting the role of parental skills in facilitating children's second language development. Cross-linguistic transfers between Chinese and English were found between reading and writing after controlling for age, grade, non-verbal intelligence, and socioeconomic status. The results were replicated with another twin sample.

Intercultural communicative competence (ICC) has been widely acknowledged as a core element of today's foreign language education. However, even though the importance of intercultural language teaching is commonly recognized among adult learners and at the post-secondary level, teachers of adolescent English learners often find it hard to effectively incorporate culture into English learning because of the lack of an instructional model facilitating their students' intercultural development and English learning experiences. Therefore, this study aims to investigate whether integrating intercultural learning into an online EFL curriculum can elevate teenage EFL students' L2 motivation, intercultural communicative competence, and English proficiency. The researchers used a quasi-experimental design by randomly selecting two eighth-grade classes in a secondary school in northern Thailand, with one class designated as the experimental group (N = 31) and the other as the control group (N = 28). The effects of this teaching experiment were then examined using both quantitative and qualitative data. The findings demonstrated that the students in the experimental group showed a greater improvement compared with those of the students in the control group after an 8-week, interculturally embedded English curriculum. The results suggested that ICC is conducive to adolescent EFL students' intercultural development as well as their English learning motivation and outcome. The applications of ICC-based EFL instruction in similar contexts are discussed.

This study is a meta-analytical study that examines the effectiveness of gamification in learning performance in educational settings (n = 29; year-span = 2011-2019). Specifically, it aimed to investigate (a) whether gamification could improve learning performance, and (b) whether peer interaction (i.e., peer competition and peer collaboration) moderated the effectiveness of gamification in learning performance. Results from random-effects models showed significant effects of gamification in learning performance (g = 0.595, 95% CI [0.432, 0.758], N = 3515). This effect remained robust after excluding outliers and was stable in a sub-split analysis that excludes studies with low methodological rigor (i.e., studies with pre-post test design). Subgroup analyses revealed a moderating effect of peer competition in gamification in learning, suggesting that competitive games were better than non-competitive games for promoting learning performance in educational settings. However, this effect was not robust and no evidence of subgroup differences were found in the sub-split analysis. Peer collaboration did not moderate the effectiveness of gamification in learning as no subgroup differences were found between collaborative games and non-collaborative games. The effectiveness of games that were both competitive and collaborative did not differ from those that were only competitive. Other moderators such as education level and research design were also investigated. No subgroup differences were found for these two moderators. Educational implications and limitations were further discussed.

This study aimed to examine the learning progression (LP) model of scientific imagination among undergraduate students using the Scientific Imagination Test-Verbal (SIT-Verbal) and investigated the influence of students' demographic characteristics including gender, age, and discipline on their scientific imagination. Six hundred and sixteen undergraduates from a university in southern Taiwan participated in this study. The SIT-Verbal covered four key components of the scientific imagination process: brainstorming, association, transformation/elaboration, and conceptualization/organization/formation. The multiple validities of SIT-Verbal were assessed via a Rasch partial credit model. The results indicated that the SIT-Verbal had good model-data fit, supporting that undergraduate students' scientific imagination in four stages from brainstorming, association and transformation/elaboration to conceptualization/organization/formation. Additionally, the results showed that the SIT-Verbal was suitable for measuring students' scientific imagination at the university level. The study also provided abundant evidence verifying the SIT-Verbal and supported the learning progression for undergraduate students' scientific imagination.

Therapeutic strategies are being clinically tested either to eradicate latent HIV reservoirs or to achieve virologic control in the absence of antiretroviral therapy. Attaining this goal will require a consensus on how best to measure the numbers of persistently infected cells with the potential to cause viral rebound after antiretroviral-therapy cessation in assessing the results of cure-directed strategies in vivo. Current measurements assess various aspects of the HIV provirus and its functionality and produce divergent results. Here, we provide recommendations from the BEAT-HIV Martin Delaney Collaboratory on which viral measurements should be prioritized in HIV-cure-directed clinical trials.

Despite effective antiretroviral therapy, HIV-1-infected cells continue to produce viral antigens and induce chronic immune exhaustion. We propose to identify HIV-1-suppressing agents that can inhibit HIV-1 reactivation and reduce HIV-1-induced immune activation. Using a newly developed dual-reporter system and a high-throughput drug screen, we identified FDA-approved drugs that can suppress HIV-1 reactivation in both cell line models and CD4+ T cells from virally suppressed HIV-1-infected individuals. We identified 11 cellular pathways required for HIV-1 reactivation as druggable targets. Using differential expression analysis, gene set enrichment analysis, and exon-intron landscape analysis, we examined the impact of drug treatment on the cellular environment at a genome-wide level. We identified what we believe to be a new function of a JAK inhibitor, filgotinib, that suppresses HIV-1 splicing. First, filgotinib preferentially suppresses spliced HIV-1 RNA transcription. Second, filgotinib suppresses HIV-1-driven aberrant cancer-related gene expression at the integration site. Third, we found that filgotinib suppresses HIV-1 transcription by inhibiting T cell activation and by modulating RNA splicing. Finally, we found that filgotinib treatment reduces the proliferation of HIV-1-infected cells. Overall, the combination of a drug screen and transcriptome analysis provides systematic understanding of cellular targets required for HIV-1 reactivation and drug candidates that may reduce HIV-1-related immune activation.

Proliferation of CD4+ T cells harboring HIV-1 proviruses is a major contributor to viral persistence in people on antiretroviral therapy (ART). To determine whether differential rates of clonal proliferation or HIV-1-specific cytotoxic T lymphocyte (CTL) pressure shape the provirus landscape, we performed an intact proviral DNA assay (IPDA) and obtained 661 near-full-length provirus sequences from 8 individuals with suppressed viral loads on ART at time points 7 years apart. We observed slow decay of intact proviruses but no changes in the proportions of various types of defective proviruses. The proportion of intact proviruses in expanded clones was similar to that of defective proviruses in clones. Intact proviruses observed in clones did not have more escaped CTL epitopes than intact proviruses observed as singlets. Concordantly, total proviruses at later time points or observed in clones were not enriched in escaped or unrecognized epitopes. Three individuals with natural control of HIV-1 infection (controllers) on ART, included because controllers have strong HIV-1-specific CTL responses, had a smaller proportion of intact proviruses but a distribution of defective provirus types and escaped or unrecognized epitopes similar to that of the other individuals. This work suggests that CTL selection does not significantly check clonal proliferation of infected cells or greatly alter the provirus landscape in people on ART.

Understanding HIV-1-host interactions can identify the cellular environment supporting HIV-1 reactivation and mechanisms of clonal expansion. We developed HIV-1 SortSeq to isolate rare HIV-1-infected cells from virally suppressed, HIV-1-infected individuals upon early latency reversal. Single-cell transcriptome analysis of HIV-1 SortSeq+ cells revealed enrichment of nonsense-mediated RNA decay and viral transcription pathways. HIV-1 SortSeq+ cells up-regulated cellular factors that can support HIV-1 transcription (IMPDH1 and JAK1) or promote cellular survival (IL2 and IKBKB). HIV-1-host RNA landscape analysis at the integration site revealed that HIV-1 drives high aberrant host gene transcription downstream, but not upstream, of the integration site through HIV-1-to-host aberrant splicing, in which HIV-1 RNA splices into the host RNA and aberrantly drives host RNA transcription. HIV-1-induced aberrant transcription was driven by the HIV-1 promoter as shown by CRISPR-dCas9-mediated HIV-1-specific activation and could be suppressed by CRISPR-dCas9-mediated inhibition of HIV-1 5' long terminal repeat. Overall, we identified cellular factors supporting HIV-1 reactivation and HIV-1-driven aberrant host gene transcription as potential therapeutic targets to disrupt HIV-1 persistence.

A stable latent reservoir for HIV-1 in resting CD4+ T cells is the principal barrier to a cure1-3. Curative strategies that target the reservoir are being tested4,5 and require accurate, scalable reservoir assays. The reservoir was defined with quantitative viral outgrowth assays for cells that release infectious virus after one round of T cell activation1. However, these quantitative outgrowth assays and newer assays for cells that produce viral RNA after activation6 may underestimate the reservoir size because one round of activation does not induce all proviruses7. Many studies rely on simple assays based on polymerase chain reaction to detect proviral DNA regardless of transcriptional status, but the clinical relevance of these assays is unclear, as the vast majority of proviruses are defective7-9. Here we describe a more accurate method of measuring the HIV-1 reservoir that separately quantifies intact and defective proviruses. We show that the dynamics of cells that carry intact and defective proviruses are different in vitro and in vivo. These findings have implications for targeting the intact proviruses that are a barrier to curing HIV infection.

Dissolved organic carbon (DOC) released from submerged leaf litter is an important allochthonous carbon source for heterotrophic bacteria that enhances the detrital food chain in freshwater ecosystems. In contrast, nitrogen (N), phosphorus (P) and micronutrients released during the leaching process support primary production and enhance the grazing food chain in these ecosystems. This study investigated how DOC and dissolved N and P from the leaf litter of two temperate broadleaf and two coniferous tree species affect heterotrophic and autotrophic biomass by incubating the freshwater microbial community in the culture media prepared from the leaf litter leachate. The results showed that heterotrophic biomass, including bacteria, heterotrophic‐flagellated protists and fungal zoospores, was the highest in the leaf litter leachate, which contained the highest DOC concentration relative to dissolved N and P (Japanese hemlock). In contrast, autotrophic biomass, including small algae, autotrophic flagellated protists and cyanobacteria, was associated with concentrations of dissolved N and P in the leaf litter leachates. Amendment of basal nutrients other than N and P, including micronutrients, trace elements and vitamins, increased autotrophic biomass in two leaf litter leachates (oak, Siebold's beech), suggesting that some elements other than N and P were deficient relative to the requirements of autotrophic organisms. These results indicated that the elemental composition of leaf litter leachates influences microbial community structure and heterotrophic versus autotrophic biomass in freshwater ecosystems. More importantly, the present results suggest that changes in forest vegetation surrounding freshwater ecosystems alter detritus‐based heterotrophic production relative to autotrophic production through the changes in the elemental composition (i.e. stoichiometry) of leaf litter inputs. Dissolved organic matter and nutrients released from leaf litter are important cross‐ecosystem resources supporting freshwater food webs. Dissolved organic matter supports heterotrophic organisms in freshwater communities. However, the role of nutrients released from leaf litter in the autochthonous production in aquatic ecosystems is not well understood. Therefore, we investigated how dissolved nutrients released from leaf litter affect algal growth, biomass production, and cellular elemental ratios. Specifically, we experimentally examined the response of green algae to nitrogen (N) and phosphorus (P) released from leaf litter of 11 temperate tree species and the degree of deficiency of nutrient elements other than N and P relative to algal demand in these litter leachates by supplementing these elements. We found that algal growth did not significantly increase with dissolved N or P in the leaf leachates. In contrast, algal biomass production increased with dissolved N concentration, regardless of amendment. Algal growth and production limitation by deficiency of elements other than N and P was found only in the leachate of Japanese hemlock, indicating that the concentrations or release efficiencies of these elements in this leaf litter were lower than those of N and P relative to algal requirements. More importantly, leaf litter leachates from different tree species altered algal cell quotas and C:N:P ratios, which would affect secondary production. These results suggest that variations in leaf litter leachate stoichiometry caused by vegetation change would affect the abundance and chemical composition of phytoplankton, and thus the trophic transfer efficiency between phytoplankton and herbivorous zooplankton.
