6. Evaluation of the Effect of Low-Temperature Plasma Treatment on Seed Germination of Long-Term Stored Genetic Resources.

Abstract

Low-temperature plasma (LTP) is currently one of the non-invasive and environmentally friendly methods of seed treatment and is massively tested on various types of crops. For the needs of gene banks, the use of LTP technology represents the treatment of seeds before sowing to improve the germination and emergence of long-term stored seed samples. Seeds of four genotypes of wheat, oats, flax, and rapeseed stored in the gene bank for 1, 10, and 20 years were plasma treated for 20, 25, and 30 min. Standard germination parameters (SG3, SG7, GR, MGT, and GI), as well as predictive models, were used to evaluate the effect of plasma treatment on seeds, and the effect on seed metabolism was assessed by superoxide dismutase (SOD) activity. The plasma treatment had different effects on germination and on the enzymatic activity of the tested species, and the result was influenced by both the duration of the treatment and the crop species/genotype. The plasma treatment has a positive effect on germination parameters in flax and rapeseed; in some variants, as in wheat, oats generally reacted negatively. SOD activity was variable in wheat, while higher activity with increasing treatment time was found in other crops. The results of this first study focused on long-term stored seeds and showed the potential of plasma treatment of seeds of plant genetic resources, the possibility of stimulating the germination of stored PGRs, and the need to optimize treatment conditions for individual genotypes. Abstract

Water deficit (drought) is an important environmental factor affecting physiological processes in plants. The present work focuses on the study of changes in physiological responses of juvenile plants (plants in the vegetative phase of growth BBCH 14-16) of selected sorghum genotypes Dokok, 30485, Barnard Red and Ruzrok to water deficit and after rehydration. Water deficit affected the observed physiological parameters-gas exchange and chlorophyll fluorescence. Genotypic differences were also confirmed, with Dokok appearing to be the more sensitive genotype and Ruzrok and Barnard Red appearing to be tolerant. Following rehydration, these parameters increased but did not reach the levels of the control plants. A significant decrease in photosynthetic rate (Pn), transpiration (E) and fluorescence compared to the control was found in the water-deficient variant twice for 10 days and 6 days between rehydration periods. Only in the variant where water deficit (14 days) was followed by irrigation (10 days) transpiration increased in genotype 30485. Chlorophyll fluorescence (Fv /Fm) also decreased significantly in this cultivar. The results suggest that a rehydration period of 14 days is insufficient to restore the photosynthetic functions of stressed sorghum plants. [ABSTRACT FROM AUTHOR]

Abstract

A comparative analysis of various parameters that characterize plant morphology, growth, water status, photosynthesis, cell damage, and antioxidative and osmoprotective systems together with an iTRAQ analysis of the leaf proteome was performed in two inbred lines of maize (Zea mays L.) differing in drought susceptibility and their reciprocal F1 hybrids. The aim of this study was to dissect the parent-hybrid relationships to better understand the mechanisms of the heterotic effect and its potential association with the stress response. The results clearly showed that the four examined genotypes have completely different strategies for coping with limited water availability and that the inherent properties of the F1 hybrids, i.e. positive heterosis in morphological parameters (or, more generally, a larger plant body) becomes a distinct disadvantage when the water supply is limited. However, although a greater loss of photosynthetic efficiency was an inherent disadvantage, the precise causes and consequences of the original predisposition towards faster growth and biomass accumulation differed even between reciprocal hybrids. Both maternal and paternal parents could be imitated by their progeny in some aspects of the drought response (e.g., the absence of general protein down-regulation, changes in the levels of some carbon fixation or other photosynthetic proteins). Nevertheless, other features (e.g., dehydrin or light-harvesting protein contents, reduced chloroplast proteosynthesis) were quite unique to a particular hybrid. Our study also confirmed that the strategy for leaving stomata open even when the water supply is limited (coupled to a smaller body size and some other physiological properties), observed in one of our inbred lines, is associated with drought-resistance not only during mild drought (as we showed previously) but also during more severe drought conditions.
