We have previously reported that a major quantitative trait locus (QTL) responsible for susceptibility to salt-induced stroke in the stroke-prone spontaneously hypertensive rat (SHRSP) is located in a 3-Mbp region on chromosome 1 covered by SHRSP.SHR-(D1Rat23-D1Rat213)/Izm (termed Pr1.31), a congenic strain with segments from SHRSP/Izm introduced into the stroke-resistant SHR/Izm. Here, we attempted to narrow down the candidate region on chromosome 1 further through analyses of subcongenic strains constructed for the target region. Simultaneously, salt-induced kidney injury was evaluated through the measurement of urinary albumin and the gene expression of renal tubular injury markers (Kim-1 and Clu) to explore a possible mechanism leading to the onset of stroke. All subcongenic strains examined in this study showed lower susceptibility to salt-induced stroke than SHRSP. Interestingly, Pr1.31 had the lowest stroke susceptibility when compared with newly constructed subcongenic strains harboring fragments of the congenic sequence in Pr1.31. Although Kim-1 and Clu expression after 1 week of salt loading in Pr1.31 did not differ significantly from those in SHRSP, the urinary albumin level of Pr1.31 was significantly lower than those of the other subcongenic strains and that of SHRSP. The present results indicated that, although the congenic fragment in Pr1.31 harbored the gene(s) related to salt-induced organ damages, further genetic dissection of the candidate region was difficult due to multiple QTLs suggested in this region. Further analysis using Pr1.31 will unveil genetic and pathophysiological mechanisms underlying salt-induced end organ damages in SHRSP.

Leukotriene B4 receptor 1 (BLT1), a high-affinity G-protein-coupled receptor for leukotriene B4 (LTB4 ), is expressed on various inflammatory cells and plays critical roles in several inflammatory diseases. In myocardial infarction (MI), various inflammatory cells are known to be recruited to the infarcted area, but the function of BLT1 in MI is poorly understood. Here, we investigated the role of BLT1 in MI and the therapeutic effect of a BLT1 antagonist, ONO-4057, on MI. Mice with infarcted hearts showed increased BLT1 expression and LTB4 levels. BLT1-knockout mice with infarcted hearts exhibited attenuated leukocyte infiltration, proinflammatory cytokine production, and cell death, which led to reduced mortality and improved cardiac function after MI. Bone-marrow transplantation studies showed that BLT1 expressed on bone marrow-derived cells was responsible for the exacerbation of inflammation in infarcted hearts. Furthermore, ONO-4057 administration attenuated the inflammatory responses in hearts surgically treated for MI, which resulted in reduced mortality and improved cardiac function after MI. Our study demonstrated that BLT1 contributes to excessive inflammation after MI and could represent a new therapeutic target for MI.

Malignant mesothelioma (MM) is one of the most lethal tumors in humans. The onset of MM is linked to exposure to asbestos, which generates reactive oxygen species (ROS). ROS are believed to be derived from the frustrated phagocytosis and the iron in asbestos. To explore the pathogenesis of MM, peritoneal MM was induced in rats by the repeated intraperitoneal injection of iron saccharate and nitrilotriacetate. In the present study, we used microarray techniques to screen the microRNA (miR) expression profiles of these MM. We observed that the histological subtype impacted the hierarchical clustering of miR expression profiles and determined that miR‐199/214 is a distinctive feature of iron saccharate‐induced sarcomatoid mesothelioma (SM). Twist1, a transcriptional regulator of the epithelial‐mesenchymal transition, has been shown to activate miR‐199/214 transcription; thus, the expression level of Twist1 was examined in iron‐induced and asbestos‐induced mesotheliomas in rats. Twist1 was exclusively expressed in iron saccharate‐induced SM but not in the epithelioid subtype. The Twist1‐miR‐199/214 axis is activated in iron saccharate‐induced and asbestos‐induced SM. The expression levels of miR‐214 and Twist1 were correlated in an asbestos‐induced MM cell line, suggesting that the Twist1‐miR‐199/214 axis is preserved. MeT5A, an immortalized human mesothelial cell line, was used for the functional analysis of miR. The overexpression of miR‐199/214 promoted cellular proliferation, mobility and phosphorylation of Akt and ERK in MeT5A cells. These results indicate that miR‐199/214 may affect the aggressive biological behavior of SM., Malignant mesothelioma (MM) is one of the most lethal tumors. Background Intrahepatic artery pseudoaneurysms are mostly iatrogenic and result from hepatobiliary interventions. The incidence of intrahepatic artery pseudoaneurysms within liver tumors without prior intervention is extremely rare. We presented herein the first report of a case of an intratumoral pseudoaneurysm within a liver metastasis of gastric cancer without any prior intervention during chemotherapy. Case presentation A 59-year-old male patient underwent a distal gastrectomy and D2 lymph node dissection for gastric cancer. He was treated in the emergency room for right abdominal pain following the 4th cycle of nivolumab administration as second-line chemotherapy after adjuvant chemotherapy with S-1 and first-line chemotherapy for a liver metastasis of gastric cancer with ramucirumab plus paclitaxel. CT showed a 72-mm metastatic liver tumor containing a 9-mm pseudoaneurysm and fluid collection around the hepatic edge. Intrahepatic artery pseudoaneurysm within the metastatic liver tumor was diagnosed, with the surrounding fluid indicating potential, active bleeding. An emergency angiography confirmed the presence of a pseudoaneurysm in the intrahepatic artery, which was embolized using microcoils. The contributory causes of the intratumoral pseudoaneurysm were assumed to be the following: (1) tumor necrosis leading to encasement, erosion of the vessel wall, and subsequent arterial wall weakening; and (2) inhibition of vascular endothelial growth by ramucirumab resulting in a vessel wall breach and pseudoaneurysm formation. Conclusion It is necessary to recognize that pseudoaneurysms can arise within a metastatic liver tumor during chemotherapy.

Conventional EMS technology cannot stimulate deep muscles to induce muscle contraction using surface electrodes. Several treatments use electrical stimulation for various neurological conditions, including stroke and spinal cord injury. One such treatment is functional electrical stimulation (FES), a form of rehabilitation in which electrical muscle stimulation (EMS) is provided while the muscles are being moved. Here, we show whether two interfering electrical stimulation pulses could stimulate the deep muscles of the forearm to control muscle contraction. The results showed that the strongest torques were generated across the subjects when the reference frequency was mid-frequency (4,000 Hz) and the beat frequencies were low (20 Hz, 40 Hz, 80 Hz, 160 Hz and 320 Hz). This study is the first counterexample to demonstrate that it is possible to control muscle contraction in the deep muscles of the forearm using surface electrodes, which was previously thought to be impossible.

In a previous study, a mushroom was shown to digest milk protein to a mixture of oligopeptides and free amino acids. The aim of this study was to examine effects of this mixture, i.e., mushroom-fermented milk, on blood pressure and stroke susceptibility in the stroke-prone spontaneously hypertensive rats (SHRSP).Rats were fed mushroom-fermented milk with or without 1 % salt water. Blood pressure was monitored either by the tail-cuff method or the telemetry system. Symptoms of stroke were examined every day to determine the stroke latency.Mushroom-fermented milk at 120 mg/Kg BW/day (estimated as a peptides/amino acids content) did not ameliorate hypertension in SHRSP. In contrast, mushroom-fermented milk significantly improved stroke susceptibility under salt-loading. The effects were replicated using milk fermented with three different mushrooms. To elucidate the effective components in mushroom-fermented milk, spermidine (3 mM), one of major components of mushroom-fermented milk, and a mixture of amino acids (0.8 g/L) was examined, both of which showed no significant effects on stroke susceptibility. Intake of mushroom-fermented milk did not affect sodium content significantly either in feces or in urine of the rats given 1% salt water. This observation indicated sodium absorption by the digestive system was not inhibited by intake of mushroom-fermented milk.Despite that the mechanisms were not elucidated, intake of mushroom-fermented milk effectively prevented stroke in SHRSP. Mushroom-fermented milk would be a new candidate for a supplemental nutrient supporting the cardiovascular health.

Oxidative stress has been implicated in the pathophysiology of cerebral stroke. As NADPH oxidases (NOXs) play major roles in the regulation of oxidative stress, we hypothesized that reduction of NOX activity by depletion of p22phox, an essential subunit of NOX complexes, would prevent cerebral stroke. To investigate this, we used the stroke-prone spontaneously hypertensive rat (SHRSP) and the p22phox-deleted congenic SHRSP. Although p22phox depletion reduced blood pressure under salt loading, it did not ameliorate oxidative stress or reduce the incidence of salt-induced stroke in SHRSPs. Additional pharmacological reduction of oxidative stress using antioxidant reagents with different mechanisms of action was necessary to prevent stroke, indicating that NOX was not the major target in salt-induced stroke in SHRSPs. On the other hand, oxidative stress measured based on urinary isoprostane levels showed significant correlations with blood pressure, stroke latency and urinary protein excretion under salt loading, suggesting an important role of oxidative stress per se in hypertension and hypertensive organ damage. Overall, our results imply that oxidative stress from multiple sources influences stroke susceptibility and other hypertensive disorders in salt-loaded SHRSPs.

Oxidative stress is involved in the pathogenesis of hypertension and hypertensive organ damage. Our previous study suggested that stroke-prone spontaneously hypertensive rats (SHRSP) exhibited greater oxidative stress than SHR and that the stroke incidence was significantly greater in SHRSP than SHR. Therefore, we hypothesized that oxidative stress was responsible for the stroke susceptibility in SHRSP. The present study constructed Prdx2 (a gene coding an antioxidative enzyme)-knockout (KO) SHR to examine whether Prdx2 knockout would make SHR more vulnerable to hypertensive organ damage, including stroke. Prdx2-KO SHR were created using CRISPR/CAS9 for genome editing. Eight-week-old male SHR and Prdx2-KO SHR were fed 1% NaCl for 2 months to induce blood pressure (BP) changes and stroke occurrence. The baseline BP was significantly greater in KO SHR, and this difference disappeared after salt loading. The life span of KO SHR was significantly reduced compared to that of SHR despite no differences in BP under salt-loading. However, no stroke was observed in KO SHR. The severity of hypertensive renal and cardiac injuries did not differ significantly between the two strains, but oxidative stress, evaluated using urinary isoprostane excretion and DHE staining, was greater in KO SHR. These results indicated that the Prdx2-depletion caused a shorter life span and modest BP increase in SHR via increased oxidative stress. The pathophysiological roles of oxidative stress in this model should be clarified in future studies.

Recently, ICT equipment has been introduced to classrooms in Japan. Moreover, effectiveness of active learning that engages students as active participants in their learning has been discussed. In order to merge the usefulness of ICT and the effectiveness of active learning, we developed interactive floor interface (IFI) that enables users to see teaching materials displayed on a floor and to control it with their gestures. We also developed an interactive teaching material of wave interference designed for IFI. To confirm the effectiveness of IFI, we gave examinations before and after learning to two groups each of which learns wave interference by using either a usual textbook or IFI. As a result, it was found that average scores of examinations have significantly increased in both groups. Although an average increase of scores for IFI was higher than that of usual textbook, we could not show its significant difference. It is important to improve teaching materials for IFI by giving further interactivity, user-friendliness, and so on.

The blood-brain barrier (BBB) comprises three cell types: brain capillary endothelial cells (BECs), astrocytes, and pericytes. Abnormal interaction among these cells may induce BBB dysfunction and lead to cerebrovascular diseases. The stroke-prone spontaneously hypertensive rat (SHRSP) harbors a defective BBB, so we designed the present study to examine the role of these three cell types in a functional disorder of the BBB in SHRSP in order to elucidate the role of these cells in the BBB more generally. To this end, we employed a unique in vitro model of BBB, in which various combinations of the cells could be tested. The three types of cells were prepared from both SHRSPs and Wistar Kyoto rats (WKYs). They were then co-cultured in various combinations to construct in vitro BBB models. The barrier function of the models was estimated by measuring transendothelial electrical resistance and the permeability of the endothelial monolayer to sodium fluorescein. The in vitro models revealed that (1) BECs from SHRSPs had an inherent lower barrier function, (2) astrocytes of SHRSPs had an impaired ability to induce barrier function in BECs, although (3) both pericytes and astrocytes of SHRSPs and WKYs could potentiate the barrier function of BECs under co-culture conditions. Furthermore, we found that claudin-5 expression was consistently lower in models that used BECs and/or SHRSP astrocytes. These results suggested that defective interaction among BBB cells-especially BECs and astrocytes-was responsible for a functional disorder of the BBB in SHRSPs.

An electric wheelchair controlled through seat pressure mapping was developed to accomplish hands-free operation. The seat pressure mapping resulting from a change in posture was measured using a pressure sensor array seated on the wheelchair in real time. The movements of the upper body were discriminated using template matching. The speed and direction can be controlled based on the similarities between the measured pressure distribution and five templates of neutral, forward, backward, left, and right movements. The developed interface was built into a commercial electric wheelchair. As the results of an experiment show, the proposed wheelchair can be controlled in any direction and velocity.

[CuCl(PPh3)2(4-Mepy)] (PPh3 = triphenylphosphine, 4-Mepy = 4-methylpyridine), a highly blue-luminescent mononuclear copper(I) complex, shows reversible emission colour change ranging from blue-green to red by vapour exposure to N-heteroaromatics, such as pyridine, pyrimidine, and 2-methylpyrazine. The remarkable colour changes occur due the ligand exchange reaction between the solid and gas. Such ligand exchange reactions would be applicable to various vapours with coordination ability and be useful for the sensing of such volatile organic compounds as well as a new convenient methodology for the preparation of luminescent complexes.

The stroke-prone spontaneously hypertensive rat (SHRSP) suffers from severe hypertension and hypertensive organ damage such as cerebral stroke and kidney injury under salt-loading. By a quantitative trait locus (QTL) analysis between SHRSP and SHR (the stroke-resistant parental strain of SHRSP), two major QTLs for stroke susceptibility were identified on chromosomes 1 and 18 of SHRSP, which were confirmed in congenic strains constructed between SHRSP and SHR. As the progression of renal dysfunction was suggested to be one of the key factors inducing stroke in SHRSP, we examined effects of the stroke-related QTLs on kidney injury using two congenic strains harboring either of SHRSP-derived fragments of chromosomes 1 and 18 in the SHR genome. The congenic strains were challenged with 1% NaCl solution for 4 weeks; measurement of systolic blood pressure and urinary isoprostane level (a marker for oxidative stress) and evaluation of renal injury by quantification of genetic marker expression and histological examination were performed. We found that the congenic rats with SHRSP-derived fragment of chromosome 18 showed more severe renal damage with higher expression of Col1α-1 (a genetic marker for renal fibrosis) and higher urinary isoprostane level. In contrast, the fragment of chromosome 1 from SHRSP did not give such effects on SHR. Blood pressure was not greater in either of the congenic strains when compared with SHR. We concluded that the QTL region on chromosome 18 might deteriorate salt-induced renal injury in SHR through a blood pressure-independent mechanism.

Thiolato-bridged dinuclear, tetranuclear, and hexanuclear Cu(I) complexes [Cu2(P∧S)2(PPh3)2], [Cu4(P∧S)4(CH3CN)2], and [Cu6(P∧S)6] (abbreviated as Cu2, Cu4, and Cu6, respectively, P∧S = 2-(diphenylphosphino)benzenethiolate, PPh3 = triphenylphosphine) were synthesized and characterized by elemental analyses and single-crystal X-ray diffraction measurements. These complexes had {Cu2S2}, {Cu4S4}, and {Cu6S6} cluster cores and exhibited strong luminescence at room temperature in solid states. Different luminescence properties were observed depending on the core structure. Cu2, Cu4, and Cu6 exhibited blue-green (λmax = 482 nm, Φem = 0.52), green (λmax = 526 nm, Φem = 0.19), and yellow (λmax = 553 nm, Φem = 0.49) luminescence, respectively, at 298 K in the solid state; among them, only Cu6 showed luminescence thermochromism. We previously revealed that two major quantitative trait loci (QTLs) for stroke latency of the stroke-prone spontaneously hypertensive rat (SHRSP) under salt-loading were located on chromosome (Chr) 1 and 18. Here, we attempted further dissection of the stroke-QTLs using multiple congenic strains between SHRSP and a stroke-resistant hypertensive rat (SHR). Cox hazard model among subcongenic strains harboring a chromosomal fragment of Chr-1 QTL region showed that the most promising region was a 2.1 Mbp fragment between D1Rat177 and D1Rat97. The QTL region on Chr 18 could not be narrowed down by the analysis, which may be due to multiple QTLs in this region. Nonsynonymous sequence variations were found in four genes (Cblc, Cxcl17, Cic, and Ceacam 19) on the 2.1 Mbp fragment of Chr-1 QTL by whole-genome sequence analysis of SHRSP/Izm and SHR/Izm. Significant changes in protein structure were predicted in CBL-C and CXCL17 using I-TASSER. Comprehensive gene expression analysis in the kidney with a cDNA microarray identified three candidate genes (LOC102548695 (Zinc finger protein 45-like, Zfp45L), Ethe1, and Cxcl17). In conclusion, we successfully narrowed down the QTL region on Chr 1, and identified six candidate genes in this region.

A series of mononuclear Cu(I)–halide complexes, [CuX(PPh3)2(L)] (X = Cl−, Br−, I−; PPh3 = triphenylphosphine; L = pyridine (py), isoquinoline (iq), 1,6-naphthyridine (nap)), were synthesized. The emission color of [CuX(PPh3)2(L)] varies from blue to red by changing the L ligands and the halide ions, and all the complexes exhibit high emission quantum yields (0.16–0.99) in the crystals. The emission studies revealed that the emissive states of [CuX(PPh3)2(L)] differ depending on the L ligand. Complexes [CuX(PPh3)2(py)] and [CuX(PPh3)2(nap)] mainly emit from the singlet metal-to-ligand charge transfer mixed with the halide-to-ligand charge transfer (1(M + X)LCT) state at room temperature. In contrast, emissions from [CuX(PPh3)2(iq)] at room temperature originate from both 3(M + X)LCT and 3ππ* states. These results indicate that N-heteroaromatic ligands play an important role in the emission properties of mononuclear Cu(I)–halide complexes.

Objectives: The stroke-prone spontaneously hypertensive rat (SHRSP) is a well-known model for essential hypertension and cerebral stroke. In the previous studies, we have identified a major blood pressure (BP) QTL on chromosome (chr) 1 of SHRSP and have explored gene(s) on the BP QTL responsible for hypertensive phenotypes of this strain through physiological analyses using reciprocal congenic strains between SHRSP and a normotensive control (Wistar-Kyoto rat, WKY). In a recent observation, we unexpectedly found that a SHRSP-based congenic strain, harboring a 0.3-Mbp fragment of the chr1 QTL, has developed stroke earlier than SHRSP. Here, we investigated pathological basis of this congenic strain and attempted to identify gene(s) related to early stroke occurrence. Methods: SHRSP and a SHRSP-based congenic strain SHRSP.WKY-( D1Smu13 )/Izm, abbreviated as SPwch1.71 were used in this study. The rats were housed 12-hour light phase-controlled environment with freely accessible SP diet (Funabashi Farm) and drinking water until at least one suggestive sign of stroke (diminished motor activity, paralytic gait, and so on) was found. MRI by MRmini SA 1508 (DS Pharma Biomedical) was employed to confirm stroke occurrence. BP measurement was performed by the tail-cuff method. Cerebral cortex, brainstem, kidney, adrenal gland and heart obtained from 14 weeks old rats (n=5) were snap-frozen for RNA extraction. Quantitative RT-PCR (qRT-PCR) was performed about 15 of candidate genes located on the chr1 QTL covered by SPwch1.71. Nonsynonymous SNPs were scanned based on the whole-genome sequence of SHRSP and WKY. Results: SPwch1.71 showed significantly high stroke susceptibility compared with SHRSP stroke-free rate until 22 weeks old; SPwch1.71: 0% (0/7), SHRSP: 62.5% (5/8), p p p Pde2a , Inppl1 and Folr1 . Pathological significance of expression/sequence variations in the genes remain unclear.

We have synthesized two luminescent mononuclear Cu(I) complexes, [Cu(PPh2Tol)(THF)(4Mepy)2](BF4) (1) and [Cu(PPh2Tol)(4Mepy)3](BF4) (2) (PPh2Tol = diphenyl(o-tolyl)phosphine, 4Mepy = 4-methylpyridine, THF = tetrahydrofuran), and investigated their crystal structures, luminescence properties, and vapor-induced ligand exchange reactions in the solid state. Both coordination complexes are tetrahedral, but one of the three 4Mepy ligands of complex 2 is replaced by a THF solvent molecule in complex 1. In contrast to the very weak blue emission of the THF-bound complex 1 (wavelength of emission maximum (λem) = 457 nm, emission quantum yield (Φem) = 0.02) in the solid state at room temperature, a very bright blue-green emission was observed for 2 (λem = 484 nm, Φem = 0.63), suggesting a contribution of the THF ligand to nonradiative deactivation. In stroke-prone spontaneously hypertensive rats (SHRSP/Izm), ischemia induces swelling of astrocytes, a process that subsequently leads to neuronal death. Following ischemic insult, arginine vasopressin (AVP) can induce edema and l-serine released by astrocytes supports the survival of neuronal cells. The purpose of this study was to examine whether AVP contributed to the regulation of l-serine production following ischemic stroke. Here, we used cultured astrocytes from SHRSP/Izm rats and Wistar Kyoto rats (WKY/Izm) to examine whether AVP changed the production of l-serine and/or altered gene expression levels of the neural amino acid transporter (Slc1a4), 3-phosphoglycerate dehydrogenase (Phgdh) and serine racemase (SRR). Furthermore, using astrocytes from the congenic rat SHRpch1_18 strain having quantitative trait loci (QTL) of stroke, we examined expression of those genes under conditions of hypoxia and reoxygenation (H/R). The expression levels of ASCT1 protein, the genes described above and l-serine levels were determined by Western blotting (WB), RT-PCR, real-time quantitative RT-PCR and HPLC. AVP increased the production of l-serine and the expression of Slc1a4 in WKY/Izm and SHRSP/Izm astrocytes. The production of l-serine and the expression of Slc1a4 were lower in SHRSP/Izm than in WKY/Izm cells. This difference was not seen with Phgdh. In the SHRpch1_18 strain, the expression of Slc1a4 and Phgdh significantly decreased after H/R. AVP-mediated enhanced expression of ASCT1 was blocked by the addition of bumetanide. These results suggest that the AVP-mediated attenuated expression of ASCT1 in astrocytes is associated with reduced l-serine production in SHRSP/Izm astrocytes. We hypothesize that reduction of gene expression by AVP might be related to the induction of stroke in the SHRpch1_18 rat strain.

Simple mononuclear Cu(I)-halide complexes, [CuX(PPh3)2(4-Mepy)] (X = Cl(-), Br(-), I(-); PPh3 = triphenylphosphine; 4-Mepy = 4-methylpyridine), were prepared. They exhibit blue light emission, with extremely high photoluminescence quantum yields approaching 100% in the crystals. Emission lifetime analyses and density functional theory calculations revealed that the bright blue light emission at room temperature is mainly delayed fluorescence originating from the singlet metal-to-ligand charge transfer (MLCT) state combined with the halide-to-ligand charge transfer (XLCT) state, ((1)(M + X)LCT), while that at 77 K is phosphorescence from the (3)(M + X)LCT transition state, due to the small singlet-triplet energy differences (ΔE = 940-1170 cm(-1)). The ternary ligand systems consisting of halide, bulky phosphine, and N-heteroaromatic ligands constitute inexpensive pure-blue-light-emitting materials, which can be fabricated by facile procedures such as simple manual grinding.

Myocardial infarction (MI) results in the generation of dead cells in the infarcted area. These cells are swiftly removed by phagocytes to minimize inflammation and limit expansion of the damaged area. However, the types of cells and molecules responsible for the engulfment of dead cells in the infarcted area remain largely unknown. In this study, we demonstrated that cardiac myofibroblasts, which execute tissue fibrosis by producing extracellular matrix proteins, efficiently engulf dead cells. Furthermore, we identified a population of cardiac myofibroblasts that appears in the heart after MI in humans and mice. We found that these cardiac myofibroblasts secrete milk fat globule-epidermal growth factor 8 (MFG-E8), which promotes apoptotic engulfment, and determined that serum response factor is important for MFG-E8 production in myofibroblasts. Following MFG-E8-mediated engulfment of apoptotic cells, myofibroblasts acquired antiinflammatory properties. MFG-E8 deficiency in mice led to the accumulation of unengulfed dead cells after MI, resulting in exacerbated inflammatory responses and a substantial decrease in survival. Moreover, MFG-E8 administration into infarcted hearts restored cardiac function and morphology. MFG-E8-producing myofibroblasts mainly originated from resident cardiac fibroblasts and cells that underwent endothelial-mesenchymal transition in the heart. Together, our results reveal previously unrecognized roles of myofibroblasts in regulating apoptotic engulfment and a fundamental importance of these cells in recovery from MI.

Oxidative stress in the rostral ventrolateral medulla (RVLM), a sympathetic center in the brainstem, was implicated in the regulation of sympathetic activity in various hypertensive models including stroke-prone spontaneously hypertensive rats (SHRSP). In this study, we evaluated the role of the NADPH oxidases (NOX) in the blood pressure (BP) regulation in RVLM in SHRSP. The P22PHOX-depleted congenic SHRSP (called SP.MES) was constructed by introducing the mutated p22phox gene of Matsumoto Eosinophilic Shinshu rat. BP response to glutamate (Glu) microinjection into RVLM was compared among SHRSP, SP.MES, SHR and Wistar Kyoto (WKY); the response to Glu microinjection was significantly greater in SHRSP than in SP.MES, SHR and WKY. In addition, tempol, losartan and apocynin microinjection reduced the response to Glu significantly only in SHRSP. The level of oxidative stress, measured in the brainstem using lucigenin and dihydroethidium, was reduced in SP.MES than in SHRSP. BP response to cold stress measured by telemetry system was also blunted in SP.MES when compared with SHRSP. The results suggested that oxidative stress due to the NOX activation in RVLM potentiated BP response to Glu in SHRSP, which might contribute to the exaggerated response to stress in this strain.

Halide-bridged rhombic dicopper(I) complexes, [Cu2(μ-X)2(DMSO)2(PPh3)2] (X = I(-), Br(-); DMSO = dimethyl sulfoxide; PPh3 = triphenylphosphine), were synthesized, the iodide complex of which exhibited interesting photochromic luminescence driven by photoirradiation and by exposure to DMSO vapor in the solid state. Single-crystal X-ray diffraction measurements revealed that the iodo and bromo complexes (abbreviated Cu2I2-[O,O] and Cu2Br2-[O,O]) were isomorphous, and that the two DMSO ligands were coordinated to the Cu(I) ion via the O atom in both complexes. Both complexes exhibited bright blue phosphorescence at room temperature (λ(em) = 435 nm, Φ(em) = 0.19 and 0.14 for Cu2I2-[O,O] and Cu2Br2-[O,O], respectively) with a relatively long emission lifetime (τ(em) ~ 200 μs at 77 K) derived from the mixed halide-to-ligand and metal-to-ligand charge transfer ((3)XLCT and (3)MLCT) excited state. Under UV irradiation, the blue phosphorescence of Cu2Br2-[O,O] disappeared uneventfully and no new emission band appeared, whereas the blue phosphorescence of Cu2I2-[O,O] rapidly disappeared with simultaneous appearance of a new green emission band (λ(em) = 500 nm). On further irradiation, the green emission of the iodide complex gradually changed to bright yellowish-green (λ(em) = 540 nm); however, this change could be completely suppressed by lowering the temperature to 263 K or in the presence of saturated DMSO vapor. The initial blue phosphorescence of Cu2I2-[O,O] was recovered by exposure to DMSO vapor at 90 °C for a few hours. IR spectroscopy and theoretical calculations suggest that the DMSO ligand underwent linkage isomerization from O-coordination to S-coordination, and both the occurrence of linkage isomerization and the removal of DMSO result in contraction of the rhombic Cu2(μ-I)2 core to make the Cu···Cu interaction more effective. In the contracted core, the triplet cluster-centered ((3)CC) emissive state is easily generated by thermal excitation of the (3)XLCT and (3)MLCT mixed transition state, resulting in the green to yellowish-green emission. In contrast, the Cu···Cu distance in Cu2Br2-[O,O] is considerably longer than that of Cu2I2-[O,O], which destabilizes the (3)CC emissive state, resulting in the nonemissive character.

SummaryBackground Ultraviolet B (UVB) irradiation induces serious damage to the skin. Collagen hydrolysate and collagen-derived peptides have effects on skin function in vivo and in vitro. However, few studies have investigated changes in the epidermal barrier or dermal elasticity caused by UVB. Here, we investigated the loss of epidermal barrier function and skin elasticity induced by UVB irradiation in hairless mice fed collagen hydrolysate. Methods Mice were orally administered collagen hydrolysate, in a single dose (20 mJ/cm2) or repeated doses (10–30 mJ/cm2, 3 times/week for 6 weeks), and the dorsal skin was exposed to UVB. Skin measurements and histological and analytical studies were performed. Results In control mice, a single UVB irradiation induced epidermal barrier dysfunction including an increase in transepidermal water loss (TEWL), epidermal hyperplasia, and a decrease in stratum corneum water content. Administration of collagen hydrolysate significantly decreased TEWL and epidermal thickness and increased stratum corneum water content. Repeated UVB irradiation decreased skin elasticity and dermal hyaluronic acid (HA) content in control mice, whereas collagen hydrolysate significantly suppressed both the increase in TEWL and the decrease in stratum corneum water content and improved skin elasticity and dermal HA content. Conclusions Collagen hydrolysate administration affects epidermal barrier function and dermal skin elasticity.

According to the report of FAO（Food and Agriculture Organization of United Nations), two-thirds of world population will face shortage of fresh water in 2025, and it is required to save the usage of fresh water for any industrial department. Reflecting this social problem, in order to save fresh water knowledge about the utilization of concrete mixed with seawater is required for concrete industry. In this thesis, the durability of concrete mixed with seawater and reinforced with steel bar against carbonation was evaluated by carbonation rate coefficient, distribution of Cl and corrosion behavior of steel bar using electrochemical measurement and visual observation methods. In the results, below results were revealed. When to use seawater as mixing water of concrete, carbonation rate of concrete mixed with seawater was almost the same as carbonation rate of concrete mixed with fresh water, concentration of internal chloride atom into the front part of carbonation depth occurred due to carbonation, and steel bar corrodes when carbonation depth reaches the covering depth of steel bar. According to these results, at the case of using seawater as mixing water of concrete the possibility of saving corrosion of steel bar embedding steel bar in the deep area of concrete was suggested.国連食糧農業機関の報告によると2025年に世界人口の約2/3が淡水不足の危機に陥るとされ、淡水の節制が求められており、建設分野においても関連する検討が必要である。本研究では、上記の一環として、練混ぜ水として海水を用いたコンクリート（海水練りコンクリート）の中性化に対する耐久性を、中性化速度係数、塩素元素分布、目視観察と電気化学的計測による鉄筋の腐食挙動の観点から評価した。その結果、海水練りの場合、①淡水練りと同程度の中性化速度となること、②中性化による塩化物イオンの濃縮が起きること、③中性化領域が鉄筋位置に到達した時点で鉄筋腐食が生じることが確認された。

We have newly synthesized two solution-stable luminescent dinuclear copper(I) complexes, [Cu2(μ-I)2(dpppy)2] (Cu-py) and [Cu2(μ-I)2(dpppyz)2] (Cu-pyz), where dpppy = 2,3-bis(diphenylphosphino)pyridine and dpppyz = 2,3-bis(diphenylphosphino)pyrazine, using chelating diphosphine ligands composed of N-heteroaromatic rings. X-ray analysis clearly indicates that the molecular structures of Cu-py and Cu-pyz are almost identical with that of the parent complex, [Cu2(μ-I)2(dppb)2] [Cu-bz; dppb = 2,3-bis(diphenylphosphino)benzene]. Complexes Cu-py and Cu-pyz exhibit luminescence [emission quantum yield (Φem) = 0.48 and 0.02, respectively] in the solid state at 298 K. A wide emission color tuning, from 497 to 638 nm (energy = 0.55 eV, with an emission color ranging from green to reddish-orange), was achieved in the solid state by the introduction of pyridinic N atoms into the bridging phenyl group between the two diphenylphosphine groups. Density functional theory calculations suggest that the emission could originate from the effective combination of the metal-to-ligand charge-transfer excited state with the halide-to-ligand charge-transfer excited state. Thus, the emission color change is due to stabilization of the π* levels of the central aryl group in the diphosphine ligand. Furthermore, these copper(I) complexes exhibit thermally activated delayed fluorescence at 298 K because of the small singlet-triplet energy difference (ΔE = 523 and 564 cm(-1) for Cu-py and Cu-pyz, respectively). The stability of these complexes in chloroform, due to the rigid bonds between the diphosphine ligands and the Cu(I) ions, enables the preparation of emissive poly(methyl methacrylate) films by the solution-doping technique.

Exposure to asbestos is a risk for malignant mesothelioma (MM) in humans. Among the commercially used types of asbestos (chrysotile, crocidolite, and amosite), the carcinogenicity of chrysotile is not fully appreciated. Here, we show that all three asbestos types similarly induced MM in the rat peritoneal cavity and that chrysotile caused the earliest mesothelioma development with a high fraction of sarcomatoid histology. The pathogenesis of chrysotile-induced mesothelial carcinogenesis was closely associated with iron overload: repeated administration of an iron chelator, nitrilotriacetic acid, which promotes the Fenton reaction, significantly reduced the period required for carcinogenesis; massive iron deposition was found in the peritoneal organs with high serum ferritin; and homozygous deletion of the CDKN2A/2B/ARF tumour suppressor genes, the most frequent genomic alteration in human MM and in iron-induced rodent carcinogenesis, was observed in 92.6% of the cases studied with array-based comparative genomic hybridization. The induced rat MM cells revealed high expression of mesoderm-specific transcription factors, Dlx5 and Hand1, and showed an iron regulatory profile of active iron uptake and utilization. These data indicate that chrysotile is a strong carcinogen when exposed to mesothelia, acting through the induction of local iron overload. Therefore, an intervention to remove local excess iron might be a strategy to prevent MM after asbestos exposure. Copyright © 2012 Pathological Society of Great Britain and Ireland. The stroke-prone spontaneously hypertensive rat (SHRSP) is a unique genetic model of severe hypertension and cerebral stroke. SHRSP, as well as the spontaneously hypertensive rat, the parental strain of SHRSP, has made a tremendous contribution to cardiovascular research. However, the genetic mechanisms underlying hypertension and stroke in these rats have not yet been clarified. Recent studies using whole-genome sequencing and comprehensive gene expression analyses combined with classical quantitative trait loci analyses provided several candidate genes, such as Ephx2, Gstm1 and Slc34a1, which still need further evidence to define their pathological roles. Currently, genome-wide association studies can directly identify candidate genes for hypertension in the human genome. Thus, genetic studies in SHRSP and other rat models must be focused on the pathogenetic roles of 'networks of interacting genes' in hypertension, instead of searching for individual candidate genes.
