Abstract

The 46th Japanese Antarctic Research Expedition (JARE-46) conducted field operations on geological and paleomagnatic studies in the regions of Lutzow-Holm Bay, Prince Olav Coast and western Enderby Land during December 17, 2004 through February 17, 2005, under a part of the SEAL II (Structure and Evolution of East Antarctic Lithosphere II) project. A small helicopter was used to support field parties during the summer operations conducted by JARE-46. This report summarizes preparations for field operations and activities of the geological and paleo-magnetic team, and points out some problems for the future planning.

Abstract

Cloning of murine glycosyl phosphatidylinositol anchor attachment protein, GPAA1. Am J Physiol Cell Physiol 279: C205-C212, 2000.--Glycosyl phosphatidylinositols (GPIs) are used to anchor many proteins to the cell surface membrane and are utilized in all eukaryotic cells. GPI anchoring units are attached to proteins via a transamidase reaction mediated by a GPI transamidase complex. We isolated one of the components of this complex, mGPAA1 (murine GPI anchor attachment), by the signal sequence trap method. mGPAA1 cDNA is about 2 kb in length and encodes a putative 621 amino acid protein. The mGPAA1 gene has 12 small exons and 11 small introns, mGPAA1 mRNA is ubiquitously expressed in mammalian cells, and in situ hybridization analysis revealed that it is abundant in the choroid plexus, skeletal muscle, osteoblasts of rib, and occipital bone in mouse embryos. Its expression levels and transamidation efficiency decreased with differentiation of embryonic stem cells. 24. Early Palaeozoic orogenic collapse and voluminous late-tectonic magmatism in Dronning Maud Land and Mozambique : insights into the partially delaminated orogenic root of the East African-Antarctic Orogen?

Abstract

The late tectonic history of the southern part of the Late Neoproterozoic–Early Palaeozoic East African–Antarctic Orogen (EAAO) is characterized by lateral extrusion, extensional collapse and large volumes of high-temperature A2-type granitoids. This late-tectonic igneous province covers an area more than 15 000 km2 of the EAAO in Dronning Maud Land (East Antarctica) and its northerly continuation as the Nampula Complex of NE Mozambique. The magmatic province is bounded in the north by the Lurio Belt. New secondary ionization mass spectrometry (SIMS) U–Pb analyses of zircons from two major late-tectonic granitoid intrusions from Dronning Maud Land indicate crystallization ages of 501+7 and 499+4 Ma, whereas a major extensional shear zone was dated at 507+9 Ma. New SIMS zircon U–Pb analyses of late-tectonic granitoid sheets and plutons from the Nampula Province indicate ages of 512+4, 508+4, 508+2 and 507+3 Ma. Consequently, the late-tectonic magmatism can be bracketed between c. 530 and 485 Ma. It started with small gabbro bodies emplaced at c. 530–520 Ma, culminated with the intrusion of major granite–charnockite plutons at c. 510–500 Ma and terminated with the introduction of small volumes of sheet-like granite at c. 485 Ma. The new dates demonstrate that extensional shearing and granitoid intrusion are synchronous, and that orogenic collapse and the magmatism are related. We ascribe the distribution, structural style, geochemical composition and age of the late magmatic province to a process of partial delamination of the orogenic root in the southern third of the EAAO. It remains to be tested whether there is a relationship between orogenic collapse–granitoid magmatism and south-directed escape tectonics in the southernmost EAAO.

48. Context-dependent transcriptional cooperation mediated by cardiac transcription factors Csx/Nkx-2.5 and GATA-4.

Abstract

Although the cardiac homeobox gene Csx/Nkx-2.5 is essential for normal heart development, little is known about its regulatory mechanisms. In a search for the downstream target genes of Csx/Nkx-2. 5, we found that the atrial natriuretic peptide (ANP) gene promoter was strongly transactivated by Csx/Nkx-2.5. Deletion and mutational analyses of the ANP promoter revealed that the Csx/Nkx-2.5-binding element (NKE2) located at -240 was required for high level transactivation by Csx/Nkx-2.5. We also found that Csx/Nkx-2.5 and GATA-4 displayed synergistic transcriptional activation of the ANP promoter, and in contrast to previous reports (Durocher, D., Charron, F., Warren, R., Schwartz, R. J., and Nemer, M. (1997) EMBO J. 16, 5687-5696; Lee, Y., Shioi, T., Kasahara, H., Jobe, S. M., Wiese, R. J., Markham, B., and Izumo, S (1998) Mol. Cell. Biol. 18, 3120-3129), this synergism was dependent on binding of Csx/Nkx-2.5 to NKE2, but not on GATA-4-DNA interactions. Although GATA-4 also potentiated the Csx/Nkx-2.5-induced transactivation of the artificial promoter that contains multimerized Csx/Nkx-2.5-binding sites, Csx/Nkx-2.5 reduced the GATA-4-induced transactivation of the GATA-4-dependent promoters. These findings indicate that the cooperative transcriptional regulation mediated by Csx/Nkx-2.5 and GATA-4 is promoter context-dependent and suggest that the complex cis-trans interactions may fine-tune gene expression in cardiac myocytes.

Abstract

We have recently shown that mechanical stress induces cardiomyocyte hypertrophy partly through the enhanced secretion of angiotensin II (ATII). Endothelin-1 (ET-1) has been reported to be a potent growth factor for a variety of cells, including cardiomyocytes. In this study, we examined the role of ET-1 in mechanical stress-induced cardiac hypertrophy by using cultured cardiomyocytes of neonatal rats. ET-1 (10(-8) approximately 10(-7) M) maximally induced the activation of both Raf-1 kinase and mitogen-activated protein (MAP) kinases at 4 and 8 min, respectively, followed by an increase in protein synthesis at 24 h. All of these hypertrophic responses were completely blocked by pretreatment with BQ123, an antagonist selective for the ET-1 type A receptor subtype, but not by BQ788, an ET-1 type B receptor-specific antagonist. BQ123 also suppressed stretch-induced activation of MAP kinases and an increase in phenylalanine uptake by approximately 60 and 50%, respectively, but BQ788 did not. ET-1 was constitutively secreted from cultured cardiomyocytes, and a significant increase in ET-1 concentration was observed in the culture medium of cardiomyocytes after stretching for 10 min. After 24 h, an approximately 3-fold increase in ET-1 concentration was observed in the conditioned medium of stretched cardiomyocytes compared with that of unstretched cardiomyocytes. ET-1 mRNA levels were also increased at 30 min after stretching. Moreover, ET-1 and ATII synergistically activated Raf-1 kinase and MAP kinases in cultured cardiomyocytes. In conclusion, mechanical stretching stimulates secretion and production of ET-1 in cultured cardiomyocytes, and vasoconstrictive peptides such as ATII and ET-1 may play an important role in mechanical stress-induced cardiac hypertrophy.
