15. Molecular phylogeny of two coelomycetous fungal genera with stellate conidia, Prosthemium and Asterosporium, on Fagales trees

Abstract

Prosthemium (teleomorph Pleomassaria) and Asterosporium (teleomorph unknown) are coelomycetous genera with stellate conidia on Fagales trees. Their morphological resemblance suggests their close relationship, but phylogenetic relatedness remains unknown. They have been distinguished on the basis of either conidiomatal morphology (pycnidia in Prosthemium and acervuli in Asterosporium) or their differing conidial septation (euseptate in Prosthemium and distoseptate in Asterosporium). To reveal their phylogenetic affinities and clarify reliable distinguishing phenotypical characters, five species of Prosthemium and two species of Asterosporium were investigated using sequences of the small subunit, large subunit, and internal transcribed spacer region of nuclear ribosomal DNA and b-tubulin gene from 43 isolates of these species. The analyses revealed the following: (i) Asterosporium typified by Asterosporium asterospermum on Fagus is a member of the Sordariomycetes and is distinct from Prosthemium belonging to the Dothideomycetes; (ii) Asterosporium betulinum on Betula species should be excluded from Asterosporium and transferred to Prosthemium as Prosthemium neobetulinum nom.nov.; (iii) conidial septation does not seem to have a diagnostic value, whereas conidiomatal morphology is useful in distinguishing both genera; (iv) the number and length of conidial arms are useful criteria in distinguishing Prosthemium species. A new species, Prosthemium intermedium sp.nov., found on both Betula ermanii and Betula maximowicziana, is described and illustrated. Les Prosthemium (teleeomorphe Pleomassaria)et Asterosporium (teleomorphe inconnu) constituent des genres de Coelomycetes possedant des conidies etoileees et venant sur des arbres appartenant aux Fagales. Leur ressemblance morphologique suggere une etroite relation, mais le lien phylogeneetique demeure inconnu. On les a distingues soit sur la base de la morphologie des conidiomata (pycnidies chez les Prosthemium et acervules chez les Asterosporium), ou sur celle des differences de leurs septations (eusepteee chez les Prosthemium et distosepteee chez les Asterosporium). Afin de reveler leurs affinites phylogenetiques et d'etablir des caracteres phenotypiques distinctifs, les auteurs ont examine cinq especes de Prosthemium et deux especes d'Asterosporium, en utilisant les sequences petite sous-unite, grande sous-unite, l'espaceur interne transcrit du l'ADN ribosomal, et le gene de la b-tubuline, a partir de 43 isolats de ces especes. Abstract

A new species, Massariosphaeria websteri, on submerged decaying culms of a grass (possibly Phragmites karka) in freshwater in Pakistan is described, illustrated and compared with closely related taxa in the genus Massariosphaeria. It is characterized by immersed, scattered, subglobose to conical, ostiolate ascomata each with a papillate beak; 20–32 µm thick, 5–7 layers, polygonal to rectangular peridial cells; branched and anastomosed pseudoparaphyses; relatively large (170–245 × 26.5–35 µm), fissitunicate, cylindrical asci; and ascospores large-sized (av. 52.2 × 13.5 µm), narrowly fusiform to clavate, (7–) 8–11-septate, surrounded by a gelatinous sheath. Additionally, six other members of the Pleosporales, Massariosphaeria typhicola, Lophiostoma caulium, Lophiostoma compressum, Lophiostoma quadrinucleatum, Nodulosphaeria aquilana, and Trichometasphaeria culmifida are reported.

26. Freshwater Ascomycetes: Minutisphaera(Dothideomycetes) revisited, including one new species from Japan

Abstract

During investigations of freshwater ascomycetes we found one interesting taxon from Aomori (Japan), as well as three additional taxa from North Carolina (USA), which were morphologically similar to Minutisphaera, a recently described freshwater fungus in the Dothideomycetes. The ascomata of all the collections bore dark hair-like structures around the ostiolar region, obovoid to obclavate bitunicate asci, and one to three septate hyaline to brown ascospores with a sheath (in material from Japan), and with both sheath and appendages (in material from the USA). The apothecial ascomata of these taxa, however, differ from those of the type species of the genus, which are perithecial. Two collections of Minutisphaera-like fungi from the USA were morphologically quite similar but differed in ascospore size. To assess the phylogenetic affinities of Minutisphaera-like taxa with the type species, M. fimbriatispora, we sequenced 18S and 28S nrDNA of five newly collected strains of Minutisphaera. We also sequenced the nrDNA for the entire internal transcribed spacer region of 10 strains to assess interspecific and intraspecific variation with M. fimbriatispora. Additionally we examined the secondary metabolite profiles of two strains from USA. Based on maximum likelihood and Bayesian analyses of combined 18S and 28S, and separate ITS sequences, as well as examination of morphology, we describe and illustrate a new species, M. japonica. One collection from North Carolina is confirmed as M. fimbriatispora, while two other collections are Minutisphaera-like fungi that had a number of similar diagnostic morphological characters but differed only slightly in ascospore sizes. The phylogeny inferred from the internal transcribed spacer region suggested that two out of the three North Carolina collections may be novel and perhaps cryptic species within Minutisphaera. Organic extracts of Minutisphaerafrom USA, M. fimbriatispora(G155-1) and Minutisphaera-like taxon (G156-1), revealed the presence of palmitic acid and (E)-hexadec-9-en-1-ol as major chemical constituents. We discuss the placement of the Minutisphaeraclade within the Dothideomycetes. The description of the genus Minutisphaerais emended to accommodate M. japonicawithin Minutisphaera.

27. Freshwater ascomycetes: two new species of Lindgomyces(Lindgomycetaceae, Pleosporales, Dothideomycetes) from Japan and USA

Abstract

During independent surveys of freshwater ascomycetes in Japan and USA two new species of Lindgomyceswere collected from submerged wood in freshwater. These species are described and illustrated based on morphological data and phylogenetic relationships based on analyses of nuclear ribosomal sequence data (partial SSU and LSU, and ITS). Lindgomyces apiculatus, collected in Japan, is characterized by immersed to erumpent, globose to subglobose ascomata; fissitunicate, cylindrical to clavate asci; and fusiform, one-septate ascospores with acute ends and short terminal appendages. Lindgomyces lemonweirensis, collected in Wisconsin, USA, differs from L. apiculatusin having clavate to cymbiform asci and oblong to fusiform ascospores that are distinctively multiguttulate and surrounded by an oval, ephemeral gelatinous sheath. The new species formed a strongly supported clade within the family Lindgomycetaceae (Pleosporales, Dothideomycetes) based on analyses of combined SSU and LSU sequence data. In addition phylogenetic analyses with ITS sequence data support the establishment of the new taxa as separate species within Lindgomycesbecause they were separated from each other and other Lindgomycesspecies based on maximum likelihood bootstrap and Bayesian analyses.

28. Micellar‐Promoted Stereospecific Hydrolysis of Amino Acid Esters Catalyzed by Peptide‐Like Nucleophiles

Abstract

The efficiency of peptide‐type nucleophiles (Z‐L‐Leu‐L‐His (2a), Z‐D‐Leu‐L‐His (2b, Z‐L‐His‐L‐Leu (2c), Z‐L‐Ala‐L‐His‐OMe (2d), Z‐L‐Leu‐L‐His‐OMe (2e), Z‐L‐Phe‐L‐His‐OMe (2f), Z‐L‐Leu‐L‐His‐L‐Leu (3a), Z‐D‐Leu‐L‐His‐L‐Leu (3b), and Z‐L‐Leu‐L‐Leu‐L‐His (3c)) in the stereospecific hydrolysis of p‐nitrophenyl N‐acyl‐phenylalanates (4a‐b) was examined at 10–30°C (pH 7.68) in an solution of hexadecyltrimethylammonium bromide (5). The hydrophobic and chiral amino acid group which is adjacent to the nucleophilic L‐histidine part in 2a‐f (or 3a‐c) was found to play an important role in determining the closeness of approach between the reactive nucelophile and substrate. The nucleophiles 2aand 3agave high reactivity and stereospecificity in the hydrolysis of 4bpossessing a long N‐acyl chain, and the maximum enantiomer rate ratio (L/D), 16.1 (25°C) or 26.2 (10°C), was obtained with mixed micelles of 3a and 5. The structural effect of the nucleophiles on the stereospecific esterolysis of 4a‐b is also discussed in detail on the basis of kinetic parameters (Kb/N, km, ΔH‡, and ΔS‡).
