17. Heterotypic contact inhibition of locomotion can drive cell sorting between epithelial and mesenchymal cell populations

Abstract

Interactions between different cell types can induce distinct contact inhibition of locomotion (CIL) responses that are hypothesised to control population-wide behaviours during embryogenesis. However, our understanding of the signals that lead to cell-type specific repulsion and the precise capacity of heterotypic CIL responses to drive emergent behaviours is lacking. Using a new model of heterotypic CIL, we show that fibrosarcoma cells, but not fibroblasts, are actively repelled by epithelial cells in culture. We show that knocking down EphB2 or ERK in fibrosarcoma cells specifically leads to disruption of the repulsion phase of CIL in response to interactions with epithelial cells. We also examine the population-wide effects when these various cell combinations are allowed to interact in culture. Unlike fibroblasts, fibrosarcoma cells completely segregate from epithelial cells and inhibiting their distinct CIL response by knocking down EphB2 or ERK family proteins also disrupts this emergent sorting behaviour. These data suggest that heterotypic CIL responses, in conjunction with processes such as differential adhesion, may aid the sorting of cell populations.

Abstract

We present the performance of a feedforward noise canceller (FNC) for the PR1 [(1+D)] channel combined with an 8/10 DC free code. It appears the PR1 channel can he superior to PR4 [(1-D)(1+D)] and EPR4 [(1-D)[(1+D).sup.2]] for high density magnetic recording due to a characteristic of high frequency suppression. Simulation results show that PR1 with FNC is better than EPR4 for K[greater than or equal to] 2.5 [K=[PW.sub.50/T]. We adapted PR1 with FNC into an actual DDS3 (Digital Data Storage 3) system and we got an improved result. Applying FNC to DDS3 system can improve the error rate by around [10.sup.2].

Abstract

The present work has been undertaken to improve the thermal stability of an RhPt-NaTi/Al2O3 catalyst, which is one of promising lean NOx trap catalysts for practical NOx conversion. For this purpose, we mainly directed our attention to the component of Na that acts as NOx trapping sites and intended to improve the thermal stability of this Na species by using various additives. After screening of various RhPt-Naα/Al2O3 (α = Ti, Si, Mg, Ca, Mn, Co, Ni, La, and Nd) catalysts, Mn addition was found to be effective for improving thermal stability of the catalyst. Moreover, the modification of Al2O3 support with Ce additive was indicated to be effective, and the most active and thermally stable catalyst was RhPt-NaMn-Ce/Al2O3, which was able to function actively even after a severe heat treatment at 800℃. The reasons for the high thermal stability of this catalyst were examined by evaluating NO oxidation activity of a model catalyst including Mn and characterizing features of the RhPt-NaMn-Ce/Al2O3 catalyst with FTIR of adsorbed NO and chemisorption of CO and CO2. An important factor is that the addition of Mn prevents the Na species from undesired sintering during heat treatments, resulting in a larger surface area of exposed Na species, namely NOx trapping sites. This catalyst can also function in a commercial vehicle engine under lean-burn conditions and should be a promising catalyst for practical use.

Abstract

Magnetic tunnel junctions (MTJs) of perpendicularly magnetized L10-CoPt/Co2MnSi/MgO/FePt structure were fabricated. In-plane x-ray diffraction measurements and element specific evaluation of magnetic properties using soft x-ray magnetic circular dichroism were carried out to examine quite thin Co2MnSi (CMS) inserted layer between CoPt and MgO interface. Ordered B2-CMS was successfully fabricated onto L10-CoPt as thin as 1 nm thick, and CMS layer shows perpendicular magnetic anisotropy below 3 nm thick via exchange coupling with CoPt layer. In the MTJ-stacking, epitaxial growth was confirmed except for partial misalignment in the upper FePt layer, and coercive field difference clearly appeared between the bottom and the top ferromagnetic electrodes. 35. Hotspots of MLV integration in the hematopoietic tumor genome

Abstract

Extensive research has been performed regarding the integration sites of murine leukemia retrovirus (MLV) for the identification of proto-oncogenes. To date, the overlap of mutations within specific oligonucleotides across different tumor genomes has been regarded as a rare event; however, a recent study of MLV integration into the oncogene Zfp521 suggested the existence of a hotspot oligonucleotide for MLV integration. In the current review, we discuss the hotspots of MLV integration into several genes: c-Myc, Stat5a and N-myc, as well as ZFP521, as examined in tumor genomes. From this, MLV integration convergence within specific oligonucleotides is not necessarily a rare event. This short review aims to promote re-consideration of MLV integration within the tumor genome, which involves both well-known and potentially newly identified and novel mechanisms and specifications.

49. ZFP521contributes to pre-B-cell lymphomagenesis through modulation of the pre-B-cell receptor signaling pathway

Abstract

ZFP521was previously identified as a putative gene involved in induction of B-cell lymphomagenesis. However, the contribution of ZFP521to lymphomagenesis has not been confirmed. In this study, we sought to elucidate the role of ZFP521in B-cell lymphomagenesis. To this end, we used a retroviral insertion method to show that ZFP521was a target of mutagenesis in pre-B-lymphoblastic lymphoma cells. The pre-B-cell receptor (pre-BCR) signaling molecules BLNK, BTKand BANK1were positively regulated by the ZFP521gene, leading to enhancement of the pre-BCR signaling pathway. In addition, c-mycand c-junwere upregulated following activation of ZFP521. Stimulation of pre-BCR signaling using anti-Vpreb antibodies caused aberrant upregulation of c-mycand c-junand of Ccnd3, which encodes cyclin D3, thereby inducing the growth of pre-B cells. Stimulation with Vpreb affected the growth of pre-B cells, and addition of interleukin (IL)-7 receptor exerted competitive effects on pre-B-cell growth. Knockdown of BTKand BANK1, targets of ZFP521, suppressed the effects of Vpreb stimulation on cell growth. Furthermore, in human lymphoblastic lymphoma, analogous to pre-B-cell lymphoma in mice, the expression of ZNF521, the homolog of ZFP521in humans, was upregulated. In conclusion, our data showed that the ZFP521gene comprehensively induced pre-B-cell lymphomagenesis by modulating the pre-B-cell receptor signaling pathway.
