Abstract

To examine the possible involvement of gelatinase B in human hepatocellular carcinoma (HCC), cellular localization of transcripts and protein of gelatinase B were studied by using in situ hybridization and immunohistochemistry. Transcripts for gelatinase B were observed in tumor cells in 22 cases of 27 HCCs and also in dysplastic nodules. However, there was no significant difference in the expression among histological grades of HCC. The expression was mostly homogeneous, but the intensity varied with the nodules. Of 13 cases with capsular invasion, 12 expressed gelatinase B, whereas 10 of 14 without capsular invasion did (P < 0.05). Gelatinase B transcripts were commonly observed in the sinusoidal cells of the hepatic lobules, in mesenchymal cells both in fibrous capsules and around the necrosis, and also in some undefined cells of the portal tracts of noncancerous liver. Localization of gelatinase B protein was mostly similar to but sometimes different from that of the transcripts in cancer nodules. In conclusion, the expression of gelatinase B appears to be an important characteristic of malignant transformation of hepatocytes. The findings suggest that gelatinase B synthesized by cancer cells plays an important role in the growth and invasion of HCC by degrading surrounding extracellular matrices.

30. Elevated urokinase-type plasminogen activator plasma levels are associated with deterioration of liver function but not with hepatocellular carcinoma.

Abstract

We measured urokinase-type plasminogen activator (u-PA) plasma levels in patients with various chronic liver diseases, including hepatocellular carcinoma (HCC), also measuring these levels in healthy volunteers. Plasma u-PA levels in the group of patients with decompensated liver cirrhosis (mean modified Pugh score of 14 points) were markedly elevated and significantly higher than those in the patients with decompensated liver cirrhosis with HCC (modified Pugh score of 10 points), those with compensated liver cirrhosis with HCC, and those with compensated liver cirrhosis. Patients in all these three latter groups had moderately and significantly elevated u-PA levels compared to levels in the chronic hepatitis group and the healthy volunteers, but the levels were not significantly different from each other. There was no relationship between u-PA plasma level and the type of HCC tumor invasion or number or size of tumors. Significant correlations were found between u-PA plasma levels and the results of seven different liver function tests in three groups without associated HCC; u-PA antigen and prothrombin time (%), hepaplastin test (%), serum cholinesterase, serum albumin, serum total cholesterol, and indocyanine green clearance correlated negatively, while u-PA antigen and serum total bilirubin correlated positively. These results suggest that plasma u-PA is associated with deterioration of liver function but not with HCC invasion. [ABSTRACT FROM AUTHOR]

31. Ultrastructural organization of the transverse tubules and the sarcoplasmic reticulum in a fish sound-producing muscle

Abstract

The ultrastructural basis for the extremely rapid contraction-relaxation cycle (up to 300 s-1) in the swim-bladder muscle (SBM) of a scorpionfish (Sebastiscus marmoratus), producing characteristic sounds for communication, was investigated by electron microscopy. The SBM fibres contained well-developed sarcoplasmic reticulum (SR) showing triadic contacts with well-organized transverse tubules (T tubules). It was newly found that different types of triadic contacts were present within the single SBM fibre. In the middle region of the fibre (∼54% of the fibre length), the triadic contacts were located around the level of boundary between the A- and I-bands (AI-type triad). However, in the two end regions of the fibre (∼21% and ∼12% of the fibre length), the triadic contacts were seen around the level of the Z-band (Z-type triad). Between the middle and end regions of the fibre, T tubule-SR contacts exhibited the form of pentads composed of a pair of T tubules and three SR elements, and newly found heptads composed of three T tubules and four SR elements. The fractional volume of SR relative to the fibre volume was estimated to be ∼26% in the middle region of the fibre with the AI-type triads and ∼15% in the fibre ends with the Z-type triads. These results are discussed in connection with the mechanism, by which the mechanical activity of the SBM muscle is neurally controlled.

34. A pathogenic point mutation reduces stability of mitochondrial mutant tRNA(Ile).

Abstract

Point mutations in mitochondrial tRNA genes are responsible for individual subgroups of mitochondrial encephalomyopathies. We have recently reported that point mutations in the tRNA(Leu)(UUR) and tRNA(Lys) genes cause a defect in the normal modification at the first nucleotide of the anticodon. As part of a systematic analysis of pathogenic mutant mitochondrial tRNAs, we purified tRNA(Ile) with a point mutation at nucleotide 4269 to determine its nucleotide sequence, including modified nucleotides. We found that, instead of causing a defect in the post-transcriptional modification, a pathogenic point mutation in the mitochondrial tRNA(Ile) reduced the stability of the mutant tRNA molecule, resulting in a low steady-state level of aminoacyl-tRNA. The reduced stability was confirmed by examining the life-span of the mutant tRNA(Ile) both in vitro and in vivo, as well as by monitoring its melting profile. Our finding indicates that the mutant tRNA(Ile) itself is intrinsically unstable.

Abstract

1. The effect of acetylcholine (ACh) on the membrane current components in guinea‐pig papillary muscle was studied by the single sucrose gap voltage clamp method. 2. The slow inward current (is) elicited by various depolarizations from the holding potential of ‐40 mV was evidently diminished by ACh greater than 5 x 10(‐7) M. The amplitude of is, estimated as the difference between the peak of is and the current at 150 msec, was reduced to 56.8 +/‐ 12.9% (mean +/‐ S.D., n = 34) by 2 x 10(‐8) M‐ACh when estimated at the membrane potential where the amplitude was maximal. 3. is was suppressed almost completely by 2 mM‐Co2+, is obtained by subtracting the current remaining in 2 mM‐Co2+ was also diminished to the same extent by 2 x 10(‐6) M‐Ach with little change in its time course; the degree of decrease was not dependent on the membrane potential. 4. The time‐dependent outward current during the depolarizing pulse was also diminished by 2 x 10(‐6) M‐Ach. Correspondingly, the positive tail current obtained when the membrane was repolarized to the holding potential of ‐40 mV after various 1 sec depolarizations was reduced in a proportionate manner to 68.6 +/‐ 7.3% (n = 10). 5. The time‐independent inward rectifying outward current was not affected by 2 x 10(‐6) M‐Ach. 6. Atropine (10(‐6) M) restored is and the time‐dependent outward current which had been previously depressed by 2 x 10(‐6) M‐ACh. 7. ACh may decrease tension development by depressing is, thereby preserving the duration of the action potential with relatively little change, since, different from atrial muscle, ACh does not increase the outward current in mammalian ventricular muscle.
