Abstract

We report about the design and the initial performances of the pathfinder mission for a possible large scale neutrino telescope named "STRings for Absorption length in Water" (STRAW). In June 2018 STRAW has been deployed at the Cascadia Basin site operated by Ocean Network Canada and has been collecting data since then. At a depth of about 2600 meters, the two STRAW 120 meters tall mooring lines are instrumented by three "Precision Optical Calibration Modules" (POCAM) and five Digital Optical Sensors (sDOM). Abstract

Fundamental to all living organisms is the capacity to coordinate cell division and cell differentiation to generate appropriate numbers of specialized cells. Whereas eukaryotes use cyclins and cyclin-dependent kinases to balance division with cell fate decisions (1), equivalent regulatory systems have not been described in bacteria. Moreover, the mechanisms used by bacteria to tune division in line with developmental programs are poorly understood. Here we show that Caulobacter crescentus, a bacterium with an asymmetric division cycle, uses oscillating levels of the second messenger cyclic diguanylate (c-di-GMP) to drive its cell cycle. We demonstrate that c-diGMP directly binds to the essential cell cycle kinase CckA to inhibit kinase activity and stimulate phosphatase activity. An upshift of c-di-GMP during the G1-S transition switches CckA from the kinase to the phosphatase mode, thereby allowing replication initiation and cell cycle progression. Finally, we show that during division, c-di-GMP imposes spatial control on CckA to install the replication asymmetry of future daughter cells. These studies reveal c-di-GMP to be a cyclin-like molecule in bacteria that coordinates chromosome replication with cell morphogenesis in Caulobacter. The observation that c-di-GMP-mediated control is conserved in the plant pathogen Agrobacterium tumefaciens suggests a general mechanism through which this global regulator of bacterial virulence and persistence coordinates behaviour and cell proliferation., To enable tissue homeostasis, metazoans tightly regulate the balance between cell proliferation and differentiation (2). The statement ""Several dozen compounds with antimicrobial properties have been isolated from nematophilic bacteria, but these do not hit specific targets or do not appear to have drug-like properties"" has been changed to ""Several dozen compounds with antimicrobial properties have been isolated from nematophiles, but only the odilorhabins (produced by Xenorhabdus nematophila) hit a specific target and show efficacy in certain animal models of infection"", and the two new references have been added as references 34 and 35 at the end of the new sentence. The subsequent references have been renumbered accordingly.

Abstract

Active Control can help to meet future engine requirements by an active improvement of the component characteristics. The concept is based on an intelligent control logic, which senses actual operating conditions and reacts with adequate actuator action. This approach can directly improve engine characteristics as performance, operability, durability and emissions on the one hand. On the other hand active control addresses the design constrains imposed by unsteady phenomena like inlet distortion, compressor surge, combustion instability, flow separations, vibration and noise, which only occur during exceptional operating conditions. The feasibility and effectiveness of active control technologies have been demonstrated in lab-scale tests. This chapter describes a broad range of promising applications for each engine component. Significant efforts in research and development remain to implement these technologies in engine rig and finally production engines and to demonstrate today s engine generation airworthiness, safety, reliability, and durability requirements. Active control applications are in particular limited by the gap between available and advanced sensors and actuators, which allow an operation in the harsh environment in an aero engine. The operating and performance requirements for actuators and sensors are outlined for each of the gas turbine sections from inlet to nozzle.

Abstract

Mitochondria cannot form de novo but require mechanisms allowing their inheritance to daughter cells. In contrast to most other eukaryotes Trypanosoma brucei has a single mitochondrion whose single unit genome is physically connected to the flagellum. Here we identify a beta barrel mitochondrial outer membrane protein termed tripartite attachment complex 40 (TAC40) that localizes to this connection. TAC40 is essential for mitochondrial DNA inheritance and belongs to the mitochondrial porin protein family. However it is not specifically related to any of the three subclasses of mitochondrial porins represented by the metabolite transporter voltage dependent anion channel (VDAC) the protein translocator of the outer membrane 40 (TOM40) or the fungi specific MDM10 a component of the endoplasmic reticulum mitochondria encounter structure (ERMES). MDM10 and TAC40 mediate cellular architecture and participate in transmembrane complexes that are essential for mitochondrial DNA inheritance. In yeast MDM10 in the context of the ERMES is postulated to connect the mitochondrial genomes to actin filaments whereas in trypanosomes TAC40 mediates the linkage of the mitochondrial DNA to the basal body of the flagellum. However TAC40 does not colocalize with trypanosomal orthologs of ERMES components and unlike MDM10 it regulates neither mitochondrial morphology nor the assembly of the protein translocase. TAC40 therefore defines a novel subclass of mitochondrial porins that is distinct from VDAC TOM40 and MDM10. However whereas the architecture of the TAC40 containing complex in trypanosomes and the MDM10 containing ERMES in yeast is very different both are organized around a beta barrel protein of the mitochondrial porin family that mediates a DNA cytoskeleton linkage that is essential for mitochondrial DNA inheritance.

Abstract

The clinical value of imaging is well established for the follow-up of many lymphoid malignancies but not for chronic lymphocytic leukemia (CLL). A meta-analysis was performed with the dataset of 3 German CLL Study Group phase 3 trials (CLL4, CLL5, and CLL8) that included 1372 patients receiving first-line therapy for CLL. Response as well as progression during follow-up was reassessed according to the National Cancer Institute Working Group1996 criteria. A total of 481 events were counted as progressive disease during treatment or follow-up. Of these, 372 progressions (77%) were detected by clinical symptoms or blood counts. Computed tomography (CT) scans or ultrasound were relevant in 44 and 29 cases (9% and 6%), respectively. The decision for relapse treatment was determined by CT scan or ultrasound results in only 2 of 176 patients (1%). CT scan results had an impact on the prognosis of patients in complete remission only after the administration of conventional chemotherapy but not after chemoimmunotherapy. In conclusion, physical examination and blood count remain the methods of choice for staging and clinical follow-up of patients with CLL as recommended by the International Workshop on Chronic Lymphocytic Leukemia 2008 guidelines.
