Abstract

The cost-effective deployment of floating offshore wind turbines is faced with a multitude of technical challenges. Several key areas of the life-cycle of floating offshore wind turbines have been discussed within this work. The key challenges and potential solutions have been presented, with state-of-the-art literature summarised to enable further work to build off. It is concluded that all areas of the use of this technology requires further maturing to de-risk this technology to entice wide-spread commercial use.

Abstract

H-NMR analysis of plastocyanins explains a high-resolution solution structure of parsley plastocyanin. This structure shows that E53, E85 and E95 compensate for the charge residue substitution of 59 and 60 sites of the parsley plastocyanins, giving the plastocyanin an acidic patch. Kinetic studies imply that the reactivity of parsley plastocyanin is similar to that of higher plant plastocyanins.

Abstract

We study how people's predisposition towards altruism, as measured by tools developed by psychologists, affects their behavior in a voluntary contributions public good experiment. Earlier experiments provide evidence against the strong free rider hypothesis; however, contributions to the public good decrease with repetition. We investigate whether a high level of contributions can be sustained in groups of subjects who have been pre-selected on the basis of their altruistic inclinations. In the first stage of the experiment, each subject responds to a psychology questionnaire that measures various dimensions of one's personality. The subjects are then matched in groups according to their altruism scores, and engage in a voluntary contribution game. We consider whether the levels and dynamics of group contributions differ significantly between the groups with altruists and non-altruists. We find that subjects' altruism has a weak but positive effect on group behavior in the public good game.

Abstract

Objectives To determine whether preoperative dexamethasone reduces postoperative vomiting in patients undergoing elective bowel surgery and whether it is associated with other measurable benefits during recovery from surgery, including quicker return to oral diet and reduced length of stay. Design Pragmatic two arm parallel group randomised trial with blinded postoperative care and outcome assessment. Setting 45 UK hospitals. Participants 1350 patients aged 18 or over undergoing elective open or laparoscopic bowel surgery for malignant or benign pathology. Interventions Addition of a single dose of 8 mg intravenous dexamethasone at induction of anaesthesia compared with standard care. Main outcome measures Primary outcome: reported vomiting within 24 hours reported by patient or clinician. Secondary outcomes: vomiting with 72 and 120 hours reported by patient or clinician; use of antiemetics and postoperative nausea and vomiting at 24, 72, and 120 hours rated by patient; fatigue and quality of life at 120 hours or discharge and at 30 days; time to return to fluid and food intake; length of hospital stay; adverse events. Results 1350 participants were recruited and randomly allocated to additional dexamethasone (n=674) or standard care (n=676) at induction of anaesthesia. Vomiting within 24 hours of surgery occurred in 172 (25.5%) participants in the dexamethasone arm and 223 (33.0%) allocated standard care (number needed to treat (NNT) 13, 95% confidence interval 5 to 22; P=0.003). Abstract

Injury to the central nervous system initiates an uncontrolled inflammatory response that results in both tissue repair and destruction. Here, we showed that, in rodents and humans, injury to the spinal cord triggered surface expression of CD95 ligand (CD95L, FasL) on peripheral blood myeloid cells. CD95L stimulation of CD95 on these cells activated phosphoinositide 3-kinase (PI3K) and metalloproteinase-9 (MMP-9) via recruitment and activation of Syk kinase, ultimately leading to increased migration. Exclusive CD95L deletion in myeloid cells greatly decreased the number of neutrophils and macrophages infiltrating the injured spinal cord or the inflamed peritoneum after thioglycollate injection. Importantly, deletion of myeloid CD95L, but not of CD95 on neural cells, led to functional recovery of spinal injured animals. Our results indicate that CD95L acts on peripheral myeloid cells to induce tissue damage. Thus, neutralization of CD95L should be considered as a means to create a controlled beneficial inflammatory response.

Abstract

Adult neurogenesis persists in the subventricular zone and the dentate gyrus and can be induced upon central nervous system injury. However, the final contribution of newborn neurons to neuronal networks is limited. Here we show that in neural stem cells, stimulation of the "death receptor" CD95 does not trigger apoptosis but unexpectedly leads to increased stem cell survival and neuronal specification. These effects are mediated via activation of the Src/PI3K/AKT/mTOR signaling pathway, ultimately leading to a global increase in protein translation. Induction of neurogenesis by CD95 was further confirmed in the ischemic CA1 region, in the naive dentate gyrus, and after forced expression of CD95L in the adult subventricular zone. Lack of hippocampal CD95 resulted in a reduction in neurogenesis and working memory deficits. Following global ischemia, CD95-mediated brain repair rescued behavioral impairment. Thus, we identify the CD95/CD95L system as an instructive signal for ongoing and injury-induced neurogenesis.

Invasion of surrounding brain tissue by isolated tumor cells represents one of the main obstacles to a curative therapy of glioblastoma multiforme. Here we unravel a mechanism regulating glioma infiltration. Tumor interaction with the surrounding brain tissue induces CD95 Ligand expression. Binding of CD95 Ligand to CD95 on glioblastoma cells recruits the Src family member Yes and the p85 subunit of phosphatidylinositol 3-kinase to CD95, which signal invasion via the glycogen synthase kinase 3-beta pathway and subsequent expression of matrix metalloproteinases. In a murine syngeneic model of intracranial GBM, neutralization of CD95 activity dramatically reduced the number of invading cells. Our results uncover CD95 as an activator of PI3K and, most importantly, as a crucial trigger of basal invasion of glioblastoma in vivo.

Abstract

The Orbital Transfer Vehicle (OTV) is to be employed for the delivery of a paylod to a high earth orbit, such as a geosynchronous orbit. Subsequently, the OTV is to return to a low earth parking orbit. The present investigation is concerned with an aeroassisted OTV (AOTV) which achieves the required reduction in velocity on its return to the parking orbit through aerodynamic braking. An adaptive guidance logic is employed to control and AOTV as it passes through the earth's upper atmosphere. Attention is given to details regarding the adaptive guidance logic, and a performance evaluation. It is found that the performance of the adaptive guidance logic is satisfactory for the considered conditions.

Abstract

Operations conducted from a space station in low Earth orbit which have different constraints and opportunities than those conducted from direct Earth launch were examined. While a space station relieves many size and performance constraints on the space shuttle, the space station's inertial orbit has different launch window constraints from those associated with customary Earth launches which reflect upon upper stage capability. A performance requirements analysis was developed to provide a reference source of parametric data, and specific case solutions and upper stage sizing trade to assist potential space station users and space station and upper stage developers assess the impacts of a space station on missions of interest.
