Anthropogenic activity impacts stream ecosystems, resulting in a loss of diversity and ecosystem function; however, little is known about the response of aquatic microbial communities to changes in land use. Here, microbial communities were characterized in 82 headwater streams across a gradient of urban and agricultural land uses using 16S rRNA gene amplicon sequencing and compared to a rich data set of physicochemical variables and traditional benthic invertebrate indicators. Microbial diversity and community structures differed among watersheds with high agricultural, urban, and forested land uses, and community structure differed in streams classified as being in good, fair, poor, and very poor condition using benthic invertebrate indicators. Microbial community similarity decayed with geodesic distance across the study region but not with environmental distance. Stream community respiration rates ranged from 21.7 to 1,570 mg O2 m-2 day-1 and 31.9 to 3,670 mg O2 m-2 day-1 for water column and sediments, respectively, and correlated with nutrients associated with anthropogenic influence and microbial community structure. Nitrous oxide (N2O) concentrations ranged from 0.22 to 4.41 μg N2O liter-1; N2O concentration was negatively correlated with forested land use and was positively correlated with dissolved inorganic nitrogen concentrations. Our findings suggest that stream microbial communities are impacted by watershed land use and can potentially be used to assess ecosystem health.IMPORTANCE Stream ecosystems are frequently impacted by changes in watershed land use, resulting in altered hydrology, increased pollutant and nutrient loads, and habitat degradation. Macroinvertebrates and fish are strongly affected by changes in stream conditions and are commonly used in biotic indices to assess ecosystem health. Similarly, microbes respond to environmental stressors, and changes in community composition alter key ecosystem processes. The response of microbes to habitat degradation and their role in global biogeochemical cycles provide an opportunity to use microbes as a monitoring tool. Here, we identify stream microbes that respond to watershed urbanization and agricultural development and demonstrate that microbial diversity and community structure can be used to assess stream conditions and ecosystem functioning.

Species distribution modelling can be useful for the conservation of rare and endangered species. Freshwater mussel declines have thinned species ranges producing spatially fragmented distributions across large areas. Spatial fragmentation in combination with a complex life history and heterogeneous environment makes predictive modelling difficult., A machine learning approach (maximum entropy) was used to model occurrences and suitable habitat for the federally endangered dwarf wedgemussel, Alasmidonta heterodon, in Maryland's Coastal Plain catchments. Landscape-scale predictors (e.g. land cover, land use, soil characteristics, geology, flow characteristics, and climate) were used to predict the suitability of individual stream segments for A. heterodon., The best model contained variables at three scales: minimum elevation (segment scale), percentage Tertiary deposits, low intensity development, and woody wetlands (sub-catchment), and percentage low intensity development, pasture/hay agriculture, and average depth to the water table (catchment). Despite a very small sample size owing to the rarity of A. heterodon, cross-validated prediction accuracy was 91%., Most predicted suitable segments occur in catchments not known to contain A. heterodon, which provides opportunities for new discoveries or population restoration. These model predictions can guide surveys toward the streams with the best chance of containing the species or, alternatively, away from those streams with little chance of containing A. heterodon., Developed reaches had low predicted suitability for A. heterodon in the Coastal Plain. Urbanization is associated with substantial losses to stream biological diversity throughout the United States' mid-Atlantic. Stream restoration has been used to improve stream conditions and, in part, to ameliorate these losses. However, the relationship between restoration and recovery of biological diversity is unclear. Our objective was to critically examine the efficacy of urban stream restorations with regard to biological diversity. We compared restored urban streams to urban nonrestored, nonurban, and reference (minimally degraded) streams using five measures each of fish and benthic macroinvertebrate diversity. Both multivariate and univariate statistical analyses show biological diversity of restored urban streams to be similar to nonrestored urban streams and lower than nonurban and reference streams. Restored urban sites showed no apparent increase in biological diversity through time, while diversity decreased at two of the reference streams coincident with increased urban development within their catchments. Our results indicate that restoration approaches commonly used regionally as in these urban streams are not leading to recovery of native stream biodiversity. Evidence from several sources indicates a need for dramatic changes in restoration approach, and we argue for a watershed-scale focus including protection of the least impacted streams and adopting other land-based actions within the watershed where possible.
