Abstract

BACKGROUND: Estimation of glomerular filtration rate using equations based on creatinine is widely used to manage chronic kidney disease. In the UK, the Chronic Kidney Disease Epidemiology Collaboration creatinine equation is recommended. Other published equations using cystatin C, an alternative marker of kidney function, have not gained widespread clinical acceptance. Given higher cost of cystatin C, its clinical utility should be validated before widespread introduction into the NHS. OBJECTIVES: Primary objectives were to: (1) compare accuracy of glomerular filtration rate equations at baseline and longitudinally in people with stage 3 chronic kidney disease, and test whether accuracy is affected by ethnicity, diabetes, albuminuria and other characteristics; (2) establish the reference change value for significant glomerular filtration rate changes; (3) model disease progression; and (4) explore comparative cost-effectiveness of kidney disease monitoring strategies. DESIGN: A longitudinal, prospective study was designed to: (1) assess accuracy of glomerular filtration rate equations at baseline (n = 1167) and their ability to detect change over 3 years (n = 875); (2) model disease progression predictors in 278 individuals who received additional measurements; (3) quantify glomerular filtration rate variability components (n = 20); and (4) develop a measurement model analysis to compare different monitoring strategy costs (n = 875). SETTING: Primary, secondary and tertiary care. PARTICIPANTS: Adults (≥ 18 years) with stage 3 chronic kidney disease. Abstract

Biospecimen Sharing Programs (BSPs) have been organized by NASA Ames Research Center since the 1960s with the goal of maximizing utilization and scientific return from rare, complex and costly spaceflight experiments. BSPs involve acquiring otherwise unused biological specimens from primary space research experiments for distribution to secondary experiments. Here we describe a collaboration leveraging Ames expertise in biospecimen sharing to magnify the scientific impact of research informing astronaut health funded by the NASA Human Research Program (HRP) Human Health Countermeasures (HHC) Element. The concept expands biospecimen sharing to one-off ground-based studies utilizing analogue space platforms (e.g., Hindlimb Unloading (HLU), Artificial Gravity) for rodent experiments, thereby significantly broadening the range of research opportunities with translational relevance for protecting human health in space and on Earth. In this presentation, we will report on biospecimens currently being acquired from HHC Award Head-Down Tilt as a Model for Intracranial and Intraocular Pressures, and Retinal Changes during Spaceflight, and their availability. The BSP add-on to the project described herein has already yielded for HHC-funded investigators more than 4,700 additional tissues that would otherwise have been discarded as waste, with additional tissues available for analysis. Young (3-mo old) male and female rats and Older (9-mo old) male rats are being exposed to HLU for either 7, 14, 28, or 90 days. Additional groups are exposed to 90 days of unloading followed by either 7, 14, 28 days or 90 days of recovery (normal loading). Comparisons are made with non-suspended controls. Unused tissues are: Skin, Lungs, Thymus, Adrenals, Kidneys, Spleen, Hindlimb Muscles (Soleus, Extensor Digitorum Longus, Tibialis Anterior, Plantaris Gastrocnemius), Fat Pads, Reproductive Organs, and Intestines. Tissues are harvested, weighed, preserved then archived (with metadata) using a sample tracking system (CryoTrack). Preservation techniques include snap-freezing and RNALatersnap-freezing. Specimens were weighed at the time of dissection, and organ mass:body mass ratios analyzed to determine unloading effects across conditions and durations. The results corroborate previously reported effects of short-term exposure to microgravity or unloading exposure on various organs, and provide new insights into adaptation to long-duration unloading relevant to sustained spaceflight exposures on ISS.

Abstract

The Secretary of State for the Department of Digital, Culture Media and Sport has granted Scheduled Monument Consent for the work with the proviso that the applicant engages an archaeological contractor to supervise and monitor the works in accordance with a Written Scheme of Investigation which has been approved by Historic England (Scheduled Monument Consent: S00130289). The Scheduled Monument Consent required that the applicant engages an archaeological contractor to supervise and monitor the works in accordance with a Written Scheme of Investigation which has been approved by Historic England. The fieldwork specified with WSI was intended to ensure that the works will not have an impact on buried archaeological remains as well as to ensure no unnecessary collateral damage to the monument occurs. De Montfort University has appointed structural engineers to carry out a survey to investigate the cause of cracking to the external walls of the brick Ancillary Block which abuts the John of Gaunt's Yard on the south side of the Great Hall of Leicester Castle. The work is required to provide information to inform a remedial strategy and will include intrusive investigation work, including the excavation of a trial hole (outside south-east corner to 1m depth), a core hole, arboricultural surveys, and provision of a temporary tower scaffold This report details the results of an archaeological watching brief that was carried out by University of Leicester Archaeological Services (ULAS) at Leicester Castle Business School Store, Leicester (SK 5823 0414). The work was carried out in November 2021 on behalf of De Montfort University during groundworks in accordance with the Ancient Monuments and Archaeological Areas Act 1979. De Montfort University has appointed structural engineers to carry out a survey to investigate the cause of cracking to the external walls of the brick Ancillary Block which abuts the John of Gaunt's Yard on the south side of the Great Hall of Leicester Castle. The field work was undertaken to ensure that the works will not have impact on the buried archaeological remains. The site is located within Leicester Castle and the Magazine Gateway Scheduled Monument (List Entry Number 1012147) located within Leicester city centre on the eastern bank of the canalised River Soar. It includes the earthwork and buried remains of a motte and bailey castle, the 12th century northern extension to the bailey, and the northern part of the fortified enclosure known as The Newarke, including the Turret Gateway and the Magazine Gateway. The work is required to provide information to inform a remedial strategy and include intrusive investigation work, including the excavation of a trial hole outside south-east corner the brick Ancillary Block, and two internal core hole, surveys, were bored into the floor of the block. The outside trial pit was hand excavated to a depth 0.90m and encountered flat sandstone thought to be a potential remnants of foundation, which was sealed under garden soils. Two core holes were excavated within the block encountered substantial resistance or obstruction at depth 1.40m sealed by potential brick floor. The reason for the obstruction below the brick floor was not determined but the approximate depth coincided with level for the top the John of Gaunt's cellar structure located directly to the west. The brick floor was sealed under demolition layers and is either associated with an earlier structure or is a buried earlier floor level within the existing ancillary block. The site archive will be deposited with Leicestershire City Council Museums Service with the accession number A3.2022.

Abstract

Evidence is increasing that untreated wastewater is a good indicator of the presence of the virus in a population. The ability to detect the current SARS-CoV-2 in wastewater is increasingly being reported independently by various research groups as a possible way to better quantify and understand its approximate overall presence in the population. Upon the first confirmation of the virus RNA appearing in stools of COVID-19 patients, research groups in the Netherlands, Australia, United States, France, Italy, Austria and elsewhere have successfully established a relationship between the virus’s concentration in influents to wastewater treatment plant and the level of infection in the population in question. Thus, wastewater surveillance of SARS-CoV-2 eventually combined with the monitoring of suitable tracers such as cross-assembly phage, pepper mild mottle virus or chemicals tracers related to human activities (e.g. food ingredients, inorganic wastewater parameters, pharmaceuticals in particular those used in the treatment of COVID-19, or other contaminants of emerging concern) is a valuable and efficient tool to monitor virus circulation in EU cities and towns. This report addresses the investigation of the tool in fast track collaborative effort and ad hoc exercise with stakeholders representing Academia as well as the Water and Public Health Sector. The assessment reveals insight in methodologies but also entailed costs for rollout of a European Sewage Sentinel System for SARSCoV-2. It also shares the findings of two experimental assessment conducted to link existing and ongoing, national, regional or local surveillance programs. It moreover shares the findings of the accompanying knowledge brokering and transfer events organized to have a rolling exchange of information and review of advances, such coping with the speed challenge of the rapid dynamics of this pandemic.
