Abstract

The active sites of single-chain and two-chain tissue plasminogen activator (sct-PA, tct-PA) in solution were directly compared by the application of a series of nitroxide spin labels and fluorophores. These effective and sensitive probes are useful for monitoring small protein structure changes. Further analysis was carried out by the creation of molecular models of the protease domains of act-PA and tct-PA.

Abstract

Kinetic data was obtained for both the recombinant human tissue plasminogen activator (t-PA) and the 'KHRR' t-PA mutant in which the sequence from amino acids 296 to 299 was substituted with tetra-alanine. Previous studies have found that the region affects the ability of the fibrinogen and fibrin to increase plasminogen activation rate of t-PA. The results suggest that 'KHRR' t-PA is a thrombolytic agent that is not inducible by fibrinogen-related molecules found systemically, but is activated in the presence of clot components.

Abstract

A new congenital dysfibrinogenemia, designated fibrinogen Petoskey, which was traced through four generations of a Michigan family, was found to exhibit an abnormally slow rate of release of fibrino-peptide A upon treatment with thrombin and batroxobin. Batroxobin only partially hydrolyzed and polymerized the fibrinogen from affected individuals, suggesting that these patients had both normal and abnormal fibrinogen in their circulation and that batroxobin was not capable of releasing fibrinopeptide A from the abnormal fibrinogen. Polymerization of fibrin monomers from fibrinogen Petoskey and plasmin mediated digestion of fibrinogen Petoskey were normal. The Factor XIIIa-catalyzed cross-linking of fibrinogen Petoskey was slightly delayed at low (but not at high) concentrations of thrombin. This delayed cross-linking appeared to be a secondary effect of the lower rate of release of fibrinopeptide A.

Abstract

Anal incontinence and defecatory difficulties are common and have a considerable impact on quality of life and health cost. Conservative management should be the first line management of anorectal dysfunction because there is minimal risk and a high rate of success with completion of therapy. This article aims to describe the principles of the physiotherapy assessment and the diagnostic process and to summarise published literature focusing on level 1 evidence of the physiotherapy strategies utilised as first-line conservative management of the following anorectal disorders: anal incontinence, defecatory difficulties and functional anorectal pain.

Abstract

Glutathione S-transferases (GST, E.C.2.5.1.18) comprise a family of detoxification enzymes. Elevated levels of specific GST isozymes in tumor cells are thought responsible for resistance to chemotherapeutics, which renders selective GST inhibitors potentially useful pharmaceutical agents. We discuss the development of a structure activity model that rationalizes the isozyme selectivity observed in a series of 12 glutathione (GSH) analogues. Enzymatic activity data was determined for human P1-1, A1-1, and M2-2 isozymes, and these data were then considered in light of structural features of these three GST proteins. A survey of all GST structures in the PDB revealed that GSH binds to these proteins in a single “bioactive” conformation. To focus on differences between binding sites, we exploited our finding of a common GSH conformation and aligned the GST x-ray structures using bound ligands rather than the backbones of the different proteins. Once aligned, binding site lipophilicity and electrostatic potentials were computed, visualized, and compared. Docking and energy minimization exercises provided additional refinements to a model of selectivity developed initially by visual analysis. 47. Dysfibrinogenemias: Probes for Establishing Relationships Between the Structure of Fibrinogen and Its Function in Blood Coagulation.

Abstract

Inherited abnormal fibrinogens that exhibit altered behavior in one or more of the steps in the conversion of fibrinogen to fibrin should be useful for determining structure-function relationships important in the final stages of blood coagulation. This thesis contains the functional characterization of three new fibrinogen variants and the structural characterization of one of these on a molecular level. The fibrinogen variant fibrinogen Ann Arbor was shown to exhibit altered end-to-end aggregation of fibrin monomers, as well as a more rapid rate of (gamma)-dimer formation. Fibrinogen Gr and Rapids was associated with hypofibrinogenemia and was shown to exhibit abnormal aggregation of fibrin monomers. Fibrinogen Petoskey exhibited a decreased initial rate of fibrinopeptide A (FPA) and fibrinopeptide B (FPB) release. Using a high performance liquid chromatography (HPLC) method for the separation of human fibrinopeptides, an abnormal FPA (FPA Petoskey) along with normal FPA and FPB was shown to be released upon incubation of fibrinogen Petoskey with thrombin. The molar ratios of released fibrinopeptides were one FPA Petoskey, one normal FPA, and two FPB. Amino acid analyses and carboxypeptidase Y digestion of purified FPA Petoskey indicated that it was identical to normal FPA, except that Arg-16 in normal FPA was replaced by a histidyl residue in FPA Petoskey. The thrombin-catalyzed cleavage of fibrinogen Petoskey reported here is the first demonstration of thrombin-catalyzed hydrolysis at a peptide bond other than at an arginyl or lysyl residue. Batroxobin, unlike thrombin, was incapable of cleaving the unusual His-A(alpha)16-Gly-A(alpha)17 bond in fibrinogen Petoskey. Less than one-quarter of the fibrinogen from an affected individual aggregated following incubation with batroxobin, suggesting the presence of heterodimers containing one normal and one abnormal chain. The removal of both FPA molecules from a fibrinogen molecule appears necessary for the molecule to aggregate properly. The HPLC separation of human fibrinopeptides developed in this work allows a simple and direct measurement of the action of thrombin on fibrinogen. The application of this method for characterizing the thrombin mediated release of fibrinopeptides from normal fibrinogen and fibrinogen Petoskey and its usefulness for determining the mechanism of thrombin-fibrinogen interaction is discussed.
