Neurostimulation is an essential technique to trigger and modulate the spatiotemporal activity of local neuronal circuits. Current stimulation methods have a trade-off relationship among aiming precision, temporal resolution, and noninvasiveness, making it difficult to stimulate and monitor a single target neuron for a long term. Here, we show that a method using two needle electrodes in combination of micropatterning techniques provides new possibilities for targeting and stimulating a single neuron selectively. Results of physiological experiments as well as analog circuit simulation reveal that two needle electrodes can stimulate a target neuron selectively by placing the two needle electrodes in proximity to and to straddle the target neuron, and that the steepness of voltage applied to two needle electrodes is important for the target neuron to fire at a low voltage. The proposed method enables a noninvasive stimulation suitable for measuring long-term activity of local neuronal circuits.

The spontaneous activity pattern of cortical neurons in dissociated culture is characterized by burst firing that is highly synchronized among a wide population of cells. The degree of synchrony, however, is excessively higher than that in cortical tissues. Here, we employed polydimethylsiloxane (PDMS) elastomers to establish a novel system for culturing neurons on a scaffold with an elastic modulus resembling brain tissue, and investigated the effect of the scaffold's elasticity on network activity patterns in cultured rat cortical neurons. Using whole-cell patch clamp to assess the scaffold effect on the development of synaptic connections, we found that the amplitude of excitatory postsynaptic current, as well as the frequency of spontaneous transmissions, was reduced in neuronal networks grown on an ultrasoft PDMS with an elastic modulus of 0.5 kPa. Furthermore, the ultrasoft scaffold was found to suppress neural correlations in the spontaneous activity of the cultured neuronal network. The dose of GsMTx-4, an antagonist of stretch-activated cation channels (SACs), required to reduce the generation of the events below 1.0 event/min on PDMS substrates was lower than that for neurons on a glass substrate. This suggests that the difference in the baseline level of SAC activation is a molecular mechanism underlying the alteration in neuronal network activity depending on scaffold stiffness. The reconstitution of ion-channel proteins in artificially formed bilayer lipid membranes (BLMs) forms a well-defined system for the functional analysis of ion channels and screening of the effects of drugs that act on these proteins. To improve the efficiency of the BLM reconstitution system, we report on a microarray of stable solvent-free BLMs formed in microfabricated silicon (Si) chips, where micro-apertures with well-defined nano- and micro-tapered edges were fabricated. Sixteen micro-wells were manufactured in a chamber made of Teflon&reg, and the Si chips were individually embedded in the respective wells as a recording site. Typically, 11 to 16 BLMs were simultaneously formed with an average BLM number of 13.1, which corresponded to a formation probability of 82%. Parallel recordings of ion-channel activities from multiple BLMs were successfully demonstrated using the human ether-a-go-go-related gene (hERG) potassium channel, of which the relation to arrhythmic side effects following drug treatment is well recognized.

Niemann-Pick C1-Like 1 (NPC1L1) is a cholesterol importer and target of ezetimibe, a cholesterol absorption inhibitor used clinically for dyslipidemia. Recent studies demonstrated that NPC1L1 regulates the intestinal absorption of several fat-soluble nutrients, in addition to cholesterol. The study was conducted to reveal new physiological roles of NPC1L1 by identifying novel dietary substrate(s). Very low-density lipoprotein and low-density lipoprotein (VLDL/LDL) are increased in Western diet (WD)-fed mice in an NPC1L1-dependent manner, so we comprehensively analyzed the NPC1L1-dependent VLDL/LDL components. Apolipoprotein M (apoM), a binding protein of sphingosine-1-phosphate (S1P: a lipid mediator), and S1P were NPC1L1-dependently increased in VLDL/LDL by WD feeding. S1P is metabolized from sphingomyelin (SM) and SM is abundant in WD, so we focused on intestinal SM absorption. In vivo studies with Npc1l1 knockout mice and in vitro studies with NPC1L1-overexpressing cells revealed that SM is a physiological substrate of NPC1L1. These results suggest a scenario in which dietary SM is absorbed by NPC1L1 in the intestine, followed by SM conversion to S1P and, after several steps, S1P is exported into the blood as the apoM-bound form in VLDL/LDL. Our findings provide insight into the functions of NPC1L1 for a better understanding of sphingolipids and S1P homeostasis.

Gaseous nanobubbles (NBs), with their unique physicochemical properties and promising applications, have become an important research topic. Generation of monodispersed bulk NBs with specified gas content remains a challenge. We developed a simple method for generating bulk NBs, using porous alumina films with ordered straight nano-scaled holes. Different techniques, such as nanoparticle tracking analysis (NTA), atomic force microscopy (AFM), and infrared absorption spectroscopy (IRAS), are used to confirm NB formation. The NTA data demonstrates that the minimum size of the NBs formed is less than 100 nm, which is comparable to the diameter of nanoholes in the porous alumina film. By generating NBs with different gases, including CO2, O2, N2, Ar, and He, we discovered that the minimum size of NBs negatively correlated with the solubility of encapsulated gases in water. Due to the monodispersed size of NBs generated from the highly ordered porous alumina, we determined that NB size is distributed discretely with a uniform increment factor of sqrt(2). To explain the observed characteristic size distribution of NBs, we propose a simple model in which two NBs of the same size are assumed to preferentially coalesce. This characteristic bubble size distribution is useful for elucidating the basic characteristics of nanobubbles, such as the long-term stability of NBs. The electrical impedance of cell membranes is important for excitable cells, such as neurons, because it strongly influences the amount of membrane potential change upon a flow of ionic current across the membrane. Here, we report on an investigation of how neuronal morphology affects membrane impedance of cultured hippocampal neurons. Microfabricated substrates with patterned scaffolding molecules were used to restrict the neurite growth of hippocampal neurons, and the impedance was measured via whole-cell patch-clamp recording under the inhibition of voltage-dependent ion channels. Membrane impedance was found to depend inversely on the dendrite length and soma area, as would be expected from the fact that its electrical property is equivalent to a parallel RC circuit. Moreover, we found that in biological neurons, the membrane impedance is homeostatically regulated to impede changes in the membrane area. The findings provide direct evidence on cell-autonomous regulation of neuronal impedance and pave the way towards elucidating the mechanism responsible for the resilience of biological neuronal networks.

Artificial bilayer lipid membranes (BLMs) provide well-defined systems for investigating the fundamental properties of membrane proteins, including ion channels, and for screening the effect of drugs that act on them. However, the application of this technique is limited due to the low stability and low reconstitution efficiency of the process. We previously reported on improving the stability of BLM based on the fabrication of microapertures having a tapered edge in SiO2/Si3N4 septa and efficient ion channel incorporation based on vesicle fusion accelerated by a centrifugal force. Although the BLM stability and incorporation probability were dramatically improved when these approaches were used, some BLMs were ruptured when subjected to a centrifugal force. To further improve the BLM stability, we investigated the effect of modifying the surface of the SiO2/Si3N4 septa on the stability of BLM suspended in the septa. New cell sources for the research and therapy of organ failure could significantly alleviate the shortage of donor livers that are available to patients who suffer from liver disease. Liver carcinoma derived cells, or hepatoma cells, are the ideal cells for developing bioartificial liver systems. Such cancerous liver cells are easy to prepare in large quantities and can be maintained over long periods under standard culture conditions, unlike primary hepatocytes. However, hepatoma cells possess only a fraction of the functions of primary hepatocytes. In a previous study, by transducing cells with liver-enriched transcription factors that could be inducibly overexpressed-hepatocyte nuclear factor (HNF)1α, HNF1β, HNF3β [FOXA2], HNF4α, HNF6, CCAAT/enhancer binding protein (C/EBP)α, C/EBPβ and C/EBPγ-we created mouse hepatoma cells with high liver-specific gene expression called the Hepa/8F5 cell line. In the present study, we performed functional and genetic analyses to characterize the Hepa/8F5 cell line. Further, in three-dimensional cultures, the function of these cells improved significantly compared to parental cells. Ultimately, these cells might become a new resource that can be used in basic and applied hepatic research.

Synaptic dysfunction and neuronal death are responsible for cognitive and behavioral deficits in Alzheimer's disease (AD). It is well known that such neurological abnormalities are preceded by long-term exposure of amyloid β-peptide (Aβ) and/or hyperphosphorylated tau prior. In addition to the neurological deficit, astrocytes as a major glial cell type in the brain, significantly participate in the neuropathogenic mechanisms underlying synaptic modulation. Although astrocytes play a significant key role in modulating synaptic transmission, little is known on whether astrocyte dysfunction caused by such long-term Aβ exposure affects synapse formation and function. Here, we show that synapse formation and synaptic transmission are attenuated in hippocampal-naive neurons co-cultured with astrocytes that have previously experienced chronic Aβ1-40 exposure. In this abnormal astrocytic condition, hippocampal neurons exhibit decrements of evoked excitatory post-synaptic currents (EPSCs) and miniature EPSC frequency. Furthermore, size of readily releasable synaptic pools and number of excitatory synapses were also significantly decreased. Contrary to these negative effects, release probability at individual synapses was significantly increased in the same astrocytic condition. Taken together, our data indicate that lower synaptic transmission caused by astrocytes previously, and chronically, exposed to Aβ1-40 is attributable to a small number of synapses with higher release probability.

Background noise in biological cortical microcircuits constitutes a powerful resource to assess their computational tasks, including, for instance, the synchronization of spiking activity, the enhancement of the speed of information transmission, and the minimization of the corruption of signals. We explore the correlation of spontaneous firing activity of ≈ 100 biological neurons adhering to engineered scaffolds by governing the number of functionalized patterned connection pathways among groups of neurons. We then emulate the biological system by a series of noise-activated silicon neural network simulations. We show that by suitably tuning both the amplitude of noise and the number of synapses between the silicon neurons, the same controlled correlation of the biological population is achieved. Our results extend to a realistic silicon nanoelectronics neuron design using noise injection to be exploited in artificial spiking neural networks such as liquid state machines and recurrent neural networks for stochastic computation.

The hippocampal formation with tightly packed neurons, mainly at the dentate gyrus, CA3, CA2, and CA1 regions, constitutes a one-way neural circuit, which is associated with learning and memory. We previously showed that the cell adhesion molecules nectins and its binding protein afadin play roles in the formation of the mossy fiber synapses which are formed between the mossy fibers of the dentate gyrus granule cells and the dendrites of the CA3 pyramidal cells. We showed here that in the afadin-deficient hippocampal formation, the dentate gyrus granules cells and the CA3, CA2, and CA1 pyramidal cells were abnormally located; the mossy fiber trajectory was abnormally elongated; the CA3 pyramidal cells were abnormally differentiated; and the densities of the presynaptic boutons on the mossy fibers and the apical dendrites of the CA3 pyramidal cells were decreased. These results indicate that afadin plays roles not only in the formation of the mossy fiber synapses but also in the formation of the cellular architecture of the hippocampus and the dentate gyrus.

An artificial cell membrane that is composed of bilayer lipid membranes (BLMs) with transmembrane proteins incorporated within them represents a well-defined system for the analysis of membrane proteins, especially ion channel proteins that are major targets for drug design. Because the BLM system has a high compatibility with recently developed cell-free expression systems, it has attracted attention as a next-generation drug screening system. However, three issues associated with BLM systems, i. e., their instability, the need for non-volatile organic solvents and a low efficiency of ion channel incorporation, have limited their use as a drug screening platform. In this personal account, we discuss our recent approaches to address these issues based on microfabrication. We also discuss the potential for using the BLM system combined with cell-free expression systems as a drug screening system for future personalized medicine.

Reservoir computing is a machine learning paradigm that was proposed as a model of cortical information processing in the brain. It processes information using the spatiotemporal dynamics of a large-scale recurrent neural network and is expected to improve power efficiency and speed in neuromorphic computing systems. Previous theoretical investigation has shown that brain networks exhibit an intermediate state of full coherence and random firing, which is suitable for reservoir computing. However, how reservoir performance is influenced by connectivity, especially which revealed in recent connectomics analysis of brain networks, remains unclear. Here, we constructed modular networks of integrate-and-fire neurons and investigated the effect of modular structure and excitatory-inhibitory neuron ratio on network dynamics. The dynamics were evaluated based on the following three measures: synchronous bursting frequency, mean correlation, and functional complexity. We found that in a purely excitatory network, the complexity was independent of the modularity of the network. On the other hand, networks with inhibitory neurons exhibited complex network activity when the modularity was high. Our findings reveal a fundamental aspect of reservoir performance in brain networks, contributing to the design of bio-inspired reservoir computing systems.

Multielectrode arrays (MEAs) are versatile tools that are used for chronic recording and stimulation of neural cells and tissues. Driven by the recent progress in understanding of how neuronal growth and function respond to scaffold stiffness, development of MEAs with a soft cell-to-device interface has gained importance not only for in vivo but also for in vitro applications. However, the passivation layer, which constitutes the majority of the cell-device interface, is typically prepared with stiff materials. Herein, a fabrication of an MEA device with an ultrasoft passivation layer is described, which takes advantage of inkjet printing and a polydimethylsiloxane (PDMS) gel with a stiffness comparable to that of the brain. The major challenge in using the PDMS gel is that it cannot be patterned to expose the sensing area of the electrode. This issue is resolved by printing 3D micropillars at the electrode tip. Primary cortical neurons are grown on the fabricated device, and effective stimulation of the culture confirms functional cell-device coupling. The 3D MEA device with an ultrasoft interface provides a novel platform for investigating evoked activity and drug responses of living neuronal networks cultured in a biomimetic environment for both fundamental research and pharmaceutical applications.

We considered a modular network with a binomial degree distribution and related the analytical relationships of the network properties (modularity, average clustering coefficient, and small-worldness) with structural parameters that define the network, i.e., number of nodes, number of modules, average node degree, and ratio of intra-modular to total connections. Even though modular networks are universally found in real-world systems and are consequently of broad interest in complex network science, the relationship between network properties and structural parameters has not yet been formulated. Here, we show that a series of equations for predicting the network properties can be related using a mean-field connectivity matrix that is defined on the basis of the structural parameters in the network generation algorithm. The theoretical results are then compared with values calculated numerically using the original connectivity matrix and are found to agree well, except when the connections between modules are sparse. Representation of the structure of the network using simple parameters is expected to be conducive for elucidating the structure–dynamics relationship.

Westernization of dietary habits increases lipid intake and is responsible for increased numbers of patients with atherosclerotic diseases. Niemann-Pick C1-Like 1 (NPC1L1)-a cholesterol importer-plays a crucial role in dietary cholesterol absorption in the intestine and is closely associated with several lipid-related diseases, including atherosclerosis. NPC1L1 is highly expressed in the liver and intestine in humans, whereas NPC1L1 expression is low in the rodent liver. Due to species differences in the tissue distribution of NPC1L1, there are limited studies on the pathophysiological role of hepatic NPC1L1, a cholesterol reabsorber from bile. In the present study, to explore whether hepatic NPC1L1 is involved in the development/progression of atherosclerosis, we compared four kinds of atherosclerosis mouse models with different expression levels of NPC1L1 in the intestinal and liver tissues in a genetic background of dysfunctional low-density lipoprotein receptor mutation. Western diet (WD)-induced hyperlipidemia and atherosclerotic plaque formation were more severe in mice expressing NPC1L1 in both the liver and intestine (plasma cholesterol, 839.5 mg/dl; plaque area, 29.5% of total aorta), compared with mice expressing NPC1L1 only in the intestine (plasma cholesterol, 573.1 mg/dl; plaque area, 13.3% of total aorta). Such hepatic NPC1L1-mediated promotion of hyperlipidemia and atherosclerosis was not observed in mice not expressing intestinal NPC1L1 and mice treated with ezetimibe, an NPC1L1 inhibitor used clinically for dyslipidemia. These results suggested that hepatic NPC1L1 promotes WD-induced dyslipidemia and atherosclerosis in concert with intestinal NPC1L1. Our findings provide novel insights into the pathophysiological importance of hepatic NPC1L1 in development/progression of atherosclerosis. SIGNIFICANCE STATEMENT: Niemann-Pick C1-Like 1 (NPC1L1) protein, a cholesterol importer and a molecular target of ezetimibe clinically used for dyslipidemia, is highly expressed not only in the intestine, but also in the liver in humans, although the pathophysiological importance of hepatic NPC1L1 in atherosclerotic diseases remained unclear. By using novel mouse models to separately analyze the effects of hepatic and intestinal NPC1L1 on the development/progression of atherosclerosis, we first demonstrated that hepatic NPC1L1 accelerates Western diet-induced atherosclerotic plaque formation in an intestinal NPC1L1-dependent and an ezetimibe-sensitive manner.

As in many naturally formed networks, the brain exhibits an inherent modular architecture that is the basis of its rish operability, robustness and integration-segregation capacity.

Single neurons in an autaptic culture exhibit various types of firing pattern with different firing durations and rhythms. However, a neuron with autapses has often been modeled as an oscillator providing a monotonic firing pattern with a constant periodicity because of the lack of a mathematical model. In the work described in this study, we use computational simulation and whole-cell patch-clamp recording to elucidate and model the mechanism by which such neurons generate various firing pattens. In the computational simulation, three types of spontaneous firing pattern, i.e., short, long-lasting, and periodic burst firing patterns are realized by changing the combination ratio of N-methyl-d-aspartate (NMDA) to α-amino-3-hydroxyl-5-methyl-4-isoxazole-propionate (AMPA) conductance. These three types of firing patterns are also observed in the experiments where neurons are cultured in isolation on micropatterned substrates. Using the AMPA and NMDA current models, we discuss that, in principle, autapses can regulate rhythmicity and information selection in neuronal networks.

BACKGROUND: Electrical stimulation of neurons is essential to control neuronal activities. Thus far, direct stimulation by whole-cell patch-clamp methods [1] and extracellular stimulation using multi-electrode arrays (MEAs) [2] have been performed commonly. While the whole-cell patch-clamp method merits from the precise control of stimulation, the use of MEAs takes advantage of maintaining neuronal activity over a long period owing to its non-invasiveness. An optogenetic technique [3] has a targeting precision of a single-cell level, but this has difficulties in applying fast stimulation patterns due to the slow photoresponse of opsin [4]. Alternative non-invasive methods capable of targeting a single neuron with arbitrary stimulation patterns open new possibility of controlling and monitoring neuronal activities over a long duration. In this presentation, we propose a novel method to combine cell-patterning techniques [5] with electrical stimulation of a target cell with a pair of needle electrodes [6]. We show that the firing of a single neuron cultured on the micropattern can be controlled by the proposed method. We also show the controllability of the stimulation by analogue circuit simulation of a neuron stimulated electrically by a pair of needle electrodes via culture medium. EXPERIMENTAL METHODS: Neurons were cultured on a micropatterned glass substrates in isolation. A pair of tungsten needles except for their tip apexes were coated with an insulating polymer and mounted on manipulators. Two needles were immersed in the medium so that the target neuron is placed in-between. The distance from the neuron to the needle tip was controlled to be several tens to 100 μm. A bipolar square pulse was applied between electrodes, and the neuronal activity was visualized by Ca imaging. SIMULATION: The membrane potential response of a neuron to extracellular stimulation was analysed using LTspice simulator [7]. The circuit model is shown in Fig. 1a. The integrate-and-fire neuron was connected electrically with two needle electrodes via the electric double layers and solution resistances. The key of the circuit simulation is to divide the neuron into two compartments, each is stimulated with either of two needle electrodes. We conjecture that the neuron fires if the membrane potential either of two compartments exceeds the threshold upon the electrical stimulation. We investigated the membrane potential response when the resistance corresponding to the relative distance from the neuron to the electrode, the capacitive component of the needles, and the shape of voltage input pulses were changed. RESULTS AND DISSCUSSION: As shown in Fig. 1b, the neuron fires if the amplitude of the bipolar square pulse exceeds 5 Vp-p. The amplitude required for the neuron firing decreases with increasing distance from the neuron to the needle electrodes. The LTspice simulation provided the following results: (i) The positive electrode hyperpolarizes the membrane potential, whereas the negative electrode depolarizes it, indicating the membrane potential of two compartments changes to inverse direction even if a bipolar pulse was applied, as shown in Fig. 1c. (ii) The closer the electrode is to the cell, the more the membrane potential of the neuron changes and the voltage drop in the solution resistance is comparatively small. (iii) The main factor of neuronal firing is the transient potential fluctuation caused by the capacitive component of the electrical double layer and the neuronal membrane. (iv) The membrane potential of both the compartments is sensitive to transient change of tip voltage and does not change if the system reaches the steady state. This is also true if the DC voltage is continuously applied. These results indicate that applying a voltage pulse of appropriate amplitude can stimulate only cells in the vicinity of needle electrodes. Only a single neuron in neuronal culture can be stimulated by the needle electrodes by placing the target neuron between two needles.
