Resumen tomado de la publicación Respuesta psicofisiológica a estresores sociales: relevancia del sexo y la edad. Antecedentes: entender los factores involucrados en la respuesta psicofisiológica ante situaciones de estrés agudo es crucial para prevenir y tratar problemas de salud relacionados con el estrés. El objetivo es integrar los resultados de estudios en humanos relacionados con el papel del sexo y la edad en las diferencias individuales en la respuesta de estrés en diferentes biomarcadores. Metodología: se integran los principales resultados de nuestro grupo de investigación y de otros laboratorios centrados en las diferencias debidas al sexo y la edad en la respuesta psicofisiológica a situaciones de estrés psicosocial. Resultados: se observan diferencias en la respuesta de estrés entre hombres y mujeres en presión sanguínea y en niveles de cortisol, pero no en frecuencia cardíaca, variabilidad de la frecuencia cardíaca y alfa-amilasa. Además, la edad influye en los niveles de cortisol y en la actividad basal del sistema nervioso simpático. Conclusiones: los resultados ponen de manifiesto la importancia de tener en cuenta el sexo y la edad de los participantes para poder entender la respuesta de estrés y sus posibles efectos en salud. The literature has been relatively silent about post-conflict processes. However, understanding the way humans deal with post-conflict situations is a challenge in our societies. With this in mind, we focus the present study on the rationality of cooperative decision making after an intergroup conflict, i.e., the extent to which groups take advantage of post-conflict situations to obtain benefits from collaborating with the other group involved in the conflict. Based on dual-process theories of thinking and affect heuristic, we propose that intergroup conflict hinders the rationality of cooperative decision making. We also hypothesize that this rationality improves when groups are involved in an in-group deliberative discussion. Results of a laboratory experiment support the idea that intergroup conflict ¿associated with indicators of the activation of negative feelings (negative affect state and heart rate)¿ has a negative effect on the aforementioned rationality over time and on both group and individual decision making. Although intergroup conflict leads to sub-optimal decision making, rationality improves when groups and individuals subjected to intergroup conflict make decisions after an in-group deliberative discussion. Additionally, the increased rationality of the group decision making after the deliberative discussion is transferred to subsequent individual decision making.

Con el envejecimiento de la población es importante comprender los cambios neurofisiológicos que se producen. En este contexto, las quejas subjetivas de la memoria (QSM) se están convirtiendo en un tema de interés ya que se han considerado un factor de riesgo para desarrollar trastornos relacionados con demencias como el deterioro cognitivo leve y enfermedad de Alzheimer. Por lo tanto, es importante investigar los biomarcadores tempranos para la detección y la planificación de los tratamientos. En este sentido, el procesamiento de emociones faciales es una importante función cognitiva comúnmente afectada en personas con trastornos relacionados con demencias que podría ayudar a analizar cambios cerebrales funcionales en las personas con QSM. El procesamiento de expresiones faciales puede ser investigado usando una técnica relativamente económica, no invasiva y con alta resolución temporal como el electroencefalograma (EEG). En este sentido, se ofrece una visión del procesamiento de las emociones faciales en personas jóvenes y mayores con y sin QSM, así como los principales potenciales relacionados con eventos (PREs) utilizados junto con medidas conductuales de tiempo de reacción (RT) y precisión. Otra forma de explorar cambios en la funcionalidad del cerebro en las personas con QSM es registrando los ritmos electroencefalográficos en estado de reposo, la reactividad del EEG y la frecuencia del pico de alpha. Estos parámetros se registran en el cuero cabelludo de las personas durante períodos breves (es decir, minutos) con los ojos cerrados y los ojos abiertos. En resumen, el objetivo general de esta tesis ha sido avanzar en el conocimiento de algunos factores neurofisiológicos que pudieran permitir identificar el deterioro y los cambios cognitivos antes de que sean observados en las pruebas neuropsicológicas. El capítulo uno presenta una introducción general. El capítulo dos incluye los principales objetivos e hipótesis. En el capítulo tres se examina si el procesamiento de las emociones faciales es diferente en los jóvenes con y sin QSM que fueron expuestos a caras positivas, negativas y neutras. Para ello, los dos componentes (N170 y LPP) usualmente utilizados en esta tarea fueron examinados, así como los datos conductuales tiempo de reacción (TR) y precisión. En cuanto a los datos conductuales, ni el TR ni la precisión fueron significativamente diferentes entre los grupos con y sin QSM. En lo que respecta a los PREs, encontramos que los participantes con QSM obtuvieron amplitudes más bajas en el componente N170 en comparación con los participantes sin QSM. El capítulo cuatro determina si existen diferencias en los correlatos neuronales y las medidas conductuales del procesamiento de las emociones faciales entre las personas mayores con y sin QSM, así como una posible asociación entre las medidas de PREs y rendimiento ejecutivo. Los resultados mostraron que las personas mayores con QSM eran más lentas y menos precisas que los controles. A nivel de los PREs, los hombres con QSM revelaron latencias más largas en el componente N170 para caras positivas, negativas y neutras que las mujeres con QSM. El capítulo cinco presenta los análisis de la potencia espectral de las bandas de frecuencia del estado de reposo del EEG y la reactividad del EEG en personas mayores y jóvenes con y sin QSM. Además, exploramos si existe una correlación entre las potencias espectrales específicas de cada región y medidas neuropsicológicas. Los resultados mostraron que, en comparación con los controles, las personas mayores con QSM tenían una mayor potencia espectral en la banda theta y una sutil pérdida de reactividad en alpha en la apertura de ojos. El capítulo seis investiga los parámetros de alpha (pico de frecuencia y amplitud), en jóvenes y mayores con QSM y en las personas del grupo control. Por último, se analiza la asociación entre reserva cognitiva (RC) y los parámetros de alpha. Los resultados no mostraron diferencias en el pico de frecuencia de alpha o la amplitud entre las personas mayores y jóvenes con QSM y los controles. Además, no se detectaron diferencias significativas por sexo. El capítulo siete contiene una discusión general y los principales hallazgos, las limitaciones y los puntos fuertes de esta tesis, y las direcciones para futuras investigaciones. El capítulo ocho presenta las principales conclusiones. Por último, el capítulo nueve, incluye un resumen general en español.

El estrés ha sido considerado uno de los principales problemas de la sociedad actual, ya que se ha relacionado con diversas condiciones de salud mental y física, así como con enfermedades propias de la edad y problemas de memoria. Sin embargo, existen diferencias individuales en la forma en que las personas enfrentan situaciones estresantes. En este sentido, algunas personas, a pesar de estar expuestas a los mismos niveles de estrés, no desarrollan patologías, lo que sugiere que ciertos factores psicológicos podrían actuar como protectores de los efectos del estrés. La resiliencia, que engloba pensamientos positivos y optimistas, ha sido entendida como la capacidad de afrontar la adversidad y recuperarse de situaciones estresantes. Más importante aún, se ha propuesto como un factor importante asociado con una mejor salud y un envejecimiento exitoso. Entre los trastornos relacionados con la edad, se ha prestado mayor atención al deterioro cognitivo y la detección temprana de la demencia. La prioridad actual es estudiar qué factores podrían prevenir o retrasar el deterioro cognitivo. En este sentido, las Quejas Subjetivas de Memoria (QSM) a lo largo del tiempo, o el Deterioro Cognitivo Subjetivo (SCD), se han sugerido indistintamente como un proxy para la detección temprana de demencia en personas mayores. Sin embargo, estas QSM también aparecen en individuos jóvenes, lo que aumenta la necesidad de determinar qué factores podrían estar asociados con este fenómeno en las diferentes edades. Todo ello pone de relieve la importancia de estudiar cómo determinados factores psicológicos, especialmente los protectores, podrían estar relacionados con el estrés y las SMC a lo largo de la vida, pero especialmente en las personas mayores por ser una etapa de mayor vulnerabilidad tanto al estrés como a la pérdida de memoria. En esta tesis doctoral se abordan los dos principales temas de preocupación en las sociedades actuales, es decir, el estrés y la detección precoz de las demencias y cómo pueden afectarles diferentes factores psicológicos. Para ello, se han relacionado factores psicológicos de resiliencia (positividad, optimismo) y vulnerabilidad (pesimismo) con diferentes biomarcadores de estrés (saliva y cabello) y contextos (respuesta a un estresor y estrés crónico) en personas mayores sanas. En segundo lugar, la relación entre las QSM y diferentes factores de vulnerabilidad (ansiedad y depresión), factores protectores (resiliencia y positivismo), estrés (funcionamiento basal del eje hipotalámico pituitario suprarrenal [eje HPA]) y biomarcadores neurodegenerativos de la enfermedad de Alzheimer (EA) y Se han abordado las enfermedades cerebrovasculares (ECV). Este segundo objetivo general también se ha abordado en diferentes grupos de edad (es decir, jóvenes sanos, personas de mediana edad y mayores). En definitiva, esta tesis doctoral ayuda a clarificar el papel de algunos indicadores psicológicos y de estrés, con el fin de potenciar y fomentar futuras investigaciones e intervenciones enfocadas a la mejora de la salud y el envejecimiento saludable. Stress has been considered one of the main problems in today’s society, given that it has been related to several mental and physical health conditions, as well as age-related diseases and memory problems. However, there are individual differences in the way people face stressful situations. In this sense, some people, despite being exposed to the same levels of stress, do not develop pathologies, suggesting that certain psychological factors could act as protectors from the effects of stress. Resilience, which encompasses positive and optimistic thoughts, has been understood as the ability to face adversity and recover from stressful situations. More importantly, it has been proposed as an important factor associated with better health and successful aging. Among the age-related disorders, greater attention has been paid to cognitive decline and early dementia detection. The current priority is to study which factors could prevent or delay cognitive impairment. In this regard, Subjective Memory Complaints (SMCs) over time, or Subjective Cognitive Decline (SCD), have been indistinctly suggested as a proxy for early dementia detection in older people. However, these SMCs also appear in young individuals, increasing the need to determine which factors could be associated with this phenomenon at different ages. All of this highlights the importance of studying how certain psychological factors, especially protective factors, could be related to stress and SMCs across the lifespan, but especially in older people because it is a stage of greater vulnerability to both stress and memory decline. In this doctoral thesis, the two main issues of concern in today's societies are addressed, that is, stress and early detection of dementia and how different psychological factors can affect them. For this purpose, psychological factors of resilience (positivity, optimism) and vulnerability (pessimism) have been related to different stress biomarkers (saliva and hair) and contexts (response to a stressor and chronic stress) in healthy older people. Secondly, the relationship between SMCs and different vulnerability factors (anxiety and depression), protective factors (resilience and positivism), stress (basal functioning of the Hypothalamic Pituitary Adrenal axis [HPA axis]), and neurodegenerative biomarkers of Alzheimer’s disease (AD) and Cerebrovascular Disease (CVD) have been addressed. This second general objective has also been addressed in different age groups (i.e., healthy young, middle-aged, and older people). In sum, this doctoral thesis helps to clarify the role of some psychological and stress indicators, in order to enhance and encourage future research and interventions focused on improving health and healthy aging.

El sentiment de solitud és un dels principals problemes de les societats occidentals. En els últims anys els avanços en internet ens han portat a ser una societat més connectada que mai, a pesar d’açò, aquest sentiment s’està incrementant. Per sobreviure i prosperar, els éssers humans necessitem dels altres per satisfer les necessitats d’afecte, pertinença i reconeixement. D’aquesta manera, la solitud sorgeix de la discrepància entre les relacions íntimes i socials desitjades i les reals. La investigació indica que la solitud està relacionada amb l’estrès i amb diverses conseqüències en la salut tant física com psicològica. Aquesta es pot experimentar al llarg de la vida, encara que és més freqüent entre la població gran. Tot açò emfatitza la importància d’estudiar-la i profunditzar en la seua influència en el benestar de la nostra societat. En aquesta tesi doctoral, la solitud s’aborda en relació a diferents estressors (estrès als primers anys de vida (ELS, per les seues sigles en anglès) i estrès durant la vida adulta) i diferents indicadors de salut subjectius (salut psicològica, salut física i declivi cognitiu subjectiu (SCD, per les seues sigles en anglès)), objectius (mesures del funcionament de l’eix hipotàlem-hipòfisi-adrenal (HPA, per les seues sigles en anglès) i biomarcadors de patologia en la Malaltia D’Alzheimer (AD, per les seues sigles en anglès) i en la Malaltia Cerebrovascular (CVD, per les seues sigles en anglès)) en persones sanes joves, de mitjana edat i grans. El primer capítol és una introducció general als estudis inclosos en aquesta tesi doctoral. Breument, es descriu l’actual literatura sobre solitud i la seua relació amb l’estrès i la salut. En primer lloc, es presenta la relació entre les experiències adverses en etapes primerenques de la vida i el sentiment de solitud durant l’etapa adulta. Igualment, s’assenyala que aquest sentiment es pot entendre com un factor estressant, i es presenten alguns estudis que informen dels seus efectes sobre la salut física i psicològica. A més, es revisa la literatura sobre la relació tant de l'ELS com de la solitud amb el funcionament basal de l'eix HPA ja que és un important sistema endocrí implicat en la salut i l'estrès. Finalment, el capítol descriu les darreres investigacions sobre el possible paper de la solitud en el context de l’SCD, suggerida com l'etapa preclínica més primerenca de l’AD, i la seua relació amb la patologia cerebral precoç de l’AD i CVD. L'última part del capítol inclou els principals objectius i hipòtesis d'aquesta tesi doctoral. Els tres capítols següents descriuen els tres estudis que en formen part. Cadascun d'aquests capítols conté una breu introducció que descriu la literatura relacionada més important, la metodologia específica utilitzada i els resultats trobats, i una discussió de les principals troballes. En el segon capítol, s’explica el primer estudi. Aquest estudi examina el paper de la solitud com a mediadora entre l'ELS i la seua relació amb la percepció de l'estrès i el funcionament basal de l'eix HPA durant l'edat adulta en persones joves i de mitjana edat sanes. En el tercer capítol, es descriu el segon estudi, que analitza la solitud i la seua associació amb la salut subjectiva psicològica i física i el funcionament basal de l'eix HPA en persones sanes de mitjana edat i gent gran i si aquestes associacions difereixen en funció del sexe. Finalment, en aquest capítol, s'explora si el funcionament basal de l'eix HPA funciona com a mecanisme subjacent en la relació entre la solitud i la salut. El quart capítol inclou el tercer estudi. Aquest estudi investiga el paper d’aquest sentiment en el context de l’SCD com a possible indicador primerenc de l'AD i la CVD en persones grans no deteriorades cognitivament. El cinquè capítol analitza les principals conclusions dels estudis empírics esmentats anteriorment. També es discuteixen les limitacions i els punts forts d'aquests estudis, així com les direccions per a futures investigacions. El sisè capítol inclou les principals conclusions. Finalment, el setè capítol consta d'un resum de la present tesi doctoral en valencià. Loneliness is one of the main issues in Western societies. Internet advances in recent years have made us a more connected society than ever, but despite this, loneliness is increasing. For survival and advancement, human beings need others to satisfy their needs for affection, belonging, and recognition. Thus, loneliness arises from the discrepancy between desired and actual social and intimate relationships. Research indicates that feelings of loneliness can be understood as a stressor, and it is related to several adverse consequences for physical and psychological health. This feeling can be experienced throughout life, although it is more frequent among the older population. All of this emphasizes the importance of studying loneliness and its influence on the well-being of our society. In this doctoral thesis, loneliness is addressed in relation to different stressors (Early Life Stress (ELS) and stress during adulthood) and different subjective (psychological, physical, and subjective cognitive decline (SCD)) and objective (basal Hypothalamic-Pituitary-Adrenal axis (HPA axis) functioning measures and biomarkers of Alzheimer’s Disease (AD) and Cerebrovascular Disease (CVD) pathology) health indicators in young, middle-aged, and older healthy people. The first chapter is a general introduction to the studies included in this thesis. Briefly, it describes the current literature on loneliness and its relationship with stress and health. In the first place, it presents the relationship between adverse experiences in the early stages of life and feelings of loneliness during adulthood. Likewise, it points out that loneliness can be understood as a stressor, and some studies that report its effects on physical and psychological health are presented. In addition, the literature on the relationship of both ELS and loneliness with the basal HPA axis functioning is reviewed because it is an important endocrine system involved in health and stress. Finally, the chapter describes the latest research on the possible role of loneliness in the context of SCD, suggested as the earliest preclinical stage of AD, and its relationship with early brain pathology of AD or CVD. The last part of the chapter includes the main aims and hypotheses of this doctoral thesis. The following three chapters describe the three studies in this doctoral thesis. Each of these chapters contains a brief introduction that describes the most important related literature, the specific methodology used and results found, and a discussion of the main findings. In the second chapter, the first study is explained. This study examines the role of loneliness as a mediator between early life stress and its relationship with the perception of stress and the basal HPA axis functioning during adulthood in healthy young and middle-aged people. In the third chapter, the second study is described, which analyzes loneliness and its association with subjective psychological and physical health and basal HPA axis functioning in healthy middle-aged and older people and whether these associations differ depending on sex. Finally, in this chapter, we explore the basal HPA axis functioning as an underlying mechanism in the relationship between loneliness and health. The fourth chapter includes the third study. This study investigates the role of loneliness in the context of SCD as a possible early indicator of AD and CVD in cognitively unimpaired older people. The fifth chapter discusses the main findings of the empirical studies mentioned above. The limitations and strengths of these studies are also discussed, as well as directions for future studies. The Sixth chapter includes the main conclusions. Finally, the seventh chapter consists of a summary of the present doctoral thesis in Valencian.

The world population is getting older, and this age group faces important health challenges, among which age-related cognitive decline stands out. Therefore, it is important to identify vulnerability and protective health factors in order to understand ways to prevent cognitive decline. This thesis aimed to address the relationship between the Hypothalamic–Pituitary–Adrenal (HPA) axis and cognitive functioning during aging, taking into account genetic (Apolipoprotein E, ApoE, polymorphism), situational (loneliness), and individual factors (personality traits). To do this, a group of healthy older people over 55 years of age, men and women, were evaluated. A neuropsychological battery was administered to assess global cognition, attention, executive function, working and declarative verbal memory. Participants also provided saliva samples to measure the HPA-axis functioning (cortisol levels) and the ApoE genotype. In addition, participants completed the revised UCLA loneliness scale, the NEO-Five Factor Inventory to measure the big five personality traits, the Cognitive Reserve Questionnaire, and the Short Form Health Survey (SF-36) to assess subjective physical and mental health. Furthermore, different objective health measures were also obtained: Body Mass Index (BMI), Waist-Hip Ratio (WHR), Glycated Hemoglobin (HbA1c), Low-Density Lipoprotein (LDL) and High-Density Lipoprotein (HDL) Cholesterol, and Pulse Pressure. The main results/conclusions of this thesis are, first, that the ApoE-ε2 allele has been shown to be a protective factor for learning ability. The ApoE-ε3 allele was related to a more adaptive HPA-axis response, with beneficial effects on cognitive performance, whereas the ApoE-ε4 allele could be a vulnerability factor in the adverse effects of HPA-axis dysregulation on cognition. In addition, loneliness was associated with a dysregulation of the HPA-axis. Furthermore, loneliness was not associated directly with cognitive function in healthy older adults, but it was indirectly related to worse cognitive performance via higher bedtime cortisol levels. Therefore, the dysregulation of the HPA-axis appears to be one of the mechanisms underlying the relationship between loneliness and cognitive impairment. Concerning personality dimensions, our results confirm that neuroticism is an important health risk factor in aging because it is associated with a dysregulation of the HPA-axis, decline in attention via a dysregulation of the HPA-axis, and worse objective and subjective health. In contrast, conscientiousness appears as a robust protective factor for health in older people because it is associated with healthier HPA-axis functioning and better objective and subjective health; however, it is not a predictor of cognitive change in healthy older adults. Regarding extraversion, it could protect against decline in executive function and delayed memory recall, but favor decline in immediate memory recall, which could be explained by cognitive reserve. Openness and agreeableness did not seem important predictors of health outcomes in older people, but they are protective factors against decline in immediate recall via cognitive reserve, and delayed recall, respectively. Finally, it is worth noting that regarding to the sex/gender, our results suggest that neuroticism is a greater risk factor for decline in working memory in men, and for health in women, whereas extraversion is a protective factor for health in women, but a risk factor in men.
