Abstract

This article describes the health-care access experiences of Indigenous members of a Gulf Coast, non-federally recognized tribe. Research exploring the experiences of non-federally recognized tribes is needed, as these tribes lack resources available to federally recognized tribes. Using a qualitative description research approach and through partnership with a community advisory board, thirty-one semi-structured life-course interviews were conducted with women tribal members. A qualitative descriptive analytic approach revealed the following key themes: First Health-Care Experiences; Going to Family Members for Health Care; Going to Indigenous Healers for Health Care and; Generational Changes in the Transmission of Traditional Knowledge. Our findings highlight the role that family members and Indigenous healers play in addressing health-care gaps and needs for tribal members. In addition, results suggest that Indigenous healers are respected, valued members of the community, and there is concern that healing knowledge is not being passed down to future generations. This research addresses a gap in the need for holistic understandings of Indigenous women’s reproductive and sexual health care, which is required for the development of interventions which not only address social justice issues and weaknesses in the health-care system, but that also promote the existing strengths and resources in Indigenous communities.

10. The Influence of Alcohol and Weapon Presence on Eyewitness Memory and Confidence

Abstract

In this quasi-experimental field study, bar drinkers (0.00-0.23% blood alcohol content) viewed a photographic sequence in which a male took a laptop from a helpdesk assistant, either on loan or at gunpoint. Following a brief retention period, participants answered 20 multiple-choice questions about the male, his actions, and details of the scene, then attempted to identify him from a simultaneous target-present or target-absent line-up. Alcohol was associated with a reduction in correct identifications and an increase in false identifications. Surprisingly, the presence of a weapon in the scene enhanced identification accuracy, though wider scene memory was not influenced by alcohol or the weapon. Findings offer some support for the view that alcohol restricts face encoding, perhaps through the narrowing of attention to salient external features (e.g., hair). We also suggest that curiosity about mock-crime perpetrators may produce weapon focus reversals, although the factors that might elicit such curiosity remain unclear.

Abstract

Introduction: Metabolic adaptability, including glucose metabolism, enables cells to survive multiple stressful environments. Glycogen may serve as a critical storage depot to provide a source of glucose during times of metabolic demand during the metastatic cascade; therefore, understanding glycogen metabolism is critical. Our goal was to determine mechanisms driving glycogen accumulation and its role in metastatic (MCF10CA1a) compared to nonmetastatic (MCF10A-ras) human breast cancer cells. Methodology:13C6-glucose flux analysis in combination with inhibitors of the gluconeogenic pathway via phosphoenolpyruvate carboxykinase (PCK), the anaplerotic enzyme pyruvate carboxylase (PC), and the rate-limiting enzyme of the pentose phosphate pathway (PPP) glucose 6-phosphate dehydrogenase (G6PD). To determine the requirement of glycogenolysis for migration or survival in extracellular matrix (ECM) detached conditions, siRNA inhibition of glycogenolysis (liver glycogen phosphorylase, PYGL) or glycophagy (lysosomal enzyme α-acid glucosidase, GAA) enzymes was utilized. Results: Metastatic MCF10CA1a cells had 20-fold greater glycogen levels compared to non-metastatic MCF10A-ras cells. Most glucose incorporated into glycogen of the MCF10CA1a cells was in the five13C-containing glucose (M+5) instead of the expected M+6 glycogen-derived glucose moiety, which occurs through direct glucose conversion to glycogen. Furthermore,13C6-glucose in glycogen was quickly reduced (~50%) following removal of13C-glucose. Incorporation of 13C6-glucose into the M+5 glucose in the glycogen stores was reduced by inhibition of PCK, with additional contributions from flux through the PPP. Further, inhibition of PC reduced total glycogen content. However, PCK inhibition increased total unlabeled glucose accumulation into glycogen, suggesting an alternative pathway to glycogen accumulation. Inhibition of the rate-limiting steps in glycogenolysis (PYGL) or glycophagy (GAA) demonstrated that both enzymes are necessary to support MCF10CA1a, but not MCF10A-ras, cell migration. GAA inhibition, but not PYGL, reduced viability of MCF10CA1a cells, but not MCF10A-ras, in ECM detached conditions. Conclusion: Our results indicate that increased glycogen accumulation is primarily mediated through the gluconeogenesis pathway and that glycogen utilization is required for both migration and ECM detached survival of metastatic MCF10CA1a cells. Abstract

Objective: This study sought to understand the roles of partners in pregnancy, childbirth, and childrearing in Indigenous communities. It explores supportive and unsupportive attitudes in the relationships mothers experienced, and how these relationships affected their lives. Background: Settler colonialism has negatively impacted Indigenous communities. However, little research has explored how it has influenced partner and social support during the perinatal time period. Methods: Through 31 semistructured interviews with women from a tribe in the southeastern United States, participants described their experiences with supportive and unsupportive partners and how that shaped their experiences during pregnancy, childbirth, and childrearing. Results: Participants described themes of (a) supportive and unsupportive partner relationships, (b) importance of partner support in childbirth, and (c) mothers as primary caregivers. Conclusion: Results suggest that women experienced a wide range of partner relationships, and they relied more on their female family members to provide support during this time, rather than counting on their male counterparts. Implications: Health care providers may want to take these findings into consideration when approaching their patient's care. 13. NURTURE THROUGH NATURE – THE IMPACT OF THE ENVIRONMENT ON PROSPECTIVE MEMORY

Abstract

There is a need for brief, accessible interventions to improve cognitive functioning, including prospective memory. Previous research has found that exposure to nature is linked to benefits for cognition and mood, including improved executive function, working memory, creativity, attention, and mood. However, there is a dearth of literature exploring the relationship between nature exposure and prospective memory. Additionally, benefits for cognition and mood have been observed with direct nature exposure and indirect exposure through videos and images. The present study sought to evaluate the impact of natural setting versus urban setting video exposure on prospective memory, creativity, hope, and affect. Data was gathered from 122 participants, and data included participants’ demographics and scores on tests of prospective memory, creativity, affect, and hope. Participants were quasi-randomly assigned to either the natural setting or urban setting group. All participants were asked to watch a 20-minute video of either a natural or urban setting, immediately followed by a battery of tests. Results indicated a statistically significant difference between groups on time-based and event-based prospective memory scores, with participants in the natural setting group demonstrating better performance. No significant differences were found between groups for creativity, affect, or hope. The observed benefits to prospective memory are novel and contribute to a growing body of literature outlining benefits of nature exposure on cognitive function. The present study also contributes to existing literature supporting the use of video exposure to nature to improve cognition. Overall, the current study demonstrates the feasibility of a brief, accessible intervention, through video exposure to nature, to improve prospective memory.

28. The Prevalence of PTSD Among an Online Sample of US Familial Dementia Caregivers

Abstract

The increased prevalence of dementia has increased the number of caregivers. Familial caregivers can suffer emotionally as a result of caregiving. Despite caregiver organizations campaigning for awareness of caregiver trauma, more research examining PTSD among US familial dementia caregivers is needed. The present study surveyed an online sample of US familial dementia caregivers to gather a prevalence rate of PTSD. We hypothesized that these caregivers experience an equal or greater prevalence of PTSD compared to the general US population, nurses, emergency medical responders, and veterans. An online survey containing the PTSD-Civilian Checklist (PCL-C) was administered to familial dementia caregivers across the US. Data (n = 23) revealed that 26.1% of participants surveyed screened positive for PTSD criteria. Consistent with our hypothesis, this percentage is greater than the prevalence rates for the general US population and is similar to the prevalence rates for emergency first responders, nurses, and even US military veterans. These findings can greatly benefit US dementia caregivers by spreading awareness and helping allocate resources to assist those struggling with PTSD.

42. THE ROLE OF GLYCOGEN ACCUMULATION AND UTILIZATION IN METASTTIC BREAST CANCER PROGRESSION

Abstract

Breast cancer is a significant public health concern being the second leading cause of cancer-related death in women, with a projected 43,250 deaths in the US in 2022. However, cancer progression to metastatic sites is the primary cause of death in breast cancer patients. A hallmark of cancer is the dysregulation of cellular metabolism. Cancer cells have the ability to hijack their metabolism and drive cellular processes supporting cancer progression. As cancer cells continue through the metastatic cascade, they are challenged with various bioenergetic processes that can be supported by the influx of glucose. Thus, altering glycogen accumulation, where glucose is stored in cells, may be beneficial in supporting cancer progression. In this study, we aim to determine what drives glycogen accumulation in metastatic cells and if it is utilized to support cancer progression. We employed the non-metastatic MCF10A-ras and the metastatic MCF10CA1a cells for these studies. Our results demonstrate that metastatic MCF10CA1a have 20-fold accumulation of glycogen compared to the MCF10A-ras cells. Utilizing 13C6-glucose flux analysis, surprisingly, most of the glucose incorporated into glycogen of the MCF10CA1a cells was in the M+5 glucose labeling pattern instead of the expected M+6 pattern which occurs when glucose is directly converted to glycogen. We showed that glycogen was accumulated due to increased gluconeogenesis through cataplerosis (PEPCK) utilizing inhibitors of the enzyme. Additionally, in a pulse-chase experiment using 13C6-glucose flux analysis, there was an approximate 50% reduction in labeled glucose in glycogen, 3 hours after removing the label, suggesting that the MCF10CA1a cells also have a rapid turnover of glycogen. Glucose can be released through two mechanisms, glycogenolysis or glycophagy. Utilizing siRNAs to a rate limiting steps in each pathway, results suggest both glycogenolysis (PYGL) and glycophagy (GAA) are necessary to support cell migration, a critical step in metastasis of the MCF10CA1a cells. Thus, glycogen metabolism is dysregulated in the MCF10CA1a breast cancer cells such that they have increased glycogen accumulation and that glycogen is required to support cell migration. Further understanding the mechanism by which glucose is accumulated and released in a specific cancer and in specific steps or stressors in cancer progression may contribute to potential therapeutic targets to help mitigate metastasis, and potentially breast cancer mortality.
