Abstract

BackgroundEver since the first atomic structure of an enzyme was solved, the discovery of the mechanism and dynamics of reactions catalyzed by biomolecules has been the key goal for the understanding of the molecular processes that drive life on earth. Despite a large number of successful methods for trapping reaction intermediates, the direct observation of an ongoing reaction has been possible only in rare and exceptional cases.ResultsHere, we demonstrate a general method for capturing enzyme catalysis "in action" by mix-and-inject serial crystallography (MISC). Specifically, we follow the catalytic reaction of the Mycobacterium tuberculosis β-lactamase with the third-generation antibiotic ceftriaxone by time-resolved serial femtosecond crystallography. The results reveal, in near atomic detail, antibiotic cleavage and inactivation from 30 ms to 2 s.ConclusionsMISC is a versatile and generally applicable method to investigate reactions of biological macromolecules, some of which are of immense biological significance and might be, in addition, important targets for structure-based drug design. With megahertz X-ray pulse rates expected at the Linac Coherent Light Source II and the European X-ray free-electron laser, multiple, finely spaced time delays can be collected rapidly, allowing a comprehensive description of biomolecular reactions in terms of structure and kinetics from the same set of X-ray data.

20. Isles of Boshen : Edward Lear's literary nonsense in context

Abstract

This thesis investigates three major areas in the background of Edward Lear's literary nonsense: the parodic relationship with text and genre of early children's literature, the trends behind Lear's innovative illustration style, and the "nonsense" child construct manifest within the genre, which I claim is, in many ways, an expression of the Romantic conception of the child. The first chapter explores the parodic basis of nonsense. Most literary nonsense is referential; it often begins by inhabiting a genre or individual work, but what it does to the original is debatable. Some critics see nonsense as parody, while others claim that nonsense precludes parody in its intentional purposelessness. In this chapter I explore the critical debate surrounding parody in nonsense, and parody in general. I then examine the works of Lear, and some Carroll, looking first at their genuine, clear parodies. Next, I look at the many borderline cases of parody which use nonsense as a device but are not overshadowed by it. Finally, I discuss the more "pure" literary nonsense which, I argue, goes beyond parody to establish a new genre. The next chapter looks at the background of Lear's nonsense illustration. His style of illustration was a widely original combination of devices which are best seen in the context of the children's book illustrations of his day. With Bewick's innovations in woodcuts, the quality of children's illustrations had drastically improved. Diverging from this trend, Lear's illustrations hearken back to the rough chapbooks which he probably read as a child. His child-like style, coupled with an expert draughtsman's eye, began a rival tradition of children's book illustration. His illustrations are in way caricatures of chapbooks. His text and illustrations, like those of Blake and Hood, are integral, and their self-reflexiveness with the verses places them in an altogether different class of illustration.

Abstract

Despite their fundamental and applied importance, a general model to predict the natural breakup length of steady capillary jets has not been proposed yet. In this work, we derive a scaling law with two universal constants to calculate that length as a function of the liquid properties and operating conditions. These constants are determined by fitting the scaling law to a large set of experimental and numerical measurements, including previously published data. Both the experimental and numerical jet lengths conform remarkably well to the proposed scaling law. This law is explained in terms of the growth of perturbations excited by the jet breakup itself.

Abstract

The Dynamic Cryptographic Context Management (DCCM) project efficiently provides security for very large, dynamically changing groups of participants. The DCCM system has two novel distinguishing characteristics. First, policy plays a key role in DCCM. Groups at all levels have policies. These policies are represented, they are negotiated, and a cryptographic context - an unambiguous set of mechanisms and configuration - is created to make particular interactions possible subject to these policies. Second, DCCM implements a scalable key management system based on One-way Function Trees (OFT) that can handle group sizes up to 100,000 members and can dynamically handle members entering and leaving groups. This report culminates the research of the project by presenting the integrated architecture that has been developed and implemented to demonstrate the fulfillment of our research goals.
