This study aims to determine the effect of administration of ciplukan (Physalis angulate Linn.) leaf extract on the variables of testicular weight, seminiferous tubules diameter, and spermatozoa plasma membrane integrity of rats (Rattus norvegicus) exposed to heat as model animals. Rats were divided randomly into five groups. In the NC group, rats were not exposed to heat and were only given 1% Na-CMC. In the PC, T1, T2, and T3 rats were exposed to heat followed by administration of 1% Na-CMC, 100, 200, and 400 mg/kg bw of ciplukan leaf extract (CLE) in 1% Na-CMC. Heat exposure at 40°C was conducted for 60 minutes every day for 21 days. On day-22, all rats were sacrificed for testicular evaluation. The results showed that all variables in the PC group were lower (p <0.05) than in the NC group. All variables in the T3 group were higher (p <0.05) than in the PC group. Testicular weight and spermatozoa plasma membrane integrity in the T3 group were not significantly different (p >0.05) from the NC group. However, the diameter of the seminiferous tubules in the T3 group were lower (p <0.05) than in the NC group. This study revealed that the administration of ciplukan leaf extract as an antioxidant increased testicular weight, seminiferous tubules diameter, and spermatozoa with intact plasma membrane in rats exposed to heat. New antimicrobial materials have drawn research and development attention due to antimicrobial resistance. Antimicrobial resistance is expected to pose a significant challenge to life in the future. This review comprehensively elucidates the potential of Myrmecodia spp. as an antibacterial agent by systematically selecting and reviewing the majority of relevant studies published in the past 10 years and retrieved from Scopus, PubMed, Web of Science, and related books. Myrmecodia spp. is a non-parasitic plant that grows as an epiphyte. These essential nutrients for the body, including flavonoids, alkaloids, polyphenols, tannins, and saponins, are found within plants. Myrmecodia spp.'s compound functions as an antibacterial agent. Anxiety disorder is one of the important public health problems. This study aimed to find out whether patchouli (Pogostemon cablin (Blanco) Benth.) essential oil (PEO) has an anxiolytic-like effect in mice. A total of 20 mice were divided into five treatment groups i.e. (C1) was treated with 1 mg/kg of saline intraperitoneally; (C2) was treated with 1 mg/kg of diazepam intraperitoneally; (T1) 1% PEO inhalation, (T2) 2.5% PEO inhalation, and (T3) 5% PEO inhalation. All experimental animals were put in the Elevated Plus-Maze (EPM), observed for five minutes, and investigated the following parameters i.e. the total number of squares that are transverse in the open arms (STO), the number of visits to the open arms (VOA), the number of visits to the closed arms (VCA), the time spent in the open arms (TOA), and the time spent in the closed arms (TCA). In results, T1 and T2 groups had a minor impact on the VOA. T1 group also had little effect on TOA. Closed-arm activities (VCA and TCA) were unaffected by any of the treatments. This study aimed to determine the effect of the administration of ethanolic extract of Moringa oleifera leaves on the number of spermatogenic and Leydig cells of gentamicin-induced rats (Rattus norvegicus). This study used 25 white male rats divided randomly into five groups. The rats were injected with Gentamicin 5 mg/kg bw/day subcutaneously for 14 days, except for the negative control group (NCG). During the next 14 days, the rats were given oral moringa leaf extract of 200, 316, and 500 mg/kg bw/day respectively for E200, E316, and E500 groups. The data were analyzed with oneway ANOVA followed by Duncan's test. The results showed that the number of spermatogonia, spermatocytes, and spermatids in NCG was higher (p <0.05) than in the other groups. The number of spermatogonia, spermatocytes, and spermatids in the E316 group was higher (p <0.05) than in PCG. The number of spermatogonia in the E500 group was higher (p <0.05) than in the E316 group, but the number of spermatocytes and spermatids in the E316 group was similar (p >0.05) to the E500 group. The number of PCG Leydig cells was the smallest (p <0.05) compared to the other groups. The administration of Moringa leaves extract in the E200 and E316 groups increased (p <0.05) the number of Leydig cells compared to PCG. The number of Leydig PCG cells in the E500 group was not significantly different (p >0.05) from that of the NCG. It could be concluded that administration of Moringa oleifera leaves extract restored the number of spermatogenic and Leydig cells of gentamicin-induced rats. [ABSTRACT FROM AUTHOR]

This study aims to determine the effect of adding melon (Cucumis melo L.) juice to Lactated Ringer's-egg yolk extender on the intact plasma membrane (IPM) and morphological abnormalities of native rooster spermatozoa. Five ejaculate each from two native roosters (Gallus gallus domesticus) were divided into four equal volumes. Ejaculate was extended 1:10 (v/v) in Lactated Ringer's-egg yolk without or with the addition of of 2, 4, and 8% (v/v) melon flesh juice, respectively for groups T0, T1, T2, and T3. Extended semen was stored in a refrigerator (5°C) and evaluated every two hours. The results showed that IPM of all groups decreased (p <0.05) consistently, lasting more than 40% for six hours in the T0 and T1 groups and four hours in the T2 and T3 groups. The percentage of spermatozoa morphological abnormalities in all groups stored for six hours was higher (p <0.05) than before storage. However, it was not significantly different (p >0.05) from that which had been stored for two and four hours. It was concluded that native rooster semen extended in Lactated Ringer's-egg yolk without (group T0) or with 2% melon flesh juice (group T1) and stored at 5°C retained spermatozoa plasma membrane integrity of more than 40% and morphological abnormalities of less than 20% for six hours. While the addition of 4 % (T2) and 8% (T3) melon flesh juice maintained the percentage of IPM and spermatozoa morphological abnormalities for only four hours. [ABSTRACT FROM AUTHOR]

Lead is a harmful pollutant from engine exhaust that causes free radicals and has detrimental effects on the testicular tissue. This study aimed to determine the effects of tomato juice on the number of Leydig cells and the diameter of the seminiferous tubules of mice exposed to lead acetate. Twentyfive male mice were divided into five groups. Mice in the control (C-) group were given placebos. Meanwhile, mice in C+, T1, T2, and T3 groups were exposed to lead acetate at a dose of 100 mg/kg BW/day for 14 days and given tomato juice respectively at 0, 0.16, 0.32, and 0.64 mL/day from day 8 to day 35. On day 36, all mice were sacrificed, and the testes were collected for histological preparation. The result showed that lead exposure in the C+ group decreased (p <0.05) in the number of Leydig cells and the diameter of the seminiferous tubules compared to the mice in group C-. Administration of tomato juice in groups T1, T2, and T3 increased (p <0.05) the number of Leydig cells and the diameter of seminiferous tubule compared to the mice in the C+ and C-groups. However, tomato juice administration to the T3 group decreased the number of Leydig cells and the diameter of the seminiferous tubules (p <0.05) compared to the T2 group. In conclusion, an effective dose of 0.32 mL/day of tomato juice restored Leydig cell number and seminiferous tubules diameter in mice exposed to lead acetate. [ABSTRACT FROM AUTHOR]

The purpose of this study was to determine the preventive effect of Apis dorsata honey (AH) on the spermatogenic and Sertoli cells count of mice (Mus musculus) exposed to monosodium glutamate (MSG). This study used 25 male BALB/c mice which were divided into five treatments. The negative control (C-) received aquadest, the positive control (C+) received MSG 4 mg/gBW, T1 received AH 2.7 mg/gBW + MSG 4 mg/gBW, T2 received AH 5.4 mg/gBW + MSG 4 mg/gBW, and T3 received AH 8.1 mg/gBW and MSG 4 mg/gBW. All groups were treated for 52 days. The testes were then prepared as histopathology slides and examined under a microscope. The results of this study showed that there was a significant difference in spermatogenic cells and Sertoli cells between C-, C+, T1, and T2 (p<0.05) and showed no significant difference (p>0.05) in spermatid and Sertoli cells count between C-(90.28 ± 1.361 and 13.60 ± 0.374) and T3 treatment group (88.04 ± 1.212 and 13.04 ± 0.434). The best preventive dosage of AH was the T3 with 8.1 mg/gBW. It can be concluded that administration of AH can maintain spermatogenic and Sertoli cells count in mice exposed to MSG. Background: Varanus salvator is one of the reptiles being hunted by human beings for several purposes, including traditional medicine. The studies about reproductive biology aspects were limited. Aim: This study aimed to determine the morphology, histology, and histometry of V. salvator paryphasmata and hemibaculum based on Snout-Vent Length (SVL) as an indicator of sexual maturity. Methods: This study examined 18 pairs of hemipenis of V. salvator with SVL more and less than 40 cm in equal number. Paryphasmata and hemibaculum parts were observed visually and micro-sliced, then stained with Hematoxylin-Eosin (HE). The histological observation was conducted under a 40x, 100x, and 400x magnification of a light microscope. The histometry of the paryphasmata was examined using 13 Megapixels Coolpad and OptiLab Plus for microscopic pictures. The chondrocyte cell area was measured using the Optilab Plus and Image Raster three applications. Results: The sizes of glans of hemipenis, paryphasmata, and hemibaculum increased according to the increasing of SVL. The average paryphasmata row number, epidermis, and loose connective tissue thickness were not significantly different (p > 0.05). However, dense connective tissue was thicker (p < 0.05), which corresponds to SVL. Hemibaculum was composed of fibrous and hyaline cartilage characterized by chondrocyte cells. The SVL also affects (p < 0.05) the ossification of hyaline in hemipenis, while the chondrocyte cell area followed the equation -1.87E7 + 7.09E5* SVL. Conclusion: The SVL size of V. salvator affects the paryphasmata, hemibaculum, thickness of dense connective tissue of paryphasmata, and the area of chondrocyte cells. [ABSTRACT FROM AUTHOR]

This study aimed to determine the function of motoric reflexes and memory as a result of the necrosis of neuron cells in the cerebellum of infant mice (Mus musculus) from their carbofuran exposed mothers during lactation periods. The decreased motoric reflexes as an indicator of the reduced memory ability and concentration ability in infant mice. This experimental laboratory study used 36 mice and carbofuran injected using gavage method with the fraction of LD50 in mother mice in the dose of 5 mg/kg body weight (BW). The mother mice were exposed with carbofuran during lactation period from Day 0 to Day 9 and the 10-days-old infant mice were then terminated for microscopic examination by counting the necrosis of neuron cell by applying HE staining and the motoric-reflex function tests (neurobehavioral test) on the 10-days-old infant mice which included: surface righting reflexes, swimming and a memory test employing an Eight-arm radial maze at 20 days-old. The calculation results were analyzed using the Kruskal-Wallis test and the Mann Whitney test. The results of this study showed a significant increase in the necrosis of neuron cell, a fundamental decreased motoric reflex in infant mice in these activities; such as surface righting reflex and swimming ability (the head angle position and swimming direction) and eight-arm radial maze memory test. Conclusion: in sum, the exposure of carbofuran in the mother mice during lactation period causes the increase in necrosis of neuron cell, the decrease of motoric reflexes and the memory ability for infant mice. The aim of this study was to find cytotoxicity effect of Curcuma aeruginosa extract on fibroblast with MTT assay method. This study used experimental laboratory with post test only control group design. Fibroblast cell lines culture divided into ten groups with four replication in each group. The positif control group was the group contained fibroblas cells in RPMI media. The negatif control group was the group contained media without cells. There were eight treatment groups given Curcuma aeruginosa extract in different concentration ( 1 ppm, 2 ppm, 2.5 ppm, 5 ppm, 10 ppm, 15 ppm, 20 ppm, 25 ppm). After exposure of the extract, the cells were incubated for twenty hours. The MTT reagen was given on the cells and then was incubated for four hours. After that the optic density was read with microplate reader in 630 wavelength. The result showed that the percentage of viable cells was more fifty percent in treatments groups. The Anova analysis showed that there was significantly decrease (p<0,05) of percentage of viable cell between positif control group and treatment groups. The conclusion was the Curcuma aeruginosa extract did not show cytotoxicity effect on fibroblast cells but the percentage of viable cell was decrease significantly in treatment groups. This study aimed to determine the mechanism of autophagy, apoptosis and necrosis in the neurons of infant mice (Mus musculus) whose mothers were exposed to carbofuran during the lactation period. This experimental study included 20 mice; carbofuran was administered at LD50 fractions by gavage to mice at the doses of 1.25 mg/kg body weight (BW) (1/4 LD50), 0.625 mg/kg BW (1/8 LD50), and 0.3125 mg/kg BW (1/16 LD50). Mothers were exposed to carbofuran during lactation on Days 1-9. On Day 10, infant mice were sacrificed in order to determine the number of neuron cells expressing protein kinase B (Akt) and the mammalian target of rapamycin complex 1 (mTORC1) as autophagy pathway using immunohistochemistry, apoptosis using the Tunel Assay, and necrosis using haematoxylin and eosin staining. The results of Akt, mTORC1, apoptosis, and cell necrosis were analyzed by analysis of variance (ANOVA) and Duncan tests. The results of the study showed that exposing the mothers to carbofuran during lactation caused an increase in necrosis and apoptosis of neuronal cells but did not cause autophagy in neuron cells via the Akt/mTOR pathway of infant mice. The increase in apoptotic neurons opens up opportunities for the prevention and handling of the effects of reactive oxygen species activities due to carbofuran exposure during lactation periods. Background: The Trichuris eggs are collected from naturally infected sheep. Natural antihelmintics such as herbal medicines are needed as an alternative, such as natural compounds from endemic plants., Aim: This present study aims to evaluate the ovicidal activity and cytotoxicity effects of ethanolic extract of Curcuma longa (EECL) and Camelia sinensis (EECS) as a biological anthelmintic against the egg of Trichuris sp ., Methods: The Trichuris eggs are collected from naturally infected sheep. CMC-Na solution 1% was used as a control. The treatments were 0.12% EECL; 0.24% EECL; 0.15% EECS; 0.30% EECS; a combination of 0.12% EECL and 0.30% EECS; a combination of 0.24% EECL; and 0.15% EECS. Ovicidal activity testing by microscopic examination of eggs treated using different concentrations of EECL extract, EECS, and a combination of them. They were exposed for various times (7, 14, 21, and 28 days) and incubated at room temperature., Results: The study shows that a combination of C. longa extract and tea extract exhibits good ovicidal anthelmintic activity against Trichuris sp . in sheep. Cytotoxicity examination using the 3-(4,5-dimethylthiazol-2-yl) 2,5-diphenyl tetrazolium bromide (MTT) test. Based on MTT data processed using regression analysis, the obtained LC 50 from the administration of EECL, EECS, and a combination of both in a ratio of 1:1, 2:2, 1:2, and 2:1. The combination of EECL extract and EECS with the highest concentration produced cell viability of 28.46%, 17.25%, 56.01%, and 46.47%, respectively., Conclusion: It can be concluded that the most cytotoxic ingredient is found in the combination of EECL and EECS (2:2) at 17.25% and the safest is in the ratio (1:2) at 56.01%., Competing Interests: The authors declare that there is no conflict of interest.

Background and Purpose: This study aimed to determine the potency of kebar grass ethanol extract to overcome an increase in cerebellar neuronal cell necrosis, which has an impact on decreasing motor reflex function and spatial memory of mice from lactating mothers exposed to carbofuran., Experimental Approach: Forty lactating mice were divided into four groups, 10 each; including control, T1 (carbofuran 0.0125 mg/day), T2 (vitamin C 5 mg + carbofuran 0.0125 mg/day), T3 (kebar grass extract 3.375 mg + carbofuran 0.0125 mg/day). The mice were orally administered with carbofuran, vitamin C, and kebar grass extract on days 0 to 14 postnatal. On the 15 th day, brains of the mice were necropsied to measure the levels of malondialdehyde (MDA), superoxide dismutase (SOD), and glutathione (GSH), H&E staining; motor reflex tests were performed on 10-day-old mice, and the mice aged 30 days were tested on their swimming and spatial memory., Findings / Results: Carbofuran caused an increase in MDA, GSH, neuronal cell necrosis, surface righting reflex, a decrease in SOD, swimming ability, and spatial memory. Kebar grass extract and vitamin C administration decreased MDA, GSH, neuron necrosis, surface righting reflex, and increased SOD, swimming ability, and spatial memory., Conclusion and Implications: Exposing to carbofuran in lactating mice caused brain oxidative stress, impaired motor reflexes, and spatial memory in mice offspring. Kebar grass extract and vitamin C administration prevented brain oxidative stress and inhibited disorders in motor reflexes, and spatial memory in mice offspring. Currently, there are increasing numbers of dermal and transdermal dosage forms of both natural and synthetic compounds on the market. Therefore, it is necessary to have a method that can measure the release and penetration of the compound into the skin. This chapter presents a current method for evaluating the skin penetration of a Curcuma heyneana rhizome extract in vivo using histological parameters. We also evaluate a liposome delivery system of the same extract to determine any differences in penetration due to changes in the drug delivery system., (© 2022. We present a histomorphometric analysis to enable quantitative measurement of anti-aging activity of topical substances in rat skin. In this method, the measured parameters were closely related to changes that occur on the skin caused by exposure to UV light. We used this protocol to determine the anti-aging activities of an ethanolic extract of Curcuma heyneana rhizome and retinoic acid by a measurement of the changes in epidermal thickness, the number of sunburned cells, the number of fibroblasts, and the space between collagen fibers.
