Abstract

Reactivation of Trypanosoma cruzi transmission by native vectors with different domiciliation capabilities is a major concern for Chagas disease control programs. T. cruzi transmission via intra-domestic Rhodnius prolixus was certified as interrupted by the Pan American Health Organization in Miraflores municipality (Boyacá, Colombia) in 2019. However, Triatoma venosa, a native vector infected with T. cruzi has been increasingly found inside human dwellings across rural areas. In this study, the aim was to describe the eco-epidemiological aspects of T. cruzi transmission in the rural area of Miraflores. For this, we designed a comprehensive, multi-faceted study in 6 rural villages and performed: (i) A cross-sectional serological and molecular study enrolling 155 people and 58 domestic dogs living within 80 households, (ii) a domestic entomological survey, (iii) a determination of the natural infection and blood meal source in collected triatomine bugs, and (iv) an evaluation of synanthropic mammal infection by parasitological and molecular tools. The T. cruzi seroprevalence rates in humans and dogs were 9.03% (14/155) and 22.4% (13/58), respectively. Most infected humans were adults between the ages of 55 and 85 years old. No evidence of T. cruzi DNA was found using qPCR in human blood samples, but we found high parasitemia levels in the infected dogs. In total, 38 triatomine bugs were collected inside dwellings and peridomestic areas: 68.4% (26/38) Triatoma venosa, 29% (11/38) Panstrongylus geniculatus, and 2.6% (1/38) P. rufotuberculatus. Natural infection prevalence was 88% (22/25) for T. venosa, 100% (12/12) for P. geniculatus, and 100% (1/1) P. rufotuberculatus: only TcI was found. No evidence of R. prolixus was found in the area. Two feeding sources were identified in T. venosa (humans and cats), while P. geniculatus fed on cows and bats. Lastly, seven D. marsupialis were captured in peridomestic areas, three were infected with T. cruzi (TcI). The results suggest the existence of T. cruzi transmission cycle between triatomines, dogs, and opossums representing a risk of infection for the human population in rural areas of Miraflores. Despite PAHO declaring Miraflores municipality, Colombia an area of T. cruzi transmission interruption in 2019, this study documents evidence of a secondary vector establishing in domestic settings. T. venosa entomological surveillance is warranted to evaluate prospective human transmission risk in an otherwise 'no-risk' perceived Chagas disease region. Author summary: A major concern for Chagas disease control programs is transmission reactivation by native vectors that are not commonly found living within the domestic environment. In Colombia, transmission by primary vector species was certified as interrupted in Miraflores municipality in 2019, however other secondary native species have been found entering human dwellings in rural areas, emphasizing the need for entomological surveillance. In this study we collected and analyzed human and animal blood samples, and secondary vector species from six rural villages to detect Trypanosoma cruzi infections. The results show evidence of active infection among three native vector species, domestic canines, and peridomestic opossums. Our results are important, as vector species ecology continues to adapt warranting increased entomological surveillance to assess human transmission risk in an area perceived as free of Chagas disease transmission. 22. Examining the gut microbiota from several human-biting tick species in Northwestern Spain

Abstract

Tick-borne diseases have increased significantly in Europe and Spain in recent years. One strategy explored for tick surveillance and control is the study of the microbiota. The focus is on understanding the relationships between pathogens and endosymbionts within the microbiota and how these relationships can alter these arthropods’ vectorial capacity. Thus, it is pivotal to depict the bacterial communities composing the microbiota of ticks present in specific territories. This work aimed to describe the microbiota present in 29 adult individuals of 5 tick species collected from 4 provinces of Castilla y Leon in northwestern Spain from 2015 to 2022. DNA extraction and sequencing of the V4 hypervariable region of 16S-rRNA was performed on the tick samples, with subsequent analysis of diversity, taxonomic composition, and correlations between genera of microorganisms. There were no differences in the alpha diversity of microbiota by tick species, nor were compositional changes evident at the phylum level for microorganisms. However, interindividual differences at the microbial genus level allowed spatial differentiation of the 5 tick species included in the study. Correlation analyses showed complex interactions between different genera of microbiota members. These findings provide an initial insight into the composition of the gut microbiota of various tick species in northwestern Spain, which can contribute to establishing surveillance and control measures to reduce diseases such as rickettsiosis, Lyme disease, and Crimean-Congo hemorrhagic fever.

Abstract

A medida que la pandemia del Síndrome Respiratorio Agudo Severo Coronavirus 2 (SARS-CoV-2) continúa expandiéndose, los recursos de atención médica a nivel mundial se han reducido. Ahora, la enfermedad es extendiéndose rápidamente por América del Sur, con consecuencias mortales en áreas con ya sistemas de salud pública debilitados. La región amazónica es particularmente susceptible a la devastación generalizada de la enfermedad por coronavirus 2019 (COVID-19) debido a sus habitantes amerindios nativos inmunológicamente frágiles y vulnerabilidades epidemiológicas. Aquí, nosotros discutir la situación actual y el impacto potencial de COVID-19 en la región amazónica y cómo una mayor propagación de la ola epidémica podría resultar devastadora para muchas personas amerindias que viven en la selva amazónica As the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) pandemic continues to expand, healthcare resources globally have been spread thin. Now, the disease is rapidly spreading across South America, with deadly consequences in areas with already weakened public health systems. The Amazon region is particularly susceptible to the widespread devastation from Coronavirus disease 2019 (COVID-19) because of its immunologically fragile native Amerindian inhabitants and epidemiologic vulnerabilities. Abstract

La enfermedad de Chagas es causada por el protozoo parasitario Trypanosoma cruzi, el cual es transmitido por triatominos de la familia Reduviidae. Psammolestes arthuri es un triatomino selvático que se alimenta de aves y se ha encontrado infectado con T. cruzi. En Colombia, Ps. arthuri se ha encontrado en las viviendas, de manera que es importante evaluar la presencia del parásito y de qué se están alimentando. Por tal razón, en el presente estudio evaluamos la presencia de T. cruzi y las fuentes alimenticias de Ps. arthuri como una primera aproximación para elucidar nuevos posibles escenarios de la tansmisión de T. cruzi en Colombia.
