This study aims to determine the effect of administration of ciplukan (Physalis angulate Linn.) leaf extract on the variables of testicular weight, seminiferous tubules diameter, and spermatozoa plasma membrane integrity of rats (Rattus norvegicus) exposed to heat as model animals. Rats were divided randomly into five groups. In the NC group, rats were not exposed to heat and were only given 1% Na-CMC. In the PC, T1, T2, and T3 rats were exposed to heat followed by administration of 1% Na-CMC, 100, 200, and 400 mg/kg bw of ciplukan leaf extract (CLE) in 1% Na-CMC. Heat exposure at 40°C was conducted for 60 minutes every day for 21 days. On day-22, all rats were sacrificed for testicular evaluation. The results showed that all variables in the PC group were lower (p <0.05) than in the NC group. All variables in the T3 group were higher (p <0.05) than in the PC group. Testicular weight and spermatozoa plasma membrane integrity in the T3 group were not significantly different (p >0.05) from the NC group. However, the diameter of the seminiferous tubules in the T3 group were lower (p <0.05) than in the NC group. This study revealed that the administration of ciplukan leaf extract as an antioxidant increased testicular weight, seminiferous tubules diameter, and spermatozoa with intact plasma membrane in rats exposed to heat. This study aims to determine the effect of adding duck egg yolk to skim milk-fructose extender on the motility and viability of Muscovy duck (Cairina moschata) spermatozoa at 5°C storage. Muscovy duck ejaculate was divided into four equal portions. The first portion was for the control group, which was extended in skim milk-fructose, while the second, third, and fourth portions were for treatment groups T1, T2, and T3, which were extended in skim milk-fructose with the addition of 2.5, 5.0, and 7.5% (v/v) duck egg yolk. Sperm motility and viability were evaluated every two hours until the motility percentage reached 40%. Data were analyzed using ANOVA and Duncan’s test. The results showed that spermatozoa motility and viability were higher (p <0.05) in groups T1, T2, and T3 compared to those of group T0 after 10 hours storage at 5°C. The highest percentage of spermatozoa motility and viability was in the T2 group compared to other groups (p <0.05). It could be concluded that the Muscovy duck semen extended in skim milk-fructose containing 2.5 to 7.5% (v/v) duck egg yolk met the requirements for artificial insemination when stored for up to 10 hours at 5°C. This study aims to determine the effect of the administration of white guava (Psidium guajava L.) fruit juice on spermatozoa plasma membrane integrity (PMI), morphological abnormality, viability, and motility of lead acetate induced rats (Rattus norvegicus). Twenty-five male rats were divided into five groups: NC (negative control) group, rats were administered with distilled water twice daily at four-hour intervals; T0 (positive control) group, rats were administered daily with lead acetate 50 mg/kg bw and distilled water four hours later; T1, T2, and T3 groups, rats were administered daily with lead acetate 50 mg/kg bw and 0.5 mL of 25, 50, and 100% white guava fruit juice four hours later. The treatment of the rats was conducted for 14 days, and on day 15, all rats were sacrificed to assess the spermatozoa quality. Data was analyzed using ANOVA followed by Duncan's multiple range test at a confidence level of 95%. The results showed that exposure to lead acetate (T0) caused lower spermatozoa PMI, viability, and motility as well as higher spermatozoa morphological abnormalities (p <0.05) compared to those of the T0 group. Administration of white guava fruit juice starting at a dose of 25% (T1) resulted in higher spermatozoa motility, viability, and PMI as well as lower spermatozoa morphological abnormalities (p <0.05) compared to rats in the T0 group. Pote goats are kept by the rural community in Bangkalan regency, Madura island, Indonesia. Symptoms of estrus in goats are usually unclear, causing low fertility. This study aims to evaluate intracervical artificial insemination performed in early and mid-estrus on Pote does. Twenty four does were randomly divided into two groups and their estrous was synchronized using two injections of 200 µg PGF2a intramuscularly with 11 days interval. In the T1 group, Pote does that were in estrus were inseminated at early estrus (0-12 hours after signs of estrus appeared), while in the T2 group, Pote does that were in estrus were inseminated at mid-estrus (12-24 hours after signs of estrus appeared). The insemination conducted artificially using frozen-thawed Boer semen. The observed variables were non-return rate at day-21 post-insemination (NRR21), and pregnancy rate (PR) based on abdominal palpation diagnosis. The result showed that frozen-thawed Boer semen was qualified (spermatozoa motility more than 40%) for artificial insemination. All Pote does were in estrus after two intramuscular injection of PGF2a with an interval of 11 days. The onset of estrus was 48-72 hours after the second injection PGF2a. The NRR21 and PR of the T2 group (66.7 and 100%) were higher than those of the T1 group (37.5 and 66.7%), respectively. It could be concluded that intracervical artificial insemination in Pote does in mid-estrus was more effective, resulting in higher NRR21 and PR than in early estrus. [ABSTRACT FROM AUTHOR]

This study aims to determine the effect of adding melon (Cucumis melo L.) juice to Lactated Ringer's-egg yolk extender on the intact plasma membrane (IPM) and morphological abnormalities of native rooster spermatozoa. Five ejaculate each from two native roosters (Gallus gallus domesticus) were divided into four equal volumes. Ejaculate was extended 1:10 (v/v) in Lactated Ringer's-egg yolk without or with the addition of of 2, 4, and 8% (v/v) melon flesh juice, respectively for groups T0, T1, T2, and T3. Extended semen was stored in a refrigerator (5°C) and evaluated every two hours. The results showed that IPM of all groups decreased (p <0.05) consistently, lasting more than 40% for six hours in the T0 and T1 groups and four hours in the T2 and T3 groups. The percentage of spermatozoa morphological abnormalities in all groups stored for six hours was higher (p <0.05) than before storage. However, it was not significantly different (p >0.05) from that which had been stored for two and four hours. It was concluded that native rooster semen extended in Lactated Ringer's-egg yolk without (group T0) or with 2% melon flesh juice (group T1) and stored at 5°C retained spermatozoa plasma membrane integrity of more than 40% and morphological abnormalities of less than 20% for six hours. While the addition of 4 % (T2) and 8% (T3) melon flesh juice maintained the percentage of IPM and spermatozoa morphological abnormalities for only four hours. [ABSTRACT FROM AUTHOR]

This study aims to determine the effect of red dragon (Hylocereus polyrhizus) fruit peel extract (RDFPE) on spermatozoa motility and viability of hypercholesterolemic rats (Rattus norvegicus) as a model. Twenty male rats were randomly divided into negative control (NC), positive control (PC), treatment 1 (T1), treatment 2 (T2), and treatment 3 (T3) groups. All rats were given 2 mL of high cholesterol feed orally every day for 28 days. On day-15, all rats were measured for their blood cholesterol levels, followed by treatment for 14 days. Rats in the NC, PC, T1, T2, and T3 groups were treated with 1% Na-CMC, Simvastatin 10 mg/kg BW, and RDFPE of 500, 750, and 1000 mg/kg BW, respectively. On day-29, all rats were sacrificed to evaluate spermatozoa viability and motility. The results showed that spermatozoa viability and motility in the hypercholesterolemic rats (NC) group were the lowest (p <0.05) among the groups. Treatment of hypercholesterolemic rats with Simvastatin 10 mg/kg BW (PC) group showed higher (p <0.05) spermatozoa viability and motility compared to the NC group. RDFPE dose of 1000 mg/kg BW (T3 group) resulted in higher (p <0.05) spermatozoa viability and motility compared to other RDFPE doses (T1 and T2) and the control (NC) groups, and it was similar (p >0.05) compared with the Simvastatin treated (PC) group. It could be concluded that the administration of 1000 mg/kg BW ethanolic extract of red dragon fruit (Hylocereus polyrhizus) peel increased the viability and motility of spermatozoa of hypercholesterolemic rats (Rattus norvegicus) which were the same as the group of rats given Simvastatin 10 mg/kg BW. This study aims to determine the motility and viability of kampung rooster (Gallus domesticus) spermatozoa in 5% Dextrose Ringer's solution as an extender with or without the addition of various egg yolk concentrations at 5°C during storage. Semen was collected by massage in the morning. In this study, four types of extenders were used, i.e., 5% Dextrose Ringer's extender without egg yolk, 5% Dextrose Ringer's extender with 5, 7.5, and 10% egg yolk for T0, T1, T2, and T3, respectively. Then, 1000 IU Penicillin and 1 mg Streptomycin were added per mL extender. Fresh semen from each rooster was evenly divided into four volumes to be diluted 11 times in each group extender. Extended semen from all groups was stored at 5°C. Data were analyzed using ANOVA and Duncan's new multiple range test. The result showed a significant difference (p <0.05) between each treatment group for spermatozoa motility and viability. In conclusion, this study revealed that the addition of various concentrations of egg yolk into 5% Dextrose Ringer's solution as an extender maintained the motility and viability of kampung rooster spermatozoa. This study aimed to determine the effect of the addition of cherry tomato, california papaya, and honey melon juice into skim milk extender in maintaining the quality of Sapudi ram semen at chilled temperature (5oC) storage. Five replication of ejaculates were divided equally into four groups. In control group (T0) semen was diluted in skim milk extender, while in T1, T2, and T3 groups semen were diluted in skim milk extender contained 20% of cherry tomato, california papaya, and honey melon juice, respectively. The extended semen was stored at a chilled temperature, and semen quality (based on sperm motility, viability, and plasma membrane integrity) was observed daily for five days. The result showed that semen quality declined day by day during the five days of storage. Based on the minimum standard of post-thawed semen motility (40%), the spermatozoa of the control group only lasted by the second day. Meanwhile, in the groups with the addition of fruit juice motility could last up to the third day, with the highest motility (p <0.05) on the addition of california papaya juice. This study concluded that the addition of 20% (v/v) california papaya juice in skim milk extender could maintain the percentage of spermatozoa motility up to three days at a chilled temperature. Background: Malnutrition can cause an increase in oxidative stress as it triggers the expression of heat shock protein70 (HSP70), a chaperon molecule that is needed to repair damaged cells within optimal levels. Honey is a source of feed that can stimulate HSP70 expression, which can be given to the malnourished in the animal trial. Aim: The purpose of this study was to prove that Kaliandra honey can improve testosterone levels, diameter, and epithelial thickness of the seminiferous tubule of rat testes (Rattus norvegicus) due to malnutrition through stimulation of HSP70, which is expressed immunohistochemically. Methods: This study used 40 male rats, which were divided into four treatment groups: T0 (negative control): normal rats and not given honey; T1 (positive control): malnourished rats and not given honey; T2 (treatment 2): malnourished rats and given 30% Kaliandra honey (v/v) for 10 days; T3 (treatment 3), malnourished rats and given 50% Kaliandra honey (v/v) for 10 days. The condition of malnutrition is carried out by fasting the feed for five consecutive days resulting in damage to the male reproductive organs, especially the testes. Results: The results showed that Kaliandra honey at a dose of 50% (v/v) had a significant effect in improving testosterone levels, diameter, and epithelial thickness of seminiferous tubule of malnourished male rats through stimulation of HSP70 expression. The HSP70 expression scores by IHC at T0, T1, T2, and T3 were 0.15a ± 0.5, 3.15c ± 0.4, 2.95c ± 0.35, and 1.75b ± 0.15, sequentially. enzyme-linked immunosorbent assay indirect testosterone levels at T0, T1, T2, and T3 (in µg/dl) were 36.39c ± 0.35, 6.12a ± 0.51, 7.45a ± 0.15, 25.27b ± 0.63, sequentially. The diameter and epithelial thickness of the seminiferous tubule of the testes (in µm) in the four treatments T0, T1, T2, and T3 were 362.40c ± 4.71, 248.46a ± 3.90, 255.22a ± 2.34, 318.37b ± 4.23 and 117.60d ± 11.30, 3.86a ± 1.57, 9.72b ± 3.96, 29.84c ± 4.02 sequentially. Conclusion: The conclusion of the study showed that Kaliandra honey at a dose of 50% (v/v) had a significant effect in improving testosterone levels, diameter, and epithelial thickness of the seminiferous tubule of malnourished rats through stimulation of HSP70, although not significantly the same as negative control (T0). Kacang goats (Capra hircus) are an indigenous Indonesian livestock species that can be reared by local communities as an additional source of income to prevent poverty and to provide animal protein. Artificial insemination techniques could use to increasing the population size, preserving genetically valuable offspring, and improving reproductive performance. This study was the first to combine the antioxidant addition in skim milk-egg yolk (SM-EY) extender and different equilibration period to obtain a higher quality post-thawed Kacang buck semen. The present study aimed to determine the effect of green tea (Camellia sinensis L. Kuntze) extract (GTE) on the quality of frozen Kacang goat sperm equilibrated for 1 and 2 h. The pool of Kacang buck ejaculate was divided into four portion equally and diluted in a SM-EY extender that contained four doses of 0, 0.05, 0.10, and 0.15 mg of GTE/100 mL for T0, T1, T2, and T3 groups, respectively. The aliquots were treated with a 1 or 2 h equilibration period before further processing as frozen semen. Post-thawed semen's quality was evaluated for sperm viability, progressive motility, intact plasma membrane, malondialdehyde concentration, and DNA fragmentation. The results showed that post-thawed semen of T2 was the highest quality than other groups. Meanwhile, 1 h equilibration period had better quality than that of 2 h equilibration. It could be concluded that a combination of 0.1 mg GTE/100 mL extender as an antioxidant and 1 h equilibration period provided the best result for frozen Kacang buck semen. [ABSTRACT FROM AUTHOR]

This study was conducted to determine the effect of aluminum silicate on the quality of sperm, plasma membrane and seminiferous tubules of mice exposed to the fungus Fusarium graminearum. Twenty-five healthy, 8-10 week old, 20-25 g, BALB/c adult male mice were used. They were divided equally into five groups: negative control (C-) was not given F. graminearum and aluminum silicate; positive control (C+) received 0.5 ml F. graminearum suspension (228 x 108 spores/ml) but without aluminum silicate. Treatments 1, 2, 3 were each given the same amount of fungus and aluminum silicate with doses 0.5 mg (T1), 1 mg (T2), and 2 mg (T3) for 21 days. Testes and spermatozoa were obtained from the epididymal duct to examine for the said parameters. Motility (%) was highest in C-, but not significantly different from T3 (P>0.05). Viability (%) was highest in C-, which was significantly different with all treatments (P<0.05), T3 was not significantly different from T2 and T1 (P>0.05). Meanwhile, abnormalities (%) were highest in C+, and significantly different from all treatments. Plasma membrane integrity was highest in C-, but not significantly different from T3. Likewise, seminiferous tubule quality was highest in C- but lowest in C+. All parameters, except the latter, were lowest in C-. Results of the study suggest that using aluminum silicate can maintain the quality of sperm in mice exposed to F. graminearum. [ABSTRACT FROM AUTHOR]

Background and Aim: The use of frozen goat semen for artificial insemination frequently results in a decline in sperm quality following thawing, which can be attributed to cold shock from cryopreservation, reduced motility, and possible DNA damage. Freezing may compromise mRNA stability due to the presence of free radicals. Despite strong post-thaw motility and no visible DNA fragmentation, sperm can still exhibit altered gene expression patterns. To reduce the damaging impact of free radicals during cryopreservation, antioxidants are typically added to the freezing medium. This study assessed the impact of adding coenzyme Q10 (CoQ10) to frozen sperm diluent on the ATP5F1A and CPT2 gene expression, sperm motility, and viability post-thawing., Materials and Methods: CoQ10 was added to sperm at six different concentrations: 0 mg/dL (P0), 6.25 mg/dL (P1), 12.5 mg/dL (P2), 25 mg/dL (P3), 50 mg/dL (P4), and 100 mg/dL (P5). The Statistical Package for the Social Sciences (SPSS) software version 22 was used to conduct comparative tests using one-way analysis of variance followed by Duncan's test for motility and viability and Kruskal-Wallis test followed by pairwise comparison test for membrane integrity and gene expression., Results: The addition of CoQ10 to semen diluent has a notable impact on the post-thawed quality of sperm. The most significant outcomes were observed with a 25 mg/dL dosage (P3) for cell viability, membrane integrity, and ATP5F1A gene expression, and with a 50 mg/dL dosage (P4) for sperm motility, membrane integrity, and CPT2 gene expression., Conclusion: Incorporating CoQ10 into frozen semen diluent improves gene expression and prevents deterioration of the cell quality of thawed goat spermatozoa. While the study demonstrates the benefits of CoQ10, the precise molecular mechanisms through which CoQ10 enhances gene expression and cell quality were not fully elucidated. Further investigation is needed to understand these mechanisms in detail.
