Recent studies have discussed the importance of snakes, going beyond the context of Public Health (snakebites). Promising research demonstrates the utility of snake venom in several fields, including oncology, diagnosis, anticoagulant therapy, and pain treatment. Nevertheless, there are still gaps in the basic knowledge on these animals, specifically regarding the histological characterization of the gastrointestinal tract, which ultimately hinder comparative histology and pathology studies. In this context the present study intends to contribute with the advance of current knowledge on snake histology by describing and analyzing histological samples of the gastrointestinal tracts of Bothrops jararaca and Crotalus durissus. Samples were collected from 12 individuals (six from each species), up to 6 months of age and equally distributed between males and females. Histological slides were prepared from the organs collected and were stained with hematoxylin and eosin, periodic acid-Schiff and alcian blue. The slides were photographed with a high-resolution camera to create a portfolio representative of the histology of the gastrointestinal systems of these animals. Na atualidade, a importância das serpentes extravasa a conotação em saúde pública (acidentes ofídicos), e pesquisas promissoras têm demonstrado a utilidade dos venenos das serpentes em diversas áreas como oncologia, diagnóstico, terapia anticoagulante e tratamento da dor. Ainda assim, existem lacunas no conhecimento básico sobre esses animais, tais como a caracterização histológica do seu sistema gastrointestinal, que se tornam obstáculos para estudos nos campos da histologia e patologia comparadas. Nesse contexto, o presente trabalho pretende contribuir com o avanço do conhecimento sobre histologia de serpentes a partir da descrição e análise histológica de amostras do trato gastrointestinal de exemplares das espécies Bothrops jararaca e Crotalus durissus. Para tanto, foram coletadas amostras de 12 indivíduos com até seis meses de idade, seis por espécie (distribuídos igualmente entre machos e fêmeas). A partir dos órgãos colhidos, lâminas histológicas foram preparadas e coradas pela técnica de hematoxilina e eosina, bem como PAS com Alcian Blue. As lâminas foram fotografadas por câmera de alta resolução, resultando em um portfólio representativo da histologia do sistema gastrointestinal desses animais.

At present the ostrich (Struthio camelus) breeding has been showing a great economical potential, although yet there are not distinct patterns about the histology of its liver, which is an organ of key importance in terms of metabolism. The knowledge of its histology can help the detection of diseases and nutritional deficiencies of this bird. Twenty four ostriches with an average age of 12-18 months and average weight of 80-100 Kg, proceeding from Don Pig Abatteur, located at Botucatu, São Paulo, were used. The birds were slaughtered with and to bleeding. Liver samples were processed for light and electron transmission microscopic studies. Hematoxilin and eosin (HE), picrosirius, Gordon and Sweets, Sudan black and Schiff periodic acid (PAS) were respectively used to observe the liver morphology, collagen, reticular fibers, lipids and glycogen. The liver portal spaces were determined. An average of 5.68% of glycogen was observed. The lipidic content with an average of 9.83%. Collagen fibers at an average of 14.71%. Reticular fibers with an average of 5.96%. Through transmission electron microscopy we noticed in the hepaticyte cytoplasm the presence of numerous mithocondria, glycogen, several lipidic droplets, some lysosomes, granular endoplasmatic reticulum around the mithocondria, some stellate cells, erythrocytes, nucleus and degenerating cells, besides the central biliary canaliculus. The suggestive steatotic results might result from the animals nutritional status Our results demonstrated that ostrich and other birds hepatocytes are very similar. A criação de avestruz (Struthio camelus, Linnaeus 1758) é uma atividade de grande potencial, porém não existem padrões definidos sobre a histologia do seu fígado, que é um órgão de grande importância no metabolismo. O conhecimento de sua histologia pode contribuir para a detecção de doenças e deficiências nutricionais no animal. Para este estudo foram utilizados 24 avestruzes com idades entre 12 e 18 meses (com peso médio em torno de 80 a 100 kg), provenientes do abatedouro Don Pig, em Botucatu no estado de São Paulo. Os animais foram abatidos com pistola pneumática e, após a sangria, as amostras do fígado foram processadas e observadas em microscopia de luz e microscopia eletrônica de transmissão (MET). A hematoxilina e eosina (H.E), picrossírius, Gordon e Sweets, Sudan black e o ácido peródico de Schiff (PAS) são colorações usadas respectivamente para observar a morfologia do fígado, colágeno, fibras reticulares, gordura e glicogênio. Foram encontrados os espaços portahepáticos.O glicogênio mostrou média de 5,68%, o conteúdo lipídico, média de 9,83%, o colágeno média de 14,71% e as fibras reticulares média de 5,96%. Quanto à MET notou-se no citoplasma dos hepatócitos, numerosas mitocôndrias, glicogênio, muitas gotas de gordura, alguns lisossomos, retículo endoplasmático granular ao redor das mitocôndrias, algumas células estreladas, célula em degeneração e o canalículo biliar ao centro. Provavelmente o quadro sugestivo de esteatose é resultante do estado nutricional dos animais. Estes resultados demonstraram que os hepatócitos dos avestruzes são muito similares às outras aves, como também muito semelhantes à estrutura e ultraestrutura das células do fígado de mamíferos.

Orientador: Sarah Arana Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Atrazina é um herbicida muito usado em agricultura intensiva e encontrado com alta freqüência em recursos hídricos na região do Pantanal Mato-grossense. Assim, devido aos riscos que a atrazina pode trazer à ictiofauna da região, foi proposta deste trabalho determinar a CL50 da atrazina em alevinos de pacu (Piaractus mesopotamicus). A determinação da CL50 em 96 h em sistema estático, realizada em duplicata, foi conduzida em aquários de vidro com 8 peixes cada, de peso médio de 5,06±0,31g, avaliando-se as seguintes concentrações nominais de atrazina: 0; 13,2; 17,6; 22,0; 26,4; 30,8; 35,2; 39,6 mg L-1, realizando-se também análise comportamental e análise anatomopatológica. Experimento de intoxicação aguda foi realizado em duplicata, nas mesmas condições do anterior, com a concentração da CL50 obtida (28,58 mg L-1), sendo empregados 6 exemplares de peso médio 6,68±0,36g. Amostras hepáticas e mesonéfricas foram colhidas e processadas para análise de microscopia de luz (ML), realizando-se nestas amostras análise semi-quantitativa das alterações encontradas, e para análise de microscopia eletrônica de transmissão (MET). Quanto à avaliação comportamental, foram observados nos grupos tratados: o escurecimento da pigmentação da pele, várias alterações na intensidade do movimento, perda de equilíbrio e presença da ação de boquejamento. Na avaliação anatomopatológica, foram observadas nos grupos tratados: dilatação da região ventral, exoftalmia, protrusão labial, hiperemia no opérculo e em todas as nadadeiras, presença de ar e/ou água no estômago e ascite sanguinolenta. Quanto à análise histopatológica, o grupo controle apresentou a típica morfologia hepática e renal para a espécie. No grupo tratado, severas alterações histopatológicas foram observadas em ML, a mais significativa em fígado foi a presença de inclusões hialinas no citoplasma dos hepatócitos e em rim a degeneração do túbulo proximal. À MET demonstrou que a atrazina causou severas alterações de organelas membranosas, sugestivas de estresse oxidativo e peroxidação lipídica, sendo as alterações mais freqüentes no fígado: inclusão lipídica nuclear e citoplasmática, tumefação do retículo endoplasmático rugoso e de mitocôndrias, degeneração do canalículo biliar com redução na quantidade de microvilos, e em rim, no túbulo proximal (TP): vacúolos citoplasmáticos e aumento do espaço intercelular na porção basolateral, figuras de mielina, tumefação mitocondrial, e, raramente, degeneração do TP. Estas alterações são compatíveis com intoxicação química, demonstrando que estes órgãos são bons biomarcadores de contaminação aquática em peixes. E por fim, o valor da CL50 sugere que a atrazina é levemente tóxica para o pacu, porém pelos demais resultados observados se inferem que só analisar a mortalidade não é o suficiente para determinar o dano causado por agrotóxicos em peixes, assim, recomenda-se o emprego de vários biomarcadores, tais como: análises comportamentais, histopatológicas, bioquímicas, etc. Abstract: Atrazine is a widely used herbicide in intensive agriculture and a frequent contaminant of waterways in the Pantanal region of Brazil. In view of the potential risks of atrazine to the ichthyofauna of this region, in this work we examined the LC50 of atrazine in pacu (Piaractus mesopotamicus) fingerlings. For this study, fish (5.06 ± 0.31g; mean±SD) were housed eight per glass aquarium and exposed to various concentrations of atrazine (0, 13.2, 17.6, 22.0, 26.4, 30.8, 35.2 and 39.6 mg L-1) for 96 h in static system and this experiment was performed in duplicate, after which the LC50 was determined. Changes in fish behavior and anatomopathological analysis were monitored throughout the experiment. The histopathological alterations caused by atrazine were examined in six fish (6.68 ± 0.36g; mean±SD) using the LC50 (28.58 mg L-1) calculated from the concentration-response curve obtained above. This experiment was done in 96h in static system and performed in duplicate. Liver and mesonephric samples were processed for light microscopy (LM), performing in these samples the semi-quantitative analysis of the changes found, and for transmission electron microscopy (TEM) analysis. Exposure to atrazine caused darkening of the skin, alterations in the intensity of movements, loss of balance and an increase in the frequency of gasping. Anatomopathological assessment revealed dilation of the ventral region, exophthalmia, lip protrusion, skin hyperemia in the opercular region and in all fins, the presence of air and/or water in the stomach, and bloody ascites after the herbicide exposition. The histopathological analysis revealed the typical hepatic and renal morphology for the specie, in control group. Several histopathological changes were observed in exposed fish, but the changes most significant were: in liver, the presence of hyaline inclusions in the cytoplasm of hepatocytes, and proximal tubule (PT) degeneration in the kidney. The ultrastructure showed that the atrazine caused several membrane alterations suggestive of oxidative stress and lipid peroxidation. The most frequent in liver, TEM findings, were: nuclear and cytoplasmic lipid inclusions, swollen rough endoplasmic reticulum and mitochondria, degeneration in the bile canaliculi with a loss in the number of microvilli, and for kidney: an increase in the intercellular space of the basolateral region, myelin figures, swollen mitochondria, and, rarely, degeneration of PT. These lesions are consistent with chemical intoxication and indicate that the liver and kidney are good biomarkers of aquatic contamination in pacu. And, finally, the LC50 value suggests that atrazine is not highly toxic to pacu, however, use of mortality index in acute toxicity as the sole marker in ecotoxicological assays is inadequate and should be completed with other biomarkers such as behavioral, histopathology, biochemistry analyses, etc. Mestrado Histologia Mestre em Biologia Celular e Estrutural

Orientador: Francisco Javier Hernandez Blazquez Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Engenharia de Alimentos Resumo: Dois produtos comerciais distintos foram utilizados para avaliar os efeitos dos probióticos quanto à imunoestimulação, proteção contra as diarréias e às possíveis alterações na mucosa intestinal de leitões. Um deles veicula cepas de Lactobacillus acidophilus inativadas; o outro é composto por cultivos mistos de L. acidophilus, Enterococcus faecium e Bifidobacterium bifidum, viáveis. Os animais foram separados em 3 grupos: 1 - grupo de controle; 2 - grupo tratado com probiótico inativado, Grofactor; e 3 - grupo tratado com probiótico composto por células viáveis, Biobac. Os grupos foram tratados durante 30 dias, com 2 doses semanais, exceto o grupo de controle, que recebeu leite materno, ração e água. Ao final do período, foi coletado sangue de todos os animais avaliados para verificação dos níveis de IgA no soro pelo método de ELISA. De cada grupo foram sacrificados 3 animais e procedeu-se à coleta de fragmentos do intestino delgado e linfonodos mesentéricos para estudo morfológico das vilosidades intestinais e identificação das células IgApositivas. Os animais que receberam o probiótico com células viáveis (Biobac) apresentaram o teor de IgA no soro mais elevado que o grupo suplementado com bactérias inativadas (Grofactor) e o grupo controle. A altura das vilosidades intestinais também foi maior no grupo que recebeu bactérias viáveis durante o período de aleitamento. Esses dados sugerem que a adição de probióticos ativos na dieta dos leitões recém-nascidos e/ou após o desmame melhora o desempenho e a saúde dos animais. Abstract: The immunostimulant and antidiarrheic role of two probiotic products were evaluated. Their effect in the intestinal mucosa of piglets was also investigated. One of the products contains inactivated strains of Lactobacillus acidophilus while the other one is composed by live mixed cultures of L. acidophilus, Enterococcus faecium, and Bifidobacterium bifidum bacteria. Three groups of animals were created: 1 - control group; 2 - animais that received the inactivated probiotic product; 3 - animals that received the probiotic product with live cells. The products were given twice weekly for 30 days with the exception of the control group. At the end of the experimental period, blood samples were taken and the 19A serum levels were measured by ELISA method. Three animals from each group were sacrificed and fragments of the small intestine and mesenteric Iymphonodes were microscopically examined to investigate the size of intestinal glands and villosities and the presence of IgA - positive cells. The animais that received live cells showed higher 19A serum levels, longer intestinal villosities and more IgA - positive cells in the Iymphonodes and intestinal mucosa than the other animals. Their body weight was also higher. These data suggest that live probiotics, when given to newbom pos - weaning piglets, are more efficient than inactivated probiotics in inducing intestinal changes that help to improve the health and performance of swine.
