Abstract

We estimate the mass of the compact object in the gamma-ray binaries LSI+61303 and MWC 148, using the latest data for the inclination, orbital motion and assuming that the orbital plane coincides with the equatorial plane of the Be star. For LSI+61303 we find the mass of the compact object to be most likely in the range 1.3 M_sun < M_2 < 2.0 M_sun, which means that it is probably a neutron star. For MWC 148, we find the mass of the compact object in a higher range, 2.1 M_sum < M_2 < 7.3 M_sun, which increases the chances for this system to host a black hole companion., Comment: 4 pages, submitted

Abstract

Granulocytic myeloid-derived suppressor cells (G-MDSCs) promote tumor growth and immunosuppression in multiple myeloma (MM). However, their phenotype is not well established for accurate monitoring or clinical translation. We aimed to provide the phenotypic profile of G-MDSCs based on their prognostic significance in MM, immunosuppressive potential, and molecular program. The preestablished phenotype of G-MDSCs was evaluated in bone marrow samples from controls and MM patients using multidimensional flow cytometry; surprisingly, we found that CD11b+CD14−CD15+CD33+HLADR− cells overlapped with common eosinophils and neutrophils, which were not expanded in MM patients. Therefore, we relied on automated clustering to unbiasedly identify all granulocytic subsets in the tumor microenvironment: basophils, eosinophils, and immature, intermediate, and mature neutrophils. In a series of 267 newly diagnosed MM patients (GEM2012MENOS65 trial), only the frequency of mature neutrophils at diagnosis was significantly associated with patient outcome, and a high mature neutrophil/T-cell ratio resulted in inferior progression-free survival (P < .001). Upon fluorescence-activated cell sorting of each neutrophil subset, T-cell proliferation decreased in the presence of mature neutrophils (0.5-fold; P = .016), and the cytotoxic potential of T cells engaged by a BCMA×CD3-bispecific antibody increased notably with the depletion of mature neutrophils (fourfold; P = .0007). Most interestingly, RNA sequencing of the 3 subsets revealed that G-MDSC–related genes were specifically upregulated in mature neutrophils from MM patients vs controls because of differential chromatin accessibility. Taken together, our results establish a correlation between the clinical significance, immunosuppressive potential, and transcriptional network of well-defined neutrophil subsets, providing for the first time a set of optimal markers (CD11b/CD13/CD16) for accurate monitoring of G-MDSCs in MM.

Abstract

© The Author(s) 2020., Detecting persistent minimal residual disease (MRD) allows the identification of patients with an increased risk of relapse and death. In this study, we have evaluated MRD 3 months after transplantation in 106 myeloma patients using a commercial next-generation sequencing (NGS) strategy (LymphoTrack®), and compared the results with nextgeneration flow (NGF, EuroFlow). The use of different marrow pulls and the need of concentrating samples for NGS biased the applicability for MRD evaluation and favored NGF. Despite that, correlation between NGS and NGF was high (R2 = 0.905). The 3-year progression-free survival (PFS) rates by NGS and NGF were longer for undetectable vs. positive patients (NGS: 88.7% vs. 56.6%; NGF: 91.4% vs. 50%; p < 0.001 for both comparisons), which resulted in a 3-year overall survival (OS) advantage (NGS: 96.2% vs. 77.3%; NGF: 96.6% vs. 74.9%, p < 0.01 for both comparisons). In the Cox regression model, NGS and NGF negativity had similar results but favoring the latter in PFS (HR: 0.20, 95% CI: 0.09–0.45, p < 0.001) and OS (HR: 0.21, 95% CI: 0.06–0.75, p = 0.02). All these results reinforce the role of MRD detection by different strategies in patient prognosis and highlight the use of MRD as an endpoint for multiple myeloma treatment.

Abstract

Airborne observations of fluorescent aerosol were made aboard an airship during CloudLab, a series of flights that took place in September and October of 2013 and covered a wideband of longitude across the continental U.S. between Florida and California and between 28 and 37 N latitudes. Sampling occurred from near the surface to 1000 m above the ground. A Wideband Integrated Bioaerosol Sensor (WIBS-4) measured average concentrations of supermicron fluorescent particles aloft (1 µm to 10 µm), revealing number concentrations ranging from 2.1 ± 0.8 to 8.7 ± 2.2 × 10[superscript 4] particles m[superscript −3] and representing up to 24% of total supermicron particle number. We observed distinct variations in size distributions and fluorescent characteristics in different regions, and attribute these to geographically diverse bioaerosol. Fluorescent aerosol detected in the east is largely consistent with mold spores observed in a laboratory setting, while a shift to larger sizes associated with different fluorescent patterns is observed in the west. Fluorescent bioaerosol loadings in the desert west were as high as those near the Gulf of Mexico, suggesting that bioaerosol is a substantial component of supermicron aerosol both in humid and arid environments. The observations are compared to model fungal and bacterial loading predictions, and good agreement in both particle size and concentrations is observed in the east. In the west, the model underestimated observed concentrations by a factor between 2 and 4 and the prescribed particle sizes are smaller than the observed fluorescent aerosol. Abstract

Persistence of minimal residual disease (MRD) after treatment for myeloma predicts inferior outcomes, but within MRD-positive patients there is great heterogeneity with both early and very late relapses. Among different MRD techniques, flow cytometry provides additional information about antigen expression on tumor cells, which could potentially contribute to stratify MRD-positive patients. We investigated the prognostic value of those antigens required to monitor MRD in 1265 newly diagnosed patients enrolled in the GEM2000, GEM2005MENOS65, GEM2005MAS65 and GEM2010MAS65 protocols. Overall, CD19pos, CD27neg, CD38lo, CD45pos, CD81pos, CD117negand CD138loexpression predicted inferior outcomes. Through principal component analysis, we found that simultaneous CD38lowCD81posCD117negexpression emerged as the most powerful combination with independent prognostic value for progression-free survival (HR:1.69; P=0.002). This unique phenotypic profile retained prognostic value among MRD-positive patients. We then used next-generation flow to determine antigen stability throughout the course of the disease, and found that the expression of antigens required to monitor MRD is mostly stable from diagnosis to MRD stages, except for CD81 whose expression progressively increased from baseline to chemoresistant tumor cells (14 vs 28%). Altogether, we showed that the phenotypic profile of tumor cells provides additional prognostic information, and could be used to further predict risk of relapse among MRD-positive patients.

Abstract

Microelectronic industry is driven by the continuous miniaturization process conducing to the introduction of new materials. These materials are subjected to stresses mainly due to thermal mismatch, microstructural changes or process integration which can be in the origin of mechanical reliability issues. To study these phenomena and even electromigration a good mechanical characterization of the materials is needed. This work aims at developing tests to assess fracture or elastoplastic behavior of thin and very thin metallic and polymeric films. The tests developed are based on indentation combined with sample preparation using mostly FIB. Among other techniques, different test geometries for microbeams have been evaluated and quantitative data have been obtained combining experimental results with analytical or numerical models, depending on the material under study. For the particular case of Cu cantilevers, a strong dependence of their plastic behavior on the orientation of the grains close to the fixed end has been detected. Abstract

Composts and vermicomposts from a municipal composting plant in northwestern Patagonia, both having undergone a thermophilic phase, (with the vermicompost being inoculated with earthworms after the thermophilic stage) and a nonthermophilic backyard vermicompost were studied. Their effects on soil biological and biochemical properties and plant growth were evaluated in laboratory incubations and a greenhouse trial, using a degraded volcanic soil amended at rates of 20 and 40 g kg-1 of vermicompost or compost. Between the two municipal products, the vermicompost had significantly larger nutrient concentrations than the compost; when mixed with the soil, the vermicompost also had higher microbial populations size and activity, and produced increased ryegrass yields. Compared to the municipal compost, the backyard vermicompost had similar or higher nutrient concentrations but its effects on soil microbial biomass, soil microbial activity and ryegrass yields were lower. Our results suggest that no generalization can be made regarding the higher quality of vermicomposts vs. composts, because the product quality depends both on the original materials and the technology employed. 40. Urinary prostaglandins (PGE2 and PGI2) in hyporeninaemic hypoaldosteronism in diabetic patients with chronic renal failure.

Abstract

The present work studies the urinary excretion of PGE and PGI (6-keto PGF 1α) in 11 insulin-dependent diabetic patients with chronic renal failure with a glomerular filtration rate of 33.9 ±9.03 ml/min who had hyporeninaemic hypoaldosteronism to evaluate the influence of these prostaglandins on the appearance of this latter process. The results obtained in this group of patients were compared with those of a control group of healthy individuals, another group of nine non-diabetic patients with CRF, and a last group of eight insulin-dependent diabetic patients with normal renal functión to evaluate to what extent the possible variations in prostaglandin excretion could be related to the diabetes, CRF, or a conjunction of both processes. The results of the groups of diabetic patients with CRF were C 33.9 ±9.03 ml/min, decreased (< 0.0001) with respect to the control group and with no difference with the CRF group without diabetes; plasma potassium (4.7 ±0.4 mEq/l), increased <0.005) with respect to the values found in the control group; plasma bicarbonate (17.8 ± 1.8 mEq/l), decreased (P< 0.005) with respect to the control group and also, though not significantly, with respect to the group of non-diabetic patients with CRF. Plasma aldosterone (pg/ml): resting 44.3±14.9; standing 65.7 ±63.5 and post-frusemide 65.5 ±58.6, decreased (P<0.01) with respect to the other three groups. Plasma renin activity (PRA) (ng/ml/h): resting 0.34±0.3; standing 0.6 ± 0.4, post-fmsemide 0.9 ±0.5, decreased significantly with respect to the other three groups. PGE (pg/mg Cr): basal 1720±397; post-frusemide 2636±462, increased (P<0.05) with respect to the control group and that of the diabetic patients without CRF, but with no differences compared with the non-diabetic patients with CRF. PGI (pg/mg cr): basal 369 ±45 and post-frusemide 699 ± 103, increased (P<0.01) with respect to the controls and diabetic patients with normal renal function and with no differences compared with the non-diabetic patients with CRF. Our findings indicate that patients with diabetes mellitus and CRF showing hyporeninaemic hypoaldosteronism have high urinary excretion of PGE and PGI. The increase in the urinary prostaglandins is related to CRF. The data rule out the hypothesis that the hyporeninaemic hypoaldosteronism of these patients is due to a deficit of prostaglandins.
