Embryos can be produced from prepubertal donor animals using laparoscopic ovum pickup and in vitro embryo production technologies (LOPU-IVEP). Together, these tools can shorten the interval between generations, rapidly accelerating the rate of genetic gain. Here, we assessed the impact of different gonadotropin stimulation protocols in Mediterranean water buffalo heifer calves aged between 2 and 6 months old. Following gonadotropin stimulation, LOPU was performed at two-week intervals, with animals receiving different protocols on subsequent LOPUs. After collection, the cumulus-oocyte complexes (COCs) were matured and fertilized in vitro, and embryos were cultured to the blastocyst stage followed by transfer into synchronized adult recipients. The number and size of follicles aspirated during LOPU, the number and quality of COCs recovered, as well as cleavage, embryo development and pregnancy rates were assessed. First, we evaluated the impact of using FSH with and without eCG (administered 24-h prior to LOPU) and found that a combination of FSH and eCG was able to significantly improve embryo development rates (20.6 ± 2.0% vs. 9.0 ± 3.6%; P 0.05). Second, we compared this protocol to a slow-release formulation of FSH reconstituted in hyaluronan. In addition to requiring less work to prepare the animals for LOPU, this slow-release formulation yielded numerically higher, but not statistically different, average number of recovered COCs (14.4 ± 2.1 vs. 10.3 ± 2.0; P 0.05) and embryo development rates (22.9 ± 4.7% vs. 14.1 ± 5.2%; P 0.05) compared to FSH given every 12 h. Next, we compared the length of gonadotropin treatment over 3-, 4- and 5-days prior to LOPU and found that as the length of gonadotropin treatment increased, although the number of COCs recovered steadily decreased (14.1 ± 2.4 vs. 8.7 ± 1.0 vs. 6.9 ± 0.7; P 0.05), the embryo development rates steadily increased (14.4 ± 3.9 vs. 27.3 ± 4.4 vs. 35.9 ± 7.0; P 0.05), presumably due to an increase in the proportion of large follicles at the time of LOPU. Numerically, the 4-day treatment yielded more transferrable embryos per donor per LOPU (2.70 ± 0.5) than 3-day (1.94 ± 0.6) and 5-day (2.25 ± 0.5) treatments. Finally, following embryo transfer, 26 of 90 recipient females became pregnant (28.9%). Pregnancies were established from all treatments, which suggests that post-implantation development was not affected among the gonadotropin treatments assessed.

Porcine in vitro fertilization often results in low embryo development rates compared to other livestock species, which is often associated with either a low fertilization rate or high incidence of polyspermy. Since the quality of oocyte maturation is known to play a significant role in oocyte competence, we investigated the impact of supplementing in vitro maturation (IVM) medium containing porcine follicular fluid (pFF) with the growth factors FGF2, LIF and IGF1 (FLI), along with different combinations of cysteine, melatonin and ITS, on cumulus cell expansion, oocyte meiotic maturation, fertilization outcome, embryo development and blastocyst cell numbers. Maturation medium containing pFF yielded the greatest cumulus expansion. Compared to pFF and FLI individually, using pFF and FLI together resulted in the best embryo development rates over total oocyte placed in IVF (12.5% vs. 15.0% vs. 26.6%, respectively). Supplementation of IVM medium containing pFF and FLI with either cysteine, melatonin or insulin-transferrin-selenium, revealed that cysteine was essential to improve embryo development, while melatonin and ITS had a limited impact on improving blastocyst rates. Finally, we observed that pig oocytes matured in medium supplemented with pFF, FLI, cysteine and melatonin had a high proportion of monospermic zygotes (68.2%) and low proportion of polyspermic zygotes (15.9%) following IVF and yielded superior cleavage (78.2%) and blastocyst (32.0%) rates.

CRISPR/Cas9 technology is a powerful tool used for genome manipulation in different cell types and species. However, as with all new technologies, it still requires improvements. Different factors can affect CRISPR/Cas efficiency in zygotes, which influence the total cost and complexity for creating large-animal models for research. This study evaluated the importance of zygote cell cycle stage between early-injection (within 6 h post activation/fertilization) versus late-injection (14–16 h post activation/fertilization) when the CRISPR/Cas9 components were injected and the inhibition of the homologous recombination (HR) pathway of DNA repair on gene editing, embryo survival and development on embryos produced by fertilization, sperm injection, somatic cell nuclear transfer, and parthenogenetic activation technologies. Injections at the late cell cycle stage decreased embryo survival (measured as the proportion of unlysed embryos) and blastocyst formation (68.2%; 19.3%) compared to early-stage injection (86.3%; 28.8%). However, gene editing was higher in blastocysts from late-(73.8%) vs. early-(63.8%) injected zygotes. Inhibition of the HR repair pathway increased gene editing efficiency by 15.6% in blastocysts from early-injected zygotes without compromising embryo development. Our finding shows that injection at the early cell cycle stage along with HR inhibition improves both zygote viability and gene editing rate in pig blastocysts.

Combining somatic cell nuclear transfer (SCNT) with genome editing technologies has emerged as a powerful platform for the creation of unique swine lineages for agricultural and biomedical applications. However, successful application of this research platform is still hampered by the low efficiency of these technologies, particularly in attaining complete cell reprogramming for the production of cloned pigs. Treating SCNT embryos with histone deacetylase inhibitors (HDACis), such as Scriptaid, has been routinely used to facilitate chromatin reprogramming after nuclear transfer. While increasing histone acetylation leads to a more relaxed chromatin configuration that facilitates the access of reprogramming factors and DNA repair machinery, it may also promote the expression of genes that are unnecessary or detrimental for normal embryo development. In this study, we evaluated the impact of inhibiting both histone deacetylases and RNA synthesis on pre- and post-implantation development of pig SCNT embryos. Our findings revealed that transcription can be inhibited for up to 40 h of development in porcine embryos, produced either by activation, fertilization or SCNT, without detrimentally affecting their capacity to form a blastocyst and their average number of cells at this developmental stage. Importantly, inhibiting RNA synthesis during HDACi treatment resulted in SCNT blastocysts with a greater number of cells and more abundant transcripts for genes related to embryo genome activation on days 2, 3 and 4 of development, compared to SCNT embryos that were treated with HDACi only. In addition, concomitant inhibition of histone deacetylases and RNA synthesis promoted the full reprograming of somatic cells, as evidenced by the normal fetal and full-term development of SCNT embryos. This combined treatment may improve the efficiency of the genome-editing + SCNT platform in swine, which should be further tested by transferring more SCNT embryos and evaluating the health and growth performance of the cloned pigs.

Laparoscopic Ovum Pick-Up (LOPU) is the most reliable and efficient technique for collecting high quality oocytes from live animals in certain species or age groups, allowing its use for In Vitro Embryo Production (IVEP) and Somatic Cell Nuclear Transfer (SCNT). In order to maximize the number and quality of oocytes collected by donor, it is necessary to synchronize estrus and stimulate follicular growth using hormonal protocols that vary according to species. There are 2 big categories of applications for the LOPU-IVEP technology in production animals, those in which it acts as an alternative to MOET (competitive applications) and those in which it doesn’t compete with MOET as it cannot be done in those categories (non competitive applications). In wild animals, LOPU can play an important role in conservation programs for endangered species when associated with effective IVEP and has been done in several species. It has commercial application in sheep, goats, cattle and buffaloes calves. Repeating LOPU procedures in the same female does not cause sequels with impact on the female's reproductive life, even when performed on prepubertal or wild animals.

Laparoscopic ovum pick-up (LOPU) conducted on bovine and buffalo calves of 2-6-month of age, followed by in vitro embryo production and transfer into synchronous adult recipients, is a powerful tool for accelerated genetic gain and early dissemination of top genetics. In its current state, the technology is characterized by higher oocyte recovery rates, lower oocyte-to-embryo yields, and similar pregnancy and term development rates compared with adult counterparts. Improvements in oocyte competence have been made in recent years mainly through gonadotropin stimulation protocols tailored for prepubertal donors. These advances have brought the technology to the point of been apt for commercial application. However, future research must focus on increasing the proportion of fully competent oocytes recovered from calves thereby further empowering the role this technology platform can play in programs for accelerated dissemination of superior genetics.

The use of oocytes recovered from prepubertal donors for in vitro embryo production has great potential for accelerating the rate of genetic gain in the dairy industry. However, these oocytes are known to be less developmentally competent than those from adult donors. In this study, we investigated the effect of age and gonadotropin stimulation in Holstein heifers subjected to oocyte collection every two weeks between 2 and 6 months of age. In order to assess the effect of gonadotropin stimulation, animals were subjected to one of three treatments, namely Short (ST; 36-42 h), Long (LT; ≥72 h) and No Treatment (NT) prior to laparoscopic ovum pick-up (LOPU). Our results show that the LT significantly improved the proportion of large follicles (5 mm diameter) present in the ovary (LT 34.0% vs. ST 11.2% vs. NT 2.4%, P 0.05), as well as the percentage of good-quality cumulus oocyte complexes (COCs) recovered (LT 95.3 ± 18% vs. ST 85.4 ± 22% vs. NT 82.2 ± 14%, P 0.05) and blastocyst rate (LT 36.7 ± 26% vs. ST 18.3 ± 15% vs. NT 16.7 ± 9%, P 0.05). Recovery rate was affected by treatment (LT 70.4 ± 25 vs. ST 85.4 ± 29 vs. NT 72.7 ± 23, P 0.05). To assess the impact of age, data was grouped into100 days (A), 100-130 days (B) and130 days (C) of age at LOPU. We found that as animals got older, although the average number of COCs per donor per LOPU declined (A: 17.5 ± 11 vs. B: 14.7 ± 7 vs. C: 11.9 ± 8), the blastocyst rate increased (A: 12.8 ± 20% vs. B: 17.1 ± 21% vs. C: 21.8 ± 25%, P 0.05). We also evaluated the incidence of polyspermy and confirmed it is a critical limitation for IVF in calf oocytes. The incidence of polyspermy was unaffected by gonadotropin treatment, but significantly decreased with age. The capacity for full development to term of in vitro produced embryos from calf oocytes was tested by embryo transfer into 21 synchronized adult recipients, which resulted in 13 pregnancies (62%), full development to term and healthy calves born. Finally, the study allowed evaluating the safety of the procedure since, on average, each animal was subjected to 8 LOPU procedures over a period of 4 months. Our results showed that the procedure is safe (no incidents during laparoscopy), and was not harmful for the reproductive future of the animals, as those that were bred became pregnant after reaching sexual maturity.

Threatened of extinction in Brazil, the jaguar is the largest predator in Latin America, playing an important role in the ecosystem where it is inserted. Despite of some important studies in this species, its reproductive physiology needs to be better understood for the development of more effective reproductive biotechnologies. One well studied biological aspect is the occurrence of ovulation following stimulation of the vaginal floor during copulation. This mechanical stimulation is responsible for the induction of ovulation in domestic and wild cats. Thus, the objective of the present study was to evaluate whether ovulation in captive jaguars can be induced by the above-mentioned mechanical stimulation but also by other forms of sensory stimulation. Nine jaguar (Panthera onca) females and six males were divided into three groups: Group 1 (four females), in which the females were close to the males, but with no possibility of copulation; Group 2 (three females), in which the females were in the same enclosure as the males, thereby being able to copulate; and Group 3 (two females), in which the females were completely isolated from any male of the species. Follicular growth was stimulated by administration of 800 IU of eCG, conducted three or five days before LOPU. In order to have certainty about the occurrence of ovulation, the ovaries were assessed by laparoscopy allowing the visualization of ovarian structures including follicles, recent ovulations, corpora hemorrhagica (CH) and corpora lutea (CL). Multiple CH were observed in the 2 females housed with males, as well as the two females kept in proximity of males, i.e. only visual contact (Group 1). None of the females in complete isolation from males showed CH or ovulation spots (Group 3). In summary, the seven females in contact or near proximity with males had multiple ovulations, regardless of copulation or not. Based on this evidence, we have established that jaguars are capable of two forms of induced ovulation: 1) the "mechanical" coitus-induced form that is traditional and thoroughly described for felines; 2) a "sensorial" form in which the nearby presence of a male can induce ovulation through the visual, olfactory and/or auditive senses. Further research is required for establishing the mechanisms and chemical mediators of sensorial stimulation. In addition, and consistent with expected results, we confirmed that females that received the same stimulation are incapable of ovulating when not exposed to any form of stimulation by males.

In Experiment I, during the non-breeding season, after intravaginal devices containing progesterone (P4) were withdrawn (n = 28), estrous rates were greater with treatment with 400 IU eCG (P 0.05). In Experiment II (non-breeding season), ewes of one group were treated with 300 IU eCG (n = 8) and of two groups were treated with 10 mg FSH. In one FSH group, 250 μg estradiol benzoate (EB) were administered after 24 h (n = 9); in the other, 4 μg GnRH were administered after 36 h (n = 10). Serum P4 concentrations were greater in eCG-treated ewes (P 0.05). In Experiment III (breeding season), the treatments were: 300 IU eCG; 250 μg estradiol cypionate; 250 μg EB; or control (n = 22). Follicular growth was greater for eCG-treated ewes within 0−24 h and for control ewes within 48−72 h (P = 0.001). Although estrous and ovulation rates did not differ (P > 0.05), all eCG-treated ewes had ovulations. During the non-breeding season, FSH treatment promoted follicular growth but did not induce ovulations. For FTAI regimens, eCG was more effective than FSH plus GnRH and estradiol esters in inducing estrus and ovulation.

Insufficient epigenetic reprogramming is incompatible with normal development of embryos produced by somatic cell nuclear transfer (SCNT), but treatment with histone deacetylases inhibitors (HDACi) enhances development of SCNT embryos. However, the mechanisms underpinning HDACi benefits in SCNT embryos remain largely uncharacterized. We hypothesized that, in addition to enhancing reprogramming, HDACi treatment may promote expression of genes not required for early development of SCNT embryos. To test this hypothesis, RNA synthesis was inhibited by treating bovine SCNT embryos with 5,6-dichlorobenzimidazole 1-β-D-ribofuranoside (DBR), which were concomitantly treated or not with Scriptaid (Scrip; an HDACi). Development to the blastocyst stage was significantly increased by treatment with Scrip alone (26.6%) or associated with DRB (28.6%) compared to Control (17.9%). The total number of nuclei was significantly improved only in embryos that were treated with both Scrip + DRB. Nuclear decondensation after SCNT was significantly increased by DRB treatment either alone or associated with Scrip. The relative mRNA expression, evaluated during the embryo genome activation (EGA) transition, revealed that some KDMs (KDM1A, KDM3A, KDM4C and KDM6A) and DNMT1 where prematurely expressed in Scrip-treated embryos. However, treatment with Scrip + DRB inhibited early mRNA expression of those genes, as well as several other KDMs (KDM4A, KDM4B, KDM5A, KDM5B, KDM5C and KDM7A) compared to embryos treated with Scrip alone. These findings revealed that HDACi improved development in SCNT embryos compared to Control, but altered the expression of genes involved in epigenetic regulation and did not improve embryo quality. Inhibition of RNA synthesis during HDACi treatment enhanced nuclear chromatin decondensation, modulated gene expression and improved SCNT embryo quality.

Oocytes collected from prepubertal animals are known to be less developmentally competent than those from adult animals. There is evidence suggesting that acquisition of developmental competence in bovine oocytes may be linked to the expression profile of genes in the granulosa cells (GCs). Cumulus-oocyte complexes (COC) and GCs were collected from 12 Holstein heifers between 2 and 6 months of age (nine follicle-stimulating hormone [FSH] treated and three untreated) and eight FSH-treated cows. The COCs from prepubertal animals were matured, fertilized, and cultured in vitro to assess development to the blastocyst stage. The relative messenger RNA (mRNA) abundance of FSHR, StAR, CYP19A1, HSD3B1, CX43, FOXO1, and XIAP in GCs were quantified by real-time quantitative polymerase chain reaction. Results from this study revealed that GCs of prepubertal animals respond to FSH treatment by increasing mRNA levels of genes promoting estradiol synthesis and follicular growth ( FSHR and CYP19A1), and preventing cell apoptosis ( XIAP), and by decreasing mRNA levels of genes promoting progesterone production ( StAR and HSD3B1). This study also revealed that the relative mRNA abundance of FOXO1 in GCs is associated with oocyte competence to support embryo development to the blastocyst stage in prepubertal Holstein heifers.

Although fixed-time artificial insemination (FTAI) protocols are available for sheep, estrus resynchronization has not been previously reported. The objectives of this study were to evaluate the effect of estrus resynchronization with exogenous progestogen on endogenous progesterone levels and to compare pregnancy rates after two consecutive estrus synchronizations in ewes. In Experiment 1, ewes (n = 20) received an intravaginal device (IVD) containing 60 mg medroxyprogesterone acetate (MPA) for 10 days. At the IVD withdrawal (D0), ewes received 250 IU eCG and were allocated into two treatments: either no further treatment (Control; n = 10) or estrus resynchronization (Resynch; n = 10) from D12 to D19. Serum progesterone (P4) levels did not differ at D12 and D19 (P 0.05), but were greater at D15 for the Control compared with the Resynch group (P 0.05). In experiment 2, ewes (n = 250) were submitted to a first synchronization protocol followed by estrus detection and either artificial insemination (AI) or natural mating (NM). Subsequently, ewes were divided into two groups: Control (n = 104): which received no further treatment and were bred by NM; and Resynch (n = 146): which were submitted to a second synchronization starting on D14 (first IVD withdrawal = D0) and to NM after second IVD withdrawal (D20). Cumulative pregnancy rates did not differ between the Control (67.3%, 70/104) and Resynch (62.3%, 91/146) groups. In a third experiment, ewes (n = 83) were bred by two consecutive FTAI within a 20-day interval. Pregnancy rates after the first (30.1%, 25/83) and the second FTAI (36.2%, 21/58) did not differ (P 0.05). In conclusion, although exogenous progestogen supplementation reduced circulating levels of P4, pregnancy maintenance was unaffected. Estrus resynchronization in ewes is feasible, resulting in similar fertility after the first and the second services. The use of resynchronization coupled with artificial insemination using semen from genetically superior rams may potentially accelerate genetic improvement in sheep herds by allowing a higher differential selection compared with natural breeding.

International audience; Genetically engineered sheep and goats represent useful models applied to proof of concepts, large-scale production of novel products or processes, and improvement of animal traits, which is of interest in biomedicine, biopharma, and livestock. This disruptive biotechnology arose in the 80s by injecting DNA fragments into the pronucleus of zygote-staged embryos. Pronuclear microinjection set the transgenic concept into people's mind but was characterized by inefficient and often frustrating results mostly because of uncontrolled and/or random integration and unpredictable transgene expression. Somatic cell nuclear transfer launched the second wave in the late 90s, solving several weaknesses of the previous technique by making feasible the transfer of a genetically modified and fully characterized cell into an enucleated oocyte, capable of cell reprogramming to generate genetically engineered animals. Important advances were also achieved during the 2000s with the arrival of new techniques like the lentivirus system, transposons, RNA interference, site-specific recombinases, and sperm-mediated transgenesis. We are now living the irruption of the third technological wave in which genome edition is possible by using endonucleases, particularly the CRISPR/Cas system. Sheep and goats were recently produced by CRISPR/Cas9, and for sure, cattle will be reported soon. We will see new genetically engineered farm animals produced by homologous recombination, multiple gene editing in one-step generation and conditional modifications, among other advancements. In the following decade, genome edition will continue expanding our technical possibilities, which will contribute to the advancement of science, the development of clinical or commercial applications, and the improvement of people's life quality around the world.

To evaluate the feasibility of long distance transport of ovine oocytes and in vitro produced embryos. Santa Ines pluriparous sheep (n = 5) were synchronized with intravaginal devices containing 0.3 mg progesterone (CIDR®, Pfizer, Netherlands) for 10 days. Thirty-six hours prior to progesterone device removal and follicular aspiration, 0.04 mg d -cloprostenol sodium (Ciosin®, Coopers, Brasil), 200 IU FSH (Folltropin®, Bioniche, Canada) and 300 IU eCG (Novormon®, Syntex, Argentina) were administered intramuscularly. Oocyte retrieval was performed via mid-ventral laparotomy, under general anesthesia. The oocytes were evaluated and classified under a stereomicroscope and placed in cryovials with TCM 199, aerated with 5% CO2, covered with mineral oil, sealed, wrapped with parafilm and stored in a transport incubator at 38.5 °C. Oocytes were then transported via the road for 14 h to the laboratory, where the oocytes then completed more than 10 h of IVM. In vitro fertilization (IVF) was performed with frozen sperm from a single ram. On Day 3 the embryos were placed in cryovials containing SOFovine (In Vitro Brasil, Sao Paulo, Brazil), aerated with 5% CO2, covered with mineral oil, sealed, wrapped with parafilm and placed into the same portable incubator. The embryos were returned to the farm under the same transport conditions (km and h). Upon arrival at the farm, all embryos were evaluated and transferred into recipient ewes 4 days after the initial oocyte retrieval. Embryos were transferred in sets of 3 to 4, into the apex of the uterine horn, ipsilateral to the corpus luteum, by means of mid-ventral laparotomy, under general anesthesia. Pregnancy was diagnosed using transrectal ultrasonography 30 days after embryo transfer. On average 9.8 ± 5.7 oocytes (49 oocytes/5 donors) were obtained per donor, and 25 grade I morulae were transferred into 7 recipients. One pregnancy (14.3%) was confirmed resulting in a healthy lamb. These results indicate the feasibility of in vitro embryo production in sheep after maturation and embryonic development following transport over long distances.

The use of the mammary gland of transgenic goats as a bioreactor is a well established platform for the efficient production of recombinant proteins, especially for molecules that cannot be adequately produced in traditional systems using genetically engineered microorganisms and cells. However, the extraordinary demand placed on the secretory epithelium by the expression of large amounts of the recombinant protein, may result in a compromised mammary physiology. In this study, milk composition was compared between control and transgenic goats expressing high levels (1–5 g/l) of recombinant human butyrylcholinesterase in the milk. Casein concentration, as evaluated by acid precipitation, was significantly reduced in the transgenic compared with the control goats throughout lactation (P < 0.01). Milk fatty acid composition for transgenic goats, as determined by gas chromatography, was found to have significantly fewer short chain fatty acids (P < 0.01) and more saturated fatty acids (P < 0.05) compared to controls, suggesting an overall metabolic stress and/or decreased expression of key enzymes (e.g. fatty acid synthase, stearoyl-CoA desaturase). The concentration of Na+, K+, assessed by atomic absorption spectrophotometry, and serum albumin, determined by bromocresol green dye and scanning densitometry, were similar in transgenic and control goats during the first several weeks of lactation. However, as lactation progressed, a significant increase in Na and serum albumin concentrations and a decrease in K+ concentration were found in the milk of transgenic goats, while control animals remained unchanged (P < 0.01). These findings suggest that: (a) high expression of recombinant proteins may be associated with a slow-down in other synthetic activities at the mammary epithelium, as evidenced by a reduced casein expression and a decreased de-novo synthesis of fatty acids; (b) the development of permeable tight junctions may be the main mechanism involved in the premature cessation of milk secretion observed in these transgenic goats.

The production of recombinant proteins in the milk of transgenic animals has attracted significant interest in the last decade, as a valuable alternative for the production of recombinant proteins that cannot be or are inefficiently produced using conventional systems based on microorganisms or animal cells. Several recombinant proteins of pharmaceutical and biomedical interest have been successfully expressed in high quantities (g/l) in the milk of transgenic animals. However, this productivity may be associated with a compromised mammary physiology resulting, among other things, from the extraordinary demand placed on the mammary secretory cells. In this study we evaluated the lactation performance of a herd of 50 transgenic goats expressing recombinant human butyryl-cholinesterase (rBChE) in the milk. Our findings indicate that high expression levels of rBChE (range 1-5 g/l) are produced in these animals at the expense of an impaired lactation performance. The key features characterizing these transgenic performances were the decreased milk production, the reduced milk fat content which was associated with an apparent disruption in the lipid secretory mechanism at the mammary epithelium level, and a highly increased presence of leukocytes in milk which is not associated with mammary infection. Despite of having a compromised lactation performance, the amount of rBChE produced per transgenic goat represents several orders of magnitude more than the amount of rBChE present in the blood of hundreds of human donors, the only other available source of rBChE for pharmaceutical and biodefense applications. As a result, this development constitutes another successful example in the application of transgenic animal technology.

This study was aimed at developing a hormonal treatment protocol in order to optimize the proportion of pronuclear-stage embryos to be used for DNA microinjection in a goat transgenic founder production programme. A total of 46 adult BELE® and 47 adult standard goats (1–5 years old) were used as donors and recipients, respectively. They were heat-synchronized using intravaginal sponges containing 60 mg medroxyprogesterone acetate for 10 days with an injection of 125 μg cloprostenol on the morning of the eighth day. Recipients were injected with 400 IU eCG at the time of sponge removal while donors received a total of 133 mg NIH-FSH-P1 (Folltropin-V) given twice daily in decreasing doses over 3 days starting 48 h before sponge removal. Ovulation was induced in donors by injecting 100 μg of GnRH at 24 h (GnRH24) or 36 h (GnRH36) after sponge removal. Embryo recovery was performed by oviduct flushing following a standard mid-ventral laparotomy procedure. The proportion of embryos in the pronuclear stage of development was higher in the GnRH36 group (90% vs 34%, pp=0.06). One transgenic female founder was produced from the group of recipients transferred with pronuclear-stage microinjected embryos.

The objective of this study was to assess the efficacy of a novel intracytoplasmic sperm injection (ICSI) procedure, as well as the in vitro and in vivo developmental competence of goat embryos produced by ICSI. Oocyte-cumulus complexes recovered by LOPU from donors stimulated with gonadotrophins were matured in vitro. Fresh goat semen was used for ICSI following Percoll gradient washing. Tail-cut spermatozoa were microinjected into the ooplasm of goat oocytes using a piezo micropipette-driving system (PiezoDrill). In order to assess developmental competence, the ICSI-derived zygotes were cultured in one of two media systems (mTALP-mKSOM vs G1.3-G2.3) for in vitro development or were transferred into recipients for full-term development. The results suggest that cutting sperm tails using the oocyte-holding pipette coupled with the PiezoDrill is an efficient approach for goat ICSI in terms of oocyte survival, pronuclear development and initial cleavage. The mTALP-mKSOM culture system was more suitable for in vitro development of ICSI-derived goat embryos than G1.3-G2.3. This first report of full-term development of an ICSI-derived goat embryo suggests that ICSI can be applied to assisted reproduction in goats.

Oocytes collected by laparoscopic ovum pick-up (LOPU) were successfully used to produce transgenic goats by pronuclear microinjection of in vitro zygotes. Estrus cycles of 109 donor goats were synchronized using intravaginal sponges impregnated with 60 mg of medroxyprogesterone acetate and treatment with 70 mg NIH-FSH-P1 and 300 IU eCG to stimulate follicular development. Follicles were aspirated under laparoscopic observation. In vitro maturation (IVM) of oocytes was performed in M199 supplemented with hormones, kanamycin and 10% estrus goat serum. Following IVM, oocytes were cocultured with capacitated semen in TALP supplemented with 20% estrus goat serum for 15-20 h. The resulting zygotes were microinjected with a linear DNA fragment. In total, 3293 follicles were aspirated (15.7+/-9 follicles aspirated per donor) and 2823 oocytes were recovered (13.4+/-8 oocytes per donor). A total of 1366 zygotes were microinjected and transferred into 219 recipient goats by midventral laparotomy (average 6.2 embryos per recipient). A total of 150 kids were born, of which 9 (6 M: 3 F) were confirmed to be transgenic by PCR and Southern blotting analyses. These results demonstrate that acceptable transgenesis rates can be obtained in goats by DNA microinjection of in vitro produced zygotes.

The developmental potential of adult somatic nuclei after nuclear transfer (NT) into enucleated, in vitro-matured oocytes was evaluated in a dwarf breed of goat (BELE: Breed Early Lactate Early). Somatic donor cells were obtained from two different sources: 1) adult granulosa cells (GCs) and 2) fetal fibroblasts. Primary GCs were obtained from follicular aspirants after laparoscopic oocyte pick-up (LOPU) and were cryopreserved immediately. Frozen aliquots of cells were thawed and cultured until confluent and were then cultured in low serum for 4 days before use in NT. Immature oocytes were obtained by LOPU and matured before enucleation and NT. Ninety-one adult GC-derived NT embryos were transferred into eight recipients, four of which were confirmed pregnant (50%) at Day 30 by ultrasound. Fifty-four male fetal fibroblast-derived NT embryos were transferred into six recipients, one of which was confirmed pregnant (17%). All pregnancies were maintained through term. Four recipients delivered seven female kids (three sets of twins) derived from the GC cultures (7.7% of embryos transferred). The other recipient delivered two male kids (3.7% of embryos transferred). Birth weights were within the normal range for dwarf goats. One female twin and one male twin died at birth; the remaining kids appeared healthy and normal. DNA analysis confirmed that the kids were genetically identical to their respective donors. These results demonstrated that adult caprine somatic cells could direct normal development after NT.

Laparoscopic ovum pick-up (LOPU) is a convenient methodology by which oocytes can be recovered and used either for in vitro production of zygotes or as a source of cytoplasts in nuclear transfer (NT) procedures. The pregnancy and transgenesis rates achieved with IVM/IVF of LOPU-sourced oocytes followed by subsequent DNA microinjection of zygotes are similar to the rates obtained when using in vivo-produced oocytes or zygotes. Similarly, pregnancy rates and kids born by using LOPU-sourced and in vitro matured oocytes as recipient cytoplasts in NT programs are comparable with those reported by others using in vivo matured oocytes collected by oviduct flushing. The use of LOPU allows for improved control over the stage of maturation/development of the oocytes and produced zygotes, a less invasive means of recovery, thereby allowing for repeated usage of the oocyte donor animals and the ability to source the oocytes from live animals of known health status. In addition, because of large follicular responses that can be obtained from prepubertal animals, LOPU followed by IVM/IVF has demonstrated great potential for the early propagation of valuable animals, in particular, transgenic animals.

Three, genetically identical, Nigerian Dwarf bucks produced by somatic cell nuclear transfer (NT) of fetal fibroblasts were monitored for sexual maturation and fertility. Starting at four months of age, these male clones were trained to serve an artificial vagina (AV). Average age of the NT-derived bucks at first semen collection was 20 weeks, which was not different from that of other young bucks of this breed (average age at first collection = 20 weeks). Average sperm production at 5 months of age for the NT-derived bucks was 5.0 x 10(8) spermatozoa, which was comparable to that of dwarf bucks of similar age (3.4 x 10(8) spermatozoa). At seven months of age, semen collected from two NT-derived bucks was used to artificially inseminate six females (three does per buck). Five does were confirmed pregnant by ultrasound at day 42. Nine healthy kids, four males and five females, were born in March and April 2000. Viable spermatozoa were collected from one of the F1 males at 28 weeks of age. These results demonstrated that NT-derived bucks and one of their male offspring developed sexually within the normal timeframe for their breed and that the clones were fertile.

The developmental potential of caprine fetal fibroblast nuclei after in vitro transfection and nuclear transfer (NT) into enucleated, in vitro-matured oocytes was evaluated. Fetal fibroblasts were isolated from Day 27 to Day 30 fetuses from a dwarf breed of goat (BELE: breed early lactate early). Cells were transfected with constructs containing the enhanced green fluorescent protein (eGFP) and neomycin resistance genes and were selected with G418. Three eGFP lines and one nontransfected line were used as donor cells in NT. Donor cells were cultured in Dulbecco minimum Eagle medium plus 0.5% fetal calf serum for 4-8 days prior to use in NT. Immature oocytes were recovered by laparoscopic ovum pick-up and matured for 24 h prior to enucleation and NT. Reconstructed embryos were transferred as cleaved embryos into synchronized recipients. A total of 27 embryos derived from transgenic cells and 70 embryos derived from nontransgenic cells were transferred into 13 recipients. Five recipients (38%) were confirmed pregnant at Day 35 by ultrasound. Of these, four recipients delivered five male kids (7.1% of embryos transferred) derived from the nontransfected line. One recipient delivered a female kid derived from an eGFP line (7.7% of embryos transferred for that cell line). Presence of the eGFP transgene was confirmed by polymerase chain reaction, Southern blotting, and fluorescent in situ hybridization analyses. Nuclear transfer derivation from the donor cells was confirmed by single-strand confirmation polymorphism analysis. These results demonstrate that both in vitro-transfected and nontransfected caprine fetal fibroblasts can direct full-term development following NT.

Synchronization of estrus and ovulation are of paramount importance in modern livestock improvement programs. These methods are critical for assisted reproduction technologies, including artificial insemination and embryo transfer, that can increase productivity. In the current study, subcutaneous implants containing norgestomet were placed for long (14 days), medium (9 days), and short (5 days) periods of time in 70 crossbred ewes undergoing fixed-time artificial insemination. The resulting effects on estrus synchronization and conception rates were subsequently evaluated. Among the synchronized ewes, 85.7% (60/70) underwent estrus over a period of 72 h after progestagen treatment ceased. The shortest mean interval between withdrawal of the device and onset of estrus (34.2 ± 8.9 h) was observed in the G14 days of P4 group (p < 0.05). The conception rate of the G14 days of P4 group was statistically higher than that of the other groups (83.3% vs. 60.9% vs. 47.8%; p < 0.05). In conclusion, 14 days of norgestomet treatment produced higher conception rates and a greater number of pregnancies at the beginning of the breeding season.

It is well documented that incidence of fertility problems is high in lactating cows but not in heifers of the same genetic merit. Understanding the metabolic and molecular differences between fertile heifers and relatively infertile lactating cows will help us understand the pathogenesis of infertility in dairy cows. Follicular waves in lactating cows (30–50 days in milk; n=12) and heifers (n = 10) were synchronized by ultrasound-guided follicle ablation. Follicular fluid and granulosa cells of the dominant follicle were collected by ultrasound-guided aspiration along with blood sampling on Day 6 after synchronization. Dominant and subordinate follicles were larger in lactating cows than in heifers. Metabolic stress in lactating cows was evidenced by lower glucose and higher s-hydroxy butyric acid compared with heifers. Insulin-like growth factor 1 signaling was reduced in the dominant follicle in lactating cows through reduced insulin-like growth factor 1 concentrations in plasma and follicular fluid of the dominant follicle, and reduced expression of pregnancy-associated plasma protein A ( PAPPA ) in their granulosa cells. We also found increased levels of total bile acids in the follicular fluid of the dominant follicle of lactating cows compared with heifers. Granulosa cells of the dominant follicle had higher expression of SLC10A2 and GPBAR1 (bile acid transporter and receptor, respectively) in lactating cows. These novel data are indicative of increased bile acid signaling within the dominant follicles of lactating cows compared with heifers. Overall, we demonstrate in the present study the metabolic, endocrine, and molecular differences within the microenvironment of the dominant follicles in lactating cows and heifers. These differences in follicular microenvironment may contribute toward abnormal ovarian function in lactating dairy cows.

Three different gonadotrophin regimens for the stimulation of donors for laparoscopic folliculocentesis were tested in a total of 142 ewes. The recovered oocytes were subjected to in vitro maturation, fertilization, and culture (IVM/IVF/IVC) for 7 d using standard procedures for sheep. The estrous cycles of all ewes were synchronized using intravaginal sponges containing 60 mg of medroxyprogesterone acetate (MPA) inserted for 14 d. In Experiment 1, all ewes were superovulated with a total dose of 125 IU FSH and 125 IU LH. One-half of the ewes received the gonadotrophin treatment in 4 decreasing doses at 12-h intervals starting 48 h before follicle aspiration (Control), while the other half received the total dose in a single injection at -24 h before collection (Oneshot). There were no significant differences between treatments for recovery rate (81.6 +/- 5.3 vs 77.4 +/- 10.3), cleavage rate (60.6 +/- 20.8 vs 61.4 +/- 23.4), or normal development to the blastocyst stage (20.8 +/- 18.2 vs 13.1 +/- 10.3). However, a higher percentage of ewes produced at least 1 normal blastocyst in the Control group (56.4 vs 31.6%; P0.05). In Experiment 2, the control regimen was repeated in half of the ewes, while the remainder were treated with half of the FSH total dose plus 500 IU eCG in a single injection at -24 h before oocyte collection (Oneshot-eCG). The recovery rate (80.9 +/- 5.6 vs 73.3 +/- 15.3), cleavage rate (76.8 +/-19.9 vs 79.7 +/- 22.6), normal development to blastocysts (19.2 +/- 15.3 vs 23.3 +/- 10.7), and percentage of ewes producing at least 1 normal blastocyst (55.9 vs 51.6%) did not differ between treatments. The large variability observed between ewes in the production of normal blastocysts is comparable to that observed with standard MOET procedures, in which a proportion of donors fail to produce good embryos. With the in vitro procedures described here, we were able to produce normal embryos from more than half of the treated ewes, indicating that the technology is useful for the multiplication of genetically valuable animals affected by temporary or irreversible infertility.

The present study was carried out to evaluate if the addition of cysteamine to the culture medium during in vitro maturation of bovine oocytes increased the glutathione (GSH) levels in the mature oocytes, and if these changes may promote an improvement on in vitro development to the blastocyst stage. Follicular oocytes from slaughterhouse ovaries were matured in TCM 199 supplemented with 10% (v/v) fetal calf serum, hormones, and O (control), 25, 50, or 100 muM of cysteamine for 24 hr. After in vitro maturation the oocytes were fertilized and cultured for 8 days. The percentage of embryos that developed to the blastocyst stage was significantly higher (P0.01) for oocytes matured in medium containing 100 muM of cysteamine than for those matured in control medium. Moreover, the intracellular GSH levels were increased (P0.05) in oocytes matured with 100 muM of cysteamine with respect to control. No differences were observed in maturation and cleavage rates, and in the mean cell numbers per blastocyst among treatments (P0.05). These results indicate that the addition of thiol compounds such as cysteamine to maturation medium increases the efficiency of in vitro blastocyst production from immature bovine oocytes. The higher levels of GSH in oocytes matured in the presence of cysteamine suggest that the beneficial effects of cysteamine on in vitro maturation and subsequent development after in vitro fertilization are mediated by GSH.

A simple and inexpensive pipette for in vivo recovery of sheep oocytes by folliculocentesis was developed. Two experiments were conducted to assess the recovery rate. In Experiment 1, 20 Merino × Corriedale ewes were heat synchronized using intravaginal sponges containing 60 mg of medroxyprogesterone acetate (MPA) for a period of 12 days. In Experiment 2, 26 Merino × Corriedale ewes were synchronized with CIDR® devices for 12 days, with the original device being replaced by a new one on Day 10 (2 days before withdrawal). In both experiments, ewes were superovulated with a total dose of 16 mg of follicle stimulating hormone (FSH) given in six decreasing dosage injections, starting 48 h before sponge/CIDR removal. Folliculocentesis was performed 36–40 h after sponge removal, and 16–20 h after CIDR removal. Overall, an average of 12.5 follicles per ewe were punctured and 10.2 oocytes per ewe were recovered (recovery rate 81.9%). The differences between the two experiments in terms of follicles punctured per ewe (10.8 vs. 13.7) and oocytes recovered per ewe (8.5 vs. 11.5) were not statistically significant ( P >0.05). The follicle size did not significantly ( P >0.05) affect the recovery rate, although there was a tendency for higher rates of recovery from 1–5 mm follicles (88.5%) compared with follicles over 5 mm (79.5%).

Compromised lactation physiology has been observed in transgenic animals, possibly due to the excessive demand placed by the expression of complex recombinant glycoproteins in the mammary gland. In previous studies we described lactation parameters and milk composition characteristics of transgenic goats expressing recombinant human butyrylcholinesterase in milk, and we showed evidence suggesting that lactation cessation could be associated with endoplasmic reticulum stress. We now report data from immunohistochemistry studies targeting activation transcription factor 6 and caspase 12, two signal transducers associated with endoplasmic reticulum stress, designed to further elucidate potential mechanisms responsible for the disruption in mammary epithelium function previously described. We found strong evidence of endoplasmic reticulum stress associated with the premature cessation of lactation. In addition, we utilized previously generated knowledge to design and test two treatments for enhanced productivity in transgenic goats. Pre-partum treatment with reserpine and dexamethasone to stimulate mammary priming for lactation resulted in a significant increase in milk production on day 1 (573 ± 350 vs. 93 ± 92 mL; P

The expression of recombinant proteins of pharmaceutical interest in the milk of transgenic farm animals can result in phenotypes exhibiting compromised lactation performance, as a result of the extraordinary demand placed on the mammary gland. In this study, we investigated differences in the protein composition of milk from control and transgenic goats expressing recombinant human butyrylcholinesterase. In Experiment 1, the milk was characterized by gel electrophoresis and liquid chromatography/mass spectrometry in order to identify protein bands that were uniquely visible in the transgenic milk and/or at differing band densities compared with controls. Differences in protein content were additionally evaluated by computer assisted band densitometry. Proteins identified in the transgenic milk only included serum proteins (i.e. complement component 3b, ceruloplasmin), a cytoskeleton protein (i.e. actin) and a stress-induced protein (94 kDA glucose-regulated protein). Proteins exhibiting evident differences in band density between the transgenic and control groups included immunoglobulins, serum albumin, beta-lactoglobulin and alpha-lactalbumin. These results were found to be indicative of compromised epithelial tight junctions, premature mammary cell death, and protein synthesis stress resulting from transgene expression. In Experiment 2, the concentration of alpha-lactalbumin was determined using the IDRing assay and was found to be significantly reduced on day 1 of lactation in transgenic goats (4.33 +/- 0.97 vs. 2.24 +/- 0.25 mg/ml, P0.01), but was not different from non-transgenic controls by day 30 (0.99 +/- 0.46 vs. 0.90 +/- 0.11 mg/ml, P0.05). We concluded that a decreased/delayed expression of the alpha-lactalbumin gene may be the cause for the delayed start of milk production observed in this herd of transgenic goats.

Oocyte competence and reproductive biology in prepubertal heifer calves are not fully understood. Multiple publications have reported high oocyte yields recovered from calves aged 2–6 months old but low embryo development rates following in vitro embryo production. The objective of this study was to characterise the developmental competence of oocytes from young calves. We report herein the oocyte/embryo yields obtained from 6 Holstein heifer calves that were subjected to gonadotropin stimulation and laparoscopic ovum pick-up (LOPU) every 2 weeks, starting at 2 months of age and ending at 5 months of age. The LOPU was conducted under general anaesthesia with the animal lying in dorsal recumbency on a table with a 45-degree angle to facilitate the visualisation of reproductive organs. Briefly, looking through the laparoscope, the ovarian surface was exposed by pulling from the fimbria with an atraumatic grasping forceps. The follicle contents were aspirated using a 20 G needle mounted on a pipette, which was connected to a collection tube and a vacuum pump. Media and procedures for aspiration, in vitro maturation (IVM), IVF, and in vitro culture (IVC) were standard in use for commercial adult bovine embryo production. Because of the small number of animals and the multifactorial variables in play (age, number of previous treatments and aspirations, etc.), in this preliminary study we focused on the overall oocyte/embryo yield and the potential effects of LOPU on ovarian integrity. A total of 766 follicles were aspirated (avg. 17/calf per session) resulting in 625 cumulus-oocyte complexes (COC) recovered (avg. 14/calf per session; 82% recovery rate). A total of 457 (73%) COC were graded eligible for IVM, of which 353 cleaved (77%) and 109 (24%) reached a viable blastocyst stage at the end of IVC, of which 42 (38.5%) were graded as freezable. In balance, ~2 viable blastocysts/calf per session were produced. No adhesions or sequels were observed in the animals up to the last LOPU session, as well as 2 weeks after the last LOPU when the animals were evaluated by rectal palpation by an experienced OPU veterinary practitioner. Further studies will look into other aspects of oocyte developmental competence to better understand this biological process.

Dangerous organophosphorus (OP) compounds have been used as insecticides in agriculture and in chemical warfare. Because exposure to OP could create a danger for humans in the future, butyrylcholinesterase (BChE) has been developed for prophylaxis to these chemicals. Because it is impractical to obtain sufficient quantities of plasma BChE to treat humans exposed to OP agents, the production of recombinant BChE (rBChE) in milk of transgenic animals was investigated. Transgenic mice and goats were generated with human BChE cDNA under control of the goat β-casein promoter. Milk from transgenic animals contained 0.1–5 g/liter of active rBChE. The plasma half-life of PEGylated, goat-derived, purified rBChE in guinea pigs was 7-fold longer than non-PEGylated dimers. The rBChE from transgenic mice was inhibited by nerve agents at a 1:1 molar ratio. Transgenic goats produced active rBChE in milk sufficient for prophylaxis of humans at risk for exposure to OP agents.

THE PRODUCTION of the first transgenic mouse by pronuclear injection in 1981 (Gordon and Ruddle, 1981) stimulated the interest of animal scientists working with domestic livestock, since the potential of transgenic domestic livestock as bioreactors for the production of valuable biopharmaceuticals was immediately recognized. Relatively soon, transgenic sheep, pigs, and goats were produced (Wall et al., 1992); however, efficiencies of transgenic animal production in livestock were lower than that obtained in mice. Following pronuclear microinjection of in vivo derived goat zygotes, only two of 29 goats born were transgenic (Ebert et al., 1991). This rate of production has not changed despite improvements in techniques (Baldassarre et al., 2003). Cloning using nuclear transfer (NT) provides a much more efficient system than DNA microinjection for the production of transgenic goats as the donor cells can be screened for transgene incorporation into the genome, copy number, and integration sites prior to its use in NT (Keefer et al., 2001). This selection of the transgenic donor cell ensures that nearly all the progeny produced will be transgenic (Baldassarre et al., 2002). Cloning can also be used to propagate transgenic animals produced by DNA microinjection (Baguisi et al., 1999; Keefer et al., 2002a). Successful production of transgenic goats not only requires care and skill in preparation of the transgenic donor cell (Lazaris et al., in press), but also in the management of the oocyte donors and the embryo recipients. Reproductive management is important in maximizing the number and quality of the oocytes recovered from donors, as well as in the ability of the recipients to allow development to term of the NT embryos. In addition, excellent health status and animal husbandry practices are required to maximize the reproductive results and ensure compliance with regulatory guidelines and animal welfare recommendations.

With an increased interest in transgenic animal production, the caprine species offers many advantages, and the prepubertal goat is a potential source of large numbers of oocytes for in vitro embryo production. The aim of the present study was to evaluate the follicular response and recovery of oocytes from prepubertal and adult goats following ovarian stimulation and laparoscopic recovery, and their developmental competence following culture in semi-defined media. Oocytes were collected over a 15-week period from prepubertal goats (3-7 months old) and adult controls (2-4 years old) that had been subjected to estrus synchronization and ovarian stimulation. Following insemination, zygotes were cultured for 96h in G1.2 followed by an additional 120h in G2.2. Morulae and blastocysts were scored using light microscopy on Days 7 and 9 followed by fluorescent staining for cell counts on Day 9 (216h postinsemination). The mean numbers of follicles aspirated and oocytes recovered were significantly greater for prepubertal than for adult goats (P0.01). The number of oocytes recovered from prepubertal goats was observed to decline significantly with increasing age of the animals (P0.05). The proportion of oocytes that matured and cleaved did not differ significantly between prepubertal and adult goats. Furthermore, no significant differences in morulae development (percentage of those cleaved), 5% versus 4%, or blastocyst development, 6% versus 7%, were observed for prepubertal and adult derived oocytes (P0.1), respectively. Mean cell number per blastocyst also did not differ significantly. In conclusion, higher yields of oocytes were obtained from gonadotrophin-primed, prepubertal does than from adults, while in vitro development was similar.
