- Abstract
LIFE-AMDRYC4 (2021) is a VR application designed for mobile phones combining informational design and gamification to stimulate critical reflection and awareness about agricultural practices and their effects on the environment. Agile production frameworks and researchers' input in the design are discussed. A Likert-based questionnaire was employed to measure attitudes from academics (environmental sciences) and professionals (farmers). Qualitative data showed that academics were more concerned about the representational aspects of the game while professionals were more concerned about its design and performance. Positive significative correlation among items, including the dimensions of 'enjoyment' and 'realism,' was also part of the findings.

9. Japanese Media Cultures in Japan and Abroad : Transnational Consumption of Manga, Anime, and Media-Mixes

- Abstract
In the last few decades, Japanese popular culture productions have been consolidated as one of the most influential and profitable global industries. As a creative industry, Japanese Media-Mixes generate multimillion-dollar revenues, being a product of international synergies and the natural appeal of the characters and stories. The transnationalization of investment capital, diversification of themes and (sub)genres, underlying threat in the proliferation of illegal audiences, development of internet streaming technologies, and other new transformations in media-mix-based production models make the study of these products even more relevant today. In this way, manga (Japanese comics), anime (Japanese animation), and video games are not necessarily products designed for the national market. More than ever, it is necessary to reconcile national and transnational positions for the study of this cultural production.The present volume includes contributions aligned to the analysis of Japanese popular culture flow from many perspectives (cultural studies, film, comic studies, sociology, etc.), although we have emphasized the relationships between manga, anime, and international audiences. The selected works include the following topics:• Studies on audiences—national and transnational case studies;• Fandom production and Otaku culture;• Cross-media and transmedia perspectives;• Theoretical perspectives on manga, anime, and media-mixes.

- Abstract
[EN] This paper presents a method that can be used to detect and identify Salmonella spp. in fresh meat and vegetable samples using a photonic biosensor with specialized bioreceptors. Detection was based on photon transduction. Silicon-nitride-based resonant cavities were used to capture the change in light response when there is specific binding of the immobilized antibody to the sensor surface against the target antigen. A control immobilization experiment was conducted to validate the immobilization process on the biosensor surface prior to biofunctionalization for Salmonella spp. detection. This experiment involved immobilization of pre-selected antibodies on silicon nitride surfaces. Two types of antibodies were suitable. The first was a specific polyclonal antibody with superior antigen-binding capacity across a wide range of concentrations. The second was a monoclonal antibody designed for effective binding at lower concentrations. Rigorous validation was performed. The outcomes were compared with those of the habitual method used to detect Salmonella spp. (reference method). Replicates from different batches of contaminated meat and vegetable samples were analyzed. This comprehensive approach provides a methodologically robust, highly sensitive, and accurate way of rapidly detecting Salmonella spp. in food samples. It has potential implications for improved food safety and quality control

Exegesis is a common practice when discussing religious texts. It has also been employed in the analysis of cultural production to elucidate the author’s intentions. Japanese animation (anime) is a transnational industry with cases such as Neon Genesis Evangelion (1995-1997), in which the figure of an individual author, such us the filmmaker Hideaki Anno, interacts with the collaborative authorship. The extensive use of obscure Judeo-Christian terminology and iconography in this work has risen debate about the actual intentions of Japanese author(s) when referring to Western culture. Our analysis concludes that the use of this iconography is intentional. The ambiguity of the narrative, shaped using multiple references, aims to induce the feeling of a complex text in the viewer. This would reinforce previous considerations of this anime as an “open work”, in the sense defined by post-structural semiotic analysis. [ABSTRACT FROM AUTHOR]

15. Development of Optical Label-Free Biosensor Method in Detection of Listeria monocytogenes from Food.

The present work describes an alternative method for detecting and identifying Listeria monocytogenes in food samples by developing a nanophotonic biosensor containing bioreceptors and optical transducers. The development of photonic sensors for the detection of pathogens in the food industry involves the implementation of procedures for selecting probes against the antigens of interest and the functionalization of the sensor surfaces on which the said bioreceptors are located. As a previous step to functionalizing the biosensor, an immobilization control of these antibodies on silicon nitride surfaces was carried out to check the effectiveness of in plane immobilization. On the one hand, it was observed that a Listeria monocytogenes-specific polyclonal antibody has a greater binding capacity to the antigen at a wide range of concentrations. A Listeria monocytogenes monoclonal antibody is more specific and has a greater binding capacity only at low concentrations. An assay for evaluating selected antibodies against particular antigens of Listeria monocytogenes bacteria was designed to determine the binding specificity of each probe using the indirect ELISA detection technique. In addition, a validation method was established against the reference method for many replicates belonging to different batches of meat-detectable samples, with a medium and pre-enrichment time that allowed optimal recovery of the target microorganism. Moreover, no cross-reactivity with other nontarget bacteria was observed. Thus, this system is a simple, highly sensitive, and accurate platform for L. monocytogenes detection. [ABSTRACT FROM AUTHOR]

Tras la aparición de la COVID-19 se han generado una serie de cambios en la forma de pensar y actuar de los seres humanos, en este estudio se hace referencia al ámbito turístico y a las nuevas características e intereses del turista, en cuanto a su elección de destino. Estas y las tendencias observadas está vinculada a los destinos que puedan ofrecer una sensación de seguridad, estar en contacto con la naturaleza, asegurar el distanciamiento social, un lugar en que poder relajarse sin estrés y con posibilidades de tener experiencias enriquecedoras y únicas. Estas nuevas características parece que abren oportunidades a territorios como Puntallana, al tratarse de un municipio rural y con los recursos necesarios. Esta nueva realidad post-COVID-19 se postula como una oportunidad para su desarrollo. Sin embargo, la zona de estudio plantea una serie de inconvenientes en cuanto a su actual proceso de desarrollo, que dificultan el acceso a la oportunidad comentada; para ello se ha establecido una propuesta pensada para un cambio en la orientación de dicho proceso en el municipio, para que pueda tener la opción a esta oportunidad.

- Abstract
[EN] To benefit the health of consumers, bioactive compounds must reach an adequate concentration at the end of the digestive process. This involves both an effective release from the food matrix where they are contained and a high resistance to exposure to gastrointestinal conditions. Accordingly, this study evaluates the impact of trehalose addition (10% w/w) and homogenization (100 MPa), together with the structural changes induced in vacuum impregnated apple slices (VI) by air-drying (AD) and freeze-drying (FD), on Lactobacillus salivarius spp. salivarius (CECT 4063) survival and the bioaccessibility of antioxidants during in vitro digestion. Vacuum impregnated apple slices conferred maximum protection to the lactobacillus strain during its passage through the gastrointestinal tract, whereas drying with air reduced the final content of the living cells to values below 10 cfu/g. The bioaccessibility of antioxidants also reached the highest values in the VI samples, in which the release of both the total phenols and total flavonoids to the liquid phase increased with in vitro digestion. The addition of trehalose and homogenization at 100 MPa increased the total bioaccessibility of antioxidants in FD and AD apples and the total bioaccessibility of flavonoids in the VI samples. Homogenizing at 100 MPa also increased the survival of L. salivarius during in vitro digestion in FD samples.

- Abstract
[EN] Vibrio parahaemolyticus is a human food-borne pathogen with the ability to enter the food chain. It is able to acquire a viable, non-cultivable state (VBNC), which is not detected by traditional methods. The combination of the direct viable count method and a fluorescent in situ hybridization technique (DVC-FISH) makes it possible to detect microorganisms that can present VBNC forms in complex samples The optimization of the in vitro DVC-FISH technique for V. parahaemolyticus was carried out. The selected antibiotic was ciprofloxacin at a concentration of 0.75 mu g/mL with an incubation time in DVC broth of 5 h. The DVC-FISH technique and the traditional plate culture were applied to detect and quantify the viable cells of the affected pathogen in artificially contaminated food matrices at different temperatures. The results obtained showed that low temperatures produced an important logarithmic decrease of V. parahaemolyticus, while at 22 degrees C, it proliferated rapidly. The DVC-FISH technique proved to be a useful tool for the detection and quantification of V. parahaemolyticus in the two seafood matrices of oysters and mussels. This is the first study in which this technique has been developed to detect viable cells for this microorganism.

[ES] El trabajo consiste en una revisión bibliográfica de la información disponible hasta el momento sobre la posible transmisión del SRAS-CoV-2 a través de los alimentos. Se estudiará que dicen los organismos de salud y las autoridades europeas de seguridad alimentaria sobre la transmisión del virus. Por otra parte, expondremos cuáles deben ser las medidas a tomar para prevenir la contaminación de los alimentos a nivel de empresa alimentaria y a nivel de consumidor. También se recogerán las orientaciones para las autoridades responsables del control de la inocuidad de los alimentos. Por otro lado, se mostrará información de la metodología para la detección de virus en agua y alimentos mediante técnicas moleculares modernas., [EN] The work consists of a bibliographic review of the information available so far on the possible transmission of SARS-CoV-2 through food. What health organizations and European food safety authorities say about the transmission of the virus will be studied. On the other hand, we will explain which measures should be taken to take to prevent food contamination at the food enterprise level and at the consumer level. The guidelines for the authorities responsible for food safety control will also be collected. On the other hand, information on the methodology for the detection of viruses in water and food using modern molecular techniques will be shown.

- Abstract
[EN] This study evaluates the effect of trehalose addition (10% or 20%, w/w) and/or sublethal homogenisation (25-150 MPa) on antioxidants content (vitamin C, total phenols and flavonoids) and activity (measured both by ABTS-TEAC and DPPH assays), as well as on microbial counts and survival to in vitro digestion of clementine juice inoculated with Lactobacillus salivarius spp. salivarius. Particle size, vacuum impregnation parameters and anti-Helicobacter pylori effect were also measured. Incubation with the probiotic improved the antioxidant properties of the juice. Homogenisation pressures below 100 MPa following incubation increased both the probiotic counts in the juice and its antioxidants bioaccesibility. Adding 10% (w/w) of trehalose to the juice was effective in preventing these bioactive compounds deterioration under adverse conditions. Once homogenised, liquids containing 10% (w/w) of trehalose became as able as those without trehalose to enter a food solid matrix. Inhibition of Helicobacter pylori growth was evident in all probiotic beverages.

[EN] Anime is frequently the origin of media-mixes, that is, strategies of production and distribution of transmedia products in the Japanese global markets. Synergies can be formed through collaborative authorships and transmedia adaptations. While there is an important academic discussion about authorship in media, its role in the context of video games and the media mix is not that clear. In this sense, Ni No Kuni saga (2011-2018) is studied for being a unique case of intertextuality between the work of Studio Ghibli, and particularly the influential Hayao Miyazaki (1941) and Role Playing Video Games. The paper will show how individual and collective authorships consolidate common styles and narratives in the form of coherent corporate images and branding. These images survive the original authors to be replicated through the rest of the transmedia production., [ES] El anime es origen habitual del media-mix o estrategias de producción y distribución de productos transmedia en el mercado japonés de alcance global. A su alrededor se forman constantes sinergias que dan lugar a autorías colaborativas y múltiples adaptaciones transmedia. Mientras que existe una importante tradición académica acerca del papel del “autor”, no esta tan claro su papel en el contexto de los videojuegos y el media mix. Aún no siendo una adaptación sino un producto original, la saga de Ni No Kuni (2011-2018) es estudiada por ser un caso único de intertextualidad entre la obra de Studio Ghibli —particularmente el influyente Hayao Miyazaki (1941)— y los videojuegos de rol. Se demostrará como la autoría individual y colectiva consolidan estilos y narrativas comunes en forma de imágenes corporativas coherentes. Estas imágenes sobreviven a los autores originales para ser replicadas en el resto de la producción transmedia.

- Abstract
Background: oral cancer contributes with a considerable number in the morbidity figures which will increase in the coming decades. Reporting on this issue could be the starting point in the battle against this scourge of modern life. Objective: to design an educational program on oral cancer for adolescents according to identified needs. Methods: a development research was carried out with a mixed approach, educational intervention, pre-experimental, prospective and longitudinal study in the municipality of Santa Clara in the period from February/ 2016 to November / 2017. Theoretical and empirical methods were used that allowed the bibliographic foundation and the collection of information. Results: in the applied diagnosis, risk factors predominated, such as poor oral hygiene, non-protective diet consumption and insufficiency of knowledge about oral cancer among adolescents; and practice of toxic habits by parents or other family members, that´s why an educational program was designed, which was assessed by specialists and applied with very favorable results in the solution of the problems detected. Conclusions: the specialists considered it pertinent, feasible, useful, with adequate structure and with scientific and pedagogical values, and its effectiveness was proven from its application in the adolescent group for which it was designed., Fundamento: el cáncer bucal aporta un número considerable en las cifras de morbilidad las que aumentarán en los decenios venideros. Informar sobre el tema pudiera ser el punto de partida si se pretende combatir este flagelo de la vida moderna.Objetivo: diseñar un programa educativo sobre cáncer bucal para adolescentes según necesidades identificadas.Métodos: se realizó una investigación de desarrollo con enfoque mixto, de intervención educativa, prexperimental, prospectiva y de corte longitudinal en el municipio de Santa Clara en el período comprendido entre febrero/2016 a noviembre/2017. Se utilizaron métodos teóricos y empíricos que permitieron la fundamentación bibliográfica y la recogida de información.Resultados: en el diagnóstico aplicado predominaron factores de riesgos como mala higiene bucal, consumo de dieta no protectora e insuficiencias de conocimientos sobre el cáncer bucal entre los adolescentes; y práctica de hábitos tóxicos por padres u otros familiares, por lo que se diseñó un programa educativo el cual fue valorado por criterio de especialistas y aplicado con resultados muy favorables en la solución de los problemas detectados.Conclusiones: los especialistas lo consideraron pertinente, factible, útil, con adecuada estructura y valor científico y pedagógico, y se comprobó su efectividad a partir de su aplicación en el grupo de adolescentes para el cual fue diseñado.

- Abstract
[EN] Changes undergone by tomato-antioxidants during gastrointestinal digestion of raw and fried tomato, with or without presence of the probiotic Lactobacillus reuteri ATCC 55730, were studied. Frying process enhanced the extractability of antioxidant compounds, being their content higher in fried than in raw tomato. In vitro digestion led to a significant loss of antioxidant activity (65 and 75% losses for raw and fried tomato, respectively), and total lycopene (60 and 50% losses for raw and fried tomato, respectively); and promoted trans-cis lycopene isomerization initiated during frying. Bioaccessibility of the antioxidant compounds was within 10% and 30%, being higher for phenolic compounds in raw tomato but lower for total lycopene. Finally, although the presence of Lactobacillus reuteri ATCC 55730 reduced the bioaccessibility of antioxidant compounds, the results suggests that the tomato's antioxidant compounds could have a protective effect against the loss of viability of the probiotic.

- Abstract
[EN] Salmonella spp. is one of the most important causal agents of food-borne illness in developed countries and its presence in irrigation water poses a risk to public health. Its detection in environmental samples is not easy when culture methods are used, and molecular techniques such as PCR or ribosomal rRNA probe hybridization (Fluorescent zn situ Hybridization, FISH) are outstanding alternatives. The aim of this work was to determine the environmental risk due to the presence of Salmonella spp. in wastewater by culture, PCR and FISH. A new specific rDNA probe for Salmonella was designed and its efficiency was compared with the rest of methods Serotype and antibiotic resistance of isolated strains were determined. Forty-five wastewater samples (collected from two secondary wastewater treatment plants) were analysed. Salmonella strains were isolated in 24 wastewater samples (53%), two of them after disinfection treatment. Twenty-three Salmonella strains exhibited resistance to one or more antimicrobial agent. Analysis of wastewater samples yielded PCR positive results for Salmonella in 28 out of the 45 wastewater samples (62%). FISH analysis allowed for the detection of Salmonella in 27 (60%) samples. By using molecular methods, Salmonella was detected in four samples after disinfection treatment. These results show the prevalence of Salmonella in reclaimed wastewater even after U.V. disinfection, what is a matter of public health concern, the high rates of resistance to antibiotics and the adequacy of molecular methods for its rapid detection. FISH method, with SA23 probe developed and assayed in this work provides a tool for detecting Salmonella in water within few hours, with a high rate of effectiveness.

- Abstract
De Morsier syndrome dysplasia septo-optic (DSO), was described by De Morsier in 1956, who recognized the association between Agenesis or hypoplasia of the septum pellucidum and hypoplasia of the optic nerve and CHIASM and optic tracts. You may be associated with dysfunction hypothalamic-pituitary. The form of presentation is sporadic and familial cases are rare, although it has been postulated the possibility of autosomal recessive inheritance. The neurological picture associated with the syndrome is variable. It occurs in some patients with normal intelligence and no neurologic deficit. Disorders of learning and mental retardation are related to abnormalities of the septum pellucidum. Seizures are frequently described demonstrations. Objectives. Identify signs and symptoms of De Morsier syndrome in 6-year-old Guatemalan girl. Patient mestizo, of six years, a history of progressive muscular dystrophy his father, goes to consultation by nystagmus, strabismus, torticollis buffer and delay of neurodevelopmental, with a correct weight increase. Physical examination palpebral telangiectasias, vertical nystagmus, deviation from the head to the left side. Fundus: pronounced temporal pallor of both discs, atrophy macular foveal, venous tortuous generalized dull. Optic nerve hypoplasia. Computerized axial tomography shows absence of septum pellucidum. Discussion. Progressive muscular dystrophy and De Morsier syndrome have a low incidence in the population, so the presence of the same father and daughter should be designated, there is no explanation to justify the finding, not found, previous reports in the lit erature pointing it., El síndrome de De Morsier o displasia septo-óptica (DSO), fue descrito por De Morsier en 1956, quien reconoció la asociación entre la agenesia o hipoplasia del septum pellucidum e hipoplasia del nervio óptico, quiasma y tractos ópticos. Se puede asociar a disfunción hipotálamo-hipofisaria. La forma de presentación es esporádica y los casos familiares son raros, aunque se ha postulado la posibilidad de herencia autosómica recesiva. El cuadro neurológico asociado al síndrome es variable. Se presenta en algunos pacientes con inteligencia normal y sin déficit neurológico. Los trastornos del aprendizaje y el retraso mental están relacionados con las anomalías del septum pellucidum. Las convulsiones son las manifestaciones descritas con frecuencia. Como objetivo tenemos el de identificar síntomas y signos del síndrome de De Morsier en niña guatemalteca de seis años. La paciente es una niña mestiza, de seis años, antecedentes de distrofia muscular progresiva paterna, acude a consulta por nistagmo, estrabismo, tortícolis compensadora y retardo del neurodesarrollo, con un incremento ponderal correcto, en el examen físico encontramos telangiectasias palpebrales, nistagmo vertical-horizontal, desviación de la cabeza al lado izquierdo. Fondo de ojo: Palidez temporal acentuada de ambos discos, atrofia macular, sin brillo foveal, tortuosidad venosa generalizada. Hipoplasia del nervio óptico. En tomografía axial computadorizada se observa ausencia de septum pellucidum. La distrofia muscular progresiva y el Síndrome de De Morsier tienen una baja incidencia en la población, por lo que la presencia de las mismas en padre e hija debe ser señalada, no se tiene explicación para justificar el hallazgo, no se han encontrado reportes previos en la literatura que lo señalen.

[ES] En 1982, se aislaron distintas especies de bacilos acidófilos a partir de zumo de manzana, y se identificaron como una nueva clase de bacterias que podrían provocar daños en la producción, con las consiguientes pérdidas económicas. Denominadas en un principio como Bacillus acidoterrestris, se incluyeron más tarde dentro de un género nuevo denominado Alicyclobacillus spp. Dentro del género Alicyclobacillus spp., Alicyclobacillus acidoterrestris es la principal especie alterante de los zumos de frutas por producción de guayacol. Debido a la resistencia de Alicyclobacillus acidoterrestris a elevadas temperaturas (soportando tratamientos de pasteurización) y a bajos valores de pH, es considerada como una amenaza importante en la industria de zumos y bebidas de frutas, originando considerables pérdidas económicas, por la producción de sabores indeseables en el producto final. Este deterioro es difícil de determinar mediante inspección visual, ya que el zumo alterado presenta un aspecto normal. El microorganismo crece lentamente en los productos terminados y a concentraciones entre 105 -106 ufc/mL –en zumo de manzana-, produce suficiente guayacol para alterar el sabor. Hoy en día, se emplean distintas metodologías para el control de este microorganismo en la industria alimentaria, entre los que destacan los métodos clásicos microbiológicos de recuento en placa seguidos de análisis bioquímicos confirmativos que suelen durar hasta 10-12 días. Si tenemos en cuenta que en la actualidad la mayoría de las empresas tienen una producción en continuo, es interesante desarrollar y aplicar nuevos métodos que sean más rápidos y fiables, con el objetivo de obtener resultados de control microbiológico en un tiempo mucho más reducido. En el presente trabajo fin de master se propone el desarrollo de una técnica molecular basadas en la hibridación in situ (FISH) para la detección y cuantificación de Alicyclobacillus acidoterrestris, haciendo una comparativa con métodos microbiológico, [EN] In 1982, different species of acidophilus bacilli were isolated from apple juice, and identified as a new class of bacteria that could cause production damage, with consequent economic losses. Named initially as Bacillus acidoterrestris, they were later included within a new genus called Alicyclobacillus spp. Within the genus Alicyclobacillus spp., Alicyclobacillus acidoterrestris is the main altering species of fruit juices produced by guaiacol. Due to the resistance of Alicyclobacillus acidoterrestris to high temperatures (undergoing pasteurization treatments) and low pH values, it is considered a significant threat in the fruit juices and beverages industry, causing considerable economic losses due to the production of undesirable flavors in the final product. This deterioration is difficult to determine by visual inspection, since the altered juice has a normal appearance. The microorganism grows slowly in the finished products and at concentrations between 105 -106 cfu / mL - in apple juice -, produces enough guayacol to alter the taste. Nowadays, different methodologies for the control of this microorganism in the food industry are used, among them the classic microbiological methods of plate counting followed by confirmatory biochemical analyzes that usually last up to 10-12 days. If we take into account that at present most of the companies have a continuous production, it is interesting to develop and apply new methods that are faster and more reliable, in order to obtain microbiological control results in a much shorter time. In the present work, the development of a molecular technique based on in situ hybridization (FISH) for the detection and quantification of Alicyclobacillus acidoterrestris is proposed, making a comparison with classical microbiological methods, also evaluating its usefulness as a diagnostic test in the Microbiological control of different raw materials, such as concentrated juices.

Background & aims: The pathogenesis of enteritis after abdominal radiotherapy is unknown, although changes in faecal microbiota may be involved. In several studies, Lactobacillus and Bifidobacterium have proven beneficial for the host. Prebiotics stimulate the proliferation of Lactobacillus and Bifidobacterium, and this may have positive effects on the intestinal mucosa during abdominal radiotherapy. Methods: We performed a randomised double-blind, placebo-controlled trial including 31 patients with gynaecological cancer who received radiotherapy (29 sessions, 52.2 Gy) after surgery. Patients were randomised to two groups: prebiotic and placebo. The first group received a mixture of fibre (50% inulin and 50% fructo-oligosaccharide) and the second received 6 g of maltodextrin twice daily from one week before to three weeks after radiotherapy. Lactobacillus and Bifidobacterium counts were determined in faeces samples (day 7 before radiotherapy, day 15 of radiotherapy, at the end of treatment, and three weeks after radiotherapy) by culture in selective media and fluorescent in situ hybridization (FISH) using genus-specific probes. Bacterial counts by FISH were significantly higher than by culture method. Results: There were no differences in baseline microbiota between groups. At the end of radiotherapy, we observed a statistically significant decrease in Lactobacillus and Bifidobacterium counts in both groups. By cultural analysis, we observed higher numbers of Lactobacillus and Bifidobacterium three weeks after radiotherapy in the prebiotic group (5.6 vs. 6.3, p = 0.04 and 5.5 vs. 6 log cfu/g, p = 0.03). Conclusions: Abdominal radiotherapy negatively affects Lactobacillus and Bifidobacterium counts. The prebiotic mixture of inulin and fructoligosaccharide can improve the recovery of both genera after radiotherapy. This work was supported in part by grants from Vegenat (Spain).

[EN] Aims: We have developed a direct viable count (DVC)-FISH procedure for quickly and easily discriminating between viable and nonviable cells of Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus strains, the traditional yogurt bacteria. Methods and Results: direct viable count method has been modified and adapted for Lact. delbrueckii subsp, bulgaricus and Strep. thermophilus analysis by testing different times of incubation and concentrations of DNA-gyrase inhibitors. DVC procedure has been combined with fluorescent in situ hybridization (FISH) for the specific detection of viable cells of both bacteria with specific rRNA oligonucleotide probes (DVC-FISH). Of the four antibiotics tested (novobiocin, nalidixic acid, pipemidic acid and ciprofloxacin), novobiocin was the most effective for DVC method and the optimum incubation time was 7 h for both bacteria. The number of viable cells was obtained by the enumeration of specific hybridized cells that were elongated at least twice their original length for Lactobacillus and twice their original size for Streptococcus. Conclusions: This technique was successfully applied to detect viable cells in inoculated faeces. Significance and Impact of the Study: Results showed that this DVC-FISH procedure is a quick and culture-independent useful method to specifically detect viable Lact, delbrueckii subsp, bulgaricus and Strep thermophilus in different samples, being applied for the first time to lactic acid bacteria.

47. Persistence of Listeria monocytogenes strains in a frozen vegetables processing plant determined by serotyping and REP-PCR

Seventy presumptive Listeria monocytogenes strains isolated from frozen vegetable products in a processing plant during a period of six months were characterized by serology, biochemical tests (API system), multiplex PCR, virulence characteristics and REP-PCR analysis. Most isolates belonged to 1/2a (62 strains) and 1/2b (3 strains) serotypes, the most common isolated from foods. Amplification of virulence gene lmo2821 was positive in 51 of the PCR-confirmed L. monocytogenes strains. Automated REP-PCR of L. monocytogenes isolates yielded 17 different patterns, formed by 70 to 80 different bands ranging from 150 to 7000 bp. A total of three genetic groups were defined at 82% homology degree. At this level, serovars 4b and 1/2b strains were discriminated from the rest of strains. Two REP-PCR patterns were frequently found for isolates sharing the same serotype and biochemical profile. These isolates were obtained from different vegetables during the sampling period, what clearly suggests the persistence of some L. monocytogenes strains in the processing plant. While the multiplex PCR assay applied in this work provided an accurate and rapid method for species identification, REP-PCR was confirmed as a rapid and reliable method for L. monocytogenes subtyping, providing useful information for epidemiological or risk assessment studies, as well as for tracking surveys in food processing plants., This work was supported by grant AGL2008-05275-C03-02 from Ministerio de Ciencia e Innovacion, Spain.

[ES] Dada la elevada prevalencia de la infección ocasionada por Helicobacter pylori y los efectos secundarios asociados a su tratamiento con antibióticos, resulta interesante desarrollar nuevas alternativas naturales que disminuyan la colonización por parte de este patógeno. Existen estudios clínicos y modelos in vitro que atribuyen a las bacterias del género Lactobacillus (en especial, L. salivarius spp. salivarius) un efecto antagonista frente a H. pylori. Sin embargo, se tiene poca información acerca de cómo la actividad antibacteriana o la supervivencia de este probiótico al proceso de digestión se podría ver afectada por la presencia de otros compuestos, tales como la vitamina C, o por la aplicación de operaciones de procesado de alimentos, tales como la homogeneización. De acuerdo con esto, el presente trabajo se propone con el objeto de evaluar el efecto que la concentración de trehalosa (desde 0 hasta 30% peso) y la aplicación de altas presiones de homogeneización (desde 0 hasta 150 MPa) ejercen sobre las propiedades físico-químicas (aw, pH, º Brix, turbidez, pulpa suspendida y tamaño de partícula) y microbiológicas (recuento de viables, resistencia a jugos gástricos y actividad antimicrobiana) de zumo de mandarina inoculado con Lactobacillus salivarius spp. salivarius. Por lo que respecta a las propiedades físico-químicas, destacar el descenso en la actividad del agua debido a la presencia de trehalosa y el descenso en el tamaño de las partículas presentes en el zumo debido a la aplicación de altas presiones de homogeneización. En relación a las propiedades microbiológicas, destacar que el zumo de mandarina por sí solo mejoró notablemente la resistencia del probiótico a la acción de las enzimas pepsina y pancreatina, así como su actividad frente a Helicobacter pylori. Además, estas propiedades microbiológicas se vieron sustancialmente incrementadas al añadir un 10% en peso de trehalosa al zumo y posteriormente aplicar una presión de homogeneización de 50 MP, [EN] Given the high prevalence of infection caused by Helicobacter pylori and the side effects associated with treatment with antibiotics, it is interesting to develop new natural alternatives that reduce the colonization of this pathogen. There are clinical studies and in vitro models that attribute to the bacteria of the genus Lactobacillus (especially L. salivarius spp. salivarius) an antagonist effect against H. pylori. However, little is known about how the antibacterial activity or the survival of the probiotic digestion process can be affected by the presence of other compounds such as vitamin C, or by the application of food processing operations such as homogenization. Accordingly, the present study aims to assess the effect of the concentration of trehalose (from 0-30% weight) and the application of high pressure homogenization (from 0 to 150 MPa) exerted on the physical properties -chemical (aw, pH, Brix, turbidity and suspended pulp particle size) and microbiological (viable count, gastric juice resistance and antimicrobial activity) tangerine juice inoculated with Lactobacillus salivarius spp. salivarius. With regard to physicochemical properties, the noteworthy aspects were the drop in water activity due to the presence of trehalose and the decrease in the size of the particles present in the juice as a result of applying high pressure homogenization. Regarding the microbiological properties, note that the tangerine juice alone greatly improved the resistance of the probiotic action of pepsin and pancreatin, and their activity against Helicobacter pylori. Existeixen estudis clínics i models in vitro que atribueixen a les bacteries del gènere Lactobacillus (especialment, L. salivarius spp. salivarius) un efecte antagonista enfront de H. pylori. No obstant això, es té poca informació al voltant de com l’activitat antibacteriana o la supervivència d’aquest probiòtic al procés de digestió es podria veure afectada per la presència d’altres compostos, com ara la vitamina C, o per l’aplicació d’operacions de processat d’aliments , com ara l’homogeneïtzació. D’acord amb això, el present treball es proposa amb l’objectiu d’avaluar l’efecte que la concentració de trehalosa (des de 0 fins al 30% en pes) i l’aplicació d’altes pressions d’homogeneïtzació (des de 0 fins a 150 MPa) exerceixen damunt les propietats físico-químiques (aw, pH, Brix, turbidesa, polpa suspesa i mida de partícula) i microbiològiques (recompte de viables, resistència a sucs gàstrics i activitat antimicrobiana) de suc de mandarina inoculat amb Lactobacillus salivarius spp. salivarius. Pel que fa a les propietats físico-químiques, destacar el descens en l’activitat de l’aigua a causa de la presència de trehalosa i el descens en la mida de les partícules presents en el suc a causa de l’aplicació d’altes pressions d’homogeneïtzació. En relació a les propietats microbiològiques, destacar que el suc de mandarina per si mateix va millorar notablement la resistència del probiòtic a l’acció dels enzims pepsina i pancreatina, així com la seua activitat enfront d’Helicobacter pylori. A més, aquestes propietats microbiològiques es van veure substancialment incrementades en afegir un 10% en pes de trehalosa al suc i posteriorment aplicar una pressió d’homogeneïtzació de 50 MPa.

[ES] Uno de los principales problemas a los que se enfrenta una bodega, es referido a la calidad de los vinos. Uno de los parámetros de más difícil control, es el microbiológico. El más temido, es la contaminación por levaduras del género Brettanomyces, pues pueden producir muy rápidamente un deterioro acusado de la calidad del vino. Debido a los métodos de procesado y embotellado, una contaminación mínima puede rápidamente extenderse por todos los depósitos del complejo, llegando a afectar de forma crónica a los caldos de la bodega, con el subsiguiente perjuicio económico. En una bodega de tamaño medio, el continuo control que se realiza al vino, hacen posible una vigilancia del mismo en todas las etapas, tratando de identificar aquellos parámetros que pueden ser coincidentes en el aumento/disminución de las colonias de Brettanomyces, y con ello realizar un nuevo protocolo de elaboración del vino., [EN] One of the main problems facing a winery, is based on the quality of the wines. One of the more difficult parameters to control is the microbiological. The most feared, is contamination by Brettanomyces yeasts, they can produce a sharp deterioration in the quality of wine very quickly. Because the methods of processing and bottling, minimal contamination can quickly spread all deposits of the complex, affecting chronically the wines of the winery, with subsequent economic loss. Un dels paràmetres de més difícil control, és el microbiològic. El més temut, és la contaminació per llevats del gènere Brettanomyces, doncs poden produir molt ràpidament un deteriorament acusat de la qualitat del vi. A causa dels mètodes de processament i embotellat, una contaminació mínima pot ràpidament estendre's per tots els dipòsits del complex, arribant a afectar de manera crònica, als vins del celler, amb el subsegüent perjudici econòmic. En un celler de mida mitjana, el continu control que es realitza al vi, fan possible un control del mateix en totes les etapes, tractant d'identificar aquells paràmetres que poden ser coincidents en l'augment / disminució de les colònies de Brettanomyces, i amb això realitzar un nou protocol d'elaboració del vi.

[ES] En la industria agroalimentaria el análisis microbiológico es una actividad necesaria para poder comercializar productos sin riesgo para la salud del consumidor. Algunos microorganismos son capaces de provocar enfermedades muy graves que pueden causar la muerte. La septicemia o sepsis es una de estas enfermedades caracterizada por una respuesta inflamatoria sistémica del organismo ante una infección. En España afecta a unas 50.000 personas cada año, de las que mueren 17.000. En el mundo, ocho millones de pacientes mueren cada año. Las bacterias gramnegativas son, entre otros agentes, los principales responsables de la sepsis a causa de unas estructuras moleculares complejas situadas en su membrana externa conocidas como lipopolisacáridos o endotoxinas. La septicemia se combate con antibióticos como la polimixina B, sin embargo es vital disponer de herramientas analíticas sensibles para reducir la tasa de mortalidad, ya que contar con un diagnóstico rápido es imprescindible dentro del protocolo de actuación para el tratamiento de la sepsis. En este trabajo se desarrolla un sistema analítico basado en microinmunoensayos de bajo coste, rápido, portátil y sensible para la determinación de endotoxinas. Los microinmunoensayos se basan en el reconocimiento molecular específico entre la polimixina B y endotoxinas tipo lipopolisacáridos (LPS). El ensayo se ha desarrollado en formato de micromatriz sobre un disco compacto, utilizando LPS de Escherichia coli O26 como sistema modelo y un lector/grabador de discos como detector. La aproximación analítica permite 20 microinmunoensayos simultáneos para la detección de lipopolisacáridos en un tiempo inferior a 70 minutos alcanzando un límite de detección de 1,4 µg/L., [EN] In the food industry the microbiological analysis is needed to market products without risk to consumer health activity. Some microorganisms are capable of causing serious disease that can cause death. Septicaemia is one of these diseases characterized by a systemic inflammatory response of the body to an infection. In Spain it affects about 50.000 people each year, of which 17.000 die. Worldwide, eight million patients passed away each year. Gram-negative bacteria are include other agents primarily responsible for sepsis because of complex molecular structures in their outer membrane known as lipopolysaccharides or endotoxins. Sepsis is treated with antibiotics such as polymyxin B, however it is important to have sensitive analytical tools to reduce the mortality rate, as have a rapid diagnosis is essential in the protocol for the treatment of sepsis. In this work, microimmunoassays an analytical system based low cost, fast, portable and sensitive for the determination of endotoxins is developed. The microimmunoassay is based on specific molecular recognition between polymyxin B and endotoxin lipopolysaccharides type (LPS). The test was developed in microarray format on a compact disc, using Escherichia coli O26 LPS as a model system and a reader/writer drives as detector. The microarray-based analysis system enables to perform 20 microimmunoassays simultaneously for detecting lipopolysaccharide in less than 70 minutes time reaching a detection limit of 1,4 µg/L.
