This article presents a narrative inquiry of a Chinese heritage mother to theorize and explicate how historical, relational, and spatial processes impacted her negotiation with power and agency in relation to her own heritage language (HL) identity development. A narrative approach enables us to draw on participant counter-stories against master narratives that erase experiences of marginalization of Asians in Asian language education in the United States. We do this through a model of HL identity development (Zhou & Liu, 2022) supplemented by an AsianCrit lens (Iftikar & Museus, 2018). We show the importance of normalizing Chinese as a HL outside of the home in terms of language maintenance as well as the impact such normalization has on the development of an affirmative Chinese HL identity. We add that spaces for such identity development are deeply associated with language programs like dual language bilingual education (DLBE), especially as the number of DLBE opportunities grow in number and in popularity. Thus, language programs, including DLBE, have a responsibility to ensure that the language education they provide address the interests and investments of families with respect to their HL in order to decenter a primary focus on the interests of ethnolinguistic majoritized families.

Glioblastoma (GBM) is the most aggressive cancer originating in the brain, but unfortunately combination treatments with resection, radiation, and chemotherapy are relatively ineffective. Therefore, novel methods of adjuvant therapy are critically needed. Cyclotides are plant-derived circular peptides that chemosensitize drug-resistant breast cancer to doxorubicin. We analyzed naturally occurring and synthetic cyclotides (Cycloviolacin O3, Cycloviolacin O19, natural Kalata B1, synthetic Kalata B1, and Vitri E) alone and in co-exposure treatments with the drug temozolomide (TMZ) in human glioblastoma cells. The cyclotides were identified by UPLC-PDA and HPLC-UV. The synthetic Kalata B1 sequence was verified with orbitrap LC-MS, and structural confirmation was provided by NMR spectroscopy. The cyclotides displayed dose-dependent cytotoxicity (IC50 values 2.4-21.1 µM) both alone and as chemosensitizers of U-87 MG and T 98 cells to TMZ. In fact, a 16-fold lower concentration of TMZ (100 µM) was needed for significant cytotoxicity in U-87 MG cells co-exposed to synthetic Kalata B (0.5 µM). Similarly, a 15-fold lower concentration of TMZ (75 µM) was required for a significant reduction in cell viability in T 98 cells co-exposed to synthetic Kalata B1 (0.25 µM). Kalata B1 remained stable in human serum stability assays. The data support the assertion that cyclotides may chemosensitize glioblastoma cells to TMZ.

RNA-binding proteins (RBPs) modulate alternative splicing outcomes to determine isoform expression and cellular survival. To identify RBPs that directly drive alternative exon inclusion, we developed tethered function luciferase-based splicing reporters that provide rapid, scalable and robust readouts of exon inclusion changes and used these to evaluate 718 human RBPs. We performed enhanced cross-linking immunoprecipitation, RNA sequencing and affinity purification-mass spectrometry to investigate a subset of candidates with no prior association with splicing. Integrative analysis of these assays indicates surprising roles for TRNAU1AP, SCAF8 and RTCA in the modulation of hundreds of endogenous splicing events. We also leveraged our tethering assays and top candidates to identify potent and compact exon inclusion activation domains for splicing modulation applications. Using these identified domains, we engineered programmable fusion proteins that outperform current artificial splicing factors at manipulating inclusion of reporter and endogenous exons. This tethering approach characterizes the ability of RBPs to induce exon inclusion and yields new molecular parts for programmable splicing control.

Using a case study design, this research explores how two department chairs, Jan and Tammy, in community colleges implement AB 705, an assessment policy impacting California ESL programs. AB 705 seeks to centralize previous multiple measures legislation in California by requiring the use of high school data in community college course placement. An ecological perspective was adopted to highlight how ideologies at the macro national and meso state level about placement tests and student autonomy trickle down and are interpreted at the micro institutional level, and how challenges and concerns about implementation reflect the micro level affordances or lack thereof for each participant. Through critical language policy (Tollefson as cited in Ricento, 2006), I also posit that as AB 705 changes the assessment landscape in California community colleges, they create power shifts between local actors. Data were collected from a questionnaire, interviews, policy documents and online artifacts. Findings revealed that Jan and Tammy's different ideologies regarding the validity of placement tests lead to different interpretations and implementation of the policy, thus creating different power dynamics for their students. Both also encountered different challenges because of their varied local affordances, highlighting the need for top-down policies to consider implementation in situated contexts.

Quantitative analysis of complex mixtures, including compounds having similar chemical properties, is demonstrated using an automatic and high throughput approach to microcrystal electron diffraction (MicroED). Compositional analysis of organic and inorganic compounds can be accurately executed without the need of diffraction standards. Additionally, with sufficient statistics, small amounts of compounds in mixtures can be reliably detected. These compounds can be distinguished by their crystal structure properties prior to structure solution. In addition, if the crystals are of good quality, the crystal structures can be generated on the fly, providing a complete analysis of the sample. MicroED is an effective method for analyzing the structural properties of sub-micron crystals, which are frequently found in small-molecule powders. By developing and using an automatic and high throughput approach to MicroED, and with the use of SerialEM for data collection, data from thousands of crystals allow sufficient statistics to detect even small amounts of compounds reliably.

Parental involvement is well-documented in the literature. Although research suggests a strong positive association between parental involvement and children's educational outcomes, few studies have examined parental involvement at home with children who attend charter schools, especially with small immigrant groups such as the Hmong. Therefore, the purpose of this study was to examine how Hmong parents of students in charter schools were involved in their children's education at home and what barriers they faced. This study included 23 Hmong parents (9 fathers, 15 mothers) of elementary school-aged children enrolled in three Hmong-focused charter schools in Minnesota. The results showed several themes related to barriers for at-home school involvement, including work schedules, literacy barriers, lack of spousal support, and multiple children at home. Despite these barriers, Hmong parents also indicated that they sacrificed for their children's education by taking on multiple jobs, created and maintained regular family routines to monitor children's education, and connected children to appropriate resources for homework help, including older siblings. Implications for school administrators and teachers who work with Hmong parents, especially Hmong parents who have children enrolled in charter schools, are also discussed.

PURPOSE: Injectable biologics have not only been described and developed to treat dermal wounds, cardiovascular disease, and cancer, but have also been reported to treat chronic pain conditions. Despite emerging evidence supporting regenerative medicine therapy for pain, many aspects remain controversial. METHODS: The American Society of Pain and Neuroscience (ASPN) identified the educational need for an evidence-based guideline on regenerative medicine therapy for chronic pain. The executive board nominated experts spanning multiple specialties including anesthesiology, physical medicine and rehabilitation, and sports medicine based on expertise, publications, research, and clinical practice. A steering committee selected preliminary questions, which were reviewed and refined. Evidence was appraised using the United States Preventive Services Task Force (USPSTF) criteria for evidence level and degree of recommendation. Using a modified Delphi approach, consensus points were distributed to all collaborators and each collaborator voted on each point. If collaborators provided a decision of disagree or abstain, they were invited to provide a rationale in a non-blinded fashion to the committee chair, who incorporated the respective comments and distributed revised versions to the committee until consensus was achieved. RESULTS: Sixteen questions were selected for guideline development. Questions that were addressed included type of injectable biologics and mechanism, evidence in treating chronic pain indications (eg, tendinopathy, muscular pathology, osteoarthritis, intervertebral disc disease, neuropathic pain), role in surgical augmentation, dosing, comparative efficacy between injectable biologics, peri-procedural practices to optimize therapeutic response and quality of injectate, federal regulations, and complications with mitigating strategies. CONCLUSION: In well-selected individuals with certain chronic pain indications, use of injectable biologics may provide superior analgesia, functionality, and/or quality of life compared to conventional medical management or placebo. Future high-quality randomized clinical trials are warranted with implementation of minimum reporting standards, standardization of preparation protocols, investigation of dose-response associations, and comparative analysis between different injectable biologics.

Context: The prevalence of obesity was 8% in 2002 and increased to 11.5% in 2011 in Indonesia. The results of the 2018 Basic Health Research stated that 13.6% of people over the age of 18 were overweight, and 21.8% were obese. Aims: To analyze the effect of intermittent restriction diet and continuous exercise with moderate intensity on individuals exposed to a high-calorie diet. Methods: A randomized posttest control group design was used as the research design, with 6 mice in each group as the study subjects. The groups in this study were the control group with a high-calorie diet (CON+), the intermittent restriction diet group (IRD), the moderate-intensity continuous exercise group (MEX), the combined intermittent restriction diet group and the moderate-intensity continuous exercise (HYB) group. The measured variables were cardiomyocyte diameter and brain natriuretic peptide (BNP) expression. Results: The results of this study were cardiomyocyte diameter CON+ (27.94 ± 5.65 µm), IRD (20.99 ± 11.80 µm), MEX (25.08 ± 9.14 µm), HYB (24.52 ± 5.90 µm) with p=0.578. Expression scores of BNP CON+ (6.00 ± 1.50), IRD (4.67 ± 1.00), MEX (5.42±2.09), HYB (6.27 ± 1.54) with p=0.335. These results showed no significant difference, but the intermittent restriction diet showed the most optimal mean with the lowest mean cardiomyocyte diameter and BNP expression. Conclusions: There was no effect between the combination of an intermittent restriction diet and moderate-intensity continuous exercise on cardiomyocyte diameter and BNP expression. However, there is a potential that an intermittent restriction diet has the optimal effect in preventing changes in the cardiac structure.

Abstract Drug-coated balloons (DCB) are increasingly utilized in percutaneous coronary intervention (PCI), but their effectiveness in coronary artery disease (CAD) needs further exploration. This study investigates the efficacy and safety of a DCB-based strategy for de novo left anterior descending artery (LAD) disease. Patients with de novo LAD lesions treated with DCB alone or combined with drug-eluting stents (DES) and were retrospectively enrolled from 2010 to 2023 (n = 268). The comparator group consisted of patients treated with second-generation DES from a Korean multicenter registry (n = 4,147). The primary endpoint was three-year major adverse cardiovascular events (MACE), including cardiac death, myocardial infarction, target vessel revascularization, target lesion thrombosis, and major bleeding. In the DCB-based group (n = 268), 218 (81.3%) received DCB-only, while 50 (18.7%) underwent a hybrid approach. After propensity score-matching of 243 paired subjects, baseline characteristics were balanced. The DCB-based PCI reduced overall stent burden by 86.7% and significantly lowered the risk of MACE at three years compared to DES-only PCI (4.5% vs. 7.6%, HR 0.50, 95% CI 0.28–0.90; p = 0.020). The most significant reduction was in major bleeding. The DCB-based approach offers an alternative to DES-only strategy for LAD PCI by reducing three-year MACE risk, supporting its use in treating de novo CAD.

Abstract Because there is a lack of comparative studies assessing drug-coated balloon (DCB) and drug-eluting stent (DES) outcomes with respect to intraluminal (IL) and subintimal (SI) approaches in femoropopliteal (FP) total occlusive lesions, we compared the outcomes between DCB (including bailout stenting) and DES treatments for this lesion. A total of 487 limbs (434 patients) were divided into the IL (n = 344, DCB: n = 268, DES: n = 76) and SI (n = 143, DCB: n = 83, DES: n = 60) approach groups. The primary outcome was a major adverse limb event (MALE), defined as above-ankle amputation or repeat revascularization of the index limb. Secondary outcomes included clinically driven target lesion revascularization (TLR), loss of clinical patency, and all-cause death. After adjustment, in each IL and SI approach, the 2-year rates of MALE (p = 0.180 and p = 0.236, respectively), TLR, loss of clinical patency, and all-cause death were similar between the DCB and DES groups. In the DCB and DES groups, both primary and secondary outcomes were similar between the IL and SI approaches. DCB and DES strategies for patients presenting with FP total occlusive lesions demonstrated similar outcomes regardless of the IL or SI approach. Clinical Trial Registration: NCT02748226.

Framed by family language policy and children's agency, this multiple case study investigates two Hmong American children's conceptualization of Hmong and the strategies they develop and utilize in their home. Interviews, artifacts, recorded interactions, and video recorded sessions were collected to gain an understanding of how the children viewed and valued Hmong. The findings reveal that while both children understood Hmong as a family language, they differed in their conceptualization of the utility of Hmong, creating their own investment in the heritage language that drove their practices. Importantly, as they negotiate their respective FLPs, they also influence their younger sibling's heritage language investment and practices.
