Understanding the microbial genomic contributors to antimicrobial resistance (AMR) is essential for early detection of emerging AMR infections, a pressing global health threat in human and veterinary medicine. Here we used whole genome sequencing and antibiotic susceptibility test data from 980 disease causing Escherichia coli isolated from companion and farm animals to model AMR genotypes and phenotypes for 24 antibiotics. We determined the strength of genotype-to-phenotype relationships for 197 AMR genes with elastic net logistic regression. Model predictors were designed to evaluate different potential modes of AMR genotype translation into resistance phenotypes. Our results show a model that considers the presence of individual AMR genes and total number of AMR genes present from a set of genes known to confer resistance was able to accurately predict isolate resistance on average (mean F1 score = 98.0%, SD = 2.3%, mean accuracy = 98.2%, SD = 2.7%). However, fitted models sometimes varied for antibiotics in the same class and for the same antibiotic across animal hosts, suggesting heterogeneity in the genetic determinants of AMR resistance. We conclude that an interpretable AMR prediction model can be used to accurately predict resistance phenotypes across multiple host species and reveal testable hypotheses about how the mechanism of resistance may vary across antibiotics within the same class and across animal hosts for the same antibiotic.

Transaction cost economics predicts that investments in management control will enable risky interfirm transactions. Risk is rarely eliminated, because firms trade off costs of management control and expected costs of control loss (together, the "cost of control"). The resultant solution typically comprises a mix of control investments with residual performance and residual relational risks. Transaction cost economics also predicts that the control-residual risk trade-off will vary with the cost of control. We use survey data on 287 risky information technology transactions to test whether the controlresidual risk trade-off varies predictably with two partnership-specific factors that proxy for variation in the cost of control: prior ties between exchange partners and the criticality of strategic resources to the transaction. The results support the hypotheses, providing novel evidence on trade-offs that managers make when investing in management controls while also prudently accepting some risks.

Abstract Background Many control methods have been implemented to tackle onchocerciasis and great successes have been achieved, leading to a paradigm shift from control of morbidity to interruption of transmission and ultimately elimination. The mandate of the African Programme for Onchocerciasis Control (APOC) ended in 2015, and endemic countries are to plan and conduct elimination activities by themselves, with technical assistance by the Expanded Special Project for Elimination of Neglected Tropical Diseases (ESPEN). To this end, an elimination expert committee was set up in Cameroon in 2018. This committee identified the need to update the data on the current situation of onchocerciasis. The present study aims to systematically review and report all available epidemiological data, including prevalence, intensity and transmission of onchocerciasis to provide pertinent information that will be useful to design optimal strategies to achieve onchocerciasis elimination in Cameroon. Methods PubMed/MEDLINE, EMBASE and Web of Science will be searched from inception onwards. Grey literature will be identified through Google Scholar searches, dissertation databases and other relevant documents such as government reports. Eligible studies will be mostly observational, including cohort and cross-sectional surveys. No limitations will be imposed on publication status and study period. The primary outcomes will be (1) the prevalence and intensity of Onchocerca volvulus infection in humans, (2) transmission intensity and (3) impact of interventions on prevalence, intensity and transmission of onchocerciasis. Secondary outcomes will be environmental and socio-demographic factors supporting the primary outcomes. Two reviewers will independently screen all citations, full-text articles and abstract data. Potential conflicts will be resolved through discussion. Methodological quality including bias will be appraised using appropriate approaches. A narrative synthesis will describe quality and content of the epidemiological evidence. Prevalence and intensity of infection estimates will be stratified according to gender, age, geographical location and year of publication. Discussion This study will provide the health authorities as well as the scientific community with up-to-date information about the epidemiological situation of onchocerciasis in Cameroon. Understanding the spatiotemporal dynamics of the infection will help to define alternative and complementary strategies to accelerate onchocerciasis elimination in the country. Results of this review will also be used to update existing epidemiological models for onchocerciasis in order to fine-tune predictions of elimination timeframes in the country. Systematic review registration This protocol is under registration review in PROSPERO.

# Background The District Health Information Software (DHIS2), adopted as national health information system by the ministry of public health in Cameroon, did not integrate neglected tropical diseases (NTD) program data. Integrating NTD program data into the national DHIS2 might require more than technical skills. Our study aimed to explore the factors that affect acceptability and use of DHIS2 by NTD stakeholders for successful integration of NTD program data into the national DHIS2. # Methods For purposes of this mixed-methods study, the data were collected through a self-administered questionnaire targeting NTD stakeholders at different levels of the health pyramid from all the ten Regions in Cameroon. The questionnaire was based on a modified Unified Theory of Acceptance and Use of Technology (UTAUT) model, supplemented by a qualitative analysis to assess the acceptability, and use of the DHIS2 as a platform for NTD program data in Cameroon. # Results We found 81.9% (95% confidence interval, CI=0.784-0.859; standard error=0.019) of intention to use DHIS2 for NTDs program data and 18.4% (95% CI=0.130-0.289; standard error=0.041) of actual use among survey participants. Social influence (β=0.269, *P*=0.000), voluntariness of use (β=0.243, *P*=0.000), performance expectancy (β=0.186, *P*=0.010), and training adequacy (β=0.199, *P*=0.000) would positively influence intention to use DHIS2. Computer anxiety (β=-0.230, *P*=0.000) and technology experience (β=0.374, *P*=0.000) would have a significant negative and positive effect on actual use, respectively. The most critical challenges in using DHIS2 referred to facilitating conditions (conditions of the work environment), specifically electricity and internet connection, impeding actual use of DHIS2. # Conclusions Our study revealed that NTD stakeholders in Cameroon are ready to accept DHIS2 for NTD program in Cameroon. However, to ensure its successful implementation. For example, we recommend that NTD program managers plan adequate support in providing proper training, non-vendor specific 2G-3G-4G internet modems with data bundle and smartphones/laptops to ease the use of DHIS2 by NTD stakeholders. We showed that acceptability of DHIS2 studied through UTAUT model should be complemented with a qualitative analysis for richer insights.

The plant-pathogenic fungus Mycosphaerella graminicola (asexual stage: Septoria tritici) causes septoria tritici blotch, a disease that greatly reduces the yield and quality of wheat. This disease is economically important in most wheat-growing areas worldwide and threatens global food production. Control of the disease has been hampered by a limited understanding of the genetic and biochemical bases of pathogenicity, including mechanisms of infection and of resistance in the host. Unlike most other plant pathogens, M. graminicola has a long latent period during which it evades host defenses. Although this type of stealth pathogenicity occurs commonly in Mycosphaerella and other Dothideomycetes, the largest class of plant-pathogenic fungi, its genetic basis is not known. To address this problem, the genome of M. graminicola was sequenced completely. The finished genome contains 21 chromosomes, eight of which could be lost with no visible effect on the fungus and thus are dispensable. This eight-chromosome dispensome is dynamic in field and progeny isolates, is different from the core genome in gene and repeat content, and appears to have originated by ancient horizontal transfer from an unknown donor. Synteny plots of the M. graminicola chromosomes versus those of the only other sequenced Dothideomycete, Stagonospora nodorum, revealed conservation of gene content but not order or orientation, suggesting a high rate of intra-chromosomal rearrangement in one or both species. This observed "mesosynteny" is very different from synteny seen between other organisms. A surprising feature of the M. graminicola genome compared to other sequenced plant pathogens was that it contained very few genes for enzymes that break down plant cell walls, which was more similar to endophytes than to pathogens. The stealth pathogenesis of M. graminicola probably involves degradation of proteins rather than carbohydrates to evade host defenses during the biotrophic stage of infection and may have evolved from endophytic ancestors.

With the integration of Intermitted Renewables Energy (I-RE) electricity production, capacity is shifting from central to decentral. So, the question is if it is also necessary to adjust the current load balancing system from a central to more decentral system. Therefore, an assessment is made on the overall effectiveness and costs of decentralized load balancing, using Flexible Renewable Energy (F-RE) in the shape of biogas, Demand Side Management (DSM), Power Curtailment (PC), and electricity Storage (ST) compared to increased grid capacity (GC). As a case, an average municipality in The Netherlands is supplied by 100% I-RE (wind and solar energy), which is dynamically modeled in the PowerPlan model using multiple scenarios including several combinations of balancing technologies. Results are expressed in yearly production mix, self-consumption, grid strain, Net Load Demand Signal, and added cost. Results indicate that in an optimized scenario, self-consumption of the municipality reaches a level of around 95%, the total hours per year production matches demand to over 90%, and overproduction can be curtailed without substantial losses lowering grid strain. In addition, the combination of balancing technologies also lowers the peak load to 60% of the current peak load in the municipality, thereby freeing up capacity for increased demand (e.g., electric heat pumps, electric cars) or additional I-RE production. The correct combination of F-RE and lowering I-RE production to 60%, ST, and PC are shown to be crucial. However, the direct use of DSM has proven ineffective without a larger flexible demand present in the municipality. In addition, the optimized scenario will require a substantial investment in installations and will increase the energy cost with 75% in the municipality (e.g., from 0.20€ to 0.35€ per kWh) compared to 50% (0.30€ per kWh) for GC. Within this context, solutions are also required on other levels of scale (e.g., on middle or high voltage side or meso and macro level) to ensure security of supply and/or to reduce overall costs.

In many work and decision situations, realizing cooperation among individuals is important. However, decision making environments of individuals are far from stable, resulting in changes in task complexity and the social settings they encounter. We argue that past experiences with cooperative behavior can result in different cooperative norms and expectations about the behavior of others and will have an effect on an individual's subsequent behavior in new situations. This study experimentally investigates these dynamics of cooperative behavior in social dilemmas and addresses the role of communication to provide empirical evidence about a cognitive mechanism that may lead to these spillovers. Specifically, the experimental design randomly assigns subjects to one type of repetitive interactions in the first social dilemma (single partner or different partners) and we then examine how this impacts the propensity to behave cooperatively in subsequent social dilemmas with unfamiliar partners (either single or different). Because of the variety in complexity of decision-making environments in practice, we do so by examining behavioral spillovers across three different social dilemmas that vary in difficulty to make cooperation successful. Our findings show that individuals cooperate more during initial interactions with a single partner. More importantly, this has positive spillover effects for subsequent behavior and communication, even to settings without repeated interactions with a single partner. However, environmental conditions affect the ability to transfer established norms of cooperation to subsequent interactions, as an initially learned cooperative norm is gradually replaced by a more competitive attitude when individuals start to interact with unfamiliar others in a setting in which cooperative success is more difficult to achieve. Our findings illustrate the power of repeated interactions for establishing and sustaining cooperation in other settings and enhance understanding of how cooperative decisions can be shaped by both incentives and the broader behavioral context of individuals.

Clinical and model studies indicate that low nitric oxide (NO) bioavailability due in part to profound hypoargininemia contributes to cerebral malaria (CM) pathogenesis. Protection against CM pathogenesis may be achieved by altering the diet before infection with Plasmodium falciparum infection (nutraceutical) or by administering adjunctive therapy that decreases CM mortality (adjunctive therapy). This hypothesis was tested by administering citrulline or arginine in experimental CM (eCM). We report that citrulline injected as prophylaxis immediately post infection (PI) protected virtually all mice by ameliorating (i) hypoargininemia, (ii) urea cycle impairment, and (iii) disruption of blood brain barrier. Citrulline prophylaxis inhibited plasma arginase activity. Parasitemia was similar in citrulline- and vehicle control-groups, indicating that protection from pathogenesis was not due to decreased parasitemia. Both citrulline and arginine administered from day 1 PI in the drinking water significantly protected mice from eCM. These observations collectively indicate that increasing dietary citrulline or arginine decreases eCM mortality. Citrulline injected ip on day 4 PI with quinine-injected ip on day 6 PI partially protected mice from eCM; citrulline plus scavenging of superoxide with pegylated superoxide dismutase and pegylated catalase protected all recipients from eCM. These findings indicate that ameliorating hypoargininemia with citrulline plus superoxide scavenging decreases eCM mortality.
