Abstract Engineering Kondo lattice with tailored functionality is desirable for elucidating the heavy fermion physics. We realize the construction of an artificial Kondo lattice/superconductor heterojunction by growing monolayer VSe2 on bulk 2H-NbSe2 with molecular beam epitaxy. Spectroscopic imaging scanning tunneling microscopy measurements show the emergence of a new charge density wave (CDW) phase with $$\sqrt{3}\times$$ 3 × $$\sqrt{3}$$ 3 periodicity on the monolayer VSe2. Unexpectedly, a pronounced Kondo resonance appears around the Fermi level, and distributes uniformly over the entire film, evidencing the formation of Kondo lattice. Density functional theory calculations suggest the existence of magnetic interstitial V atoms in VSe2/NbSe2, which play a key role in forming the CDW phase along with the Kondo lattice observed in VSe2. The Kondo origin is verified from both the magnetic field and temperature dependences of the resonance peak, yielding a Kondo temperature of ~ 44 K. Moreover, a superconducting proximity gap opens on monolayer VSe2, whose shape deviates from the function of one-band BCS superconductor, but is reproduced by model calculations with heavy electrons participating the pairing condensate. Our work lays the experimental foundation for studying interactions between the heavy fermion liquids and the superconducting condensate.

Background Diabetic kidney disease (DKD), a prevalent complication of diabetes mellitus, is often associated with acute kidney injury (AKI). Thus, the development of preventive and therapeutic strategies is crucial for delaying the progression of AKI and DKD.Methods The GSE183276 dataset, comprising the data of 20 healthy controls and 12 patients with AKI, was downloaded from the Gene Expression Omnibus (GEO) database to analyze the AKI group. For analyzing the DKD group, the GSE131822 dataset, comprising the data of 3 healthy controls and 3 patients with DKD, was downloaded from the GEO database. The common differentially expressed genes (DEGs) in renal tubular epithelial cells (TECs) were subjected to enrichment analyses. Next, a protein-protein interaction (PPI) network was constructed using the Search Tool for the Retrieval of Interacting Genes database to analyze gene-related regulatory networks. Finally, the AKI animal models and the DKD and AKI cell models were established, and the reliability of the identified genes was validated using quantitative real-time polymerase chain reaction analysis.Results Functional analysis was performed with 40 common DEGs in TECs. Eight hub genes were identified using the PPI and gene-related networks. Finally, validation experiments with the in vivo animal model and the in vitro cellular model revealed the four common DEGs. Four DEGs that share molecular mechanisms in the pathogenesis of DKD and AKI were identified. In particular, the expression of Integrin Subunit Beta 6(ITGB6), a hub and commonly upregulated gene, was upregulated in the in vitro models.Conclusion ITGB6 may serve as a biomarker for early AKI diagnosis in patients with DKD and as a target for early intervention therapies.

Acute kidney injury (AKI) is a systemic clinical syndrome increasing morbidity and mortality worldwide in recent years. Renal tubular epithelial cells (TECs) death caused by mitochondrial dysfunction is one of the pathogeneses. The imbalance of mitochondrial quality control is the main cause of mitochondrial dysfunction. Mitochondrial quality control plays a crucial role in AKI. Mitochondrial quality control mechanisms are involved in regulating mitochondrial integrity and function, including antioxidant defense, mitochondrial quality control, mitochondrial DNA (mtDNA) repair, mitochondrial dynamics, mitophagy, and mitochondrial biogenesis. Currently, many studies have used mitochondrial dysfunction as a targeted therapeutic strategy for AKI. Therefore, this review aims to present the latest research advancements on mitochondrial dysfunction in AKI, providing a valuable reference and theoretical foundation for clinical prevention and treatment of this condition, ultimately enhancing patient prognosis.

Abstract The heavy fermion physics is dictated by subtle competing exchange interactions, posing a challenge to their understanding. One-dimensional (1D) Kondo lattice model has attracted special attention in theory, because of its exact solvability and expected unusual quantum criticality. However, such experimental material systems are extremely rare. Here, we demonstrate the realization of quasi-1D Kondo lattice behavior in a monolayer van der Waals crystal NbSe2, that is driven into a stripe phase via Se-deficient line defects. Spectroscopic imaging scanning tunneling microscopy measurements and first-principles calculations indicate that the stripe-phase NbSe2 undergoes a novel charge-density wave transition, creating a matrix of local magnetic moments. The Kondo lattice behavior is manifested as a Fano resonance at the Fermi energy that prevails the entire film with a high Kondo temperature. Importantly, coherent Kondo screening occurs only in the direction of the stripes. Upon approaching defects, the Fano resonance exhibits prominent spatial 1D oscillations along the stripe direction, reminiscent of Kondo holes in a quasi-1D Kondo lattice. Our findings provide a platform for exploring anisotropic Kondo lattice behavior in the monolayer limit.

Background The aim of this study was to evaluate the roles of interleukin (IL)-17A in risk stratification and prognosis of patients with sepsis-associated acute kidney injury (SAKI). Methods We enrolled 146 sepsis patients (84 non-SAKI and 62 SAKI patients) admitted to the emergency department from November 2020 to November 2021. Patients with SAKI were differentiated based on the severity of acute kidney injury. All clinical parameters were evaluated upon admission before administering antibiotic treatment. Inflammatory cytokines were assessed using flow cytometry and the Pylon 3D automated immunoassay system (ET Healthcare). In addition, a receiver operating characteristic (ROC) curve was utilized to determine the prognostic values of IL-17A in SAKI. Results The levels of creatinine, IL-2, IL-4, IL-6, IL-17A, tumor necrosis factor alpha, C-reactive protein, and procalcitonin (PCT) were significantly higher in the SAKI group than in the non-SAKI group (p < 0.05). The level of IL-17A revealed significant differences among stages 1, 2, and 3 in SAKI patients (p < 0.05). The mean levels of PCT, IL-4, and IL-17A were significantly higher in the non-survival group than in the survival group in SAKI patients (p < 0.05). In addition, the area under the ROC curve of IL-17A was 0.811. Moreover, the IL-17A cutoff for differentiating survivors from non-survivors was 4.7 pg/mL, of which the sensitivity and specificity were 77.4% and 71.0%, respectively. Conclusion Elevated levels of IL-17A could predict that SAKI patients are significantly prone to worsening kidney injury with higher mortality. The usefulness of IL-17A in treating SAKI requires further research.

Abstract: Seed ageing during storage or shipping can affect the yield and cost of agricultural production. Sweet corn is a naturally mutated corn cultivar with weak seed vitality during storage. In this study, we have shown that endogenous hormone level dynamics of fresh and aged sweet corn seeds were different during early geminating process. Transcriptomic assembling uncovered several novel transcripts during early stage of sweet corn germination in signal transduction and RNA processing pathways. Comparative transcriptomic profiling revealed that expression of many genes was activated earlier in aged seeds than fresh ones. Functional analyses showed that, as two major networks, plant hormone signaling and defense pathways were different between fresh and aged sweet corn seeds. Auxin, cytokinin, gibberellin, salicylic acid and jasmonic acid signaling pathways were activated earlier in aged seeds than fresh ones on the first day after imbibition. These results provided knowledge for understanding the ageing process of sweet corn seeds and several candidate genes for genetic study of seed ageing mechanism.

Abstract Background With more emergency visits, there is increasing pressure to provide emergency medical services globally and locally. This study aimed to investigate the epidemiological characteristics and the disease spectrum of patients presenting in the last three years to the Department of Emergency Medicine of Tianjin Medical University General Hospital, a tertiary hospital in Tianjin, China, to improve the services of the emergency medicine department. Methods A retrospective study was conducted on all patients in the Department of Emergency Medicine of Tianjin Medical University General Hospital from Jan 1, 2017, 00:00:00 to Dec 31, 2020, 23:59:59, including variables like medical record number, gender, age, date of admission, principal diagnosis. The data were analyzed by SPSS statistical software; statistical charts were prepared by GraphPad Prism9.0 and SPSS 20.0; statistical tables were made by Microsoft Excel. Results A total of 1,314,916 patients presented to the Department of Emergency Medicine of Tianjin Medical University General Hospital from Jan 1, 2017, 00:00:00 to Dec 31, 2020, 23:59:59. In terms of gender distribution, the male–female ratio was 0.78∶1. As for age distribution, patients aged 60–69 were the most (23.47%), and patients younger than 20 years were the least (2.80%). Concerning monthly data, the number of visits peaked during January and December. The distribution of daily visits showed the feature of three highs and a low. The top three prevalence diseases in the emergency disease spectrum were respiratory, cardiovascular, and digestive diseases. The respiratory system was the most common in patients with infectious diseases (200,912, accounting for 86.97%). Among the patients suffering from infectious diseases, the number of patients with respiratory infections peaked in 2019 (73,530) and was the lowest in 2020 (20,078). Conclusions From 2017 to 2019, the demand for emergency services in Tianjin Medical University General Hospital continued to increase, but it was greatly affected by COVID-19 in 2020. This emergency department is mainly for patients with respiratory system, circulatory system and digestive system diseases, and its treatment time is relatively centralized. The prevention of diseases for people of all ages, especially female patients and the elderly, should be strengthened, and emergency medical resources should be allocated reasonably according to the peak months and crowed periods of patients.

Background. Rhabdomyolysis (RM) refers to a clinical syndrome in which muscle cells are damaged by various causes and the clinical manifestations are mainly muscle pain, weakness, and dark urine. Acute kidney injury (AKI) is a serious complication of RM with complex mechanisms and high mortality. Therefore, understanding the pathogenesis and clinical manifestations, early diagnosis and treatment of RM are crucial to improve its prognosis. Method. Analysis of medical records of RM patients admitted to Tianjin Medical University General Hospital from October 2019 to October 2022. Statistical software SPSS 25.0 was used to analyze the data. The risk factors of RM-complicated AKI were analyzed by logistic regression. The receiver operating characteristic (ROC) curve was plotted, the area under the curve (AUC) was calculated, and the optimal cutoff value was determined by the Youden index. P

This paper focuses on the hydrodynamic interaction between the large floating body and a small transfer platform in a jettyless floating transfer system. A high-order boundary-element method combined with a direct time-domain-solution method to calculate and analyze the hydrodynamic response of the small platform while berthed with the fixed large floating body and freely floating large body under wave action was applied and compared with the hydrodynamic response of a single small transfer platform. It was found that when the large floating body and the small transfer platform were both located on the leeward side, they had little mutual influence, and the hydrodynamic response agreed well with that of the single small transfer platform and the single large floating body. While the small platform was located on the leeward side of the large floating body, it significantly affected the hydrodynamic response of the platform, resulting in a significant reduction in horizontal motion and pitch motion, meaning that the shielding effect was significant. Within a certain range of wave periods, the heave-motion amplitudes can be significantly reduced, but beyond that range, they increase. Therefore, it is important to carefully consider the relative motions of LNG transport ships and small platforms when connecting them via aerial jumper pipes in jettyless floating transfer systems.

Background: This study aimed to analyze the potential genes associated with immune cell infiltration in atherosclerosis (AS). Methods: Gene expression profile data (GSE57691) of human arterial tissue samples were downloaded, and differentially expressed RNAs (DERNAs; long-noncoding RNA [lncRNAs], microRNAs [miRNAs], and messenger RNAs [mRNAs]) in AS vs. control groups were selected. Based on genome-wide expression levels, the proportion of infiltrating immune cells in each sample was assessed. Genes associated with immune infiltration were selected, and subjected to Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. Finally, a competing endogenous RNA (ceRNA) network was constructed, and the genes in the network were subjected to functional analyses. Results: A total of 1749 DERNAs meeting the thresholds were screened, including 1673 DEmRNAs, 63 DElncRNAs, and 13 DEmiRNAs. The proportions of B cells, CD4+ T cells, and CD8+ T cells were significantly different between the two groups. In total, 341 immune-associated genes such as HBB, FCN1, IL1B, CXCL8, RPS27A, CCN3, CTSZ, and SERPINA3 were obtained that were enriched in 70 significantly related GO biological processes (such as immune response) and 15 KEGG pathways (such as chemokine signaling pathway). A ceRNA network, including 33 lncRNAs, 11 miRNAs, and 216 mRNAs, was established. Conclusion: Genes such as FCN1, IL1B, and SERPINA3 may be involved in immune cell infiltration and may play important roles in AS progression via ceRNA regulation.

With the rapid development of the construction industry, tower cranes are increasingly used in coastal engineering. However, due to the complexity of their operating environment, tower cranes are vulnerable to typhoons, thunderstorms, and other extreme natural disasters. Therefore, it is becoming increasingly important to carry out safety warnings for the tower crane structure under the action of strong winds. In this paper, a real-time monitoring system for tower responses based on the Internet of things (IoT), which realizes long-term monitoring of the whole process of tower crane operation, was built. Based on the long-term monitoring data and the machine learning algorithm, two tower response prediction models were established. During the transit of super typhoon In-fa, the maximum displacement of the tower structure was predicted in advance, based on the measured wind speed data at the site, which is in good agreement with the displacement data monitored by the IoT. The results show that under strong winds, the non-working tower has a response lag, resulting in the fact that its maximum displacement does not correspond to the maximum wind speed moment at the site. This is mainly due to the weathercock effect of the tower in the non-working condition. The prediction model proposed in this paper can provide timely and effective safety warnings for the tower structure. It also can provide useful engineering references and scientific structural safety warning suggestions for the same type of tower cranes that do not have IoT monitoring systems installed.

Abstract Background Rapamycin (RAPA) is a potent angiogenic inhibitor and the aim of this study is to identify the inhibitory effect of RAPA on retinal neovascularization (RNV) in experimental oxygen-induced retinopathy (OIR). Methods Forty-two 7-day-old C57BL/6 J mice were randomly divided into normoxia control group (14 mice), OIR group (14 mice), and rapamycin (RAPA) group. OIR model was induced in OIR and RAPA group. Vehicle and RAPA (2 mg/kg/d) was injected intraperitoneally daily from postnatal day 12 (P12) in OIR and RAPA groups, respectively. RNV was evaluated using fluorescence angiography and histopathology on P17. Non-perfused areas of retina were analyzed by Image-Pro plus 6.0 software. Retinal expression of cyclin D1 was detected both at mRNA and protein levels. Results RAPA treatment significantly decreased RNV, non-perfused areas and number of endothelial cell nuclei breaking through the internal limiting membrane (ILM) in OIR mice. Moreover, RAPA decreased activation of cyclin D1 in retina caused by OIR. Conclusion RAPA can inhibit RNV by downregulating the expression of cyclin D1, which indicates its therapeutic potential in treating RNV-related diseases.

BiFeO3 is one of the most important perovskite ferroelectric materials. It is known that cation substitution in BiFeO3 can greatly modulate its fundamental physical properties for various applications. However, the role of anion substitution in BiFeO3 is still largely unknown. In this article, using first-principles calculations, we have systematically investigated the trends of electronic, ferroelectric, and optoelectronic properties of BiFeX3 as a function of complete anion evolution. Interestingly, the calculated bandgaps of BiFeX3, in the favorable G-type antiferromagnetic configuration, decrease from 3.32 eV (X = O) to 1.27 eV (X = S) to 0.75 eV (X = Se) and to 0.27 eV (X = Te) due to the enhanced Fe d and X p orbital hybridization at the bottom of the conduction band of BiFeX3, providing an insight to explore narrow bandgap ferroelectrics. Meanwhile, it is found that the value of spontaneous polarization of BiFeX3 decreases along with the increase in dipole moment from X = O to X = Te. In addition, the ferroelectric reversal barrier of BiFeX3 increases from X = O to X = Te, indicating that an increased external field is required to flip the opposite ferroelectric phases. The role of covalency of Bi–X bonds is revealed in the formation of spontaneous polarization. Furthermore, in the visible-light energy range, the increased absorption coefficient has been observed for larger anions and significant optical anisotropy has been found in different energy ranges for different BiFeX3 systems. Our work provides a fundamental understanding on the electronic, ferroelectric, and optoelectronic properties of complete anion substitution in BiFeO3.

Loss of function of the lipid kinase diacylglycerol kinase ε (DGKε), encoded by the gene DGKE, causes a form of atypical hemolytic uremic syndrome that is not related to abnormalities of the alternative pathway of the complement, by mechanisms that are not understood. By generating a potentially novel endothelial specific Dgke-knockout mouse, we demonstrate that loss of Dgke in the endothelium results in impaired signaling downstream of VEGFR2 due to cellular shortage of phosphatidylinositol 4,5-biphosphate. Mechanistically, we found that, in the absence of DGKε in the endothelium, Akt fails to be activated upon VEGFR2 stimulation, resulting in defective induction of the enzyme cyclooxygenase 2 and production of prostaglandin E2 (PGE2). Treating the endothelial specific Dgke-knockout mice with a stable PGE2 analog was sufficient to reverse the clinical manifestations of thrombotic microangiopathy and proteinuria, possibly by suppressing the expression of matrix metalloproteinase 2 through PGE2-dependent upregulation of the chemokine receptor CXCR4. Our study reveals a complex array of autocrine signaling events downstream of VEGFR2 that are mediated by PGE2, that control endothelial activation and thrombogenic state, and that result in abnormalities of the glomerular filtration barrier.

Histones are considered potential risk factors that contribute to the development of septic acute kidney injury (SAKI) by inducing apoptosis and inflammation. This study aimed to explore the protective effects of heparin on septic acute kidney injury through the neutralization of extracellular histones (EH) and to uncover the underlying mechanism. C57BL mice (16 each) were randomly divided into the sham group, the sepsis group (established by cecal ligation and puncture operation, CLP), and the heparin intervention group. Mice in the heparin intervention group received a subcutaneous injection of unfractionated heparin (0.03 IU/g) 4 h after CLP. At 6 h after the operation, nine mice from each group were sacrificed by the removal of the eyeballs to harvest blood samples; the upper half of the right kidney was used as the study sample. Mice renal tubular epithelial cells cultivated in six-well plates were equally divided into five groups. We cultured cells treated with either histone (40 U), histone (40 U) + heparin (25 IU/ml), histone(40U) + lipopolysaccharides (LPS; 10 μg/ml), or histone (40 U) + LPS (10 μg/ml) + heparin (25 IU/ml) for 6 h. For the histone + heparin group and the histone + LPS + heparin group, histone (and LPS) were treated with heparin simultaneously. Mice in the heparin intervention group showed decreased levels of EH4, neutrophil gelatinase-associated lipocalin (NAGL), kidney injury molecule-1 (KIM-1), tumor necrosis factor-α (TNF-α), and interleukin (IL)-6 in the blood serum, longer average 72-h survival rate, significantly decreased kidney tissue edema, and a clearer glomerular structure coupled with decreased protein and mRNA expression levels of kidney apoptosis-related proteins (cleaved Caspase-3/Caspase-3 and Bax/Bcl-2) compared with those in the sepsis group at 6 h after CLP (P < 0.05). Meanwhile, cells in the heparin intervention group exhibited lower expression levels of serum EH4 and inflammatory cytokines, a lower apoptosis rate, and decreased expression of apoptosis-related proteins, both at protein and mRNA levels, than those in the histone-stimulated group at 6 h after stimulation (P < 0.05). Heparin may alleviate apoptosis and inflammation through the neutralization of histones, thus playing a protective role against septic acute kidney injury.

The growth of global energy transportation has promoted the rapid increase of large-scale LNG (liquefied natural gas) carriers, and concerns around the safety of LNG ships has attracted significant attention. Such a floating structure is affected by the external wave excitation and internal liquid sloshing. The interaction between the structure’s motion and the internal sloshing under wave actions may lead to the ship experiencing an unexpected accident. In this research, a hydrodynamic experiment is conducted to investigate the motion responses of a floating tank mooring, both close to and away from a dock. The resonance coupling effect of the internal sloshing and gap flow on the tank’s motion is considered. Based on the measured motion trajectory of the floating tank, the stability and safety of the floating tank are estimated. The results show that the sloshing resonance and narrow gap resonance are beneficial to the stability of the ship. This is helpful for controlling the motion of a berthed ship under wave action with a reasonable selection of the gap distance and the liquid level.

Water storage tanks in cities are usually large and are occasionally affected by earthquakes. A sudden earthquake can cause pressure pulses that damage water containers severely. In this study, the sloshing motion in a high filling level tank caused by seismic excitation is investigated by the numerical method in a 2D model. Two well-studied strong earthquakes are used to analyze the broadband frequency nonlinear displacement of the tank both in the longitudinal and vertical directions. Based on careful experimental verification, the free surface motion and the elevations at the side wall are captured, and the sloshing pressure response is examined. The results show that the 2D section of the cylindrical tank can be used to estimate the maximum response of the 3D sloshing, and the water motions under the seismic excitations are consistent with the modal characteristics of the sloshing. The time histories response of the water motion reflected that the sloshing response is hysteretic compared with the seismic excitation. The anti-seismic ability of the damping baffle shows that its effect on sloshing pressure suppression is limited, and further study on the seismic design of water tanks in earthquake-prone regions is needed.

Photodynamic antimicrobial chemotherapy (PACT), as a novel and effective therapeutic modality to eradicate drug resistant bacteria without provoking multidrug resistance, has attracted increasing attention. This study examined the antimicrobial efficacy of the novel cationic amino acid-porphyrin conjugate 4I with four lysine groups against two different clinical isolated strains (drug sensitive and multidrug resistant) of the Acinetobacter baumannii species and its toxicity on murine dermal fibroblasts in vitro, as well as the therapeutic effect of PACT on acute, potentially lethal multidrug resistant strain excisional wound infections in vivo. The PACT protocol exposed 4I to illumination, exhibiting high antimicrobial efficacy on two different strains due to a high yield of reactive oxygen species (ROS) and non-selectivity to microorganisms. The photoinactivation effects of 4I against two different strains were dose-dependent. At 3.9 μM and 7.8 μM, PACT induced 6 log units of inactivation of sensitive and multidrug resistant strains. In contrast, 4I alone and illumination alone treatments had no visibly antimicrobial effect. Moreover, cytotoxicity tests revealed the great safety of the photosensitizer 4I in mice. In the in vivo study, we found 4I-mediated PACT was not only able to kill bacteria but also accelerated wound recovery. Compared with non-treated mice, over 2.89 log reduction of multidrug resistant Acinetobacter baumannii strain was reached in PACT treat mice at 24 h post-treatment. These results imply that 4I-mediated PACT therapy is an effective and safe alternative to conventional antibiotic therapy and has clinical potential for superficial drug-resistant bacterial infections.

Congestions of the traffic flow within the urban traffic network have been a challenging task for all the urban developers. Many approaches have been introduced into the current system to solve the traffic congestion problems. Reconfiguration of the traffic signal timing plan has been carried out through implementation of different techniques. However, dynamic characteristics of the traffic flow increase the difficulties towards the ultimate solutions. Thus, traffic congestions still remain as unsolvable problems to the current traffic control system. In this study, artificial intelligence method has been introduced in the traffic light system to alter the traffic signal timing plan to optimize the traffic flows. Q-learning algorithm in this study has enhanced the traffic light system with learning ability. The learning mechanism of Q-learning enables traffic light intersections to release itself from traffic congestions situation. Adjacent traffic light intersections will work independently and yet cooperate with each others to a common goal of ensuring the fluency of the traffic flows within the traffic network. The simulated results show that the Q-Learning algorithm is able to learn from the dynamic traffic flow and optimize the traffic flow accordingly.

Objective: To explore the effect and mechanism of prostaglandins D2 (PGD2) on the stemness of gastric cancer stem cells (GCSCs). Methods: 7901-GCSCs were enriched by serum-free culture method; then the positivity rate of CD44, a stemness marker, was detected by flow cytometry in serum-free cultured 7901-GCSCs; the sphere-forming ability was detected by the sphere-forming assay after stimulation with different concentrations of PGD2 (2.5, 5, 10) μg/mL, and the expression of stemness-related indicators (OCT4, CD44) and autophagyrelated proteins (LC3, Beclin-1) after PGD2 stimulation was detected by the western blot assay in different concentrations. The expression of stemness-related indexes (OCT4, CD44) and autophagy-related proteins (LC3, Beclin-1) were detected by Western blot assay after stimulation with different concentrations of PGD2. The expression of autophagy-related proteins after stimulation with different concentrations of CQ (2.5, 5, 10) μM was detected by Western blot experiment. The protein expression of autophagy-related proteins (LC3, Beclin-1) and stemness-related indexes (OCT4, CD44) was detected by Western blot experiment after PGD2 as well as PGD2+CQ treatment. Results: Flow cytometry results showed that the expression of CD44 positivity was increased in serum-free cultured 7901-GCSCs compared with gastric cancer cells SGC-7901 (P<0.05), which fulfilled the needs of subsequent experiments. The results of stem cell spheroid formation assay showed that the spheroid formation ability of 7901-GCSCs in the PGD2 group was significantly weakened compared with that of the DMSO group (P<0.05).Western blot results showed that the protein expression of stemness-related indexes (OCT4, CD44) was down-regulated in the 7901-GCSCs in the PGD2 group compared with that of the DMSO group (P<0.05), and the expression of autophagy-related proteins (LC3, Beclin-1) expression increased (P<0.05).Compared with the DMSO group, the expression of autophagy-related proteins (LC3, Beclin-1) was decreased in the CQ group (P<0.05).Western blot results also showed that the expression of cellular autophagy-related proteins and stemness-related indexes in the PGD2+CQ group was not significantly changed compared with that of the DMSO group (ns: the difference was not significant),suggesting that the CQ could block the effect of PGD2 on the expression of stemness markers in 7901-GCSCs. 7901-GCSCs stemness inhibition.
