The lesser kestrel ( Falco naumanni ) is a small falcon with a Euro-Central and Asian-Mediterranean range wintering in sub-Saharan Africa. In the second half of the 20th century, the European population experienced a steep decline and was classified as at risk; thus, its biological and ecological aspects have been widely investigated. Nonetheless, data on hematology and plasma chemistry are not yet available. Therefore, hematology and biochemistry parameters were investigated in a sampling population of clinically healthy lesser kestrels (21 female and 10 male adults) from an Italian rescue center during breeding season, estimating the 95% (2.5 - 97.5th percentile) reference intervals (RI) for standard tests based on either parametric or robust statistical methods. The effect of sex on the referenced parameters was also tested and showed no statically significant differences. The established 95% RI highlighted values comparable with those of other similar raptors such as American kestrel ( Falco sparverius ) and peregrine falcon ( Falco peregrinus ). As the first recorded hematology and serum chemistry RI, these clinical data could support conservation efforts and clarify the effects of various environmental and ecological factors on the clinical and health status of lesser kestrels, although they should be reinforced with further data from healthy wild animals.

Leopard sharks ( Triakis semifasciata ) are temperate, Eastern Pacific elasmobranchs popular in public aquariums. Blood analysis is commonly used for assessing animal health, yet reference values have not been established for this species. This study analyzed T. semifasciata population data to characterize blood reference values for a collection of T. semifasciata housed at a public aquarium. Twenty-one captive leopard sharks were sampled. Blood was collected during annual health examinations from sedated animals. After collection, blood samples were anticoagulated with lithium heparin, and hematocrit and plasma biochemistry values were analyzed. The minimum-maximum ranges were hematocrit 11-31%, buffy coat 1-2%, glucose 4.94-9.38 mM/L, sodium 244-272 mM/L, potassium 3.7-5.5 mM/L, chloride 214-246 mM/L, aspartate aminotransferase 5-31 U/L, creatine kinase 36-1,136 U/L, calcium 3.65-3.95 mM/L, phosphorus 1.13-2.23 mM/L, total protein 21-38 g/L, and total CO 2 12-18 mM/L. The values identified will contribute to a better understanding of captive leopard shark physiology and to improved veterinary care for captive leopard sharks. Further research can examine the validity of machines like the Vetscan VS2, which will expand the resources available to care professionals.

Between the years 2022 and 2023, 62 red kite ( Milvus milvus ) nestlings were translocated from England to Spain to bolster declining populations in mainland Europe as part of a wider conservation initiative. Health examinations were undertaken by veterinarians ahead of translocation, including examination of hematology and biochemistry parameters from blood samples. This study aimed to establish reference values for these parameters in nestling red kites for use in future translocations or for other clinical purposes. All individuals included in the analysis were clinically healthy at the time of sampling. Biochemical reference intervals were comparable to published values for other Accipitridae, although differences in hematology were noted: PCV was generally lower; and WBC counts higher than (up to triple) those reported for related species of a similar age. It is hypothesized that these differences reflect species variations or the effects of the stress of recent capture on the immune system of the red kites. A Leucocytozoon species was identified on blood smears of six of the red kites. The reference intervals presented in this study are representative of free-living red kite nestlings in England that have recently been captured for conservation translocation purposes.

Immune-mediated hemolytic anemia (IMHA) is defined as an immune-mediated destruction of erythrocytes. Relapses are recognized, but risk factors are poorly defined. We hypothesized that a lower packed cell volume (PCV) on presentation, more transfusions during hospitalization, or a higher total bilirubin would be associated with an increased risk of relapse. IMHA was defined as a PCV less than 30% at diagnosis with two of the following identified: spherocytes, positive Coombs test, elevated total bilirubin, hemoglobinemia, or positive slide agglutination. This was a retrospective study evaluating 163 dogs between January 2005 and December 2019 from one specialty hospital. There were 13 relapses. The probability (95% confidence interval [CI]) of relapse by 3 and 12 mo was 0.05 (0.02-0.13) and 0.11 (0.06-0.22). The probability (95% CI) of relapse by 12 mo in patients who required two or more transfusions was 0.20 (0.09-0.42) compared with 0.07 (0.02-0.19) in patients who did not (P = .191). A lower PCV at diagnosis was not associated with an increased risk of relapse (hazard ratio [95% CI] 0.95 [0.86-1.04], P = .238). A higher total bilirubin was associated with a significantly increased risk of relapse (P = .003). With each increase of 1 mg/dL of total bilirubin, there was a 0.17 (95% CI 0.06-0.28) increase in the probability of relapse. Background: Feeding milk substitutes with low iron content or whole milk without iron supplementation is considered a major factor in developing iron-deficiency anemia in neonatal dairy calves. Young calves are often supplemented with iron dextran injections on the first day of life to prevent anemia. However, the effects of preventive treatment and the presence of disease on serum iron (Fe) concentrations, serum ferritin levels, and hematological blood parameters during the early neonatal stages have not been examined in detail. Therefore, we examined and evaluated the effects of iron dextran injections and health status on the development of hematocrit (Ht), red blood cells (RBC), hemoglobin concentration (Hb), erythrocyte indices (mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration), Fe, and serum ferritin concentrations in dairy calves within the first 10 days of life. The suitability of serum ferritin as a reliable indicator of anemia in very young calves was evaluated by correlating ferritin concentrations with known laboratory diagnostic parameters of anemia., Results: Iron supplementation significantly increased Fe levels (P = 0.048) but did not affect serum ferritin levels in neonatal calves. Fe concentrations were significantly lower in diseased than healthy calves (P = 0.0417). Iron supplementation significantly affected the health status, as observed in Ht (P treat =0.0057; P health =0.0097), RBC (P treat =0.0342; P health =0.0243), and Hb (P treat =0.0170; P health =0.0168). Serum ferritin levels did not significantly correlate with Fe levels. Both groups showed marked differences in ferritin levels, with the highest levels measured on day 2. Fe concentrations showed weak negative correlations with Hb and Ht levels on day 3 (ρ=-0.45; P = 0.0034 and ρ=-0.045; P = 0.0032, respectively). RBC count showed strong positive correlations with Hb and Ht levels (ρ = 0.91 and ρ = 0.93; P < 0.001)., Conclusion: Iron dextran injections increased Fe concentrations but reduced Ht level, RBC count, and Hb level. The presence of diseases led to a reduction in Fe and higher values of Ht, RBC, and Hb in moderate disease than in severe disease. This study evaluated the effect of different supportive treatments on PCV replacement of dairy calves naturally infected with tick fever (TF) agents, and treated with diminazene and enrofloxacin. Five products were tested as supportive treatments in four experiments. In these experiments, we used Girolando female calves (Gyr × Holstein, genetic ratio of 15 / 16 and 31 / 32 Holstein) four to six months old, raised in pasture, naturally infected with TF agents, and infested with R. microplus. Supportive treatment was administered once on day 0 of the study concurrently with specific treatment targeting TF agents. The animals were observed on days 0, 3, and 7. Oral or intravenous administration of a vitamin complex and mineral salts enhanced the increase in PCV and biochemical analytes present in the serum of calves naturally infected with TF agents. No positive effect on PCV values was observed with the administration of (1) invigorating tonic: calcium, casein-peptides and vitamin B12, (2) iron-based stimulant tonic and (3) metabolic tonic: vitamin A, vitamin D, and a fraction of polyunsaturated fatty acids. Supplementation by injection with Type III iron resulted in increased hemoglobin and PCV in treated animals. However, these results did not occur with iron citrate. The aim of this study was to evaluate the serum iron status and its relation to hematological indexes in cyclic mares. Blood samples were taken from 40 Spanish Purebred mares on days - 5, 0, + 5 and + 16 of their cycle. Concentration of transferrin (TRF) was significantly lower on day 0 than on days + 5 and + 16, transferrin saturation (TSAT) decreased significantly on days 0 and + 16 compared to day - 5, total iron-binding capacity (TIBC) on day + 16 was significantly higher than those on days - 5 and 0, and on day + 5 it was also significantly higher than that on day 0, unsaturated iron-binding capacity (UIBC) was reduced on day + 16 compared to days - 5 and 0, red blood cell (RBC) count on day + 16 was higher than that on days - 5 and 0 (p < 0.05), with no differences in the concentration of hemoglobin (HB) and packed cell volume (PCV). TRF and TIBC (r = 0.95), RBC and HB (r = 0.64), RBC and PCV (r = 0.78), and HB and PCV (r = 0.63) were positively and significantly correlated (P < 0.05). The estrous cycle in the Spanish Purebred mare is characterized by an increase in TRF and TIBC during the follicular phase and an increase in TSAT, UIBC and RBC in the luteal phase, without changes in other hematological parameters. The coordinated activity of these parameters guarantees an adequate iron (Fe) transfer and utilization during follicular development, ovulation, and the luteal period in the mare. Therefore, the estrous cycle must be considered in the evaluation of the mare's iron status, in light of significant changes observed both in early and at late luteal phases. The magnitude of these changes and the meaning to the physiology of the mares showed that in cyclic mares, hematological parameters and indicators of iron status evolve differently depending on the phase of the cycle, and their interpretation can help to veterinarians involved in equine practice., (© 2024. The evaluation of hematological and plasma biochemical parameters and the subsequent establishment of reference intervals facilitate the diagnosis of the health status of animals. This work aimed to determine the blood parameters of wild specimens of the stingrays Potamotrygon motoro and Potamotrygon orbignyi from the lower Solimões River region, Amazonas, Brazil. One hundred forty-one stingrays were captured, 92 specimens of P. motoro and 49 of P. orbignyi, of both sexes and at different stages of development. No effect of sex was observed on the blood parameters of juvenile animals for both species. P. motoro neonates presented a distinct hematological and biochemical profile, with significantly lower hematocrit values, hemoglobina, number of erythrocytes, mean corpuscular hemoglobin concentration, monocytes, plasma glucose, total proteins, albumin, and globulin. On the other hand, total cholesterol and urea levels were significantly higher in this same group compared to juveniles of the same species. Comparison between species revealed lower values of triglycerides and total cholesterol in P. orbignyi of both sexes. The results obtained are pioneering for these Amazonian species in white water environments and will serve as a basis for evaluating the health status of wild stingrays. No reference intervals for serum biochemistry and haematology of sea turtles in Thailand exists to assist veterinarians who are responsible for sea turtle health management and treatment. This study determined serum biochemistry and basic haematology of healthy juvenile green sea turtles (n = 92) in captivity in Thailand following the American Society for Veterinary Clinical Pathology (ASVCP), Quality Assurance and Laboratory Standards Committee (QALS) guidelines for the determination of reference intervals in veterinary species. Biochemistry tests, including blood urea nitrogen, creatinine, uric acid, alanine aminotransferase, aspartate aminotransferase and alkaline phosphatase were analysed using an IDEXX VetTest Chemistry Analyzer. Haematology parameters were measured manually using a microhaematocrit for packed cell volume (PCV), Neubauer counting chamber for red blood cell count and cyanmethemoglobin method for haemoglobin concentration. mean corpuscular volume and mean corpuscular haemoglobin concentration were calculated using the PCV, red blood cell count and haemoglobin. Turtles in this study were found to have higher mean values for PCV (28.70%), haemoglobin (92.13 g/L), mean corpuscular haemoglobin concentration (327.03 g/L), uric acid (247.15 μmol/L), alanine aminotransferase (16.53 IU/L), aspartate aminotransferase (209.44 IU/L), and alkaline phosphatase (245.08 IU/L) compared to sea turtles in Brazil. The reference intervals established using high numbers of healthy turtles in this study will assist veterinarians with diagnostic and treatment decisions when evaluating laboratory results for juvenile green sea turtles., (© 2024 The Authors. Parasitic infections with gastrointestinal nematodes are a serious problem for the health and welfare of domestic animals and negatively affect the economics of animal production. Haemonchus contortus is a haematophagous nematode of small ruminants responsible for significant mortality and morbidity. In addition, the widespread resistance to synthetic anthelmintic drugs emphasizes the urgent need of alternative treatment options against haemonchosis. This work aims to investigate the anthelmintic activity of an hydroethanolic Combretum mucronatum leaf extract (CMLE) against Haemonchus contortus in goats. Goats were artificially infected with 3500 third-stage larvae of H. contortus, and 21 days later, treated with CMLE (1000, 500, 250 mg/kg) for 4 consecutive days. Different parameters such as faecal egg count reduction, weight and haematocrit were monitored during the experimental period. The number of eggs per gram of faeces (EPG) was concentration-depended lower and significantly reduced compared to the untreated control (p < 0.0001). The effect of the highest CMLE dose (4 ×1000 mg/kg body weight) was similar to the effect of albendazole (1 ×5 mg/kg of body weight). The ED 50 and ED 90 values calculated were 189.17 and 392.33 mg/kg body weight respectively. ED 50 and ED 90 values were time-dependent. Moreover, CMLE improved haematocrit and weight of goats in dose-dependent and time-dependent manner. Background: Fluid therapy in veterinary medicine is pivotal for treating various conditions in pigs; however, standard solutions, such as Hartmann's solution, may not optimally align with pig physiology. This study explored the development and efficacy of a customized fluid therapy tailored to the ionic concentrations of pig blood, aiming to enhance treatment outcomes and safety in both healthy and diseased pigs., Results: The study involved two experiments: the first to assess the safety and stability of customized fluids in healthy pigs, and the second to evaluate the efficacy in pigs with clinical symptoms of dehydration. In healthy pigs, the administration of customized fluids showed no adverse effects, with slight alterations observed in pO2, hematocrit, and glucose levels in some groups. In symptomatic pigs, the customized fluid group did not show any improvement in clinical symptoms, with no significant changes in blood chemistry or metabolite levels compared to controls. The customized fluid group showed a mild increase in some values after administration, yet within normal physiological ranges. The study reported no significant improvements in clinical or dehydration status, attributing the observed variations in blood test results to the limited sample size and anaesthesia effects rather than fluid characteristics., Conclusions: Customized fluid therapy, tailored to mimic the ionic concentrations of pig blood, appears to be a safe and potentially more effective alternative to conventional solutions such as Hartmann's solution for treating pigs under various health conditions. Objective: To evaluate the effects of darbepoetin on platelet population and reactivity in healthy cats (HCs) and azotemic cats with remnant kidney (RK) model-induced chronic kidney disease., Animals: 12 purpose-bred domestic shorthair cats (n = 6 HCs and n = 6 RK)., Methods: In this pilot study, all cats received darbepoetin (1 µg/kg, SC) on days 0, 7, and 14. Blood was sampled at baseline and on days 3, 10, 15, 17, 20, and 21. At each time point, a CBC was performed, platelet aggregometry was assessed by impedance and optical methods, and platelet P-selectin (CD62P) was quantified before and after thrombin stimulation. Additionally, reticulated platelets were quantified using both thiazole orange staining and proprietary analysis by the CBC analyzer. For RK cats, systemic blood pressure (BP) was serially measured., Results: No adverse effects of darbepoetin were seen. There was no statistically significant change in platelet count between or within groups at any time point. Hematocrit increased significantly over time in the RK but not the HC group. RBC reticulocyte numbers in both groups increased over time. Reticulated platelet percentage did not increase in either group. Differences in platelet reactivity within or between groups were not seen in the aggregometry or flow cytometric assessments. In RK cats, indirect BP did not significantly change during the study., Clinical Relevance: This preliminary investigation did not find evidence that darbepoetin administration impacted platelet number, reactivity, nor reticulated platelet count. Anemic RK cats experienced increased hematocrit and RBC reticulocytes as expected with darbepoetin therapy.

Background: Limited studies are available on vitamin D supplementation in dogs. This study evaluates the effect of a commercial vitamin D3 supplement on serum 25-hydroxy vitamin D as well as selected biochemical and hematological parameters in healthy dogs. Eight intact male adult dogs with a mean body weight of 20 kg from mixed breeds were included in the study. After adaptation period, dogs received vitamin D3 supplement at the dose of 50 IU/kg body weight per day. Blood samples were collected on days 0, 14, 28 and 42 of supplementation. Food was used for analysis of vitamin D3 content., Results: Significant increase in serum level of 25-hydroxy vitamin D3 was detected since day 14 of supplementation. Changes in serum 25-hydroxy vitamin D3 concentration during time showed an upward significance (p < 0.05). Vitamin D3 content of the food was 2900 IU/kg dry matter. Changes in serum phosphorus levels were upward significant. No dog showed calcium or phosphorus levels above the highest reference level. Liver and kidney parameters remained in the reference range during the experiment. A gradual significant increase was observed in hemoglobin and hematocrit which was started from day 14. Vitamin D3 supplementation had no significant effect on neutrophils, monocytes and lymphocytes percent during the study., Conclusions: Vitamin D3 supplementation at 50 IU/kg BW daily, increases serum levels of 25-hydroxy vitamin D in healthy dogs fed with a diet containing proper amount of this vitamin. This study examined the effects of using mushroom mycelium to ferment tigernut and cassava pulp on the growth performance, haematology and immunology of rabbits. Seventy-five New Zealand Bulk grower rabbits were randomly distributed to four treatment groups and a control group in a completely randomized approach. The treatment groups were fed with formulated experimental diets containing one of fermented tigernut drink by-product (FT), fermented cassava sievate (FC), unfermented tigernut drink by-product (UT), or unfermented cassava sievate (UC). The control group was fed a basal diet with no additives. The proximate composition of the fermented feed was analyzed. The weight gain of the animals was, 834.5, 633, 790, 510, and 706 g for control, FT, FC, UT, and UC respectively. The packed cell volume (PCV) for animals in the control group, FT, and FC are 34.33, 37.26, and 32.29% respectively. The red blood cell (RBC) of the FT was favourably improved (5.53 × 10 12 /L) compared to those of UT (2.28 × 10 12 /L), while there was a reduction in the red blood cell count of FC group (1.02 × 10 12 /L). Conclusively, the inclusion of fermented tiger nut drink by-product in rabbit feed improved the PCV and RBC of the rabbits' understudy but did not affect their growth performance., (© 2024. Photoperiod manipulation is emerging as an effective approach for regulating physiological functions in fish. This study aimed to assess the impact of photoperiod on the growth performance, haematological responses, and economic returns of the endangered and highly valued Indian butter catfish, Ompok bimaculatus. Fish with an average body weight of 28.60 ± 4.78 g were randomly placed in six FRP tanks, each measuring 120 × 45 × 60 cm 3 . Each tank contained 20 fish exposed to a light intensity of 1500 lx under different photoperiods [24:0 light: dark (L: D), 15 L: 9D, 12 L: 12D, 9 L: 15D, 0 L: 24D and a natural photoperiod (control)], and fed at a daily rate of 2% of their body weight twice daily for 60 days. The fish in the 15 L: 9D photoperiod exhibited the highest final weight (g), percentage weight gain, specific growth rate (SGR) and survival rate, while the lowest was displayed in 24 L: 0D photoperiod group. The feed conversion ratio (FCR) was at its lowest in the catfish subjected to the 15 L: 9D photoperiod. Regarding haematological parameters, the 15 L: 9D photoperiod group showed higher total erythrocyte count, total leukocyte count, haemoglobin levels, and haematocrit values compared to the other groups. Conversely, the 0 L: 24D group, which experienced prolonged darkness, exhibited the lowest values in these parameters. Moreover, the 24 L: 0D, 9 L: 15D, and 0 L: 24D groups displayed a lower mean corpuscular volume (MCV) but higher mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC) when compared to the control group. The economic analysis revealed that O. bimaculatus reared in a moderate photoperiod (15 L: 9D) displayed better growth, feed utilization, and overall health. Background: Quality control material (QCM) for hematology in veterinary laboratories is limited, and repeat patient testing quality control (RPT-QC) is an alternative method using excess matrix-specific samples., Objectives: This study aimed to determine if median differences between RPT-QC analyses for each time interval for RBC, HGB, HCT, and WBC were the same, determine if unified RPT-QC limits can be applied to a network of veterinary laboratories, compare the performance of RPT-QC to commercial QCM for the reference analyzer and evaluate the experience over a 4 month period and design, improve and implement an automated spreadsheet for RPT-QC data management., Methods: The potential to unify individual analyzer RPT-QC limits for red blood cells (RBC), hematocrit (HCT), hemoglobin (HGB), and white blood cells (WBC) on multi-site Sysmex XT-2000-iV analyzers was explored by a difference of means test and confidence interval determination for the median difference for each network analyzer in comparison to the network reference analyzer. User experience of an automated RPT-QC data management Excel spreadsheet was collected by user feedback during monthly meetings. Numbers of out-of-control results and the root causes for these for RPT-QC were compared against those of a commercial QCM over a 4-month period., Results: Differences between individual analyzer RPT-QC limits were too large to allow for unification of network limits. The automated spreadsheet successfully highlighted out-of-control events for RPT-QC. Trends or shifts were more frequent for commercial QCM based on observed performance and a 1-2.5 s QC rule than for RPT-QC. Following routine troubleshooting, RPT-QC out-of-control events were resolved with an alternative RPT-QC sample indicating random error associated with excessive deterioration. Use of an automated spreadsheet for recording RPT-QC, documentation and troubleshooting of out-of-control events, and collating monthly summary calculations were considered an asset in laboratory quality management., Conclusions: RPT-QC can be successfully implemented and integrated into a multi-site veterinary laboratory. Individual analyzer RPT-QC limit generation is recommended. Background: Repeat patient testing-quality control (RPT-QC) uses retained patient samples as an alternative to commercial quality control material (QCM). We elected to calculate and validate RPT-QC limits for red blood cell count (RBC), hemoglobin (HBG), hematocrit (HCT), and white blood cell count (WBC)., Objectives: (1) To validate RPT-QC across a network of four harmonized Sysmex XT-2000iV hematology analyzers and determine the total error that can be controlled with RPT-QC. (2) To generate quality control (QC) limits using the standard deviation (SD) of the duplicate measurement differences and determine a suitable simple QC rule with a probability of error detection >0.85 and probability of false rejection <0.05. (3) Monitor RPT-QC using sigma metrics as a performance indicator and (4) to challenge RPT-QC to ensure acceptable sensitivity., Methods: Fresh adult canine EDTA samples with results within reference intervals were selected and run again on days 2, 3, and 4. QC limits were generated from the SD of the duplicate measurement differences. The QC limits were challenged using interventions designed to promote unstable system performance. The total error detectable by RPT-QC was determined using EZRULES 3 software., Results: In all, 20-40 data points were needed for RPT-QC calculations and validated using 20 additional data points. The calculated limits differed among the network of analyzers. The total error that could be controlled was the same or better than that of the manufacturer's commercially available quality control material using the same analyzer for all measurands except hematocrit, which required a higher total error goal than that proposed by ASVCP guidelines to achieve an acceptable probability of error detection. The challenges designed to mimic unstable system performance were successfully detected as out-of-control QC., Conclusions: The challenges for RPT-QC resulted in acceptable detection of potential unstable system performance. This initial study demonstrates that RPT-QC limits differ among the network of Sysmex XT-2000iV analyzers, indicating a requirement to customize for the individual analyzer and laboratory conditions. Background: Few studies have been conducted on haematological reference intervals (RIs) in Icelandic horses. Reference intervals have been published for Icelandic horses in Austria and a preliminary study in Iceland compared haematological values in riding horses to published RIs for other breeds as well as Icelandic horses abroad. Haematological parameters can vary greatly due to factors such as breed, gender, age, reproductive status, and training, as well as feeding, prior exercise and management method. Icelandic broodmares are kept on pasture under supervision throughout the year, with haylage provided during the winter, and it is therefore of interest to establish haematological reference intervals for pregnant broodmares in Iceland. The purpose of this study was to establish haematological RIs specific to Icelandic broodmares in the first months of pregnancy, kept on pasture. Blood samples from 183 mares, stabilised in EDTA were analysed using IDEXX ProCyte Dx and total protein was analysed in serum samples from 157 of the 183 mares, using IDEXX Catalyst One analyser. The RIs were established using the guidelines of the American Society for Veterinary Clinical Pathology., Results: The RIs for red blood cell count, haematocrit and haemoglobin were higher in pasture-kept Icelandic mares in early pregnancy, most of which were lactating, than in pregnant mares of other breeds. This was also true for white blood cell count, as well as numbers of monocytes, eosinophils, and basophils, which in some instances might illustrate problems in the automated categorisation of some leukocytes., Conclusions: As no RIs have been published for other pasture-kept Icelandic horses, future investigations should include other groups of pasture-kept Icelandic horses. Background: Ultrafiltration (UF) is an extracorporeal technique for treating fluid overload and is monitored with noninvasive blood volume monitors., Hypothesis/objectives: The primary objective was to determine the accuracy of Crit-Line® III and IV noninvasive blood volume monitors to estimate canine packed cell volume (PCV) and changes in blood volume during UF. A secondary aim was to determine accuracy of targeted ultrafiltration rates (UFR) compared to calculated delivered UFR., Methods: An ex vivo study with a single Phoenix® X36 platform and canine packed red blood cells (pRBC) was performed. Through dilution and UF, clinically applicable PCV values were obtained and compared to Crit-Line® hematocrit (Hct). Blood volume was constantly measured and compared to targeted UFR. Systematic and proportional bias were calculated using a Bayes method., Results: Crit-Line® III and IV reported Hct was significantly lower than PCV (n = 140, median 26%, range, 8%-50%) when PCV was >25% and >30%, respectively. Crit-Line® III and IV calculated change in blood volume (ΔBV%) was significantly different from measured blood volume changes at ΔBV% ±20% and >-20%, respectively. Comparing targeted and delivered UFR (mL/h), less than targeted UFR was removed at UFR100 through UFR400 and UFR0 adding volume., Conclusions and Clinical Importance: Crit-Line® III and IV monitors provide accurate estimates of canine PCV and UF volume change within specific ranges and are useful in monitoring canine UF and hemodialysis. We aimed to validate a targeted selective treatment (TST) methodology for treating parasitic gastrointestinal infections in ewes in different physiological states using parasitological and hematological parameters. Forty ewes were monitored from December 2021 to June 2022 and evaluated during various physiological stages in their life cycle. Before starting the experiment, a fecal egg count (FEC) reduction test was performed to evaluate the efficacy of the anthelmintic (AH) treatment. Weekly assessments were performed based on the Famacha© (F) system and body condition score (BCS), and ewes were subjected to AH treatment when necessary, with their physiological states recorded. Ewes were treated when they presented F ≥ 3, BCS ≤ 2.0 (when F = 2), or submandibular edema. Parasitological, i.e., FEC, and hematological, i.e., hematocrit (Ht), parameters were evaluated monthly to determine the efficiency of the TST methodology. Comparisons between the mean Ht and FEC values in ewes subjected to AH treatment and untreated ewes were performed using analysis of variance, followed by Tukey's test. Spearman's correlation was performed to determine the correlation between the variables, i.e., F scores, BCS, Ht, and FEC. All tests were performed at a significance level of 5 %. During the experimental period, 1138 evaluations were performed. The main reason for AH treatment was F ≥ 3. Ewes in early pregnancy, lactation and late pregnancy received comparatively more AH treatments than the other physiological states. Ewes in late pregnancy and lactation exhibited lower mean Ht values (23.5 % and 22.9 %) and higher mean FEC values (3269 and 1426) compared with those in early pregnancy (30.2 % and 727 EPG). In addition, a statistically significant difference was observed in the Ht and FEC values of ewes that presented submandibular edema (P < 0.001) compared with those that did not exhibit submandibular edema. The genus Haemonchus sp. showed a 96.4 % prevalence in coprocultures. A positive correlation existed between F scores and FEC (r = 0.3819) and a negative correlation between F scores and Ht (r = -0.4728). Ewes that needed AH treatment had lower mean Ht values than ewes that did not need the treatment (19.2 % × 29.3 %; P < 0.001) and higher mean FEC values (8747 × 1163; P < 0.001), confirming that these ewes needed AH treatment. Background: The regenerative response following Babesia rossi infection in dogs is mild, despite severe hemolytic anemia., Objective: We aimed to compare the admission absolute reticulocyte count (ARC) and reticulocyte indices in 103 dogs naturally infected with B. rossi with 10 dogs suffering from immune-mediated hemolytic anemia (IMHA) and 14 healthy control dogs. The regenerative response was also evaluated in five dogs experimentally infected with B. rossi., Methods: This is a retrospective observational study of records generated on the ADVIA 2120 hematology analyzer., Results: The median hematocrits (HCT) of the B. rossi and IMHA groups were significantly lower than the control group (p < .001 for both); however, no differences were seen between the B. rossi and IMHA groups. Compared with the control group, the median ARC was significantly higher in the B. rossi (p = .006) and IMHA (p = .019) groups but significantly lower in the B. rossi group than the IMHA group (p = .041). In the experimentally infected dogs, there was a sudden decrease in the ARC approximately 48 h after the detection of peripheral parasitemia, which was followed by an increase after treatment. Reticulocytes of naturally infected B. rossi dogs were larger, with more variation in cellular volume. The reticulocytes of the experimentally infected dogs decreased in size with decreasing hemoglobin concentrations as the study progressed., Conclusions: The regenerative response in dogs naturally infected with B. rossi is inadequate, given the severity of the anemia observed, and it might be a result of direct suppressive action by the parasite or host response on the bone marrow., (© 2023 The Authors. Background: Packed cell volume (PCV) is important for assessing a patient's health status. Some blood gas analyzers measure hematocrit, and the agreement with PCV varies among different analyzers., Objectives: We aimed to determine the agreement between PCV measured by microcentrifugation and hematocrit measured by the Cobas b121 blood gas analyzer in dogs, cats, and horses., Methods: Whole blood samples for PCV and blood gas analysis were collected in lithium-heparin syringes and analyzed within 10 min of collection. Agreement and association between the PCV and Cobas b121 generated hematocrit were assessed by the Bland-Altman method, Pearson's correlation, Deming regression analysis, and paired t tests. A total allowable error of 10% was used for the analysis., Results: This study included 45 dogs, 45 cats, and 33 horses. The respective mean ± SD (minimum-maximum) of PCVs and hematocrits were: dogs, 34.9 ± 9.9% (9.0-55.0) and 32.5 ± 8.8% (10.4-50.6); cats, 29.0 ± 9.6% (11.0-51.0) and 26.9 ± 9.3% (10.2-50.9); horses, 34.2 ± 6.5% (24.0-47.0) and 34.1 ± 6.0% (22.5-46.1). There were no significant differences between the methods. The bias ± SD was: dogs, -2.4 ± 2.6%; cats, -2.2 ± 2.3%; horses, -0.1 ± 2.4%. Pearson's correlation coefficients were > 0.90 for all species (P < 0.0001). In 60%, 49%, and 85% of the samples for dogs, cats, and horses, respectively, the percentage differences between the methods were within 10%., Conclusions: The Cobas b121 blood gas analyzer provided accurate estimates of PCVs in horses. Association between ovine β-globin polymorphisms and resistance against haemonchosis was described and related to the mechanism of high oxygen affinity β A ➔ β C switch during anaemia, but there are no studies regarding the involved local host responses. Phenotypic parameters and local responses were evaluated in sheep from two β-globin haplotypes naturally infected with Haemonchus contortus. Morada Nova lambs were monitored at 63, 84 and 105 days of age for faecal egg counts and packed cell volume (PCV) under natural infection with H. contortus. At 210 days of age, lambs of Hb-AA and Hb-BB β-globin haplotypes were euthanised, and the fundic region of abomasum was sampled for evaluation of microscopic lesions and relative expression of genes related to immune, mucin and lectin activities. Lambs harbouring the β A allele presented an improved resistance/resilience against clinical haemonchosis, showing higher PCV during infection. Hb-AA animals presented increased eosinophilia in the abomasum compared to Hb-BB animals, accompanied by higher Th2 profile, mucin and lectin activity transcripts, while the inflammatory response was increased in Hb-BB animals. The pirarucu (Arapaima gigas) is a fast-growing Amazonian species of high commercial value. The present study aimed to determine the dietary crude protein (CP) level to promote maximum zootechnical performance for pirarucu fingerlings and as their resistance to Aeromonas hydrophila, as well as evaluate their hematological parameters. Pirarucu fingerlings (2.4 ± 0.08 g, 6.8 ± 0.52 cm) were distributed in 18 tanks (140 L, 40 fish per tank, n = 3) and fed six experimental diets consisting of increasing levels of CP: 300, 400, 450, 500, 550 and 650 g kg -1 in a completely randomized design. Data were submitted to one-way ANOVA (p < 0.05) and the ideal CP level for weight gain was determined using polynomial regression analysis. The dietary CP levels were evaluated using a quadratic polynomial regression and the level of 595 g kg -1 was determined for the best weight gain. The hematocrit of fish fed 300 g kg -1 was higher than in the other groups. No mortalities were observed after the 15-day bacterial challenge; however, number of pirarucu with bacterial damage on the pirarucu caudal fin was higher in the group that was fed the diet with 300 g kg -1 . A dietary protein level of 618 g kg -1 is therefore recommended for providing maximum weight gain and immunological resistance in pirarucu fingerlings weighing 2.4-112.5 g., (© 2023. Aims: To establish a reference range for the canine C-ACT activated clotting time (ACT) test using a water bath and visual clot assessment technique., Methods: Healthy, privately owned dogs (n = 48) were prospectively recruited to the study. Blood samples were collected via direct jugular venipuncture for complete blood count, serum biochemistry analysis and measurement of prothrombin time (PT) and activated partial thromboplastin time (aPTT). Five animals with major abnormalities or who became agitated during phlebotomy were excluded. For the 43 remaining animals, 2 mL of blood was collected via the cephalic vein and added directly to a C-ACT tube that was shaken vigorously before being placed in a water bath at 37°C. Tubes were visually assessed for clot formation and C-ACT was recorded in seconds when the magnet within the tube lodged in the clot., Results: The nonparametric reference interval (capturing the central 95% of the data) was 50-80 seconds, with a 90% CI for the lower limit of 50-55 seconds and a 90% CI for the upper limit of 75-80 seconds. The C-ACT ACT test had a positive correlation with aPTT (0.42; 95% CI = 0.13-0.64). There was no evidence of a correlation between C-ACT ACT and age, weight, PT, haematocrit, white blood cell count, platelet count or total protein., Conclusions and Clinical Relevance: The results of this study suggest that the normal reference interval for ACT in dogs using C-ACT tubes in a 37°C water bath is 50-80 seconds. Care should be taken extrapolating the results of this study to the general population, as the smaller study design had less control for confounders than a larger study. However, when using the described analytical methods, C-ACT tube ACT test results >80 seconds should be considered prolonged in dogs and should prompt further investigation.

The objective of this study was to determine some factors that influence the haematological values of cattle reared in the humid and subhumid tropics of Mexico. Whole blood samples were taken from 1355 crossbred cattle in the years 2017 to 2019. Haematocrit (HTC, %), total plasma protein (TPP, g/dL) and peripheral eosinophils count (PEOS, × 10 3 /µL) were determined manually, and the main haematological variables were recorded with an automatic analyser. The statistical analysis considered as classification variables age, sex, season (cold, dry and rainy), year (2017, 2018 and 2019) and origin of the cattle. The mean of the haematological parameters was determined along with the confidence limits (CL) of the different categories of animals according to age. Calves younger than 1-year-old presented higher levels of HTC, red blood cell count (RBC), haemoglobin (HGB), red blood cell distribution width (RDW), platelet number (PLT), white blood cell count (WBC) and lymphocyte count (LYMF) than animals older than 2 years of age. However, they showed the lowest mean cell volume (MCV) and TPP values. In cows, the highest levels of PEOS, granulocytes (GRAN), MCV and medium cells (MID) were observed and the lowest HTC, RBC, RDW and WBC levels. Intervals were determined with the 1st quartile (Q1) or lower confidence interval (90% CI) as the minimum values and the 3rd quartile (Q3) or upper confidence interval (90% CI) as the maximum values. In human medical practice, a hematological rule of three has been validated for healthy human populations. One such formula is estimating hemoglobin (Hb) levels as 1/3rd of Packed Cell Volume (PCV). However, no such hematological formulae have been devised and validated for veterinary medical practice. The present study was devised with an aim to evaluate the relationship between hemoglobin (Hb) concentration and Packed Cell Volume (PCV) in camels (n = 215) being reared under pastoralism, and to devise a simple pen-side hematological formula for estimation of Hb from PCV. The PCV was determined through microhematocrit method whereas Hb estimation by cyanmethaemoglobin method (HbD). The Hb was also calculated as 1/3rd of PCV and was dubbed as calculated Hb (HbC). Overall HbD and HbC were significantly (P≥0.05) different. Similar results were attained for all study groups i.e. males (n = 94) and females (n = 121), and young (n = 85) and adult (n = 130) camels. The corrected Hb (CHb) was deduced through regression prediction equation attained from linear regression model. Scatterplots were drawn, linear regression was carried out, and Bland Altman chart was built for agreement of both methods of Hb estimation. A non-significant (P≥0.05) difference was noticed between HbD and CHb. Bland Altman agreement analysis revealed satisfactory agreement between HbD and CHb and the data was distributed closely around the mean difference line (Mean = 0.1436, 95% CI = 3.00, -2.72). A simplified pen-side hematological formula for deducing Hb concentration from PCV is accordingly recommended viz. Background: Despite their popularity, hematology reference intervals (RIs) have not been established in big-bellied seahorses (Hippocampus abdominalis)., Objective: The objectives of this study were to establish hematologic RIs to compare values between sex in regard to cytochemical staining of blood cells. We also sought to compare white blood cell concentrations using the Natt and Herrick technique vs blood smear estimates., Methods: Forty-three healthy individuals from the Aquarium du Québec (22 females and 21 males) were included. Normal health status was confirmed by an unremarkable physical examination in five individuals and by necropsy of five other individuals, of which all were excluded from further analyses. Venipuncture was performed from the ventral coccygeal vein in the remaining 33 individuals without anesthesia using heparinized insulin syringes. A blood volume of 0.05 to 0.1 ml was collected to prepare Wright Giemsa-stained blood smears and hematocrits immediately after venipuncture. Whole blood was stored in heparinized Eppendorf tubes to determine red and white blood cell concentrations using the Natt and Herrick technique with a hemocytometer in 10 individuals; these results were compared with blood smear estimates. Additional blood smears were stained with alkaline phosphatase substrate, periodic acid Schiff, and toluidine blue stains., Results: The reference intervals included the packed cell volume (27.4-67.5%), thrombocyte count (19.5-197.7 × 10 9 /L), and white blood cell (WBC) count (2-54.8 × 10 9 /L), including neutrophils (1.1-21.3 × 10 9 /L), lymphocytes (2.7-45.5 × 10 9 /L), and monocytes (0-2.2 × 10 9 /L). The WBC hemocytometer counts showed no correlation with blood smear estimates (Spearman's rho = 0.2). Haemonchus contortus is the most prevalent and important gastrointestinal nematode (GIN) in small ruminants. Since it reduces the packed cell volume (PCV), causing anemia, early diagnosis can be used for targeted selective treatment (TST) of sheep, reducing antiparasitic drug use and anthelmintic resistance. This study aimed to predict PCV values through near-infrared spectroscopy (NIRS) and to develop a classification and diagnosis model of H. contortus infection using PCV values, eggs per gram of feces (EPG) counts and mean daily weight gain (DWG). A total of 1728 spectra were collected from blood samples of 216 lambs with a portable NIR spectroscope. In parallel, other parameters indicative of infection were measured: PCV by hematocrit, FAffa MAlan CHArt (FAMACHA) scores, EPG and DWG. To evaluate the relationship between NIRS spectra and the evaluated parameters, principal component analysis (PCA) was used for an exploratory analysis, regression by the partial least squares method (PLS) for the prediction of PCV values via NIRS, and PCA linear discriminant analysis (PCA-LDA) as a classification model for diagnosis. The absorption peaks in the NIRS region associated with the excitation of overtones of nitrogen-hydrogen (N-H) functional groups of proteins had a strong impact on the principal components (PCs), indicating that blood proteins, especially hemoglobin, can be estimated by the NIRS technique. The model for predicting PCV by PLS presented a standard error of prediction of 2.53, root-mean-square error of 2.48, and coefficient of determination of 0.84, indicating good correlation between the PCV values predicted by the model and the PCV obtained by hematocrit. The PCA-LDA model presented 93.33% sensitivity and 82.18% accuracy, both higher than those of the FAMACHA method, as was expected for resilient Morada Nova lambs. The multivariate models associated with the NIRS technique reported here can be used in the future as a quick and versatile tool for H. contortus infection diagnosis and TST application in lambs., (Copyright © 2023 Australian Society for Parasitology. African swine fever (ASF) is a contagious viral disease that affects pigs of all ages, inducing hemorrhagic fever with high mortality and severe threat to pig production. This study investigated the hematological and serum biochemical abnormalities associated with a natural ASF infection in pigs. A total of 100 serum samples of pigs from piggery suspected of ASFV infection were screened for antibodies by ELISA. Thirty‑two blood samples from serologically positive pigs and 32 negative pigs were undergo to hematological and serum biochemical analyses following standard procedures. The results showed that the mean values of the red blood cell (RBC) count, total white blood cell (TWBC) count, absolute lymphocyte count, absolute monocyte count, serum total protein (TP) and globulin were significantly (p < 0.05) lower while the mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), absolute neutrophil count and serum gamma glutamyl transferase (GGT) were significantly (p < 0.05) higher in the infected than the healthy pigs. There were no significant differences (p > 0.05) in the mean values of the packed cell volume (PCV), hemoglobin concentration, absolute eosinophil count, cholesterol, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities between the infected and healthy pigs. Hence, natural ASFV infection may have caused alterations in the hematological and serum biochemical parameters in the infected pigs. The generated data could complement the existing laboratory diagnostic techniques such as polymerase chain reaction, direct fluorescence antibody test, indirect fluorescent antibody test and ELISA in the diagnosis of ASF in pigs.

The present study has been designed to devise a pen-side hematological formula for estimation of hemoglobin (Hb) from packed cell volume (PCV) in Cholistani breed of cattle being reared under pastoralism in Cholistan desert, Pakistan. It also aims to validate the soundness of rule of calculating Hb concentration as one-third of PCV and vice versa as being used in human medicine. Cholistani cattle (n = 364) were bled for PCV determination (microhematocrit) method and Hb estimation (hematology analyzer) (HbD) as well as through calculation being one-third of PCV (HbC). The independent-sample t-test was implied for deducing difference between HbD and HbC, and between HbD and corrected Hb (CHb) for all study groups. The CHb was deduced through regression prediction equation attained from linear regression model. Scatter-plots were drawn and linear regression was carried out between various studied hematological attributes. Significantly (P ≤ 0.01) positive correlation coefficient was noticed for all study groups being highest for female adult stock (r = 0.893; adjusted R-square = 0.79) between HbD and PCV and between HbD and HbC. The regression equation for overall results attained, i.e., Hb = 0.13 (PCV) + 6.3 was used to deduce CHb. A non-significant (P ≥ 0.05) difference was noticed between HbD and CHb. In conclusion, a convention of human clinical medicine that Hb concentration is third of PCV and vice versa cannot be implied for Cholistani cattle. A different equation, i.e., Hb (g/dL) = 0.13 (PCV) + 6.3 may provide reliable results for Hb estimation from the PCV in cattle., (© 2022. This report describes successful transspecies blood transfusion from a Magellanic horned owl ( Bubo virginianus magellanicus ) to a barn owl ( Tyto alba ). The barn owl was admitted to a wildlife rehabilitation center with severe anemia (packed cell volume [PCV] = 6.7%) from suspected anticoagulant rodenticide poisoning. Procedures performed included patient stabilization, pharmacological treatment, and persistent monitoring following the blood transfusion. The patient's PCV was measured daily, increasing progressively, and attaining a normal value for the species (PCV > 40%) on the eighth day posttransfusion. With no possibility of obtaining a same-species donor and because of the serious condition of the patient and unfavorable prognosis, a decision was made to perform the xenotransfusion. The result was a successful recovery and ultimately the release of the owl into its natural habitat. We concluded that xenotransfusion for avian species should be considered in cases with no possibility of obtaining a homologous donor.

The present work investigated the presence of Trypanosoma vivax in semen and reproductive tissues of experimentally infected cattle and evaluated changes in seminal parameters. Two groups of cattle were established: T01 - experimentally infected with T. vivax (n = 8) and T02 - not experimentally infected with T. vivax (n = 8). After infection, blood (every seven days until 182 days post-infection - DPI), semen (7, 14, 35, 56, 70, 120 and 182 DPI) and reproductive tissue (after euthanasia, 182 DPI) were collected to search for T. vivax using different techniques, including PCR, Woo and Brener. Seminal parameters, including turbulence, motility, concentration, and vigor, were also analyzed. Packed cell volume (PCV) of the animals was determined weekly and weight gain was calculated. The PCR revealed T. vivax DNA in 7/56 semen samples of post-infection T01 cattle. Trypanosoma vivax DNA was detected in the semen of 5/8 animals at 7, 14, 56, 70 and 120 DPI, in the testis of four, and in the epididymis and fat located around the testis of two others. Trypomastigote forms of T. vivax were not found in any semen sample. Sperm of T01 cattle had lower turbulence (p ≤ 0.05) at 7, 14, 35, 56, 120 and 182 DPI, lower vigor (p ≤ 0.05) at 120 DPI and more sperm abnormalities (p ≤ 0.05) than T02. Digital dermatitis was observed among T01 cattle. Animals of T01 had lower PCV values than did those of T02 for most of the evaluations performed and T02 animals gained more weight during the experiment. The results highlight the presence of T. vivax DNA in semen of infected cattle and the importance of this disease for male breeding cattle. Objectives: To investigate and compare the accuracy of two veterinary portable blood glucose metres (AccuTell; AccubioTech; and AlphaTrak2; Abbott Laboratories)., Materials and Methods: One hundred twenty venous blood samples were obtained for immediate whole blood glucose concentration measurement using two portable blood glucose metres designed for use in dogs. Plasma glucose concentration was measured by a laboratory analyser using a hexokinase reaction method. Packed cell volume was measured for each sample. Linear regression analysis was performed and Bland-Altman plots constructed with International Standards Organisation 15197:2013 standards applied to assess relationship and agreement between results of portable blood glucose metres and laboratory analyser methods, respectively. Clarke's error grid analysis was used to assess clinical accuracy and usefulness., Results: The AccuTell and the AlphaTrak2 had mean differences of -0.69 ± 0.70 mmol/L (bias ±sd) and -1.09 ± 1.22 mmol/L (bias ± sd) with the reference analyser, respectively. Eighty-eight of 120 (73.3%) samples for the AccuTell, and 73 of 120 (60.8%) samples for the AlphaTrak2 were plotted in the zone of the error grid analysis that indicated less clinical risk of misdiagnosis of glucose status. Neither device fulfilled the International Standards Organisation 15197:2013 standards for system accuracy. A statistically significant effect of packed cell volume was identified on the difference between blood glucose concentration obtained by the AlphaTrak2 and the hexokinase reaction method, but not for the AccuTell., Clinical Significance: Both devices do not meet International Standards Organisation 15197:2013 standards. Background: Hematocrit (HCT) determination is an integral part of health and disease assessments in captive and wild white rhinoceroses. Several affordable automated hematology analyzers have been developed for in-clinic and field use and have the advantage of being able to measure a large number of additional measurands. However, the accuracy of these analyzers for rhinoceros HCT measurements has not yet been investigated., Objectives: We aimed to compare the HCT results generated by the EPOC portable analyzer system and the Abaxis VetScan HM5 with the gold standard of a manual packed cell volume (PCV) measured using the microhematocrit method., Methods: Hematocrits were measured with the EPOC and the Abaxis VetScan HM5 (bovine setting) and compared with the PCVs of 69 white rhinoceros whole blood samples. Results were compared using Bland-Altman difference plots and Passing-Bablok regression analysis. A total allowable analytical error of 10% was set as the performance goal., Results: A significant positive bias, with a mean of 7.7% for the EPOC and 17.9% for the Abaxis, was found compared with the manual PCV method., Conclusions: The allowable error goal of 10% was not exceeded with the EPOC analyzer. Although not analytically equivalent to the gold standard, the EPOC results could therefore be used as approximations in critical situations where manual measurements cannot be performed. The Abaxis exceeded this allowable error and overestimated HCTs in rhinoceroses. Therefore, method-specific reference intervals should be used., (© 2022 The Authors. Background: The validity of hematologic data is crucial for correctly diagnosing certain conditions and providing accurate research results. However, delays in blood testing are inevitable., Objective: This study aimed to determine whether prolonged storage at different temperatures affects CBC measurand stability in rabbit whole blood using the impedance-based Scil Vet ABC hematology analyzer., Methods: Hematologic tests were performed on whole blood samples collected from 20 New Zealand white rabbits at <1 hour post-blood collection as the baseline and at 6, 24, 48, and 72 hours post-blood collection, with aliquots stored at 21 and 4°C. CBC measurand stability was determined by comparing the obtained values at each time point to the baseline value. Statistically significant variations were compared with current quality specifications to determine if a meaningful bias existed., Results: Hematocrit, mean corpuscular hemoglobin, and mean corpuscular hemoglobin concentration remained stable for 72 hours in blood stored at 4°C. No meaningful bias was observed for mean corpuscular volume after 48 and 72 hours of storage at 4°C. Only the white blood cell count was stable for 72 hours at 21°C. The platelet count, hemoglobin concentration, and mean platelet volume were not stable at 6 hours post-blood collection at either temperature., Conclusions: The results of this study showed that the hematocrit can be tested for 72 hours in blood stored at 4°C, without a significant change from the baseline value. Whole blood biochemistry and blood gas analysis are uncommonly used in poultry, but their use could improve the diagnosis of certain diseases or aid in monitoring flock health. To create preliminary reference intervals for selected blood analytes in broilers using the i-STAT and Vetscan VS2 (VS2) portable analyzers, we tested broilers at 7, 21, and 35 days of age. A total of 134 venous blood samples from healthy chickens of two different flocks were analyzed. There were significant age-related increases in concentration for glucose, hematocrit, ionized calcium, sodium, and carbon dioxide partial pressure on the i-STAT and for aspartate aminotransferase, creatine kinase, total calcium, phosphorus, and total protein on the VS2. Conversely, significant decreases in concentration were observed for pH, oxygen partial pressure, oxygen saturation on the i-STAT and for uric acid and albumin on the VS2. Additionally, significant differences were found on some blood parameters among the two flocks. Extremely high CK values were found on broilers after 21 days of age, indicating a possible degree of muscle injury during the grow-out. Preliminary reference intervals for all the analytes at each of the age groups were obtained. This study's data provide a starting point for the interpretation of blood analysis in broiler chickens at different ages and offer a new approach to investigate certain metabolic diseases that affect commercial poultry.

There is a large population of donkeys in Saint Kitts; however, hematological and biochemical reference intervals (RIs) are lacking. This study addressed this deficiency by following the American Society for Veterinary Clinical Pathology RI guidelines. Sixty-six healthy, gelding standard donkeys with a median and interquartile range age of 5 years (3.5 - 8 years) and a mean ± standard deviation body weighed of 156 ± 16.7 kg were used to produce a five-part differential complete blood count using an impedance-based analyzer. Clinical chemistry analytes were quantified using a photometric-based analyzer utilizing two reagent rotors that determined 14 and 11 analytes, respectively. An electrochemical-based analyzer quantified chloride, sodium and potassium. Reference intervals were computed using Reference Value Advisor. Results of analytes determined using different rotors/analyzers were assessed using Passing-Bablok regression and Bland-Altman plot analyses. Reference intervals for 43 hematological and biochemical analytes were generated. Reference intervals for hematocrit, red blood cells, white blood cells, total protein, glucose, blood urea nitrogen, and creatinine were 23.67% - 38.08%, 4.08 - 6.42 10 12 /L, 4.7 - 12.34 10 9 /L, 5.84 - 6.93 g/dL, 64.7 - 130.9 mg/dL, 11.1 - 13.4 mg/dL, and 0.67 - 1.36 mg/dL, respectively. There was good agreement between detection system for albumin, aspartate aminotransferase, gamma glutamyl transferase, total protein, globulin, and potassium, but not for blood urea nitrogen, calcium, creatinine kinase, and sodium. This study is the first to establish hematological and biochemical RIs in donkeys in Saint Kitts. Objectives: The aim of this study was to compare the characteristics of fresh and stored feline red blood cells (RBCs) after passage through an 18 μm microaggregate filter., Methods: Nine cats were recruited for a single blood donation using an open collection system. A simulated transfusion using a syringe driver and microaggregate filter was performed over 2 h with half the blood on the day of donation and the other half after 35 days of storage. Differences in haematological parameters, haemolysis percentage and osmotic fragility (OF) were compared on the day of donation pre-filter passage (D0-) vs day of donation post-filter (D0+) or day 35 storage pre-filter (D35-) and post-filter (D35+). Blood was cultured at D0+ and D35+., Results: There were no statistically significant differences in the D0- vs D0+ comparisons. There were statistically significant ( P <0.05) increases in haemolysis percentage, red cell distribution width (RDW) percentage and mean OF, and decreases in packed cell volume (PCV), RBC count, haemoglobin and haematocrit for D0- vs D35-. The same was found for D0- vs D35+ with the addition of a significant increase in mean cell haemoglobin (MCH). For D35- vs D35+ only MCH significantly increased. At day 35, 6/9 units had haemolysis percentages that exceeded 1%. This increased to 8/9 of stored units post-filter passage. All blood units cultured negative., Conclusions and Relevance: Fresh RBCs exhibited no in vitro evidence of injury following passage through an 18 μm microaggregate filter. Increased MCH was observed in the stored blood and may represent haemolysis induced by the filter. All other changes can be explained by storage lesion rather than filter passage. The findings highlight the importance of blood banking quality controls and the need for further research to assess the effects of transfusion technique, specifically filter passage, on storage lesion-affected feline blood.

The Spix's Saddleback Tamarin, Leontocebus fuscicollis is widely distributed across the Amazon region, but is endangered. This species is serving an important role in biomedical research in captivity. However, reference values for hematological and biochemical parameters are required for the proper characterization of the species. It was therefore the objective of our research to establish these parameters taking into consideration sex and body mass differences in healthy adult Spix's saddleback tamarins. Collecting 2 mL of blood from each individual, 20 animals were examined (7 males, 13 females), and hematological and biochemical parameters were determined using commercial kits. Of the sixteen variables measured, only red blood cell (RBC), hemoglobin (Hb) and hematocrit (Ht) values were significantly higher in males (7.12 ± 0.98 106/mm, 14.98 ± 1.25 g/dL and 48.71 ± 4.91%, respectively), while red cell distribution width (RDW) was higher in females (14.58 ± 1.89%). Of the biochemical parameters measured, only gamma-glutamyl transferase (GGT) enzyme showed higher activity in females (8.08 ± 4.87 U/L), and a high glucose concentration range was observed (102.0 to 521.0 mg/dL) for both sexes. These parameters established with reference ranges for healthy adults provide a reliable reference source for the interpretation of laboratory housed saddleback tamarin.

Background: A point-of-care device that can provide immediate and reliable hemoglobin (Hb) concentrations and packed cell volumes (PCVs) would be useful in veterinary medicine., Objectives: We aimed to compare the use of a human device (Mission Plus; MP) with a gold standard (GS) method for measuring Hb concentrations and PCVs in cattle blood., Methods: Blood samples from clinically healthy cattle (n = 122) were collected with or without an anticoagulant (K 2 EDTA). The GS and MP methods were compared with correlation coefficients. Passing-Bablok regression analyses were also performed, and the acceptability judgment was completed using Bland-Altman plots., Results: The CV max for Hb values obtained using the GS method, the MP device without K 2 EDTA, and the MP device with K 2 EDTA were approximately 2.70%, 1.70%, and 2.0%, respectively, whereas the CV max for PCVs was 0.90%, 1.83%, and 2.05%, respectively. A positive correlation (97.5% confidence interval) was observed between the Hb concentrations and PCV values detected using the MP and GS techniques in blood with and without K 2 EDTA. Bland-Altman plots showed agreement between the MP and GS methods. For Hb using blood collected with or without the addition of K 2 EDTA, the mean differences were -0.87 g/dL (95% CI: 1.35; -3.96) and 0.08 g/dL (95% CI: 2.16, -1.99), respectively. For PCVs using blood collected with or without the addition of K 2 EDTA, the mean differences were -3.75% (95% CI: 0.61. -8.12) and -0.88% (95% CI: 2.86, -4.62), respectively., Conclusions: The MP device can be used to analyze Hb concentrations and PCVs in bovine blood to assist in field diagnoses., (© 2021 American Society for Veterinary Clinical Pathology.)

Objectives: Haemolysis caused by the use of peristaltic infusion pumps (PIPs) has been described in human and canine packed red blood cells (pRBCs). The aim of this study was to evaluate the effects of two different linear PIPs on the haemolysis of feline pRBC units stored for a long time., Methods: Feline pRBC units stored with adenine, dextrose, mannitol and sodium chloride (SAGM) were manufactured. After 35-42 days of storage at 2-4°C, a line administration system with a 180 µm filter was attached to every pRBC bag, the system was drained by gravity alone (8 drops/min) and a 1.3 ml sample was collected (G). A NIKI V4 pump was then used at a flow rate of 25 ml/h, the flow was stopped when the infusion system was filled with blood coming from the infusion pump and another 1.3 ml sample was collected (NK). Finally, an Infusomat FmS pump was evaluated, collecting another 1.3 ml sample (IM). Packed cell volume (PCV) was measured in all samples by microhaematocrit centrifugation, total haemoglobin (HGB) was measured using a specific haemoglobin analyser and, after centrifugation, free HGB was determined by spectrophotometry. The percentage of haemolysis was calculated. Friedman's test was used to compare the samples., Results: Fifteen feline pRBC units were evaluated. The average degree of haemolysis for sample G (gravity-assisted) was 1.12%. Comparison of the degree of gravity-assisted haemolysis with haemolysis in PIP NK (1.13%) and IM (1.14%) samples revealed no significant differences, with differences of only 0.01% and 0.02%, respectively., Conclusions and Relevance: The results of this study demonstrate that the use of two common PIPs in veterinary hospitals does not produce levels of haemolysis that are significantly different than that caused by gravity alone during transfusion of feline pRBCs at a rate of 25 ml/h.

Trypanosoma brucei is one of the most pathogenic species of the genus Trypanosoma, and T. brucei brucei is one of the subspecies that is of great economic concern to animals. A large range of labouratory animals are commonly used in Trypanosoma studies. This study is aimed at exploring the possibility of using guinea fowls as experimental models for future studies and preservation of T. b. brucei. In achieving our aim, we studied the infectivity and pathogenicity of T. b. brucei in guinea fowls in relation to rabbits. The level of parasitaemia, mean body weight, mean temperature, haematological and histopathological parameters were accessed. Ten each of rabbits (Oryctolagus cuniculus) (control model) and guinea fowls (Numidia meleagris) (study model) (5 in the uninfected groups and 5 in the infected groups) were used for this study. The infected rabbits were inoculated intraperitoneally, while the infected guinea fowls were inoculated through the wing veins. Both animals were inoculated with 0.20 ml of T. b. brucei-infected blood estimated to be 1× 10 6 parasites/ ml. The infected rabbits and guinea fowls were screened daily for the presence of T. b. brucei using the haematocrit centrifugation technique (HCT). The mean weight, mean temperature and haematological parameters were accessed weekly, while the histopathological parameters were accessed at the end of the study. Trypanosoma b. brucei was detected in the blood of infected rabbits about 8 days post-infection, while there was no parasitaemia in the infected guinea fowls. The haemoflagellate exerted a significant (p < 0.05) effect on the mean body weight, mean temperature and haematological parameters of rabbits compared to guinea fowls. The pathological effects of T. b. brucei infection was seen in the liver and kidney of infected rabbits, and in the spleen of infected guinea fowls. There appears to be no successful multiplication and proliferation of T. b. brucei in the guinea fowls, making it not to be a suitable animal model for experimental studies and preservation of T. b. brucei., (Copyright © 2021 Elsevier B.V. Hematological parameters play a key role in assessing the disease states and physiological status of birds, however the reliable information of hematological reference values for Lohmann silver in the pre- and post-laying periods is limited. Thus, the aim of this study was to research the effect of age on hematological parameters and provide reliable reference intervals for commercial Lohmann silver layer. Blood samples were obtained from 231 laying hens at different ages (5-37 wk) with 4-wk sampling intervals. A total of 12 hematological parameters including leucocytes (WBC, 109/L), red blood cell (RBC,10 12 /L), hemoglobin (HGB, g/L), hematocrit (HCT,%), mean corpuscular volume (MCV, fL), mean corpuscular hemoglobin (MCH, pg), mean corpuscular hemoglobin concentration (MCHC, g/L), red cell distribution width (RDW,%), platelet count (PLT,109/L), mean platelet volume (MPV, fL), platelet volume distribution width (PDW), plateletcrit (PCT, fL) were determined with a reference value advisor. One-ANOVA and Tukey test were utilized to test for significant differences of each parameter between ages. Our findings showed that these parameters were significantly affected by age, except for HCT in the pre-laying period. And reference intervals for all parameters except MCH, MCHC, MCV, MPV, and PDW in the pre-laying period were higher than that in the post-laying period. In this study, it was aimed to investigate the effect of hip dysplasia on some biochemical parameters, oxidative stress factors and hematocrit values in dogs. Hematocrit values (HTC), serum calcium (Ca), phosphorus (P) levels, serum alkaline phosphatase (ALP), creatine kinase (CK) activities and oxidative stress factors were evaluated in a total of 27 dogs with healthy hip joints (n: 11) and hip dysplasia (n: 16). There was no statistically significant difference between the two groups in terms of HCT, Ca and P values (p˃0.05). ALP and CK activities were found to be statistically significantly increased in the group with hip dysplasia compared to the control group with a healthy hip joint (p˂0.05). While malondialdehyde (MDA) level, one of the oxidative stress factors, was increased in the group with hip dysplasia, decreased glutathione (GSH) levels, catalase (CAT) and glutathione peroxidase (GSH-Px) activities were significantly decreased. There was no significant difference between the two groups in terms of superoxide dismutase (SOD) level. Packed cell volume (PCV) is commonly used to assess and monitor red blood cell count in animals, but the results can be altered if inappropriate ratios of anticoagulant/blood are used. The purpose of this study was to determine the effect of ideally filled, overfilled, and underfilled K3 ethylenediaminetetraacetic acid (EDTA) tubes with various volumes of healthy dog blood on centrifuged PCV. Six milliliters of blood was obtained from 94 blood donors each. Initial distribution was injected into two nonheparinized microhematocrit tubes. The remainder was instilled into 1.3 mL K3 EDTA spray-dried tubes as 1.5 mL, 1.3 mL, 0.75 mL, 0.5 mL, and 0.25 mL aliquots. Normality was determined using the D'agostino-Pearson method and by visual examination of histograms. Data were analyzed using a repeated-measures analysis of variance with post hoc testing using Tukey's test. There is a statistically significant decrease in the PCV between all groups with progressive underfilling of tubes (P < .0001). The closest difference is between 1.5 and 1.3 mL (P = .0138). Our study suggested that underfilling K3 EDTA tubes significantly and negatively influences the PCV in healthy dogs. Background: The hematocrit (Hct) or packed cell volume (PCV) reflects the blood volume occupied by red blood cells. The development of point-of-care (PoC) instruments can accelerate the ease of measuring Hct/PCV compared with traditional capillary centrifugation (TCC) methods. However, no studies have compared Hct/PCV levels in cattle at high elevation with other measurement methods., Objectives: In this study, we aimed to compare methods to estimate Hcts/PCVs of rangeland cattle at high elevation. We specifically wanted to determine if Hct/PCV levels measured with a commercial PoC instrument (i-Stat with CHEM8+ cartridges [PoC i ]) were comparable to Hct/PCV levels measured with traditional laboratory methods., Methods: We assessed the Hct/PCV of 94 mature beef cattle (black Angus; Bos taurus) at ~2195 m above sea level using paired analyses of the PoC i and TCC methods from each animal. We used paired samples t-tests to compare mean Hct/PCVs. Correlation analyses relative to the line of identity and Passing-Bablok regression were used to assess systematic and proportional differences, respectively, and Bland-Altman plots were used to assess agreement between the two methods., Results: The PoC i estimated a Hct of 28.2% ± 0.7% (SE), which was lower than the TCC estimated PCV of 39.2% ± 0.5%. The Bland-Altman plot revealed poor agreement between the two methods in addition to a -11% bias for the PoC i .
