1. Production of Esters in Escherichia coli Using Citrate Synthase Variants

Abstract

Acetate esters comprise a wide range of products including fragrances and industrial solvents. Biosynthesis of esters offers a promising alternative to chemical synthesis because such routes use renewable carbohydrate resources and minimize the generation of waste. One biochemical method for ester formation relies on the ATF1 gene from Saccharomyces cerevisiae, which encodes alcohol-O-acyltransferase (AAT) which converts acetyl-CoA and an exogenously supplied alcohol into the ester. In this study, the formation of several acetate esters via AAT was examined in Escherichia coli chromosomally expressing citrate synthase variants, which create a metabolic bottleneck at acetyl-CoA. In shake flask cultures, variant strains generated more acetate esters than the strains expressing the wild-type citrate synthase. In a controlled bioreactor, E. coli GltA[A267T] generated 3.9 g propyl acetate in 13 h, corresponding to a yield of 0.155 g propyl acetate/g glucose, which is 18% greater than that obtained by the wild-type GltA control. These results demonstrate the ability of citrate synthase variants to redistribute carbon from central metabolism into acetyl-CoA-derived biochemicals.

Abstract

G quadruplexes (G4) are higher-order DNA and RNA secondary structures formed by G-rich sequences that are built around tetrads of hydrogen-bonded guanine bases. Potential G4 quadruplex sequences have been identified in G-rich eukaryotic non-telomeric and telomeric genomic regions. Upon function, G4 formation is known to involve in chromatin remodeling, gene regulation and has been associated with genomic instability, genetic diseases and cancer progression. The natural role and biological validation of G4 structures is starting to be explored, and is of particular interest for the therapeutic interventions for human diseases. However, the existence and physiological role of G4 DNA and G4 RNA in plants species have not been much investigated yet and therefore, is of great interest for the development of improved crop varieties for sustainable agriculture. In this context, several recent studies suggests that these highly diverse G4 structures in plants can be employed to regulate expression of genes involved in several pathophysiological conditions including stress response to biotic and abiotic stresses as well as DNA damage. In the current review, we summarize the recent findings regarding the emerging functional significance of G4 structures in plants and discuss their potential value in the development of improved crop varieties.

Abstract

Global interest in lignocellulosic biorefineries has increased in the recent past due to technological advancements in sustainable and cost-effective production of numerous commodity and speciality chemicals and fuels from renewable lignocellulosic biomass (LCB). As a result, the market value of biorefinery products has also increased over the time, with an estimated worth of USD 867.7 billion by 2025. However, biorefinery operations, especially enzymatic hydrolysis, suffer from many challenges that limits the cost-effectiveness of conversion of LCB. Therefore, it is essential to understand and address these challenges in future biorefineries. The paper focuses on recent trends and challenges in enzymatic hydrolysis of LCB during lignocellulosic biorefinery operation for greener synthesis of energy, fuels, chemicals and other high-value products. Insights into the gaps in knowledge and technological challenges have also been addressed together with focus on future research needs and perspectives of enzymatic hydrolysis of LCB for biorefinery applications.

Abstract

Nutraceuticals are the food ingredients which have a proven beneficial effect on human health. These include low calories sugars, proteins and vitamins B complex, etc. Microorganisms, such as Lactococcus lactis, are ideal microbial cell factories for the production of these nutraceuticals. Developments in the genetic engineering of food-grade microorganisms have been very helpful for enhanced production or overexpression of nutraceuticals. This chapter describes the use of food grade microorganisms in industrial production of nutraceuticals. The main emphasis is on industrial production of these beneficial nutraceuticals by food grade microorganism. The diversity of microbial cell types, various approaches for improved nutraceutical production through process optimization as well as strain improvement of the producing microorganisms are discussed.

Abstract

Filamentous fungi are the predominant microorganisms used to produce various industrially important products including enzymes, organic acids, etc. Thermophilic fungi have the to thrive between 45 and 55 °C and are particularly advantageous for the large-scale production of several compounds by fermentation. One such well-studied thermophilic fungi is Myceliophthora thermophila, which has strong ability to degrade biomass during its optimal growth at 45–55 °C. It produces a repertoire of biomass hydrolyzing enzymes which find many applications in biotechnology due to their higher stability and better catalytic rates at elevated temperatures during the process. In recent times, this fungus has been improved through metabolic engineering and other strain improvement strategies. The genome sequence data of M. thermophila has further helped in better understanding of its physiology for industrial exploitation. This chapter provides insights on bioprospects of M. thermophila supported by recent developments.

Abstract

Municipal solid waste (MSW) is produced globally in huge quantities and generates problems associated with its processing and final disposal in environmentally friendly and economical ways. Its judicious use, on the other hand, keeping in mind the nutrient and energy content, opens up many avenues for utilization as an economic, abundant, and sustainable source for generating renewable energy, value-added products, and chemicals in a waste biorefinery-based approach. The main aim of this chapter is to provide up-to-date information related to MSW biorefinery in a global perspective. The biotechnological advancements in MSW to energy conversion processes via biochemical route are discussed with an overview of the developed technologies. The challenges faced by current MSW biorefining technologies and their potential solutions are also highlighted in this chapter, with a focus on prospects of MSW biorefinery in the sustainable development of bioenergy.

Abstract

In the present study, adsorption isotherms, kinetics and thermodynamics of fermentation inhibitors generated during the acid hydrolysis of corncob were investigated using activated charcoal (0.5–2.0% w/v) as the adsorbent. The results indicated that lower pH and lower temperature favor the adsorption of both furans and phenolics. The adsorption kinetic regimes were found more favorable in pseudo second order kinetics than pseudo first order kinetics. The experimental values were in good correlation with the predicted values with a regression coefficient (R2) value of more than 0.99. Moreover, the negative values of enthalpy (ΔH), and free energy (ΔG) indicated that the adsorption process was spontaneous in nature and exothermic. The detoxified corncob hydrolysate on its assessment for fermentability revealed the production of 4.8 g/L and 4.3 g/L xylitol from Candida tropicalis ITCC 1830 and C. tropicalis MTCC 9038. The activated charcoal adsorption has shown differential adsorption capacity and isotherms on various inhibitors under different conditions. Hence using the optimum conditions the process efficiency can be improved to make the whole process cost competitive.

Abstract

BACKGROUND Cotton stalk (CS) was deconstructed by sequential dilute acid‐alkali pretreatment (DALP) using sulfuric acid (1%, v/v) and sodium hydroxide (3%, w/v). The enzymatic hydrolysis of DALP‐CS by cellulase was optimized by Taguchi method, employing factors like solid loading, enzyme dose, tween 80, and pH as main process variables. For bioethanol production, pretreated CS was subjected to simultaneous saccharification and fermentation (SSF) using a newly identified yeast strain Kluyveromyces marxianus JKSG‐6. RESULTS: Sequential pretreatment of CS resulted in 66% and 42% removal of hemicellulose and lignin, respectively. The same was evident from the changes in the fourier transforming infrared (FT‐IR) spectroscopy absorption spectrum. Under optimized conditions, the enzymatic hydrolysis of DALP‐CS released 375.50 ± 0.04 mg g−1 of glucose. Further, by employing K. marxianus JKSG‐6 under SSF at 42 °C, the strain produced an ethanol titer of 26.10 ± 0.05 g L−1 from pretreated CS. CONCLUSION: Sequential dilute acid‐alkali pretreatment rendered CS, a highly recalcitrant LCB feedstock for bioethanol, more accessible for hydrolysis by cellulase. SSF of the effectively pretreated CS was successfully carried out for bioethanol production by employing a newly identified yeast K. marxianus JKSG‐6. © 2021 Society of Chemical Industry (SCI). Abstract

Cellulases are an important category of enzymes, comprising three enzyme activities, namely endoglucanase, cellobiohydrolase, and β-glucosidase. Cellulase has numerous applications in different industrial sectors, including agriculture, textiles, food, biofuels, biorefineries, and bioconversions. Cellulase production for biofuels and the biorefinery industry has received a major focus in recent decades and the market for cellulase is expected to grow further. To meet the worldwide demands for the upcoming biorefinery and biofuel plants, major breakthroughs in cellulase production technologies are needed. This chapter starts with an introduction to the structure of the substrate of cellulases, that is, cellulose, and mechanism of cellulase action. The chapter mainly deals with advancements in two cellulase production technologies: solid-state and submerged fermentation. Both these methodologies are compared on the basis of technologies, bioreactors used, advantages, and disadvantages. Toward the end of this chapter, recent developments in cellulase production technologies are discussed.

Abstract

Rhizosphere had a complex dynamic ecological relationship of plant and microbe. They are involved in the secretion of wide array of chemicals that can be classified as signaling compounds, antipathogenic compounds, growth regulators, and nutrient solubilizers. Thus, unraveling these interactions and the chemicals involved in these bilateral signaling can enable us to have a sustainable agriculture. Recently, ultrahigh-throughput techniques offered the potential to explore these interactions in holistic manner. Transcriptomics and metabolomics have led to novel investigations on plant development and stress responses. Additionally, these techniques had changed scenario of PGPR and biocontrol studies that previously involved single-aspect studies through traditional techniques. In this chapter, we summarize the aspects of rhizosphere metabolite studies including their potential challenges and opportunities.

Abstract

Cellulase is a complex of three enzymes that work synergistically to hydrolyze native cellulose, these are endoglucanases (EC 3.2.1.4), exoglucanases (cellobiohydrolases EC 3.2.1.91), and β-glucosidases (EC 3.2.1.21). A large number of microorganisms, such as bacteria, actinomycetes, and fungi, are known to degrade cellulose. Cellulolytic enzymes from soft rot and white rot fungi have been studied in model fungi such as Trichoderma viride and Phanerochaete chrysosporium, respectively. Cellulase has major application in the bioconversion of lignocellulosic materials. In the recent years apart from saccharification of lignocellulose, there has been an increasing interest in the production of beverages using cereal blends based upon a complex endogenous enzymology occurring during the malting of grain, mashing of grist, and fermentation, which can be connected way back historically. The present chapter discuss the use of this enzyme cellulase in brewing industry specially focussing on the methodologies and its role in beer and wine making.

Abstract

In the present study, adsorption isotherms, kinetics and thermodynamics of fermentation inhibitors generated during the acid hydrolysis of corncob were investigated using activated charcoal (0.5–2.0% w/v) as the adsorbent. The results indicated that lower pH and lower temperature favor the adsorption of both furans and phenolics. The adsorption kinetic regimes were found more favorable in pseudo second order kinetics than pseudo first order kinetics. The experimental values were in good correlation with the predicted values with a regression coefficient (R2) value of more than 0.99. Moreover, the negative values of enthalpy (ΔH), and free energy (ΔG) indicated that the adsorption process was spontaneous in nature and exothermic. The detoxified corncob hydrolysate on its assessment for fermentability revealed the production of 4.8 g/L and 4.3 g/L xylitol from Candida tropicalis ITCC 1830 and C. tropicalis MTCC 9038. The activated charcoal adsorption has shown differential adsorption capacity and isotherms on various inhibitors under different conditions. Abstract

G quadruplexes (G4) are higher-order DNA and RNA secondary structures formed by G-rich sequences that are built around tetrads of hydrogen-bonded guanine bases. Potential G4 quadruplex sequences have been identified in G-rich eukaryotic non-telomeric and telomeric genomic regions. Upon function, G4 formation is known to involve in chromatin remodeling, gene regulation and has been associated with genomic instability, genetic diseases and cancer progression. The natural role and biological validation of G4 structures is starting to be explored, and is of particular interest for the therapeutic interventions for human diseases. However, the existence and physiological role of G4 DNA and G4 RNA in plants species have not been much investigated yet and therefore, is of great interest for the development of improved crop varieties for sustainable agriculture. In this context, several recent studies suggests that these highly diverse G4 structures in plants can be employed to regulate expression of genes involved in several pathophysiological conditions including stress response to biotic and abiotic stresses as well as DNA damage. In the current review, we summarize the recent findings regarding the emerging functional significance of G4 structures in plants and discuss their potential value in the development of improved crop varieties.
