Temperate mountain forests have experienced an increase in frequency and severity of natural disturbances (e.g., droughts, fires, windstorms and insect outbreaks) in recent decades due to climate and environmental change. Outbreaks of bark beetles have caused significant dieback of conifer forests in Central Europe and it is essential to model and predict the potential severity of future bark beetle outbreaks. However, to predict future bark beetle activity, historical baseline information is required to contextualize the magnitude of current and potential future outbreaks. A fossil beetle record from a forest hollow in the Tatra Mountains, Slovakia; one of the best-preserved national parks in Central Europe, was produced to identify insect outbreaks during the last millennia. Sub-fossil bark beetle remains were compared with parallel pollen and charcoal to assess whether peaks in conifer bark beetle remains correspond with indications of disturbance documented in historical or sedimentary fossil records. Three peaks in bark beetle remains were detected (1) post-2004, (2) AD 1140–1440, and (3) AD 930–1030. The abundance of species Pityogenes chalcographus and Pityophthorus pityographus in the two top samples can be linked directly to large bark beetle outbreaks in the High Tatra Mountains after 2004. P. chalcographus and P. pityographus are also the abundant species in the second peak (AD 1140–1440) while the third peak (AD 930–1030) consists of the species Polygraphus poligraphus. The most prominent conifer bark beetle in Central Europe, Ips typographus, was found to be present in most of the samples but always at very low numbers. It is plausible that P. chalcographus and P. pityographus fossils might be useful proxies for past conifer bark beetle outbreaks in Central Europe, as they occur together with fossils of I. typographus but appear to be well-preserved. A significant correlation was found between primary bark beetles and macroscopic charcoal densities in the sediment, highlighting the complex interactions between disturbance agents, bark beetles and fire, in this long-term regime of natural disturbances. Our 1400-year disturbance record shows how bark beetle outbreaks have been an important component of the regional natural disturbance regime for over a millennium and have intensified with increasing anthropogenic activity. Bark beetle outbreaks are likely one of the drivers promoting the future ecological stability of the temperate conifer ecosystem over decades to centuries.

AimsReconstruction of the Holocene diversity changes in a biogeographically complex region. Description of major diversity patterns, testing their predictors, and their interpretation in the palaeoecological and biogeographical context. Testing the assumption that pollen record is informative with respect to plant diversity in our study area.MethodsFossil pollen extracted from 18 high-quality profiles was used as a proxy of past plant diversity. Pollen counts of tree taxa were corrected by pollen productivity, and pollen assemblages were resampled to 100 grains per sample and 150 grains per 500-years time window. SiZer analysis was used to test and visualize multi-scale diversity patterns. Linear modelling was used to identify the best predictors. SiZer maps and pollen composition were analysed using non-metric multidimensional scaling. K-means clustering and indicator species analysis were used to interpret ordination results.ResultsMean Holocene plant diversity is significantly predicted by latitude, while its temporal pattern followed the biogeographical region and elevation. Major differences were found between the Mesic and Montane Hercynia (lower diversity, increasing only in the Late Holocene) and Pannonia, the Carpathians and Warm Hercynia (higher diversity, increasing from the Early or Middle Holocene on). Low diversity in the Middle and Late Holocene is associated with the prevalence of woody and acidophilic taxa. High diversity is associated with numerous grassland and minerotrophic wetland taxa, crops and weeds. Fossil-modern pollen diversity and modern pollen-plant diversity show significant positive relationships.ConclusionsPlant diversity and its changes during the Holocene are geographically structured across temperate Europe. Main causes appear to be differences in past dynamics of the landscape openness and vegetation composition, driven mainly by changes in climate and human impact and their different timing. Fossil pollen, if appropriately treated, is a useful proxy of past plant diversity.

Pollen is an abundant fossil and the most common proxy for plant diversity during the Holocene. Based on datasets in open, forest, and mixed habitats, we used the spatial distribution of floristic diversity to estimate the source area of pollen diversity and identify factors influencing the significance of this relationship.Our study areas are Bohemian-Moravian Highlands and White Carpathians (the Czech Republic and the Slovak Republic).Sampling 60 sites in forest and open habitats in two study regions with contrasting floristic diversity, we calculated taxonomic richness (alpha diversity) and total spatial variance (beta diversity) for pollen and floristic data along two transects, each 1 km long. Following this, we calculated the correlation between floristic and pollen diversity. We also assessed the consistency of the relationship in different habitats. Finally, we regressed local contributions of individual sites to the beta diversity of pollen and floristic data in each of the regions.There was a positive correlation between pollen and floristic richness in both habitats in both regions; open and mixed datasets were significant. The highest correlation (adjusted R2) mostly occurred within the first tens of metres (1.5–70) and then within the first hundreds of metres (250–550). Variances of pollen data significantly correlated with variances of floristic data between 100 and 250 m. Local contributions to beta diversity of pollen and plants significantly correlated in the forest and one of the mixed datasets.Floristic richness at the pollen site and position of the site within the landscape structure determine the sequence of the appearing species in the increasing distance. The number of species sets the source area of pollen richness and dissimilarity of appearing species controls the source area of pollen variance. These findings, linking pollen and floristic diversity, provide an essential stepping-stone for the reconstruction of historic plant diversity.

Investigating past changes in temperate mountain spruce forest ecosystems and the processes behind them can provide valuable information for understanding present and future ecosystem dynamics. To assess the late Holocene ecosystem change and disturbance history in mountain spruce forests, we sampled four small forest hollows from the High Tatra mountains in Slovakia. We use pollen analysis to reconstruct changes in forest composition over the last circa 5000 cal. yr BP. Fire history is analysed using macroscopic charcoal counts and charcoal area measurements. As disturbance is one of the key factors shaping mountain forest dynamics, the analysed pollen records will be processed with a new method quantifying disturbance based on plant ecological indices (Kuneš et al. 2019). These indices for disturbance will be attributed to pollen taxa and then disturbance frequency and severity for the whole community will be calculated. We assess the role of climate and human impact as potential drivers on the past forest and disturbance dynamics. The climate variable will be constructed from modelled climate data for the last 4000 years and for the past 1000 years we will use climate reconstruction from the tree-ring records from the region. We use human indicator pollen taxa as the variable for human influence on ecosystem dynamics, and to indicate human activity in the region.Preliminary results demonstrate opening of the landscape circa 800-500 cal. yr BP in connection with a change in the disturbance regime as indicated by the disturbance indices. The presence of human indicator pollen taxa in all small hollow records suggest landscape opening in connection with anthropogenic activity in the region. In addition, the charcoal records demonstrate periods of fire, which coincide with the opening of landscape and it is plausible that change in the fire regime is connected to the intensified human activity in the region. Multiproxy palaeoecological evidence from a sandstone region in northern Czech Republic was collected to explore the impact of fire disturbances on the decline of the broadleaved forests during the Late Bronze Age (3250–3050 cal. BP). It has been hypothesized that human-accelerated soil leaching affected the nutrient availability in the sandstone area, thus promoting the expansion of oligotrophic-adapted plant communities in the late-Holocene. Little is known about the mechanisms which induced such large-scale vegetation transformation. We sought to determine which driving forces were involved using independent proxy records – soil and sedimentary charcoal, pollen and fungal spores. Local fire history was derived from the variation in charcoal accumulation rates (CHAR) preserved in Eustach peatbog. The population boom-and-bust during the European Neolithic (7000–2000 BC) has been the subject of lively discussion for the past decade. Most of the research on this topic was carried out with help of summed radiocarbon probability distributions. We aim to reconstruct population dynamics within the catchment of a medium sized lake on the basis of information on the presence of all known past human activities. We calculated a human activity model based on Monte Carlo simulations. The model showed the lowest level of human activity between 4000 and 3000 BC. For a better understanding of long-term socio-environmental dynamics, we also used the results of a pollen-based quantitative vegetation model, as well as a local macrophysical climate model. The beginning of the decline of archaeologically visible human activities corresponds with climatic changes and an increase in secondary forest taxa probably indicating more extensive land use. In addition, social and technological innovations are important, such as the introduction of the ard, wheel, animal traction and metallurgy, as well as changes in social hierarchy characterizing the same period.

In the Czech Republic at least five landscape zones, differing by settlement history, can be distinguished. The main focus of this study are marginal mountainous zones: foothills, where the cultural landscape fully developed during high medieval colonization in the 13th century, and the central mountains were colonized mostly in the 16th century and later. Before colonization, these areas are thought to have been essentially wild; however, they are much less archaeologically known than the lowlands. The study focuses on the tracing of a fine-scale prehistoric human impact in two pollen diagrams from the Lusatian Mountains (foothills) and the Bohemian Forest (central mountains). The pollen records are compared with available archaeological data. A case study comparing the off-site and on-site pollen records from the Vladař hillfort (Hallstatt) is presented to demonstrate the local signal of the pollen profiles of small mires. In both studied mountainous regions, the strongest prehistoric human impact was detected during the Hallstatt/La Tene period, that is characterized by a rapid expansion of a single tree taxon ( Pinus and Abies respectively) together with the consistent occurrence of Plantago lanceolata and other indicators for anthropogenic influence. The human impact was less distinct in the pollen profile from the central part of the Bohemian Forest. In both studied regions, the presumed prehistoric human-induced vegetation changes influenced the species composition of the forest for hundreds of years and were therefore not minor. The pollen sites are small mires or larger forested peat bogs and thus mostly reflect local conditions. It is also assumed that the pre-medieval human impact on vegetation must have been limited in space. Despite the limits of recent data, we present evidence for prehistoric human influence on the vegetation. Future research should be either more extensive (thorough archaeological prospection, more well-dated pollen profiles) or focused on special cases when both archaeological and pollen sites are next to each other.

A thousand years old 105 cm deep deposit of bat guano in the Domica Cave (southern Slovakia, Slovak Karst National Park) has been discovered for science, and three samples were analysed for pollen to identify the bats’ preferred foraging habitats and for insect remains to identify their diet. The bat species concerned, Rhinolophus euryale, is rare in the area, which lies at the northern margin of its distribution. The pollen record captured alder forests between 897–1024 AD, temperate light broad-leaved oak-hornbeam forests with Quercus cerris, Fraxinus ornus, Cornus mas and Corylus avellana between 1522–1800 AD, and almost recently willow shrubs. This pattern may, however, reflect local changes in the surrounding landscape where the bats hunted. Pollen of anemophilous taxa was underrepresented (e.g. Fagus), while entomophilous taxa were overrepresented (e.g. Fraxinus ornus, Loranthus europaeus, Acer, Agrostemma githago). The phenology of the encountered pollen taxa indicates that the bats used the Domica Cave mainly as spring and summer roosts. The pollen record further indicates that the bats prefer to forage in a forest–steppe landscape with open Pannonian broadleaved forests and humid temperate riparian environments. Today, this kind of landscape does not occur further north, which may explain the northern limit of this bat species at the study site.

Aim: We provide the first European-scale geospatial training set relating the charcoal signal in surface lake sediments to fire parameters (number, intensity and area) recorded by satellite moderate resolution imaging spectroradiometer (MODIS) sensors. Our calibration is intended for quantitative reconstructions of key fire-regime parameters by using sediment sequences of microscopic (MIC from pollen slides, particles 10-500 µm) and macroscopic charcoal (MAC from sieves, particles >100 µm). Location: North-south and east-west transects across Europe, covering the mediterranean, temperate, alpine, boreal and steppe biomes. Time period: Lake sediments and MODIS active fire and burned area products were collected for the years 2012-2015. Methods: Cylinder sediment traps were installed in lakes to annually collect charcoal particles in sediments. We quantitatively assessed the relationships between MIC and MAC influx (particles/cm2/year) and the MODIS-derived products to identify source areas of charcoal and the extent to which lake-sediment charcoal is linked to fire parameters across the continent. Results: Source area of sedimentary charcoal was estimated to a 40-km radius around sites for both MIC and MAC particles. Fires occurred in grasslands and in forests, with grass morphotypes of MAC accurately reflecting the burned fuel-type. Despite the lack of local fires around the sites, MAC influx levels reached those reported for local fires. Both MIC and MAC showed strong and highly significant relationships with the MODIS-derived fire parameters, as well as with climatic variation along a latitudinal temperature gradient. Main conclusions: MIC and MAC are suited to quantitatively reconstructing fire number and fire intensity on a regional scale. However, burned area may only be estimated using MAC. Local fires may be identified by using several lines of evidence, e.g. analysis of large particles (>Â600 µm), magnetic susceptibility and sedimentological data. Our results offer new insights and applications to quantitatively reconstruct fires and to interpret available sedimentary charcoal records.

High-resolution diatom analysis was carried out to assess the limnological and climatic changes that took place at Ribains maar (French Massif Central) during the Late Pleistocene (∼131–∼105 ka BP), with a focus on the Eemian interglacial in particular. Numerical analyses were used to show that most of the variability in the fossil diatom assemblages was due to climate independently from the changes in the lake catchment vegetation (as represented by pollen data). Diatom-based quantitative reconstructions of the past limnological conditions, as well as a comprehensive literature review on the auto-ecological requirements for the principal diatom taxa, were used to interpret the record. An absolute time-scale for the sequence was derived by matching the major pollen shifts with the radiometrically dated changes in oxygen isotopes observed in Italian stalagmites. This study shows that at Ribains maar, the transition from the Riss (=Saalian) Glacial to the Eemian interglacial was marked by a gradual increase in the contribution of spring-blooming diatom species, indicating a longer growing season and milder winter/spring conditions at that time. A short cooling event interrupts this trend and may correspond to a stadial. At the start of the Eemian a peak in benthic taxa and the suppression of spring-blooming flora probably reflects the effects of deglaciation on the catchment. During the Eemian interglacial itself three main phases were distinguished within the diatom record. The first phase (∼8000 years in duration) was dominated by Stephanodiscus minutulus , which suggests that intense mixing in the water-column took place during spring. The pollen record was simultaneously dominated by Quercus and Corylus that typify this phase as the climatic optimum of the Eemian. The second phase, almost equal in duration to the first phase (∼7000 years), is generally dominated by Cyclotella taxa and suggests a less productive lake and much reduced period of spring mixing compared with the first phase. In the pollen diagram this corresponds to an interval dominated by Carpinus–Picea–Abies that indicates a cooler and wetter climate. The third and last phase of the Eemian, ∼2000 year long, saw the return to Stephanodiscus -dominated assemblages, indicating a warming that may correspond to the Dansgaard–Oeschger event 25 identified in the Greenland ice-core record. In the early stage of the Wurm Glacial (=Weichselian), assemblages in the Melisey I stadial (∼3000 year long) were dominated by either Aulacoseira subarctica or Asterionella formosa , which suggest colder spring conditions than during the late Eemian, but not as cold as the ones indicated by the pollen record. Stephanodiscus spp. again dominate during the Saint-Germain Ia interstadial (∼5000 year long) suggesting a return to the conditions that prevailed before the Melisey stadial, in agreement with the pollen record. The record ends with the Montaigu cold event, which is characterised by a Pinus peak in the pollen record, and corresponds to a large abundance of A. subarctica in the diatom sequence. Throughout the Eemian the abundance of Stephanodiscus spp., which is thought to be driven by winter conditions, show cyclic fluctuations that most likely match the cooling events identified in a pollen record from Germany. Variation in insolation throughout the Eemian may have been the driving factor behind the species succession observed in the diatom sequence. While this study demonstrates that diatom analysis of lake sediment can provide very detailed information on long-term climate change, a review of the few other diatom investigations published on European Eemian deposits shows that this technique has been so far seldom used to its full potential in this context in central and southern Europe.

Homocysteine lowering with folic acid has been shown to be ineffective in reducing vascular events in patients with established cardiovascular disease. However, it may be beneficial in the early stages of atherosclerosis particularly for the population at risk. We have investigated the effect of folic acid administration on homocysteine levels, endothelial function, and leukocyte adhesion molecules in atorvastatin-treated patients. Thirty-nine patients were randomized to folic acid 5 mg daily or no additional treatment. Plasma total homocysteine levels were determined by high-performance liquid chromatography. Biochemical and hematologic tests were performed using routine automated analyzer methods. We examined the expression of l-selectin (CD62L); α -subunits of integrins LFA-1 (CD11a), Mac-1 (CD11b), and VLA-4 (CD49d); and β -subunit of β -2 integrins (CD18) by an immunofluorescence method using single-step staining of whole blood with monoclonal antibodies. The fluorescence was quantified by flow cytometry. Reactivity of the forearm skin microvascular bed to thermal and ischemic stimuli was measured using laser Doppler flowmetry. Postocclusive and thermal reactive hyperemia tests were performed. Statistical analysis used 2-sample t test and paired t test. Homocysteine levels decreased significantly in the folic acid–treated group (11.78 ± 2.7 vs 9.75 ± 1.89 μ mol/L, P μ mol/L). There was no effect of folic acid treatment on leukocyte expression of cell adhesion molecules or microvascular reactivity. The lack of effect of folic acid on microvascular reactivity and leukocyte adhesion molecules supports the evidence against the beneficial effects of folic acid administration in the early stages of atherogenesis.

Plant cover and past vegetation development of five mires were analyzed in different orographic and mesoclimatic situations along the NW–SE transect through the Bohemian Forest, Central Europe. Bogs at various altitudes (m), precipitation total (mm) and mean annual temperature (°C) were compared among: (1) wet windward cooling upslope plains at 860 m, 807 mm and 5.1°C, (2) very wet, cold and wind–exposed summit plains at 1120 and 1060 m, 1337 and 1100 mm, 3.2°C and 3.7°C, respectively, and (3) downwind rain-shadow valleys at 750 m, 757 mm, and 6.2°C. The mires of summit plains differ from the others by both present-day plant cover and past vegetation development, which began as late as the Preboreal, 10,000 years ago. Plant cover of their margins is predominated by the krummholz Pinus x pseudopumilio and open central mire expanses are noted by remarkable biodiversity and surface patterning. Mires of lower elevations are represented by domed raised-bogs forested by Pinus rotundata. According to pollen analysis, their development in the NW followed a typical sequence of the Bohemian Forest from the Late Glacial, with steppe-tundra prevailing 11,000 years ago, to the Boreal, with open Pinus and Corylus forests. Since the Atlantic period 6000 years ago, the surrounding forests were formed mainly by Picea, then by Picea and Fagus, and later by Abies, which became dominant in the whole Bohemian Forest since the Subboreal, 4000 years ago. Pinus expanded in the last 300 years. At present, herb and moss communities with scattered Betula pubescens occupy wet lagg, and closed–canopy bog-pine forest of primeval appearance covers the entire mire. In the SE bogs, the development started earlier, 13,000 years ago. Vegetation history here was slightly different; Alnus-Betula carrs have developed since the Atlantic period. At present and in spite of NE, the very centers of these SE bogs remain open, in lagg woods of Betula pubescens with the undergrowth of Phragmites australis prevail. The analysis confirmed that development, vegetation and mire types correspond to the differences in mountain mesoclimate. In extrapolations of paleoecological data from mountain areas the diversification of biotic development has to be taken into account.

Six pollen diagrams from peat bogs in the Vltavský luh (upper Vltava river valley) provide new information about vegetation reconstruction, woodland dynamics, and local development of mires during the Late-glacial and Holocene. Vegetation development began in the Oldest Dryas/Boling with open park plant cover. In the Allerod, woodland with Pinus and Betula developed, and in the Younger Dryas there was a steppe tundra with plants of open habitats. In the Pre-boreal, woodland tundra grew. In the Boreal, Corylus spread, and a major expansion of Picea began in the early Boreal. Picea spread during the Atlantic probably by two different migration routes. Fagus immigrated earlier than in the Bayerischer Wald and Oberpfalzer Wald in the adjoining parts of Germany, and had its major expansion in the early Atlantic. Abies expanded in the late Atlantic. The great abundance of Abies in this area is remarkable, forming Abies or Abies-Fagus woods in less extreme habitats. Human occupation started in the Sub-boreal, as shown by both archaeology and palynology. However, human impact is recognized from anthropogenic indicators which appear in the early Atlantic. At the end of the later Sub-atlantic the development of natural woodland was interrupted by plantation of Picea according to historical and palynological evidence.

The penultimate temperate period, 127–110 ka before present (BP), bracketed by abrupt shifts of the global climate system initiating and terminating it, is considered as an analogue of the Holocene because of a similar low global ice‐volume. Ice core records as well as continental and marine records exhibit conflicting evidence concerning the climate variability within this period, the Last Interglacial. We present, for the first time, a high‐resolution record of oxygen isotopes in diatom opal of the Last Interglacial obtained from the Ribains Maar in France (44°50′09″N 3°49′16″E). Our results indicate that the Last Interglacial in southwestern Europe was generally a period of climatic stability. The record shows that the temperate period was initiated by an abrupt warm event followed midway by a minor climatic transition to a colder climate. An abrupt isotopic depletion that occurs simultaneously with abrupt changes in pollen and diatom assemblages marks the end of the temperate period, and is correlative with the Melisey I stadial. Variations in the isotopic composition of lake‐water related to the isotopic composition of precipitation and evaporation dominate the biogenic opal oxygen isotope record.

In the volcanic region of Velay (Massif Central, France), lake sediment sequences derived from maar craters situated close to one another (Ribains, Praclaux and Lac du Bouchet) have been correlated on the basis of tephrostratigraphical analyses. This has enabled the construction of a sequence (2980 pollen spectra) that begins during the glaciation preceding the Holsteinian and ends in the present. This sequence covers the period from 450 ka, i.e. from the end of marine oxygen isotope stage 12, to the present time, and includes five climatic cycles. The study of a second core from the Praclaux site completed the long Velay sequence. The biostratigraphy of the whole sequence is described in detail and illustrated by a synthetic and simplified pollen diagram. Thirteen temperate forest episodes (interglacials or interstadials) and as many cold periods (glacials or stadials) are defined. Each temperate episode is characterised by particular forest development and vegetation dynamics, and all of them, with the exception of the Holocene, begin and end with a Pinus forest. Abies alba (fir), a submontane tree from Central European mountains and uplands, is of special interest for palaeoecological and palaeoclimate interpretations due to its sensitivity to air and soil humidity. Its present distribution limit in the uplands of SE Poland is still a matter of debate. In the Holocene fir expanded to Poland very late, but early fir populations are supposed to occur in the Sumava Mts (Czech Republic). The study aims: to estimate pollen thresholds for fir presence/absence in Bohemia (Czech Republic) and Poland on the basis of modified Tauber pollen traps; to use these thresholds for tracing fir presence in two pollen diagrams from Poland (Slone and Bezedna lakes) in the border zone between the Roztocze region (with fir forest stands today) and Polesie (where fir has never played an important role); and to investigate how the percentage presence/absence threshold can be used to trace the occurrence and abundance of fir trees in the Sumava Mts based on the pollen diagrams of Rokytecka slat' and Mrtvý luh. The fir pollen thresholds estimated in terms of PAR (pollen accumulation rates or pollen influx) range from 843 (grains cm− 2 year− 1) (Roztocze) to 61 (Krkonose) and 49 (Sumava). Percentage thresholds range from 0.3% in Krkonose where fir trees are not present within 4 km to 22% in fir-dominated woodland of the Roztocze, providing evidence of strong underrepresentation of fir in the pollen deposition. Application of these percentage thresholds to the Slone and Bezedna pollen diagrams indicates that occurrence of fir in the region is possible from 3.5 cal ky BP onwards, though the evidence is not decisive. In the Sumava, a low representation of fir pollen (1–2%) reflecting presence of scattered fir trees was detected as early as ca. 7.0 cal ky BP.

This paper compares pollen spectra derived from modified Tauber traps and moss samples from a selection of woodland types from Bulgaria, the Czech Republic, Georgia, Greece, Poland, Switzerland and Wales. The study examines the representation of individual taxa in the two sampling media and aims to ascertain the duration of pollen deposition captured by a moss. The latter aim was pursued through the calculation of dissimilarity indexes to assess how many years of pollen deposited in a pollen trap yield percentage values that are most similar to those obtained from the moss. The results are broadly scattered; the majority of moss samples being most similar to several years of pollen deposition in the adjacent trap. For a selection of samples, a comparison of the pollen accumulation rate in pollen traps with the pollen concentration in the moss per unit surface indicates that the entrapment and/or preservation of individual pollen types in the moss differ from that in the pollen trap. A comparison of the proportion of different taxa in the moss with the pollen spectrum of 2 years of pollen deposition in the trap also revealed large differences. There is a tendency for bisaccate grains such as Pinus and Picea to have a higher representation in moss than in traps but there is considerable regional variation. The results indicate that pollen proportions from moss samples often represent the pollen deposition of one area over several years. However, bisaccate pollen grains tend to be over-represented in moss samples compared to both pollen traps and, potentially, lake sediments. peerReviewed

Annual pollen-accumulation rates (PAR) of Fagus (beech) obtained within the framework of the Pollen Monitoring Programme (PMP) were analyzed in pollen traps along a N–S transect from the Baltic Sea to the Black Sea in different European vegetation units. The study regions are situated in the lowlands of northern Poland, the uplands of SE Poland, the Czech Krkonoše Mts, the Czech Šumava Mts, the Swiss Jura Mts, the Swiss Alps, the Bulgarian Rila Mts and the Bulgarian Strandzha Mts. Most time series are 10 or 11 years long, some are 5–16 years long. Inter-annual fluctuations in Fagus PAR were analyzed and compared with seed mast years. Years with high Fagus PAR and others with low Fagus PAR occurred most frequently in parallel within each region and often in two neighbouring regions. 2006 was exceptional as it had a very high Fagus sylvatica pollen deposition in all study regions and it was also a mast year. In Bulgaria, the trend in the 5 years of Fagus orientalis PAR in the Strandzha Mts differed from that of F. sylvatica PAR in the Rila Mts. Aiming at establishing the relationship between average Fagus PAR and tree cover, differences in Fagus PAR (averaged per pollen trap) were related in each region to the proportion of beech trees in the vegetation within 2 km of the pollen traps, the distance to the nearest pollinating Fagus tree, regional or local presence of beech forests, the degree of landscape openness, and the size of forest opening in which a trap is situated. Average Fagus PAR was found to track the regional abundance of beech trees in the vegetation, not the distance of the nearest Fagus tree. Regional occurrence of beech-dominated forests was reflected by a Fagus PAR of ca. 1,400 grains cm−2 year−1, local abundance very close to pollen traps by ca. 2,400 grains, small patches of forest with admixture of Fagus by ca. 170–220 grains, and scarcity or absence of Fagus by ca. 40 grains or less. peerReviewed

Annual PAR (pollen accumulation rates; grains cm−2 year−1) were studied with modified Tauber traps situated in ten regions, in Poland (Roztocze), the Czech Republic (two regions in Krkonoše, two in Šumava), Switzerland (4 regions in the Alps), and Georgia (Lagodekhi). The time-series are 10–16 years long, all ending in 2007. We calculated correlations between pollen data and climate. Pollen data are PAR summarized per region (4–7 traps selected per region) for each pollen type (9–14 per region) using log-transformed, detrended medians. Climate data are monthly temperature and precipitation measured at nearby stations, and their averages over all possible 2- to 6-month windows falling within the 20-month window ending with August, just prior to the yearly pollen-trap collection. Most PAR/climate relationships were found to differ both among pollen types and among regions, the latter probably due to differences among the study regions in the habitats of plant populations. Results shared by a number of regions can be summarized as follows. Summer warmth was found to enhance the following year’s PAR of Picea, Pinus non-cembra, Larix and Fagus. Cool summers, in contrast, increase the PAR of Abies, Alnus viridis and Gramineae in the following year, while wet summers promote PAR of Quercus and Gramineae. Wetness and warmth in general were found to enhance PAR of Salix. Precipitation was found to be more important for PAR of Alnus glutinosa-type than temperature. Weather did not have an impact on the PAR of Gramineae, and possibly of Cyperaceae in the same year. Care is advised when extrapolating our results to PAR in pollen sequences, because there are large errors associated with PAR from sediments, due to the effects of taphonomy and sedimentation and high uncertainty in dating. In addition, in pollen sequences that have decadal to centennial rather than near-annual resolution, plant-interaction effects may easily out-weigh the weather signal. peerReviewed

Diabetes mellitus is associated with a number of prothrombotic abnormalities, and correction of these abnormalities might translate into the reduction of cardiovascular risk. Glitazones improve endothelial function and reduce inflammation, but much less is known about their effect on thrombogenic factors. We have therefore studied the effect of rosiglitazone on leukocyte and soluble thrombogenic markers in patients with type 2 diabetes mellitus. Thirty-three subjects with type 2 diabetes and 32 normal controls were included; patients were examined at baseline and after 5 months of rosiglitazone treatment (4 mg/day). We measured leukocyte-platelet aggregates and leukocyte expression of either P-selectin glycoprotein ligand 1 (PSGL-1) or receptor for urokinase-type plasminogen activator (uPAR) using flow cytometry, as well as several circulating soluble thrombogenic markers by ELISA method. Leukocyte expression of uPAR and PSGL-1 was significantly higher in patients than in controls. Leukocyte-platelet aggregates and leukocyte expression of uPAR and PSGL-1 significantly decreased after rosiglitazone. There was also significant decrease in CRP and fibrinogen levels, but there was no effect of diabetes and/or rosiglitazone on other circulating molecules. In conclusions, we observed a substantial improvement in the expression of thrombogenic markers on leukocytes after rosiglitazone treatment, suggesting the novel antithrombotic effects of rosiglitazone.

Diabetes mellitus is associated with increased inflammatory response, which may contribute to atherosclerosis progression. Experimental results demonstrated anti-inflammatory activity of glitazones; their effect on leukocyte adhesion molecules has not been studied to date. We therefore studied the effect of rosiglitazone treatment on leukocyte surface expression of adhesion molecules in patients with type 2 diabetes mellitus and compared our results with findings in healthy subjects. 33 subjects with type 2 diabetes and 32 healthy controls were included; patients were examined at baseline and after 5 months of rosiglitazone treatment (4 mg/d). Leukocyte expression of adhesion molecules LFA-1, CD18 and ICAM-1 was quantified using flow cytometry; in addition, CD14 (lipopolysaccharide receptor) expression was analyzed as a marker of nonspecific immunity. The expression of examined molecules at baseline was higher in patients compared to controls. Despite only mild decrease in blood glucose, rosiglitazone treatment induced substantial decrease of CD18 and CD14 expression and borderline decrease of LFA-1 and ICAM-1 expression (on monocytes only). We thus observed improvement in the expression of leukocyte inflammatory markers after rosiglitazone treatment. This effect is supposed to be mediated by direct effect of rosiglitazone on PPAR-γ receptors on leukocytes.

Atherogenesis involves the migration of leukocytes into vascular subendothelial space, a process mediated by endothelial and leukocyte cell adhesion molecules. Endothelial molecules are assessed indirectly via serum levels, but leukocyte molecules can be assessed directly. We have therefore hypothesized that leukocyte adhesion molecules are altered to a greater degree in hypercholesterolemia than serum endothelial adhesion molecules. We examined 29 subjects with hypercholesterolemia and 27 controls at baseline and after 12 weeks of atorvastatin treatment (20 mg/day). Expression of leukocyte integrins CD11a, CD11b, CD18, and CD49d and of L-selectin was measured by flow cytometry. Serum ICAM-1, E-selectin and von Willebrand factor were measured by ELISA. Expression of leukocyte adhesion molecules was significantly higher in patients at baseline than in the controls, except for CD11a. Expression significantly decreased after atorvastatin in most adhesion molecules except for CD11b. In contrast, there was no effect of hypercholesterolemia and/or atorvastatin on the serum endothelial molecules. Leukocyte but not endothelial adhesion molecules were influenced by hypercholesterolemia and by lipid lowering treatment. Leukocyte molecules may therefore be a more sensitive marker of atherogenesis than endothelial molecules. Our results support the role of increased leukocyte adhesiveness in atherogenesis.

Lipoprotein(a) [Lp(a)] comprises of an LDL particle and apolipoprotein(a) [apo(a)] and its elevated levels are considered a risk factor for atherosclerosis. The aim of our study was to find out whether elevated Lp(a) levels are associated with increased risk of atherosclerosis in patients with multiple other risk factors. We further tested the association of three polymorphisms of the apo(a) gene promoter with Lp(a) levels. No significant correlation was detected between Lp(a) levels and lipid and clinical parameters tested. The study demonstrated a significantly (p=0.0219) elevated Lp(a) level (mean 28±35 mg/dl, median 0.14) in patients with coronary heart disease (CHD). In a group with premature CHD the correlation was not significant anymore.
