The paper introduces an adaptive version of the stabilized Trace Finite Element Method (TraceFEM) designed to solve low-regularity elliptic problems on level-set surfaces using a shape-regular bulk mesh in the embedding space. Two stabilization variants, gradient-jump face and normal-gradient volume, are considered for continuous trace spaces of the first and second degrees, based on the polynomial families $Q_1$ and $Q_2$. We propose a practical error indicator that estimates the `jumps' of finite element solution derivatives across background mesh faces and it avoids integration of any quantities along implicitly defined curvilinear edges of the discrete surface elements. For the $Q_1$ family of piecewise trilinear polynomials on bulk cells, the solve-estimate-mark-refine strategy, combined with the suggested error indicator, achieves optimal convergence rates typical of two-dimensional problems. We also provide a posteriori error estimates, establishing the reliability of the error indicator for the $Q_1$ and $Q_2$ elements and for two types of stabilization. In numerical experiments, we assess the reliability and efficiency of the error indicator. While both stabilizations are found to deliver comparable performance,the lowest degree finite element space appears to be the more robust choice for the adaptive TraceFEM framework.

5. Are Carriers Unaffected? A Literature Review of Metabolic and Health Outcomes among Genetic Carriers of Phenylketonuria

Background: Phenylketonuria (PKU) is an autosomal recessive genetic condition that results in reduced enzymatic functioning within the phenylalanine hydroxylase (PAH) pathway, which is involved in the metabolism of phenylalanine (Phe) into tyrosine (Tyr). Without dietary intervention, individuals with PKU exhibit significantly elevated levels of Phe, which is presumed to cause severe neurological dysfunction and other associated health risks. Carriers of PKU are heterozygotes for a PAH gene mutation and are typically described in the literature as “unaffected.” However, decades of existing research challenges this classical thinking and it is plausible that these individuals currently classified as carriers may present with an intermediate phenotype or may be “moderately affected.” Summary: The purpose of this scoping review was to explore this hypothesis further, by searching for and summarizing existing literature on metabolism and health outcomes among PKU carriers. Preliminary research has suggested that some PKU carriers exhibit reduced PAH enzyme function, and relatedly, elevated circulating Phe levels compared to noncarriers. In addition, Phe dosing trials have further demonstrated that carriers have increased Phe levels and decreased Tyr levels compared to noncarriers. Because of these metabolic perturbations, it is biologically plausible for carriers to experience an intermediate phenotype in terms of metabolic consequences and clinical outcomes. While these outcomes have yet to be thoroughly explored, early research has found associations between PKU carrier status and lower IQs as well as decreased executive functioning, memory, processing speed, and inhibitory control. The PAH pathway is also involved in melanogenesis, and research has demonstrated increased melanoma risk among PKU carriers. However, there are many limitations to this research, and thus whether or not carriers are clinically impacted cannot yet be conclusively determined. Key Message: Overall, while preliminary research suggests a possible intermediate phenotype among PKU carriers, the current available research is limited and PKU carriers are still clinically considered “unaffected.” This review outlines the current literature while discussing future research endeavors related to the metabolism and health of PKU carriers.

16. SGLT2 Inhibitors Correct Fluid Overload in Adult Kidney Transplant Recipients—A Prospective Observational Study

In this longitudinal observational study, we measured urinary glucose concentration, body composition and volume status (bioimpedance spectroscopy) and plasma renin and aldosterone concentrations in n = 22 kidney transplant recipients (KTRs) initiating on SGLT2I at baseline (BL), and after 1 week and 1, 3, and 6 months. Estimated glomerular filtration rate (eGFR) decreased by −2 mL/min/1.73 m2 (IQR −10–0) after 1 week and remained stable thereafter. Urinary glucose concentration was 10 (3–24) g/g creatinine after 1 week and correlated with eGFR (r2 = 0.273; p = 0.057). SGLT2I did not affect HbA1c, fasting blood glucose, body weight, fat or lean mass. SGLT2I decreased fluid overload dependent on baseline overhydration (OH, r2 = 0.54, p = 0.0003) without occurrence of dehydration. Plasma aldosterone increased at day 7, while plasma renin did not change significantly. In conclusion, SGLT2I corrected fluid overload in patients with elevated overhydration at baseline, while in euvolemic KTRs fluid status remained stable without reduction of body water below the reference range, thus promoting the safety of SGLT2I therapy in patients following kidney transplantation. Glucosuria, together with effects of SGLT2I on blood glucose control and body weight, is attenuated in KTRs dependent on eGFR.

Rotating detonation engines (RDEs) are the subject of research in the combustion community due to the prospects of enhanced thermal efficiency and power density when compared to current deflagrative-based aerospace combustors. Many current simulations presume premixed reactants and therefore miss the important characteristics of transient mixing, wave-induced mixing, and injector design/spacing that are known to play a pivotal role in the system performance. Very ambitious large eddy simulations are being conducted, but necessarily on a limited number of realistic and complex cases, thus limiting their utility in deriving fundamental understanding. For these reasons, a two dimensional parametric study was conducted to assess propagation of a detonation across an idealized array of mixing/injection sites, parametrically characterized by the width and axial mixing profile.Under such non-premixed conditions, discrete energy release and interdependence are observed. The discrete energy release sites frequently create pressures that exceed idealized Chapman-Jouguet (CJ) predictions based on perfect and uniform mixtures. The local higher pressure is shown to be caused by delayed heat release behind the shock, near constant pressure combustion, and additional compression due to the non-uniformities present. The resulting compression and the near constant pressure combustion are accompanied by time scale separation of exothermic and endothermic reactions due to the mixing efficiencies in the non/poorly-premixed cases. In contrast, the better mixed cases show that the detonation wave is sustained but unburnt fuel and oxidizer exist behind the main combustion wave and impulse performance suffers. Results show that for the conditions modeled there exists an optimal injector spacing to maximize the impulse produced and that discrete injection impulses can exceed that of the premixed systems. These somewhat counter-intuitive results imply that the detailed mixing evolution, provoked by the passage of the wave can lead to an extended heat release zone that elevates the pressure over a longer distance along the wavefront. These revelations provide potential for optimizing injector configurations for real non-premixed systems in order to exploit these physics.

Inflammatory macrophages are key drivers of atherosclerosis that can induce rupture-prone vulnerable plaques. Skewing the plaque macrophage population towards a more protective phenotype and reducing the occurrence of clinical events is thought to be a promising method of treating atherosclerotic patients. In the current study, we investigate the immunomodulatory properties of itaconate, an immunometabolite derived from the TCA cycle intermediate cis-aconitate and synthesised by the enzyme Aconitate Decarboxylase 1 (ACOD1, also known as IRG1), in the context of atherosclerosis. Ldlr−/− atherogenic mice transplanted with Acod1−/− bone marrow displayed a more stable plaque phenotype with smaller necrotic cores and showed increased recruitment of monocytes to the vessel intima. Macrophages from Acod1−/− mice contained more lipids whilst also displaying reduced induction of apoptosis. Using multi-omics approaches, we identify a metabolic shift towards purine metabolism, in addition to an altered glycolytic flux towards production of glycerol for triglyceride synthesis. Overall, our data highlight the potential of therapeutically blocking ACOD1 with the aim of stabilizing atherosclerotic plaques.

20. Efficient distributed matrix-free multigrid methods on locally refined meshes for FEM computations

This work studies three multigrid variants for matrix-free finite-element computations on locally refined meshes: geometric local smoothing, geometric global coarsening, and polynomial global coarsening. We have integrated the algorithms into the same framework-the open-source finite-element library deal.II-, which allows us to make fair comparisons regarding their implementation complexity, computational efficiency, and parallel scalability as well as to compare the measurements with theoretically derived performance models. Serial simulations and parallel weak and strong scaling on up to 147,456 CPU cores on 3,072 compute nodes are presented. The results obtained indicate that global coarsening algorithms show a better parallel behavior for comparable smoothers due to the better load balance particularly on the expensive fine levels. In the serial case, the costs of applying hanging-node constraints might be significant, leading to advantages of local smoothing, even though the number of solver iterations needed is slightly higher., Comment: 34 pages, 17 figures

21. Robust preconditioning for a mixed formulation of phase-field fracture problems

In this work, we consider fracture propagation in nearly incompressible and (fully) incompressible materials using a phase-field formulation. We use a mixed form of the elasticity equation to overcome volume locking effects and develop a robust, nonlinear and linear solver scheme and preconditioner for the resulting system. The coupled variational inequality system, which is solved monolithically, consists of three unknowns: displacements, pressure, and phase-field. Nonlinearities due to coupling, constitutive laws, and crack irreversibility are solved using a combined Newton algorithm for the nonlinearities in the partial differential equation and employing a primal-dual active set strategy for the crack irreverrsibility constraint. The linear system in each Newton step is solved iteratively with a flexible generalized minimal residual method (GMRES). The key contribution of this work is the development of a problem-specific preconditioner that leverages the saddle-point structure of the displacement and pressure variable. Four numerical examples in pure solids and pressure-driven fractures are conducted on uniformly and locally refined meshes to investigate the robustness of the solver concerning the Poisson ratio as well as the discretization and regularization parameters.

High Energy Physics - Experiment - Abstract
The first search for the doubly heavy $\it{\Omega}_{bc}^{\rm0}$ baryon and a search for $\it{\Xi}_{bc}^{\rm0}$ baryon are performed using $pp$ collision data collected via the LHCb experiment from 2016 to 2018 at a centre-of-mass energy of $13TeV$, corresponding to an integrated luminosity of 5.2$fb^{-1}$. The baryons are reconstructed via their decays to $\it{\Lambda_{c}^{+}\pi^{-}}$ and $\it{\Xi_{c}^{+}\pi^{-}}$. No significant excess is found for invariant masses between 6700 and 7300$MeV/c^2$, in a rapidity range from 2.0 to 4.5 and a transverse momentum range from 2 to 20$GeV/c$. Upper limits are set on the ratio of the $\it{\Omega}_{bc}^{\rm0}$ and $\it{\Xi}_{bc}^{\rm0}$ production cross-section times the branching fraction to $\it{\Lambda_{c}^{+}\pi^{-}}$ ($\it{\Xi_{c}^{+}\pi^{-}}$) relative to that of the $\it{\Lambda_b^{\rm0}}$ ($\it{\Xi_b^{\rm0}}$) baryon, for different lifetime hypotheses, at $95\%$ confidence level. The upper limits range from $0.5\times10^{-4}$ to $2.5\times10^{-4}$ for the $\it{\Omega}_{bc}^{\rm0}\rightarrow\it{\Lambda_{c}^{+}\pi^{-}}$ ($\it{\Xi}_{bc}^{\rm0}\rightarrow\it{\Lambda_{c}^{+}\pi^{-}}$) decay, and from $1.4\times10^{-3}$ to $6.9\times10^{-3}$ for the $\it{\Omega}_{bc}^{\rm0}\rightarrow\it{\Xi_{c}^{+}\pi^{-}}$ ($\it{\Xi}_{bc}^{\rm0}\rightarrow\it{\Xi_{c}^{+}\pi^{-}}$) decay, depending on the considered mass and lifetime of the $\it{\Omega}_{bc}^{\rm0}$ ($\it{\Xi}_{bc}^{\rm0}$) baryon., Comment: All figures and tables, along with machine-readable versions and any supplementary material and additional information, are available at https://cern.ch/lhcbproject/Publications/p/LHCb-PAPER-2021-002.html (LHCb public pages)

Due to the increasing availability of high-performance computing over the past few decades, numerical models have become an important tool for research in geodynamics. Several generations of mantle convection software have been developed, but due to their differing methods and increasing complexity it is important to evaluate the accuracy of each new model generation to ensure published geodynamic research is reliable and reproducible. Here we explore the accuracy of the open-source, finite-element codes ASPECT and CitcomS as a function of mesh spacing using low to moderate-Rayleigh-number models in steady-state thermal convection. ASPECT (Advanced Solver for Problems in Earth's ConvecTion) is a new-generation mantle convection code that enables modeling global mantle convection with realistic parameters and complicated physical processes using adaptive mesh refinement (Kronbichler et al., 2012; Heister et al., 2017). We compare the ASPECT results with calculations from the finite-element code CitcomS (Zhong et al., 2000; Tan et al., 2006; Zhong et al., 2008), which has a long history of use in the geodynamics community. We find that the globally averaged quantities, i.e., root-mean-square (rms) velocity, mean temperature, and Nusselt number at the top and bottom of the shell, agree to within 1 % (and often much better) for calculations with sufficient mesh resolution. We also show that there is excellent agreement of the time evolution of both the rms velocity and the Nusselt numbers between the two codes for otherwise identical parameters. Based on our results, we are optimistic that similar agreement would be achieved for calculations performed at the convective vigor expected for Earth, Venus, and Mars.

46. Participation in Everyday Leisure and Its Influencing Factors for People with Intellectual Disabilities: A Scoping Review of the Empirical Findings

Background: Participation is often defined as taking part and being included in different areas of life. Leisure represents an important area of life for all people. People with disabilities have the right to experience leisure time in a self-determined manner. They have the right to participate in leisure activities on an equal basis with others. Due to various influencing factors, people with intellectual disabilities, especially those with severe to profound intellectual disabilities, are at risk of decreased participation. This is alarming because participation in leisure activities reflects quality of life. Purpose: The present study aims to review the empirical findings on leisure participation and its influencing factors in people with mild to moderate disabilities as compared to people with severe to profound intellectual disabilities. Method: A scoping review following the PRISMA-ScR checklist by Cochrane and the Joanna Briggs Institute (JBI) was conducted to examine research studies published in peer-reviewed journals between 2000 and 2022. The studies that were included relate to activities within the everyday leisure time of people with intellectual disabilities, regardless of age, gender, or severity of their cognitive disability. The categories of vacation and tourism were excluded so as to focus on everyday leisure. The sample was screened by two reviewers independently. In total, 27 articles met the inclusion criteria, with 21 articles referring to people with a mild to moderate intellectual disability and only six articles referring to people with a severe to profound intellectual disability. The evidence was summarized with a predefined standardized charting form, which was used by the two reviewers. Results: The results show that participation in leisure activities by people with intellectual disabilities can be limited, especially for those with severe to profound intellectual disabilities. This contradicts the guiding principle and human rights of inclusion and self-determination. Their participation in leisure time is extremely dependent on external factors, such as support people, leisure time availability, and form of living. Passive activities at home are often provided for people with severe to profound intellectual disabilities in particular; therefore, the need for interactive and self-determined leisure opportunities in the community is enormous. Various factors influencing leisure participation can be identified. Implications: The findings of this scoping review can be used to consider intervention, support, and barriers to enhancing leisure participation among people with disabilities as an important area of life.

High Energy Physics - Experiment - Abstract
A first search for $CP$ violation in the Cabibbo-suppressed $\Xi_c^+\rightarrow pK^-\pi^+$ decay is performed using both a binned and an unbinned model-independent technique in the Dalitz plot. The studies are based on a sample of proton-proton collision data, corresponding to an integrated luminosity of $3.0~{\rm fb^{-1}}$, and collected by the LHCb experiment at centre-of-mass energies of $7$ and $8~\rm TeV$. High Energy Physics - Experiment - Abstract
A measurement of the branching fraction of the decay $B_s^0 \to K_S^0 K_S^0$ is performed using proton-proton collision data corresponding to an integrated luminosity of 5 $fb^{-1}$ collected by the LHCb experiment between 2011 and 2016. The branching fraction is determined to be BR($B_s^0 \to K_S^0 K_S^0$) = [8.3 $\pm$ 1.6 (stat) $\pm$ 0.9 (syst) $\pm$ 0.8 (norm) $\pm$ 0.3 ($f_s/f_d$)] $\times 10^{-6}$ , where the first uncertainty is statistical, the second is systematic, and the third and fourth are due to uncertainties on the branching fraction of the normalization mode $B^0 \to \phi K_S^0$ and the ratio of hadronization fractions $f_s/f_d$. This is the most precise measurement of this branching fraction to date. Furthermore, a measurement of the branching fraction of the decay $B^0 \to K_S^0 K_S^0$ is performed relative to that of the $B_s^0 \to K_S^0 K_S^0$ channel, and is found to be $\frac{BR(B^0 \to K_S^0 K_S^0)}{BR(B_s^0 \to K_S^0 K_S^0)}$ = [7.5 $\pm$ 3.1 (stat) $\pm$ 0.5 (syst) $\pm$ 0.3 ($f_s/f_d$)] $\times 10^{-2}$., Comment: All figures and tables, along with any supplementary material and additional information, are available at https://cern.ch/lhcbproject/Publications/p/LHCb-PAPER-2019-030.html

High Energy Physics - Experiment - Abstract
A search for the decay $K^0_{\mathrm{S}}\rightarrow\mu^{+}\mu^{-}$ is performed using proton-proton collision data, corresponding to an integrated luminosity of $5.6\,\text{fb}^{-1}$ and collected with the LHCb experiment during 2016, 2017 and 2018 at a center-of-mass energy of $13\,\mathrm{TeV}$. The observed signal yield is consistent with zero, yielding an upper limit of ${\cal B}(K^0_{\mathrm{S}}\rightarrow\mu^{+}\mu^{-}) < 2.2 \times 10^{-10}$ at 90% CL. The limit reduces to ${\cal B}(K^0_{\mathrm{S}}\rightarrow\mu^{+}\mu^{-}) < 2.1 \times 10^{-10}$ at 90% CL once combined with the result from data taken in 2011 and 2012., Comment: All figures and tables, along with any supplementary material and additional information, are available at https://cern.ch/lhcbproject/Publications/p/LHCb-PAPER-2019-038.html

High Energy Physics - Experiment - Abstract
The element $|V_{cb}|$ of the Cabibbo-Kobayashi-Maskawa matrix is measured using semileptonic $B_s^0$ decays produced in proton-proton collision data collected with the LHCb detector at center-of-mass energies of 7 and 8 TeV, corresponding to an integrated luminosity of 3 fb$^{-1}$. Rates of $B_s^0 \to D_s^{-} \mu^+ \nu_{\mu}$ and $B_s^0 \to D_s^{*-} \mu^+ \nu_{\mu}$ decays are analyzed using hadronic form-factor parametrizations derived either by Caprini, Lellouch and Neubert (CLN) or by Boyd, Grinstein and Lebed (BGL). The measured values of $|V_{cb}|$ are ${(41.4 \pm 0.6 \pm 0.9 \pm 1.2)\times 10^{-3}}$ and ${(42.3 \pm 0.8 \pm 0.9 \pm 1.2)\times 10^{-3}}$ in the CLN and BGL parametrization, respectively. The first uncertainty is statistical, the second systematic, and the third is due to the external inputs used in the measurement. These results are in agreement with those obtained from decays of $B^+$ and $B^0$ mesons.
