1. Magma Transfer Processes in the NE Japan Arc: Insights From Crustal Ambient Noise Tomography Combined With Volcanic Eruption Records

Abstract

Subduction zone magmatic budgets and crustal growth processes depend on poorly constrained intrusive:extrusive ratios of arc magmas. Published ambient noise tomography data from northeast Japan reveal upper crustal low-VS bodies, indicating elevated temperatures of plutonic rocks beneath most sites of active arc-front volcanoes, with the exception of Zao volcano. Strikingly, when small eruptions (VEI 1 to VEI 3) from arc-front volcanoes in the last 2 kyr are considered, Zao has erupted most frequently and has produced a more than three times greater tephra volume than any of the other volcanoes. We propose that the frequent low-volume volcanism at Zao is fed by dikes that traverse the crustal section rapidly, fracturing through brittle lithologies. Beneath the other volcanoes, hotter and more ductile plutonic bodies have developed through repeated intrusive activity, hindering magma transport to the surface. This positive feedback mechanism ultimately results in cataclysmic caldera-forming events. We show that the number of late Cenozoic calderas is higher above the imaged low-VS crust, consistent with this interpretation. We propose that generation of plutonic bodies is facilitated by volcaniclastic materials buried in a Miocene rift graben, which dampen fracture propagation and promote repeated sill intrusion. In contrast, transcurrent faulting has moved cold Cretaceous basement of the fore-arc range into the arc-front beneath Zao. These brittle rocks instead provide fracture pathways explored by magmas rapidly rising to the surface. The combined data imply that millions of years of crustal growth and tectonic history have a direct control on present-day volcanic eruption style.

Abstract

Abstract: An insect antifreeze protein gene Mpafp698 was cloned by the RT-PCR approach from the desert beetle Microdera punctipennis. The gene was constructed and heterogeneously expressed in Escherichia coli as fusion proteins, His-MpAFP698, glutathione S-transferase (GST)-MpAFP698, and maltose-binding protein (MBP)-MpAFP698. The thermostability and thermal hysteresis activity of these proteins were determined, with the aim of elucidating the biological characteristics of this protein. The approximate thermal hysteresis (TH) value of the purified His-MpAFP698 was 0.37°C at 0.84mg/ml, and maintained approximately 95.7% of the TH activity at 100°C for 5min. Furthermore, heat incubation showed that MBP-MpAFP698 was 10°C more thermostable than MBP protein, indicating that MpAFP698 could, to some extent, improve the thermal stability of the fused partner MBP protein. This study suggests that MpAFP698 has a high thermal stability and could be used to improve the thermal stability of the less stable proteins by producing fusion proteins, which could be used for biotechnological purposes. Abstract

We developed a protocol for the fast purification of small proteins and peptides using heat incubation as the first purification step. The proteins are expressed from a new bacterial expression vector (pETM-90) fused to the C-terminus of thermostable Ftr from Methanopyrus kandleri. The vector further contains a 6xHis-tag to allow immobilised metal ion affinity purification and a TEV protease cleavage site to enable the removal of the His-tag and fusion partner. Heat incubation induces the specific denaturation and precipitation of the Escherichia coli proteins but not of the thermostable fusion protein. Using the fusion construct and the heat incubation protocol a number of fusion proteins were purified to near homogeneity. The thermostability was ensured when Ftr had a molecular weight higher than twice the target protein. The obtained purification yields were similar and, in some cases, even higher than the ones obtained by affinity purification with the same Ftr-fusion proteins or the same target proteins fused to other often used partners such as NusA, GST, or DsbA. The protocol does not depend on a specific thermostable protein as was shown by the exchange of Ftr for M. kandleri Mtd. Purification by heat incubation is a fast and inexpensive alternative to chromatographic techniques, particularly suitable for the production of antigenic sequences for which the loss of native structure is not detrimental. Abstract

Stained glasses form an important element in human history. Since stained glasses are multifunctional, they have been utilized both in religious and civil architecture. Various conservation techniques such as mechanical, chemical, and the latest one being laser-based cleaning have been utilized. This work aims at making contributions to the knowledge about laser-glass interaction. The three main laser-glass interactions are photo-thermal, photo-chemical, and photo-mechanical. Experiments were carried out in burst and in continuous mode using a 800 ps n-IR laser with a wavelength of 1064nm. A pulsed/burst mode produces a sequence of a defined number of pulses with a high repetition rate. Contemporary glasses and historical pieces were used. The results realized will be important in the establishment of a laser tool selection criterion for decontamination/conservation of actual in-situ historic stained glasses and also develop a dynamic, automated, or semi-automated laser conservation method for historic glass. Complementary analytical techniques used include scanning electron microscopy-energy dispersive spectroscopy (SEM-EDS), confocal microscopy among others. The generally expected outcomes are to develop a practical criterion to apply lasers in cultural heritage; develop skills necessary to select, operate safely and apply lasers to problems in cultural heritage materials in general.
