Purpose: This paper discusses the optimization of pharmacokinetic modelling and alternate simplified quantification method for [18F]AlF-P16-093, a novel tracer for in vivo imaging of prostate cancer. Methods: Dynamic PET/CT scans were conducted on eight primary prostate cancer patients, followed by a whole-body scan at 60 min post-injection. Time-activity curves (TACs) were obtained by drawing volumes of interest for primary prostatic and metastatic lesions. Optimal kinetic modelling involved evaluating three compartmental models (1T2K, 2T3K, and 2T4K) accounting for fractional blood volume (Vb). The simplified quantification method was then determined based on the correlation between the static uptake measure and total distribution volume (Vt) obtained from the optimal pharmacokinetic analysis. Results: In total, 17 intraprostatic lesions, 10 lymph nodes, and 36 osseous metastases were evaluated. Visually, the contrast of the tumor increased and showed the steepest incline within the first few minutes, whereas background activity decreased over time. Full pharmacokinetic analysis revealed that a reversible two-compartmental (2T4K) model is the preferred kinetic model for the given tracer. The kinetic parameters K1, k3, Vb, and Vt were all significantly higher in lesions when compared with normal tissue (P < 0.01). Several simplified protocols were tested for approximating comprehensive dynamic quantification in tumors, with image-based SURmean (the ratio of tumor SUVmean to blood SUVmean) within the 28–34 min window found to be sufficient for approximating the total distribution Vt values (R2 = 0.949, P < 0.01). Both Vt and SURmean correlated significantly with the total serum prostate-specific antigen (tPSA) levels (P < 0.01). Conclusions: This study introduced an optimized pharmacokinetic modelling approach and a simplified acquisition method for [18F]AlF-P16-093, a novel PSMA-targeted radioligand, highlighting the feasibility of utilizing one static PET imaging (between 30 and 60 min) for the diagnosis of prostate cancer. Sonodynamic therapy (SDT) has attracted considerable attention owing to its large penetration depth. However, the low efficiency of traditional sonosensitizers and the hypoxia environment of deep tumors hinder their therapeutic effect in practical application. Herein, the Au@TiO2-hemoglobin loaded liposome (Au@TiO2-Hb-Lip) complex has been constructed to improve the efficiency of SDT against hypoxic tumors. The results indicate that the generated carriers upon ultrasound activation of TiO2 nanoparticles (NPs) can be effectively trapped by Au NPs, and the production of reactive oxygen species is increased by two times compared to that of bare TiO2 NPs. Particularly, the introduction of liposomes as a carrier and hemoglobin as an oxygen supplier not only prevents the aggregation of Au@TiO2 NPs in the blood environment but also improves the adaptability of Au@TiO2-Hb-Lip to hypoxic tumors. The photodecomposition of PbI2 in the perovskite halide structure has been identified as a primary reason for the rapid degradation of metal halide perovskite solar cells (PSCs) under continuous illumination. In particular, excess PbI2 on the surface of perovskite films reduces the stability of PSCs via several mechanisms that are still unelucidated. Herein, the influence of excess PbI2 on PSCs is investigated by varying the Au deposition rate, which results in different temperatures during the fabrication of PSCs, and then they are compared with stoichiometric PSCs. It is demonstrated that the anion exchange of Br− in perovskite and I− in excess PbI2 occurs at the grain boundary and interface of the perovskite layers induced by the high heating temperature, leading to a photoluminescence peak redshift and lower power conversion efficiency. Light illumination accelerates the exchange of Br− and I− between the perovskite layer and excess PbI2 induced by carrier accumulation at the interface and grain boundary, leading to the poor stability of the PSCs. Low photoluminescence (PL) quantum yield of molybdenum disulfide (MoS2) quantum dots (QDs) has limited practical application as potential fluorescent materials. Here, we report the intercalation of aluminum ion (Al3+) to enhance the PL of MoS2 QDs and the underlying mechanism. With detailed characterization and exciton dynamics study, we suggest that additional surface states including new emission centers have been effectively introduced to MoS2 QDs by the Al3+ intercalation. The synergy of new radiative pathway for exciton recombination and the passivation of non-radiative surface traps is responsible for the enhanced fluorescence of MoS2 QDs. Our findings demonstrate an efficient strategy to improve the optical properties of MoS2 QDs and are important for understanding the regulation effect of surface states on the emission of two dimensional sulfide QDs.
