The precise rebound of a droplet upon hitting a solid surface has garnered significant attention in recent years due to its critical applications in self-cleaning, printing industries, and the design of heat exchanger surfaces, among others. This study introduces an innovative approach that combines femtosecond laser processing with a high-temperature stearic acid modification to create surfaces that feature superhydrophilic (SHL) stripes on a superhydrophobic substrate. By controlling the offset distance between the droplet's impact point and the SHL stripe, we achieved a directional and precise rebound of the droplets. Our findings indicate that the lateral displacement of the droplet increases with the offset distance and always tilts toward the direction of the SHL stripe. This study also incorporates numerical simulations to validate the findings, shedding light on the energy conversion mechanisms at the liquid–solid interface during the impact, particularly during the retraction phase. The Berkeley Drosophila Genome Project (BDGP) strives to disrupt each Drosophila gene by the insertion of a single transposable element. As part of this effort, transposons in more than 30,000 fly strains were localized and analyzed relative to predicted Drosophila gene structures. Approximately 6,300 lines that maximize genomic coverage were selected to be sent to the Bloomington Stock Center for public distribution, bringing the size of the BDGP gene disruption collection to 7,140 lines. It now includes individual lines predicted to disrupt 5,362 of the 13,666 currently annotated Drosophila genes (39%). Other lines contain an insertion at least 2 kb from others in the collection and likely mutate additional incompletely annotated or uncharacterized genes and chromosomal regulatory elements. The remaining strains contain insertions likely to disrupt alternative gene promoters or to allow gene mis-expression. The expanded BDGP gene disruption collection provides a public resource that will facilitate the application of Drosophila genetics to diverse biological problems. Finally, the project reveals new insight into how transposons interact with a eukaryotic genome and helps define optimal strategies for using insertional mutagenesis as a genomic tool.

A putative exchange factor for Rho1 GTPase is discussed. The factor is necessary for Drosophila cytokinesis initiation. Cytokinesis ensures success of the cell cycle and distribution of chromosomes, organelles and cytoplasm in daughter cells. Formation and constriction of an actomyosin contractile ring drives progression of a cleavage furrow to bring about cytokinesis. Results of a recent study show Drosophila Pebble (Pbl), the putative exchange factor, to be a RhoGEF specifically needed for cytokinesis and linked through Rho1 activity to reorganization of the actin cytoskeleton at the cleavage furrow.

To identify novel genes and to isolate tagged mutations in known genes that are required for the development of the peripheral nervous system (PNS), we have screened a novel collection of 2460 strains carrying lethal or semilethal P-element insertions on the third chromosome. Monoclonal antibody 22C10 was used as a marker to visualize the embryonic PNS. We identified 109 mutant strains that exhibited reproducible phenotypes in the PNS. Cytological and genetic analyses of these strains indicated that 87 mutations affect previously identified genes: tramtrack (n = 18 alleles), string (n = 15), cyclin A (n = 13), single-minded (n = 13), Delta (n = 9), neuralized (n = 4), pointed (n = 4), extra macrochaetae (n = 4), prospero (n = 3), tartan (n = 2), and pebble (n = 2). In addition, 13 mutations affect genes that we identified recently in a chemical mutagenesis screen designed to isolate similar mutants: hearty (n = 3), dorsotonals (n = 2), pavarotti (n = 2), sanpodo (n = 2), dalmatian (n = 1), missensed (n = 1), senseless (n = 1), and sticky ch1 (n = 1). The remaining nine mutations define seven novel complementation groups. The data presented here demonstrate that this collection of Pelements will be useful for the identification and cloning of novel genes on the third chromosome, since >70% of mutations identified in the screen are caused by the insertion of a P element. A comparison between this screen and a chemical mutagenesis screen undertaken earlier highlights the complementarity of the two types of genetic screens.

The molecular processes which control the neural development in Drosophila melanogaster is investigated using the Drosophila embryonic peripheral nervous system (PNS). About 2000 lethal p-element insertion strains are screened by identifying the PNS development controlling genes. A neural specific marker MAb 22C10 and 92 mutant strains are used for the examination of the PNS of the mutant embryos.

In our quest for novel genes required for the development of the embryonic peripheral nervous system (PNS), we have performed three genetic screens using MAb 22C10 as a marker of terminally differentiated neurons. A total of 66 essential genes required for normal PNS development were identified, including 49 novel genes. To obtain information about the molecular nature of these genes, we decided to complement our genetic screens with a molecular screen. From transposon-tagged mutations identified on the basis of their phenotype in the PNS we selected 31 P-element strains representing 26 complementation groups on the second and third chromosomes to clone and sequence the corresponding genes. We used plasmid rescue to isolate and sequence 51 genomic fragments flanking the sites of these P-element insertions. Database searches using sequences derived from the ends of plasmid rescues allowed us to assign genes to one of four classes: (1) previously characterized genes (11), (2) first mutations in cloned genes (1), (3) P-element insertions in genes that were identified, but not characterized molecularly (1), and (4) novel genes (13). Here, we report the cloning, sequence, Northern analysis, and the embryonic expression pattern of candidate cDNAs for 10 genes: astray, chrowded, dalmatian, gluon, hoi-polloi, melted, pebble, skittles, sticky ch1, and vegetable. This study allows us to draw conclusions about the identity of proteins required for the development of the nervous system in Drosophila and provides an example of a molecular approach to characterize en masse transposon-tagged mutations identified in genetic screens.

We generated a library of ~1000 Drosophila stocks in which we inserted a construct in the intron of genes allowing expression of GAL4 under control of endogenous promoters while arresting transcription with a polyadenylation signal 3’ of the GAL4. This allows numerous applications. First, ~90% of insertions in essential genes cause a severe loss-of-function phenotype, an effective way to mutagenize genes. Interestingly, 12/14 chromosomes engineered through CRISPR do not carry second-site lethal mutations. Second, 26/36 (70%) of lethal insertions tested are rescued with a single UAS-cDNA construct. Third, loss-of-function phenotypes associated with many GAL4 insertions can be reverted by excision with UAS-flippase. Fourth, GAL4 driven UAS-GFP/RFP reports tissue and cell-type specificity of gene expression with high sensitivity. We report the expression of hundreds of genes not previously reported. Finally, inserted cassettes can be replaced with GFP or any DNA. These stocks comprise a powerful resource for assessing gene function.

The development of a simple fluorescent sensor for detecting the Pb2+ heavy metal is fundamentally important. The CH3NH3PbBr3 perovskite material exhibits excellent photoluminescence properties that are related to Pb2+. Based on the effects of Pb2+ on the luminescent properties of CH3NH3PbBr3, we design a novel platform for the selective fluorescence detection of Pb2+ ions. Herein, we use a CH3NH3Br solution at a high concentration as the fluorescent probe. Incorporation of PbBr2 into the CH3NH3Br solution results in a rapid chemical reaction to form CH3NH3PbBr3. Hence, the nonfluorescent CH3NH3Br material displays a sensitive and selective luminescent response to Pb2+ under UV light illumination. Moreover, the reaction between CH3NH3Br and PbBr2 could transform Pb2+ into CH3NH3PbBr3, and therefore, CH3NH3Br may also be used to extract Pb2+ from liquid waste in recycling applications. Crude oil is one of the most widely used energy and industrial raw materials that is crucial to the world economy, and is used to produce various petroleum products. However, crude oil often spills during extraction, transportation and use, causing negative impacts on the environment. Thus, there is a high demand for products to remediate leaked crude oil. Among them, oleophilic and hydrophobic adsorbents can absorb crude oil through thermal effects and are research hotspots. In this review, we first present an overview of wettability theory, the heating principles of various thermal effects, and the theory of reducing crude oil viscosity by heating. Then we discuss adsorbents based on different heating methods including the photothermal effect, Joule heating effect, alternating magnetic field heating effect, and composite heating effect. Preparation methods and oil adsorption performance of adsorbents are summarized. Finally, the advantages and disadvantages of various heating methods are briefly summarized, as well as the prospects for future research.

Purpose: To investigate the feasibility, safety and efficacy of high intensity focused ultrasound ablation (HIFU) as a preoperative treatment for challenging hysteroscopic myomectomies., Materials and Methods: A total of 75 patients diagnosed with types 0-III of uterine fibroids were enrolled. Based on the Size, Topography, Extension of the base, Penetration and lateral Wall position (STEPW) classification scoring system, 25 cases with a score ≥ 5 points were treated with HIFU followed by hysteroscopic myomectomy (HIFU + HM group), whereas 50 cases with a score < 5 points were treated with hysteroscopic myomectomy (HM group)., Results: The median preoperative STEPW score was 7 in the HIFU + HM group and 2 in the HM group. The average non-perfused volume (NPV) ratio achieved in fibroids after HIFU was 86.87%. Patients in the HIFU + HM group underwent hysteroscopic myomectomy one to four days after HIFU, and downgrading was observed in 81.81% of fibroids. The operation time for patients in the HIFU + HM group was 73 min and the success rate of myomectomy in a single attempt was 60%. The volume of distention medium used during the operation was greater in the HIFU + HM group than in the HM group (15,500 ml vs. 7500 ml). No significant difference was observed between the two groups in terms of intraoperative blood loss, the incidence of intraoperative and postoperative complications, menstrual volume score, or uterine fibroid quality of life score., Conclusion: HIFU can be utilized as a preoperative treatment for large submucosal fibroids prior to hysteroscopic myomectomy. HIFU offers a novel approach in the management of this subset of patients.
