Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by persistent activation of immune cells and overproduction of autoantibodies. The accumulation of senescent T and B cells has been observed in SLE and other immune‐mediated diseases. However, the exact mechanistic pathways contributing to this process in SLE remain incompletely understood. In this study, we found that in SLE patients: (1) the frequency of CD4+CD57+ senescent T cells was significantly elevated and positively correlated with disease activity; (2) the expression levels of B‐lymphoma‐2 (BCL‐2) family and interferon‐induced genes (ISGs) were significantly upregulated; and (3) in vitro, the cytokine IL‐15 stimulation increased the frequency of senescent CD4+ T cells and upregulated the expression of BCL‐2 family and ISGs. Further, treatment with ABT‐263 (a senolytic BCL‐2 inhibitor) in MRL/lpr mice resulted in decreased: (1) frequency of CD4+CD44hiCD62L−PD‐1+CD153+ senescent CD4+ T cells; (2) frequency of CD19+CD11c+T‐bet+ age‐related B cells; (3) level of serum antinuclear antibody; (4) proteinuria; (5) frequency of Tfh cells; and (6) renal histopathological abnormalities. Collectively, these results indicated a dominant role for CD4+CD57+ senescent CD4+ T cells in the pathogenesis of SLE and senolytic BCL‐2 inhibitor ABT‐263 may be the potential treatment in ameliorating lupus phenotypes. Objective SIRT1, an NAD+-dependent deacetylase, is upregulated in CD4+ T cells from SLE patients and MRL/lpr lupus-like mice. This study aimed to explore the role of SIRT1 in follicular helper T (Tfh) cell expansion and its potential value as a therapeutic target for SLE. Methods Frequencies of CD4+CXCR5+PD-1+ Tfh cells in peripheral blood from SLE patients and their expression of SIRT1 and B cell lymphoma 6 (BCL-6) were determined with flow cytometry. Naïve CD4+ T cells were transfected with SIRT1 -expressing lentivirus and small interfering RNA (siRNA) targeting SIRT1 , respectively, and then cultured under Tfh-polarizing conditions to study the impact of SIRT1 on Tfh cell differentiation. This impact was also evaluated in both CD4+ T cells and naïve CD4+ T cells by treatment with SIRT1 inhibitors (EX527 and nicotinamide) in vitro. MRL/lpr mice and pristane-induced lupus mice were treated with continuous daily intake of nicotinamide, and their lupus phenotypes (including skin rash, arthritis, proteinuria and serum anti-dsDNA autoantibodies) were compared with those of controls. Results Expression of SIRT1 was elevated in Tfh cells from SLE patients and was positively correlated with Tfh cell frequencies. SIRT1 expression gradually increased during Tfh cell differentiation. Overexpression of SIRT1 by lentiviral vectors significantly promoted Tfh cell differentiation/proliferation. Reciprocally, suppressing expression of SIRT1 by siRNA and inhibiting SIRT1 activity by EX-527 or nicotinamide hindered Tfh cell expansion. Continuous daily intake of nicotinamide alleviated lupus-like phenotypes and decreased serum CXCL13 in the two mouse models. Conclusion SIRT1 overexpression contributed to the expansion of Tfh cells in SLE and may serve as a potential target for treatment. [ABSTRACT FROM AUTHOR]

Objective(s): To investigate the effects of Nonylphenol (NP) in pups from dams exposed during gestational and lactational periods on immediate early genes (c-jun, c-fos) in hippocampus and the learning and memory of F1 rats. Materials and Methods: Twenty eight pregnant dams, stratified by pregnancy date, were randomly assigned into 4 groups, which were gavaged with NP at the doses of 50 mg/kg/day, 100 mg/kg/day, 200 mg/kg/day and groundnut oil, respectively. Step-down avoidance test, and learning and memory effects of NP were evaluated on 8-weeks-old pups. The expressions of c-jun and c-fos and the activities of choline acetyltransferase (ChAT) and acetylcholinesterase (AchE) were evaluated in hippocampus of pups. Results: Compared to the control, reaction time (RT) that pups spent to jump to the platform was longer (P=0.02), the number of errors were higher (P=0.01), and the step-down latency was shorter in the 200 mg/kg/day NP-treated group (P=0.04). Exposure to NP induced a significant reduction in ChAT activity in hippocampus in the 100 (P=0.005) and 200 mg/kg/day NP-treated groups (P=0.002), whereas exposure to 200 mg/kg/day caused a significant increase in AchE activity (P=0.004); a dose–response relationship was revealed between ChAT and AchE activities and NP exposure in the hippocampus of pups (r=-0.821, P=0.01; r=0.757, P=0.04). Exposure to NP in the 100 and 200 mg/kg/day NP-treated groups exhibited an increase in number of c-fos and c-jun positive cells. Conclusion: Exposure to NP might negatively affect learning and memory ability in F1 rats, possibly due to the alterations in the expression of c-jun and c-fos, and ChAT, AchE activities in hippocampus of pups.

Carbon emissions reduction is not only physical, but also rooted in powerful economic actions. Coal-related emissions reduction strategies mainly focus on the physical quantity of carbon emissions, rather than the economic efficiency. So it's necessary to strengthen the analysis of carbon emission reduction strategies from the perspective of economic value. Based on the theory of coal supply chain management, this study builds a carbon dioxide accounting model of China's coal supply chain. The indicators for the process-based carbon emissions and carbon emissions per unit of economic value of various coal products are evaluated from the perspective of the whole coal life cycle. The results show that the processing and conversion of coal are the most important sources of carbon emissions in the coal life cycle. Coal-fueled electricity is the largest contributor to carbon emissions of all coal products. The coal-to-heat process has the highest carbon emissions per unit of economic value. Emission reduction strategies for coal supply chain are given in this research from both the physical and economic perspective. To realize emission reduction based on the physical perspective, it's helpful to decrease coal consumption by reducing the proportion of coal-fired power and the gas capture technology should be promoted to facilitate gas utilization. Implementing the coal transition to natural gas policy and developing heat recovery technology have an effect on carbon emissions reduction from the economic viewpoint of coal supply chain. • Life cycle and supply chain are integrated to study carbon footprint of coal industry. • Carbon emission accounting is creatively established by process and product branch. • Economic value is used to analyze the profitability of coal carbon footprint. • The processing and transformation is the major emission source in coal life cycle. • Coal to electricity and heat contribute to physical and economic carbon emissions.
