Abstract

Adipose-derived mesenchymal stem cells (ADSCs) and bone marrow-derived mesenchymal stem cells (BMSCs) have shown great potential in clinical applications. However, the similarities and differences between these two cell types have not been fully elucidated. Recent advances in transcriptomic and metabolomic research have provided valuable insight into the characteristics and functions of ADSCs and BMSCs. In this perspective article, we review the key findings from these studies, including cellular heterogeneity as well as differences in metabolic and secretory properties. We discuss how these insights can help guide the selection of the most suitable cell source for the clinic, and the optimization of preconditioning strategies prior to clinical deployment. Furthermore, we analyze the current landscape of products and clinical trials involving ADSCs and BMSCs, highlighting their therapeutic potential. We propose that the integration of multi-omics datasets will be crucial for establishing a comprehensive understanding of ADSC and BMSC identity and potency, and the provision of quality-assured stem cell-derived products for the clinic.

2. SpikeMOT: Event-Based Multi-Object Tracking With Sparse Motion Features

Abstract

In comparison to conventional RGB cameras, the exceptional temporal resolution of event cameras allows them to capture rich information between frames, making them prime candidates for object tracking. Yet in practice, despite their theoretical advantages, the body of work on event-based multi-object tracking (MOT) remains in its infancy, especially in real-world environments where events from complex background and camera motion can easily obscure the true target motion. To address these limitations, we introduce SpikeMOT, an innovative event-based MOT framework employing spiking neural networks (SNNs) within a Siamese architecture. SpikeMOT extracts and associates sparse spatiotemporal features from event streams, enabling high-frequency object motion inference while preserving object identities. Additionally, a simultaneous object detector provides updated spatial information of these objects at an equivalent frame rate. To evaluate the efficacy of SpikeMOT, we present DSEC-MOT, a meticulously constructed, real-world event-based MOT benchmark. This dataset features manually corrected annotations for objects experiencing severe occlusions, frequent intersections, and out-of-view scenarios commonly encountered in real-world applications. Extensive experiments on the DSEC-MOT and the FE240hz dataset demonstrate SpikeMOT’s superior tracking accuracy under demanding conditions, advancing the state-of-the-art in event-based multi-object tracking.

3. Interactive ethics teaching for students of chemistry

Abstract

Choosing an approach is a regular occurrence for chemists, be it for a new synthesis, a change in production, or out of financial necessity. Arriving at a decision which is right for the individual means weighing up options, even making ethical decisions. The interactive teaching approaches described here involve student chemists making choices in situations they may encounter. The goal is not to instruct chemistry students on what is right or wrong but to encourage them to debate issues and to reach a conclusion which they consider to be ethical. Two teaching approaches are described which are adaptable for many different scenarios; the scenarios were developed to encourage discussion about dual-use issues, issues in which chemicals can be used for good or ill. In the first exercise chemists are invited to make a drug using a simple chemical process and answer a range of questions. This is followed by role play to determine if the chemist might sell the drug to profit from it. A further exercise, using a jigsaw technique, with greater opportunity for discussion is also described. More actors, with different agendas, are introduced into the discussion and student chemists explore circumstances they may be in, make decisions and justify choices. Group discussions enable educators to discuss ethical dilemmas.

Abstract

Abstract Background This study aimed to produce Odontoglossum ringspot virus (ORSV)-free Cymbidium orchid ‘New True’ plants from ORSV-infected mother plants by culturing their meristems and successively repeating subcultures of protocorm-like bodies (PLBs) derived from the meristems. Results Initially, ORSV was confirmed as the causative agent of viral symptoms in orchid leaves via reverse transcription-polymerase chain reaction (RT-PCR) analysis. Meristems from infected plants were cultured to generate PLBs, which in sequence were repeatedly subcultured up to four times. RT-PCR and quantitative RT-PCR analyses revealed that while ORSV was undetectable in shoots derived from the first subculture, complete elimination of the virus required at least a second subculture. Genetic analysis using inter-simple sequence repeat markers indicated no somaclonal variation between regenerated plants and the mother plant, suggesting that genetic consistency was maintained. Conclusion Overall, our findings demonstrate that subculturing PLBs for a second time is ideal for producing genetically stable, ORSV-free Cymbidium orchids, thus offering a practical means of generating genetically stable, virus-free plants and enhancing plant health and quality in the orchid industry.

Abstract

The COVID-19 pandemic and the restrictions that came with it led to unprecedented disruptions in learner development and put them at risk of poor developmental outcomes. It is thus important that the extent of the disruption on their development and the support needs of learners be investigated. Researching the impact of the disruption should focus on holistic development. In this book, the authors use multiple research approaches and methods of gathering data in their respective fields to examine the impact of the disruption and determine the support needs of learners. Data gathered through the different methods are processed and findings are presented. The findings have implications for both practice and future research in the fields of inclusive education, learner support, educational psychology, movement education, Life Orientation and curriculum development.

Abstract

Background Urinary tract infections are diagnosed by general practitioners based on symptoms, dipstick tests in some and laboratory urine culture. Patients may be given inappropriate antibiotics. Point-of-care tests can diagnose urinary tract infection in near-patient settings quicker than standard culture. Some can identify the causative pathogen or antimicrobial sensitivity. Objective To assess whether point-of-care tests for people with suspected urinary tract infection have the potential to be clinically effective and cost-effective to the NHS. Design Systematic review and conceptual economic model. Results Two randomised controlled trials evaluated Flexicult Human (one against standard care; one against ID Flexicult). One trial found no evidence of a difference between groups in concordant antibiotic use (odds ratio 0.84, 95% confidence interval 0.58 to 1.20), and the other found no difference in appropriate antibiotic prescribing (odds ratio 1.44, 95% confidence interval 1.03 to 1.99). Compared with standard care, Flexicult was associated with reduced antibiotic prescribing at initial consultation (odds ratio 0.56, 95% confidence interval 0.35 to 0.88). Identifying individuals with a bacterial infection is crucial to ensure appropriate treatment. However, it is also important to avoid overprescription of antibiotics, to prevent unnecessary side effects and antimicrobial resistance. We conducted a systematic review to summarise evidence on the diagnostic accuracy of symptoms, signs and point-of-care tests to diagnose bacterial respiratory tract infection in adults, and to diagnose two common respiratory viruses, influenza and respiratory syncytial virus. Methods The primary approach was an overview of existing systematic reviews. We conducted literature searches (22 May 2023) to identify systematic reviews of the diagnostic accuracy of point-of-care tests. Where multiple reviews were identified, we selected the most recent and comprehensive review, with the greatest overlap in scope with our review question. Methodological quality was assessed using the Risk of Bias in Systematic Reviews tool. Summary estimates of diagnostic accuracy (sensitivity, specificity or area under the curve) were extracted. Where no systematic review was identified, we searched for primary studies. We extracted sufficient data to construct a 2 × 2 table of diagnostic accuracy, to calculate sensitivity and specificity. Methodological quality was assessed using the Quality Assessment of Diagnostic Accuracy Studies version 2 tool. Where possible, meta-analyses were conducted. We used GRADE to assess the certainty of the evidence from existing reviews and new analyses. Results We identified 23 reviews which addressed our review question; 6 were selected as the most comprehensive and similar in scope to our review protocol. These systematic reviews considered the following tests for bacterial respiratory infection: individual symptoms and signs; combinations of symptoms and signs (in clinical prediction models); clinical prediction models incorporating C-reactive protein; and biological markers related to infection (including C-reactive protein, procalcitonin and others). We also identified systematic reviews that reported the accuracy of specific tests for influenza and respiratory syncytial virus. No reviews were found that assessed the diagnostic accuracy of white cell count for bacterial respiratory infection, or multiplex tests for influenza and respiratory syncytial virus. We therefore conducted searches for primary studies, and carried out meta-analyses for these index tests. Overall, we found that symptoms and signs have poor diagnostic accuracy for bacterial respiratory infection (sensitivity ranging from 9.6% to 89.1%; specificity ranging from 13.4% to 95%). Accuracy of biomarkers was slightly better, particularly when combinations of biomarkers were used (sensitivity 80–90%, specificity 82–93%). The sensitivity and specificity for influenza or respiratory syncytial virus varied considerably across the different types of tests. Tests involving nucleic acid amplification techniques (either single pathogen or multiplex tests) had the highest diagnostic accuracy for influenza (sensitivity 91–99.8%, specificity 96.8–99.4%). Limitations Most of the evidence was considered low or very low certainty when assessed with GRADE, due to imprecision in effect estimates, the potential for bias and the inclusion of participants outside the scope of this review (children, or people in hospital). Future work Currently evidence is insufficient to support routine use of point-of-care tests in primary and emergency care. Further work must establish whether the introduction of point-of-care tests adds value, or simply increases healthcare costs. Plain language summary What is the problem? Respiratory infections are a common cause of illness. Currently, healthcare professionals use clinical experience to decide whether an infection is caused by a virus or bacteria, and whether antibiotics are needed. However, this is not always easy to establish. We tried to identify the effectiveness of rapid tests (with results in under 45 minutes) at distinguishing between viral and bacterial respiratory infections. What did we do? We identified and summarised all the existing reviews and studies in this area. We looked at many different tests which aim to distinguish between bacterial and viral causes of respiratory infections. In particular, we assessed: individual symptoms and signs (such as the presence of cough, or a fever) combinations of symptoms and signs (the presence or absence of multiple symptoms) various ‘biomarker’ tests (blood tests for evidence that the body has used its defence mechanisms) We also looked at specific tests for flu and respiratory syncytial virus, which are common causes of viral infection. What did we find? The reviews we found showed symptoms and signs were not able to identify bacterial infections in people accurately. The accuracy of biomarker tests was slightly better, particularly when multiple markers were used. The accuracy of rapid tests for flu and respiratory syncytial virus varied; the most accurate tests were those that detect viral genetic material. We also found studies showing that genetic tests that identify many viruses at once (multiplex tests) were very accurate. However, most of the evidence we identified was not robust. There were concerns about the conduct of some of the studies. In some cases there was uncertainty whether a test was really accurate enough to be useful. Therefore there is still doubt about whether any of these tests will be useful additions to current clinical care.

Abstract

District heating pipes are usually installed in open trenches that are refilled with backfill material. Special requirements were set for this material in standards and codes of practice and in the most cases natural sands were used in the past. Today natural sands are a scarce resource and must be protected. Furthermore, regulations of the circular economy challenge existing construction methods. In sustainable energy systems natural sands should only be used in exceptional cases. Against this background the general planning process and the significant calculation methods for district heating pipes were analysed and other material options and the hindrances for the use of alternative backfill were identified. Necessary modifications in the planning and calculation process were elaborated. It was shown, that the transition to a more circular economy approach in district heating systems is realistic, if conventional planning methods will be adjusted.

Abstract In Southeast Asia malaria elimination is targeted by 2030. Cambodia aims to achieve this by 2025, driven in large part by the urgent need to control the spread of artemisinin-resistant falciparum malaria infections. Rapid elimination depends on sustaining early access to diagnosis and effective treatment. In much of Cambodia, rapid elimination will rely on a village malaria worker (VMW) network. Yet as malaria declines and is no longer a common cause of febrile illness, VMWs may become less popular with febrile patients, as VMWs do not diagnose or treat other conditions at present. There is a risk that VMWs become inactive and malaria rebounds before the complete interruption of transmission is achieved. During 2021–23 a large-scale operational research study was conducted in western Cambodia to explore how a VMW network could be sustained by including health activities that cover non-malarial illnesses to encourage febrile patients to continue to attend. 105 VMWs received new rapid diagnostic tests (including dengue antigen–antibody and combined malaria/C-reactive protein tests), were trained in electronic data collection, and attended health education packages on hygiene and sanitation, disease surveillance and first aid, management of mild illness, and vaccination and antenatal care. In August 2023 the National Malaria Control Programme of Cambodia convened a stakeholder meeting in Battambang, Cambodia. Findings from the study were reviewed in the context of current malaria elimination strategies. The discussions informed policy options to sustain the relevance of the VMW network in Cambodia, and the potential for its integration with other health worker networks. This expansion could ensure VMWs remain active and relevant until malaria elimination is accomplished.

Pancreatic steatosis and metabolic-dysfunction-associated steatotic liver disease are characterised by fat accumulation in abdominal organs, but their correlation remains inconclusive. Recently proposed deep learning (DL) for proton density fat fraction (PDFF) estimation, which quantifies organ fat, has primarily been assessed for quantifying liver fat. This study aims to validate DL models for pancreatic PDFF quantification and compare pancreas and liver fat content. We evaluated three DL models—Non-Linear Variables Neural Network (NLV-Net), U-Net, and Multi-Decoder Water-Fat separation Network—against a reference PDFF measured using a graph-cut-based method. NLV-Net showed a strong correlation (Spearman rho) with the reference PDFF in the six-echo pancreatic head (slope: 1.02, rho: 0.95) and body (slope: 1.04, rho: 0.94) and a moderate correlation in the three-echo pancreatic head (slope: 0.44, rho: 0.40) and body (slope: 0.49, rho: 0.34). Weak correlations were found between liver and pancreatic body PDFF using graph cut in six-echo (slope: −0.041, rho: −0.12) and three-echo images (slope: 0.0014, rho: 0.073) and using NLV-Net in six-echo (slope: −0.053, rho: −0.12) and three-echo images (slope: −0.014, rho: −0.033). In conclusion, NLV-Net showed the best agreement with the reference for pancreatic fat quantification, and no correlation was found between liver and pancreas fat.

Abstract

Cynanchum auriculatum Royle ex Wight (CA) is a traditional medicinal and edible plant in China. This study aimed to isolate and characterize the phenolic compounds of C. auriculatum to identify its main antioxidant constituents. Polyphenols were extracted using an ultrasound-assisted ethanol extraction method, followed by partitioning with ethyl acetate. The ethyl acetate extract was then purified through thin-layer chromatography, silica gel column chromatography, and reverse-phase silica gel column chromatography. Three monomeric compounds—cynandione A (I), 2,5-dihydroxyacetophenone (II), and radix piperacanthone (III)—were identified through their physical and chemical properties, UV and IR spectra, and liquid chromatography–mass spectrometry (LC-MS/MS). Vitamin C (VC) and 2,4-dihydroxyacetophenone were used as controls to evaluate the antioxidant potential of the two most abundant monomers. Antioxidant assays demonstrated that 2,5-dihydroxyacetophenone and cynandione A exhibited strong antioxidant activity at lower concentrations, whereas 2,4-dihydroxyacetophenone showed significantly weaker activity. Furthermore, cynandione A displayed superior cellular antioxidant activity compared to 2,5-dihydroxyacetophenone, indicating its potential as a promising bioactive compound. In conclusion, this study provides valuable insights into the phenolic composition of C. auriculatum and highlights cynandione A as a key antioxidant, paving the way for future research on its therapeutic applications.

Analytics for behavioral habit typically model one behavior at a time, despite the fact that habit often involves multiple cooccurring behaviors, such as food choices and physical activities, where interrelated behaviors are often equally recommended. We propose a novel Mixed-Effects Dynamic hAbit model (MEDA) to simultaneously model multiple related, habitual behaviors. As an illustrative example, MEDA was applied to real-time assessments of sun protection (sunscreen, shade, hat, and protective clothing) reported twice daily by first-degree relatives of melanoma patients who are themselves at an elevated risk of skin cancer. MEDA aims to explicate habits in sun protection under varying environmental cues (e.g., sunny and hot weather). We found consistent between-group differences (e.g., men responding to weather cues more consistently than women) and interactions between cooccurring behaviors (e.g., being in shade discourages sunscreen wearing, more so in men than women). Moreover, MEDA transcends conventional methods to address longstanding challenges—how cue to action and volitional choices differ by groups or even by individual persons. Such nuances in interrelated habitual behaviors are relevant in numerous other applications, such as recommended dietary or physical activity behaviors. These methods best inform personalized behavioral interventions targeting individual preferences for precision behavioral intervention.

Introduction. The impact of renal allograft rejection treatment on infection development has not been formally defined in the literature. Methods. We conducted a retrospective cohort study of 185 rejection (case) and 185 nonrejection (control) kidney transplant patients treated at our institution from 2014 to 2020 to understand the impact of rejection on infection development. Propensity scoring was used to match cohorts. We collected data for infections within 6 months of rejection for the cases and 18 months posttransplant for controls. Results. Abstract Background The use of functional scales is crucial for the management of low back pain. Numerous pertinent outcome measures are available; however, it is unknown how much use Egyptian physiotherapists make of these scales and measurements when diagnosing and treating patients with low back pain. Aim The purpose of this study was to investigate how Egyptian physical therapists working in Egypt used functional outcome measures and scales when treating low back pain. Design and methods A cross-section study used an online web-based survey which was accessible to all Egyptian physiotherapists practicing in Egypt. Results 334 Egyptian physical therapists participated in this study. The use of the pain disability index and back pain functional scale represents the highest frequency of (always/often) of 56.3% and 53.6% respectively, whereas the use of the Quebec Back Pain Disability Scale or the Roland-Morris Disability Questionnaire represents the highest frequency of (rarely/never) 45.8% and 53%, respectively. There was no significant association between gender, years of experience, educational level, and work setting with the use of functional outcome measures (p > 0.05). Conclusion Egyptian physiotherapists more usually utilize the pain disability index and back pain functional scale (PDI and BPFS), when evaluating their patients with low back pain, although they did not commonly use the Roland-Morris Disability Questionnaire or the Quebec Back Pain Disability Scale

Abstract

Automatic speaker identification is done to tackle daily security problems. Speech data collection is an essential but very challenging task for under-resourced languages like Burmese. The speech quality is crucial to accurately recognize the speaker’s identity. This work attempted to find the optimal speech quality appropriate for Burmese tone to enhance identification compared with other more richy resourced languages on Mel-frequency cepstral coefficients (MFCCs). A Burmese speech dataset was created as part of our work because no appropriate dataset available for use. In order to achieve better performance, we preprocessed the foremost recording quality proper for not only Burmese tone but also for nine other Asian languages to achieve multilingual speaker identification. The performance of the preprocessed data was evaluated by comparing with the original data, using a time delay neural network (TDNN) together with a subsampling technique that can reduce time complexity in model training. The experiments were investigated and analyzed on speech datasets of ten Asian languages to reveal the effectiveness of the data preprocessing. The dataset outperformed the original dataset with improvements in terms of equal error rate (EER). The evaluation pointed out that the performance of the system with the preprocessed dataset improved that of the original dataset.

Abstract

Access to plant genetic resources is fundamental to the development of more resilient and nutritious crops. The efficient and effective conservation of plant genetic resources is therefore key to ensuring global food security. There are more than 1750 genebanks around the world, storing various types of plant germplasm including tissue cultures, seeds, embryos and pollen. The method of conservation depends on the seed storage behaviour and propagation requirements of the species. The germplasm of most major crops is conserved as seeds under ‘conventional’ genebank storage conditions, because the seeds tolerate drying to low moisture content and survive storage, perhaps for many decades, at low temperature. Other species are typically conserved as tissue cultures and/or as cryopreserved explants. New technologies for the routine conservation of seeds in genebanks includes evidence-based refining of procedures and the introduction of automation. Similarly, in vitro culture and cryopreservation techniques continue to be developed and improved.
