Disease is a key driver of community and ecosystem structure, especially when it strikes foundation species. In the widespread marine foundation species eelgrass (Zostera marina), outbreaks of wasting disease have caused large-scale meadow collapse in the past, and the causative pathogen, Labyrinthula zosterae, is commonly found in meadows globally. Research to date has mainly focused on abiotic environmental drivers of seagrass wasting disease, but there is strong evidence from other systems that biotic interactions such as herbivory can facilitate plant diseases. How biotic interactions influence seagrass wasting disease in the field is unknown but is potentially important for understanding dynamics of this globally valuable and declining habitat. Here, we investigated links between epifaunal grazers and seagrass wasting disease using a latitudinal field study across 32 eelgrass meadows distributed from southeastern Alaska to southern California. From 2019 to 2021, we conducted annual surveys to assess eelgrass shoot density, morphology, epifauna community, and the prevalence and lesion area of wasting disease infections. We integrated field data with satellite measurements of sea surface temperature and used structural equation modeling to test the magnitude and direction of possible drivers of wasting disease. Our results show that grazing by small invertebrates was associated with a 29% increase in prevalence of wasting disease infections and that both the prevalence and lesion area of disease increased with total epifauna abundances. Furthermore, these relationships differed among taxa; disease levels increased with snail (Lacuna spp.) and idoteid isopod abundances but were not related to abundance of ampithoid amphipods. This field study across 23° of latitude suggests a prominent role for invertebrate consumers in facilitating disease outbreaks with potentially large impacts on coastal seagrass ecosystems.

Relaxor ferroelectric polymers exhibiting a giant electrocaloric effect (ECE) can potentially be used to create next-generation solid-state coolers. Under an electric field, poly(vinylidene fluoride-trifluoroethylene-chlorofluoroethylene) terpolymer goes through a large dipolar entropy change producing a high adiabatic temperature change (ΔTECE). This work resolves the molecular origins of the large entropy change behind the electric field-induced dipole switching. A Fourier transform infrared spectroscopy equipped with a high voltage source is used to operandoly observe the characteristic molecular vibrational modes. A short-range trans (T) conformation of the CF2-CH2 dyads interrupted by a gauche (G) conformation, e.g., TTTG in the terpolymer chain, undergoes a dynamic transformation that leads to a corresponding ΔTECE whenever an electric field is applied. The molecular dynamics simulation also proves that the energy barrier that the transformation from TTTGs into a long T sequence overcomes is smaller than that for all other conformations. A mixed solvent system is used to obtain T3G-enriched terpolymer films exhibiting a 4.02 K ΔTECE at 60 MVm−1 and these films are employed to manufacture a 2-layer-cascaded cooling device that achieves a 6.7 K temperature lift, the highest reported value for a 2-layer cascaded device made of fluoropolymers.

BACKGROUND: Alcohol-associated hepatitis (AH) is one of the clinical presentations of alcohol-associated liver disease. AH has poor prognosis, and corticosteroids remain the mainstay of drug therapy. However, ~40% of patients do not respond to this treatment, and the mechanisms underlying the altered response to corticosteroids are not understood. The current study aimed to identify changes in hepatic protein expression associated with responsiveness to corticosteroids and prognosis in patients with AH. METHODS: Patients with AH were enrolled based on the National Institute on Alcohol Abuse and Alcoholism inclusion criteria for acute AH and further confirmed by a diagnostic liver biopsy. Proteomic analysis was conducted on liver samples acquired from patients with AH grouped as nonresponders (AH-NR, n = 7) and responders (AH-R, n = 14) to corticosteroids, and nonalcohol-associated liver disease controls (n = 10). The definition of responders was based on the clinical prognostic model, the Lille Score, where a score < 0.45 classified patients as AH-R and a score > 0.45 as AH-NR. Primary outcomes used to assess steroid response were Lille Score (eg, improved liver function) and survival at 24 weeks. RESULTS: Reduced levels of the glucocorticoid receptor and its transcriptional co-activator, glucocorticoid modulatory element-binding protein 2, were observed in the hepatic proteome of AH-NR versus AH-R. The corticosteroid metabolizing enzyme, 11-beta-hydroxysteroid dehydrogenase 1, was increased in AH-NR versus AH-R along with elevated mitochondrial DNA repair enzymes, while several proteins of the heat shock pathway were reduced. Analysis of differentially expressed proteins in AH-NR who survived 24 weeks relative to AH-NR nonsurvivors revealed several protein expression changes, including increased levels of acute phase proteins, elevated coagulation factors, and reduced mast cell markers. CONCLUSIONS: This study identified hepatic proteomic changes that may predict responsiveness to corticosteroids and mortality in patients with AH.

Abstract The Sc2.0 global consortium to design and construct a synthetic genome based on the Saccharomyces cerevisiae genome commenced in 2006, comprising 16 synthetic chromosomes and a new-to-nature tRNA neochromosome. In this paper we describe assembly and debugging of the 902,994-bp synthetic Saccharomyces cerevisiae chromosome synXVI of the Sc2.0 project. Application of the CRISPR D-BUGS protocol identified defective loci, which were modified to improve sporulation and recover wild-type like growth when grown on glycerol as a sole carbon source when grown at 37˚C. LoxPsym sites inserted downstream of dubious open reading frames impacted the 5’ UTR of genes required for optimal growth and were identified as a systematic cause of defective growth. Based on lessons learned from analysis of Sc2.0 defects and synXVI, an in-silico redesign of the synXVI chromosome was performed, which can be used as a blueprint for future synthetic yeast genome designs. The in-silico redesign of synXVI includes reduced PCR tag frequency, modified chunk and megachunk termini, and adjustments to allocation of loxPsym sites and TAA stop codons to dubious ORFs. This redesign provides a roadmap into applications of Sc2.0 strategies in non-yeast organisms.

Rehearsals have become an increasingly popular pedagogy to support preservice teachers in developing competency in enacting challenging instructional practices and communicating associated pedagogical commitments. To understand how best to support learners participating in rehearsals, researchers have begun to analyse the facilitation of teacher educators leading such pedagogy. These studies have revealed a variety of ways teacher educators can structure rehearsals. We contribute to this emerging body of knowledge by studying the facilitation of rehearsals in the context of professional development with experienced teachers. Our results expand researchers' understanding of the facilitation of rehearsals in three ways. First, our analysis revealed a range of new ways teacher educators can structure rehearsal interactions. This elaboration and refinement of earlier descriptions offers a variety of new tools for teacher educators to use when leading rehearsals. Second, we categorise these different structures based on their purpose and present three dimensions that contrast the different facilitation choices available. Such an organisation provides a framework that has the potential to aid teacher educators to be more purposeful as they consider the type of interactions they would like to support as well as researchers studying rehearsals to better understand the rationale and effect of such decisions. Third, our reflection about our choices in how we structured interactions when working with experienced teachers presents an example of how the facilitation of rehearsals might be adapted to better respond to the needs and strengths of in-service teachers.

Background: Atrial fibrillation (AF) is a recognized risk factor for mortality after transcatheter aortic valve replacement for severe aortic stenosis, but the impact of different types of AF on clinical outcomes remains unclear. Methods: This retrospective study included 982 patients divided into 3 groups: no AF, paroxysmal AF, and nonparoxysmal AF (persistent or permanent). Clinical outcomes were analyzed using inverse probability weighting and multivariate models. Results: There were 610, 211, and 161 patients in the no-AF, paroxysmal AF, and nonparoxysmal AF groups, respectively. For the entire cohort, the mean (SD) age was 82 (7.7) years, and the periprocedural, 1-year, and 5-year mortality rates were 2.0%, 12%, and 50%, respectively. After inverse probability weighting, the periprocedural mortality rate was higher in the nonparoxysmal AF group than in the no-AF group (odds ratio, 4.71 [95% CI, 1.24-17.9]). During 5 years of follow-up (median [IQR], 22 [0-69] months), all-cause mortality was higher in the nonparoxysmal AF group than in the no-AF group (hazard ratio [HR], 1.56 [95% CI, 1.14-2.14]; P = .006). The paroxysmal AF group was not associated with worse clinical outcomes than the no-AF group (HR, 1.02 [95% CI, 0.81-1.49]) for all-cause mortality. Stroke rates were comparable among the 3 groups. Multivariate analysis also showed increased all-cause mortality in the nonparoxysmal AF group compared with the no-AF group (adjusted HR, 1.43 [95% CI, 1.06-1.93]; P = .018), while all-cause mortality was comparable between the paroxysmal AF and no-AF groups (adjusted HR, 1.00 [95% CI, 0.75-1.33]). Conclusion: In patients undergoing transcatheter aortic valve replacement for severe aortic stenosis, having nonparoxysmal AF was associated with a higher risk of periprocedural and all-cause mortality compared with having no AF. Paroxysmal AF showed no such association.

Abstract: This paper unfolds in three parts. In Part 1, I describe how I have contended with the fraught relationships among mapping, nationalism, and colonialism in my teaching and research. When writing my first book, Contesting Race and Citizenship, I had to reckon with the positivist impulse to enumerate and map Black Italianness—and, more broadly, with the politics of visibility at a time of increasingly virulent racial nationalisms, xenophobia, and outright anti‐Black violence. In Part 2, I describe the ways my own thinking about mapping has been pushed in new directions by insights from Black geographies. Drawing on Clyde Woods, Katherine McKittrick, and Édouard Glissant—as well as essays from the edited volume The Black Geographic—I argue that Black geographic counter‐cartographies foreground insurgent Black spatial knowledges and practices that have always exceeded racial‐spatial violence, and approach mapping as both a material and poetic process. In Part 3, I conclude by advancing some tentative ideas about what it might mean to map new or alternative geographies of abolitionist struggle that attend to the interconnections between Black Atlantic and Black Mediterranean histories of racial capitalism.

INTRODUCTION: Intracranial pressure (ICP) monitoring is a very commonly performed neurosurgical procedure but there is a wide variation in how it is reported, hindering analysis of it. The current study sought to generate consensus on the reporting of ICP monitoring recording data. RESEARCH QUESTION: What should be included in an ICP monitoring report? MATERIAL AND METHODS: The exercise was completed via a modified eDelphi survey. An expert panel discussion was held from which themes were identified and used to produce a code to annotate the transcript of the discussion. Statements were generated for a further two rounds of electronic questionnaires distributed via the REDcap platform. A Likert scale was used to grade agreement with each statement in the survey. A statement was accepted if more than 70% agreement was achieved between respondents. Data was collated using Microsoft Excel and analysed using R. RESULTS: 149 relevant statements were identified from the transcript and categorised into recording parameters, waveform characteristics or reporting. A total of 22 statements were generated for the first round of the survey which was answered by 39 respondents. Following the electronic round of surveys consensus was achieved for all but one statement regarding the acceptability of automating ICP reporting. This was put forward to a second round after which 79% agreement was reached. DISCUSSION AND CONCLUSION: The themes and statements from this eDelphi can be used as a framework to allow the standardisation of the reporting of intracranial pressure monitoring data.

Precise modulation of brain activity is fundamental for the proper establishment and maturation of the cerebral cortex. To this end, cortical organoids are promising tools to study circuit formation and the underpinnings of neurodevelopmental disease. However, the ability to manipulate neuronal activity with high temporal resolution in brain organoids remains limited. To overcome this challenge, we introduce a bioelectronic approach to control cortical organoid activity with the selective delivery of ions and neurotransmitters. Using this approach, we sequentially increased and decreased neuronal activity in brain organoids with the bioelectronic delivery of potassium ions (K+) and γ-aminobutyric acid (GABA), respectively, while simultaneously monitoring network activity. This works highlights bioelectronic ion pumps as tools for high-resolution temporal control of brain organoid activity toward precise pharmacological studies that can improve our understanding of neuronal function.

Reducing methane (CH4) emissions from the oil and gas (O&G) sector is crucial for mitigating climate change in the near term. MethaneSAT is an upcoming satellite mission designed to monitor basin-wide O&G emissions globally, providing estimates of emission rates and helping identify the underlying processes leading to methane release in the atmosphere. MethaneSAT data will support advocacy and policy efforts by helping to track methane reduction commitments and targets set by countries and industries. Here, we introduce a CH4 retrieval algorithm for MethaneSAT based on the CO2 proxy method. We apply the algorithm to observations from the maiden campaign of MethaneAIR, an airborne precursor to the satellite that has similar instrument specifications. The campaign was conducted during winter 2019 and summer 2021 over three major US oil and gas basins. Analysis of MethaneAIR data shows that measurement precision is typically better than 2 % at a 20×20 m2 pixel resolution, exhibiting no strong dependence on geophysical variables, e.g., surface reflectance. We show that detector focus drifts over the course of each flight, likely due to thermal gradients that develop across the optical bench. The impacts of this drift on retrieved CH4 can mostly be mitigated by including a parameter that squeezes the laboratory-derived, tabulated instrument spectral response function (ISRF) in the spectral fit. Validation against coincident EM27/SUN retrievals shows that MethaneAIR values are generally within 1 % of the retrievals. MethaneAIR retrievals were also intercompared with retrievals from the TROPOspheric Monitoring Instrument (TROPOMI). We estimate that the mean bias between the instruments is 2.5 ppb, and the latitudinal gradients for the two data sets are in good agreement. We evaluate the accuracy of MethaneAIR estimates of point-source emissions using observations recorded over the Permian Basin, an O&G basin, based on the integrated-mass-enhancement approach coupled with a plume-masking algorithm that uses total variational denoising. We estimate that the median point-source detection threshold is 100–150 kg h−1 at the aircraft's nominal above-surface observation altitude of 12 km. This estimate is based on an ensemble of Weather Research and Forecasting (WRF) large-eddy simulations used to mimic the campaign's conditions, with the threshold for quantification set at approximately twice the detection threshold. Retrievals from repeated basin surveys indicate the presence of both persistent and intermittent sources, and we highlight an example from each case. For the persistent source, we infer emissions from a large O&G processing facility and estimate a leak rate between 1.6 % and 2.1 %, higher than any previously reported emission levels from a facility of its size. We also identify a ruptured pipeline that could increase total basin emissions by 2 % if left unrepaired; this pipeline was discovered 2 weeks before it was found by its operator, highlighting the importance of regular monitoring by future satellite missions. The results showcase MethaneAIR's capability to make highly accurate, precise measurements of methane dry-air mole fractions in the atmosphere, with a fine spatial resolution (∼ 20×20 m2) mapped over large swaths (∼ 100×100 km2) in a single flight. The results provide confidence that MethaneSAT can make such measurements at unprecedentedly fine scales from space (∼ 130×400 m2 pixel size over a target area measuring ∼ 200×200 km2), thereby delivering quantitative data on basin-wide methane emissions.

Academic advising is a common student success and retention strategy, but most research focuses on student satisfaction rather than traditional measures of student achievement, such as grade point average (GPA). The current study examined how academic advising predicts student GPA. The findings support the idea that Advisor Accountability and Advisor Empowerment are important predictors of student success, along with Study Skills and Perceived Support. Additional analyses revealed that students who scored higher on Study Skills and Perceived Support reported better grade point averages.
