Energy extraction from subsurface reservoirs is important for addressing the increasing energy demand and environmental concerns such as global warming. However, the characterization of subsurface reservoirs, particularly reservoirs dominated by fracture networks remains a challenge due to the lack of means to directly observe subsurface processes. This study explores the feasibility and efficacy of characterizing fracture flow and transport processes in an enhanced geothermal system (EGS) testbed through stochastic tracer modeling. There are two enabling factors that allow application of stochastic modeling to characterize a subsurface reservoir. First, an abundance of geological and geophysical measurements enables the development of a high-fidelity and well-constrained fracture network model. Second, high-performance computing (HPC) allows running massive realizations efficiently. Six conservative tracer tests were stochastically modeled and produced satisfactory realizations that successfully reproduce field tracer recovery data from each tracer test. The evolution of flow and transport processes in the fracture network was then analyzed from these satisfactory realizations. The present study demonstrates that stochastic tracer modeling on a high-fidelity fracture network model is feasible and can provide important insights regarding flow and transport characteristics in subsurface fractured reservoirs.

Prostate cancer has heterogeneous growth patterns, and its prognosis is the poorest when it progresses to a neuroendocrine phenotype. Using bioinformatic analysis, we evaluated RNA expression of neuroendocrine genes in a panel of five different cancer types: prostate adenocarcinoma, breast cancer, kidney chromophobe, kidney renal clear cell carcinoma and kidney renal papillary cell carcinoma. Our results show that specific neuroendocrine genes are significantly dysregulated in these tumors, suggesting that they play an active role in cancer progression. Among others, synaptophysin (SYP), a conventional neuroendocrine marker, is upregulated in prostate adenocarcinoma (PRAD) and breast cancer (BRCA). Our analysis shows that SYP is enriched in small extracellular vesicles (sEVs) derived from plasma of PRAD patients, but it is absent in sEVs derived from plasma of healthy donors. Similarly, classical sEV markers are enriched in sEVs derived from plasma of prostate cancer patients, but weakly detectable in sEVs derived from plasma of healthy donors. Over the last century, northwestern Canada experienced some of the highest rates of tropospheric warming globally, which caused glaciers in the region to rapidly retreat. Our study seeks to extend the record of glacier fluctuations and assess climate drivers prior to the instrumental record in the Mackenzie and Selwyn mountains of northwestern Canada. We collected 27 10 Be surface exposure ages across nine cirque and valley glacier moraines to constrain the timing of their emplacement. Cirque and valley glaciers in this region reached their greatest Holocene extents in the latter half of the Little Ice Age (1600–1850 CE). Four erratic boulders, 10–250 m distal from late Holocene moraines, yielded 10 Be exposure ages of 10.9–11.6 ka, demonstrating that by ca. 11 ka, alpine glaciers were no more extensive than during the last several hundred years. Estimated temperature change obtained through reconstruction of equilibrium line altitudes shows that since ca. 1850 CE, mean annual temperatures have risen 0.2–2.3 ∘ C. We use our glacier chronology and the Open Global Glacier Model (OGGM) to estimate that from 1000 CE, glaciers in this region reached a maximum total volume of 34–38 km 3 between 1765 and 1855 CE and had lost nearly half their ice volume by 2019 CE. OGGM was unable to produce modeled glacier lengths that match the timing or magnitude of the maximum glacier extent indicated by the 10 Be chronology. However, when applied to the entire Mackenzie and Selwyn mountain region, past millennium OGGM simulations using the Max Planck Institute Earth System Model (MPI-ESM) and the Community Climate System Model 4 (CCSM4) yield late Holocene glacier volume change temporally consistent with our moraine and remote sensing record, while the Meteorological Research Institute Earth System Model 2 (MRI-ESM2) and the Model for Interdisciplinary Research on Climate (MIROC) fail to produce modeled glacier change consistent with our glacier chronology. Finally, OGGM forced by future climate projections under varying greenhouse gas emission scenarios predicts 85 % to over 97 % glacier volume loss by the end of the 21st century. Subsurface environments host diverse microorganisms in fluid-filled fractures; however, little is known about how geological and hydrological processes shape the subterranean biosphere. Here, we sampled three flowing boreholes weekly for 10 mo in a 1478-m-deep fractured rock aquifer to study the role of fracture activity (defined as seismically or aseismically induced fracture aperture change) and advection on fluid-associated microbial community composition. We found that despite a largely stable deep-subsurface fluid microbiome, drastic community-level shifts occurred after events signifying physical changes in the permeable fracture network. The community-level shifts include the emergence of microbial families from undetected to over 50% relative abundance, as well as the replacement of the community in one borehole by the earlier community from a different borehole. Null-model analysis indicates that the observed spatial and temporal community turnover was primarily driven by stochastic processes (as opposed to deterministic processes). We, therefore, conclude that the observed community-level shifts resulted from the physical transport of distinct microbial communities from other fracture(s) that outpaced environmental selection. Given that geological activity is a major cause of fracture activity and that geological activity is ubiquitous across space and time on Earth, our findings suggest that advection induced by geological activity is a general mechanism shaping the microbial biogeography and diversity in deep-subsurface habitats across the globe. The identification of natural fractures and the wells they connect is crucial for the development of geological reservoirs because it may have an important impact on reservoir model construction and hydraulic fracture propagation. In this study we investigated the use of a novel data source, the microbial community composition in the reservoir formation fluids, for identification of interwell connectivity caused by natural fractures. We verified this concept at a newly developed mesoscale enhanced geothermal system (EGS) field testbed located 4,850 ft (1,478 m) beneath ground surface at the Sanford Underground Research Facility in Lead, SD. Fluids produced at or near the EGS testbed were sampled and subjected to high‐throughput 16S rRNA gene amplicon sequencing to analyze the microbial community profile therein. Despite the typically substantial heterogeneity across the community profiles of samples spatially distributed (10 m to 1.9 km apart) throughout the site, samples from two wells at the EGS testbed showed highly similar microbial community composition, suggesting the two wells intersected the same natural fracture. This evidence of natural connectivity between the two wells at the EGS testbed was later corroborated by core log analysis and sewer camera surveys into the boreholes. Besides the field case described in this study, microbial community composition as a reservoir diagnostic tool would be applicable in a much broader context such as unconventional hydrocarbon exploitation, groundwater reservoir characterization, and environmental remediation, adding valuable “hard” data capable of pinpointing the origins of fluids unambiguously.
