Abstract

Objective\ud \ud Phaeochromocytomas and paragangliomas (PPGL) are rare, but strongly heritable tumours. Variants in succinate dehydrogenase (SDH) subunits are identified in approximately 25% of cases. However, clinical and genetic information of patients with SDHC variants are underreported.\ud \ud \ud \ud Design\ud \ud This retrospective case series collated data from 18 UK Genetics and Endocrinology departments.\ud \ud \ud \ud Patients\ud \ud Both asymptomatic and disease-affected patients with confirmed SDHC germline variants are included.\ud \ud \ud \ud Measurements\ud \ud Clinical data including tumour type and location, surveillance outcomes and interventions, SDHC genetic variant assessment, interpretation, and tumour risk calculation.\ud \ud \ud \ud Results\ud \ud We report 91 SDHC cases, 46 probands and 45 non-probands. Fifty-one cases were disease-affected. Median age at genetic diagnosis was 43 years (range: 11–79). Twenty-four SDHC germline variants were identified including six novel variants. Head and neck paraganglioma (HNPGL, n = 30, 65.2%), extra-adrenal paraganglioma (EAPGL, n = 13, 28.2%) and phaeochromocytomas (PCC) (n = 3, 6.5%) were present. One case had multiple PPGLs. Malignant disease was reported in 19.6% (9/46). Eight cases had non-PPGL SDHC-associated tumours, six gastrointestinal stromal tumours (GIST) and two renal cell cancers (RCC). Cumulative tumour risk (95% CI) at age 60 years was 0.94 (CI: 0.79–0.99) in probands, and 0.16 (CI: 0–0.31) in non-probands, respectively.\ud \ud \ud \ud Conclusions\ud \ud This study describes the largest cohort of 91 SDHC patients worldwide. We confirm disease-affected SDHC variant cases develop isolated HNPGL disease in nearly 2/3 of patients, EAPGL and PCC in 1/3, with an increased risk of GIST and RCC. One fifth developed malignant disease, requiring comprehensive lifelong tumour screening and surveillance.

Abstract

Background: Lynch syndrome is a rare familial cancer syndrome caused by pathogenic variants in the mismatch repair genes MLH1, MSH2, MSH6, or PMS2, that cause predisposition to various cancers, predominantly colorectal and endometrial cancer. Data are emerging that pathogenic variants in mismatch repair genes increase the risk of early-onset aggressive prostate cancer. The IMPACT study is prospectively assessing prostate-specific antigen (PSA) screening in men with germline mismatch repair pathogenic variants. Here, we report the usefulness of PSA screening, prostate cancer incidence, and tumour characteristics after the first screening round in men with and without these germline pathogenic variants. Method(s): The IMPACT study is an international, prospective study. Men aged 40-69 years without a previous prostate cancer diagnosis and with a known germline pathogenic variant in the MLH1, MSH2, or MSH6 gene, and age-matched male controls who tested negative for a familial pathogenic variant in these genes were recruited from 34 genetic and urology clinics in eight countries, and underwent a baseline PSA screening. Men who had a PSA level higher than 3.0 ng/mL were offered a transrectal, ultrasound-guided, prostate biopsy and a histopathological analysis was done. All participants are undergoing a minimum of 5 years' annual screening. The primary endpoint was to determine the incidence, stage, and pathology of screening-detected prostate cancer in carriers of pathogenic variants compared with non-carrier controls. We used Fisher's exact test to compare the number of cases, cancer incidence, and positive predictive values of the PSA cutoff and biopsy between carriers and non-carriers and the differences between disease types (ie, cancer vs no cancer, clinically significant cancer vs no cancer). Abstract

Deletions on chromosome 15q14 are a known chromosomal cause of cleft palate, typically co-occurring with intellectual disability, facial dysmorphism, and congenital heart defects. The identification of patients with loss-of-function variants in MEIS2, a gene within this deletion, suggests that these features are attributed to haploinsufficiency of MEIS2. To further delineate the phenotypic spectrum of the MEIS2-related syndrome, we collected 23 previously unreported patients with either a de novo sequence variant in MEIS2 (9 patients), or a 15q14 microdeletion affecting MEIS2 (14 patients). All but one de novo MEIS2 variant were identified by whole-exome sequencing. One variant was found by targeted sequencing of MEIS2 in a girl with a clinical suspicion of this syndrome. In addition to the triad of palatal defects, heart defects, and developmental delay, heterozygous loss of MEIS2 results in recurrent facial features, including thin and arched eyebrows, short alae nasi, and thin vermillion. Genotype–phenotype comparison between patients with 15q14 deletions and patients with sequence variants or intragenic deletions within MEIS2, showed a higher prevalence of moderate-to-severe intellectual disability in the former group, advocating for an independent locus for psychomotor development neighboring MEIS2.

Abstract

The Western Central Atlantic (WCA) hosts a population of manta rays whose distribution and habitat preference is poorly understood. Addressing this lack of knowledge will be essential to aid the effective implementation of targeted conservation measures. Here, we use ensemble ecological niche modeling to predict the monthly distribution of manta rays in the WCA using multiple sources of species occurrence data, and consequently identify core areas, which represent suitable habitat in all months of the year. Models suggest a seasonal cycle of expansion and contraction of suitable habitat from coastal areas in spring‐summer, to open ocean in autumn‐winter. Core areas are concentrated along the coast of the USA, Mexico, and Venezuela. Only 24% of predicted suitable habitat is encompassed by the present regional network of MPAs, and is intersected by numerous, heavily trafficked shipping lanes. Manta rays are slow‐growing animals that only produce a limited number of offspring; as such, this species is sensitive to even moderate increases in mortality rates. For this reason, the existing regional network of MPAs may offer them limited protection. As manta rays in the WCA are challenged by a complex landscape of threats and protective measures, future marine conservation planning in the region should explicitly address the conservation of manta rays, focusing on areas identified as key habitats for the species. Abstract

Purpose: Germline WWOX pathogenic variants have been associated with disorder of sex differentiation (DSD), spinocerebellar ataxia (SCA), and WWOX-related epileptic encephalopathy (WOREE syndrome). We review clinical and molecular data on WWOX-related disorders, further describing WOREE syndrome and phenotype/genotype correlations. Methods: We report clinical and molecular findings in 20 additional patients from 18 unrelated families with WOREE syndrome and biallelic pathogenic variants in the WWOX gene. Different molecular screening approaches were used (quantitative polymerase chain reaction/multiplex ligation-dependent probe amplification [qPCR/MLPA], array comparative genomic hybridization [array-CGH], Sanger sequencing, epilepsy gene panel, exome sequencing). Results: Two copy-number variations (CNVs) or two single-nucleotide variations (SNVs) were found respectively in four and nine families, with compound heterozygosity for one SNV and one CNV in five families. Eight novel missense pathogenic variants have been described. By aggregating our patients with all cases reported in the literature, 37 patients from 27 families with WOREE syndrome are known. This review suggests WOREE syndrome is a very severe epileptic encephalopathy characterized by absence of language development and acquisition of walking, early-onset drug-resistant seizures, ophthalmological involvement, and a high likelihood of premature death. The most severe clinical presentation seems to be associated with null genotypes. Conclusion: Germline pathogenic variants in WWOX are clearly associated with a severe early-onset epileptic encephalopathy. We report here the largest cohort of individuals with WOREE syndrome.

Abstract

Animal migration is ubiquitous in nature with individuals within a population often exhibiting varying movement strategies. The basking shark (Cetorhinus maximus) is the world's second largest fish species, however, a comprehensive understanding of their long-term wider-ranging movements in the north-east Atlantic is currently lacking. Seventy satellite tags were deployed on basking sharks over four years (2012-2015) off the west coast of Scotland and the Isle of Man. Data from 28 satellite tags with attachment durations of over 165 days reveal post-summer ranging behaviours. Tagged sharks moved a median minimum straight-line distance of 3,633 km; achieving median displacement of 1,057 km from tagging locations. Tagged individuals exhibited one of three migration behaviours: remaining in waters of UK, Ireland and the Faroe Islands; migrating south to the Bay of Biscay or moving further south to waters off the Iberian Peninsula, and North Africa. Sharks used both continental shelf areas and oceanic habitats, primarily in the upper 50-200 m of the water column, spanning nine geo-political zones and the High Seas, demonstrating the need for multi-national cooperation in the management of this species across its range.

Abstract

Phenotyping of 137 unrelated deletion and reciprocal duplication carriers of the distal 16p11.2 220 kb BP2-BP3 interval showed that these rearrangements are associated with autism spectrum disorders and mirror phenotypes of obesity/underweight and macrocephaly/microcephaly. Such phenotypes were previously associated with rearrangements of the non-overlapping proximal 16p11.2 600 kb BP4-BP5 interval. These two CNV-prone regions at 16p11.2 are reciprocally engaged in complex chromatin looping, as successfully confirmed by 4C-seq, fluorescence in situ hybridization and Hi-C, as well as coordinated expression and regulation of encompassed genes. We observed that genes differentially expressed in 16p11.2 BP4-BP5 CNV carriers are concomitantly modified in their chromatin interactions, suggesting that disruption of chromatin interplays could participate in the observed phenotypes. We also identified cis- and trans-acting chromatin contacts to other genomic regions previously associated with analogous phenotypes. For example, we uncovered that individuals with reciprocal rearrangements of the trans-contacted 2p15 locus similarly display mirror phenotypes on head circumference and weight. Our results indicate that chromosomal contacts' maps could uncover functionally and clinically related genes.

Abstract

Current practice is to report and manage likely pathogenic/pathogenic variants in a given cancer susceptibility gene as though having equivalent penetrance, despite increasing evidence of intervariant variability in risk associations. Using existing variant interpretation approaches, largely based on full-penetrance models, variants in which reduced penetrance is suspected may be classified inconsistently and/or as variants of uncertain significance. We aimed to develop a national consensus approach for such variants within the Cancer Variant Interpretation Group UK (CanVIG-UK) multidisciplinary network.

Abstract

The future persistence of biodiversity is likely to be affected by global climate change. We propose a qualitative framework for assessing the resilience of coastal species to climate change, incorporating current and likely future breeding success, the availability of temporal and spatial climate refugia, the effect of future sea-level rise, behavioural plasticity and the level of other specific threats.We test this framework using one of theworld's largest nesting rookeries of loggerhead sea turtles (Caretta caretta) in the island of Boa Vista (Republic of Cape Verde, West Africa). We measured air, sand and nest temperatures across more than 50 km of nesting habitat and four years to model the likely sex ratios produced and predicted what future sex ratios might be under one conservative future climate change scenario (RCP 2.6). We assessed the relative threat of sea-level rise, and other threats including beachfront construction, pollution and human harvest as well as to estimate the level of foraging behaviour plasticity observed for this population. Estimated sex ratio over the study period was 79.15% female and while an increase of 2 °C air temperature would lead to 99.86% female production, incubation temperature is unlikely to be lethal even in the hottest part of the year. Only half the total suitable length of coastline is currently used for nesting and there is a size-linked dichotomy in foraging strategies of loggerhead turtles of both sexes. Under our qualitative framework,we make the surprising observation that this population of conservation concern should have remarkable resilience to climate change due to some unique features of their habitat use and preferences. This may be compromised by high levels of human harvest and planned and on-going coastal development. The framework may be of utility in assessing the threat of climate change to other climate-sensitive, coastal and mobile species such as migratory seabirds, coastal fish and butterflies.

Abstract

The Benton Visual Retention Test (BVRT) is a neuropsychological assessment of visuo-spatial and visuo-memory ability. Claims of high reliability and validity are based on solitary samples representative of a wide age range. This study validated theBVRTagainst the National Adult Reading Test (NART), a highly validated and reliable test of estimated pre-morbid IQ in an age-specific group of participants (18-35 years).\ud Using Between-subjects factorial design, fifty-three participants (24 female, 29 male) aged 18-35 years (inclusive) were administered the NART and 3 administrations of the BVRT.\ud Significant positive correlations were found betweenBVRTError scores and NART Error scores for administrations B and C of theBVRTwhich is when presented stimuli are followed by a short time delay before allowing respondents to recall. Significant negative correlations were found over these administrations forBVRTCorrect scores and NART Error scores. No significant relationship was found between depression and performance on theBVRT. However, a weak, non-significant relationship was found between anxiety andBVRTperformance.\ud The BVRTis a well-validated and highly reliable neuropsychological test of visuo-spatial and visuo-memory abilities. Abstract

The connectivity and frequency of exchange between sub-populations of migratory birds is integral to understanding population dynamics over the entire species range. True geese are highly philopatric and acquire lifetime mates during the winter, suggesting that the number of distinct sub-populations may be related to the number of distinct wintering areas. In the Bar-headed Goose Anser Indicus, a species found exclusively in Central Asia, the connectivity between breeding and wintering areas is not well known. Their migration includes crossing a broad front of the Himalaya Cordillera, a significant barrier to migration for most birds. Many Bar-headed Geese fly to breeding areas on the Tibetan-Qinghai Plateau (TQP), the highest plateau in the world. From 2005 2008, 60 Bar-headed Geese were captured and marked with satellite transmitters in Nepal (n = 2), India (n = 6), China (n = 29), and Mongolia (n = 23) to examine their migration and distribution. Distinct differences were observed in their migration corridors and timing of movements, including an apparent leap-frog migration pattern for geese from Mongolia. Measurements of geese from Mongolia were larger than their counterparts from China, providing some evidence of morphological differences. Alteration of habitats in China, including the warming effects of climate change on glaciers increasing runoff to TQP wetlands, may be changing goose migration patterns and timing. With the exception of one individual, all geese from Qinghai Lake, China wintered in the southern TQP near Lhasa, and their increasing numbers in that region may be related to the effects of climate change and agricultural development. Thus, our findings document both morphological and geographical variation in sub-populations of Bar-headed Geese, but their resilience to environmental change may be lost if migratory short-stopping results in larger congregations restricted to a smaller number of wintering areas.

Abstract

Birds that migrate across high altitude mountain ranges are faced with the challenge of maintaining vigorous exercise in environments with limited oxygen. Ruddy shelducks are known to use wintering grounds south of the Tibetan Plateau at sea level and breeding grounds north of Himalayan mountain range. Therefore, it is likely these shelducks are preforming high altitude migrations. In this study we analyse satellite telemetry data collected from 15 ruddy shelduck from two populations wintering south of the Tibetan Plateau from 2007 to 2011. During north and south migrations ruddy shelduck travelled 1481 km (range 548-2671 km) and 1238 km (range 548-2689 km) respectively. We find mean maximum altitudes of birds in flight reached 5590 m (range of means 4755-6800 m) and mean maximum climb rates of 0.45 m s-1 (range 0.23-0.74 m s-1). The ruddy shelduck is therefore an extreme high altitude migrant that has likely evolved a range of physiological adaptations in order to complete their migrations. [ABSTRACT FROM AUTHOR]

Abstract

Horror and redemption in Holocaust writing for young adults: Marcus Zusak’s The Book Thief, John Boyne’s The Boy in the Striped Pyjamas. While it has long been thought that the Holocaust is not an appropriate subject matter for young audiences, from The Diary of Anne Frank onwards it has always been part of their reading matter. Never, however, has there been so much interest as in the recent best-selling publications by Zusak and Boyne (the latter of which has been made into a film). This chapter examines the politics of crafting stories for young people about the unspeakable events of the recent past, about who has the right to ‘speak for’ the victims, and whether some genres (for example, fairy stories or fabulism) work best, given the horrific nature of the subject matter.
