Abstract

The distribution of data contributed to the Coupled Model Intercomparison Project Phase 6 (CMIP6) is via the Earth System Grid Federation (ESGF). The ESGF is a network of internationally distributed sites that together work as a federated data archive. Data records from climate modelling institutes are published to the ESGF and then shared around the world. It is anticipated that CMIP6 will produce approximately 20 PB of data to be published and distributed via the ESGF. In addition to this large volume of data a number of value-added CMIP6 services are required to interact with the ESGF; for example the citation and errata services both interact with the ESGF but are not a core part of its infrastructure. With a number of interacting services and a large volume of data anticipated for CMIP6, the CMIP Data Node Operations Team (CDNOT) was formed. The CDNOT coordinated and implemented a series of CMIP6 preparation data challenges to test all the interacting components in the ESGF CMIP6 software ecosystem. This ensured that when CMIP6 data were released they could be reliably distributed.

14. Wave-swept coralliths of Saba Bank, Dutch Caribbean

Abstract

During a recent reef coral survey at the submarine Saba Bank (Eastern Caribbean), an uncommon and diverse assemblage of unattached scleractinian corals (coralliths) was encountered, which has not been reported from the Atlantic before. Four different types of these free-living (unattached) corals were distinguished. They were observed on a relatively flat seafloor (15–20 m deep) with poor coral cover and full exposure to oceanic swell. Much of the substratum was not consolidated and consisted mainly of sand and fragments of branching coralline algae. One of the four types is the (1) anthocyathus stage in the life history of the free-living species Manicina areolata and Meandrina danae. The other three are coralliths formed as ecophenotypic varieties: (2) spheroidal–amoeboidal (= globular and (sub)massive) in Porites astreoides, Siderastrea radians, S. siderea, and Stephanocoenia intersepta; (3) tumbleweed-like (= globular and ramose) in Porites divaricata and P. furcata; and (4) discoidal (flat and circular with short branches) in Madracis decactis and possibly in M. cf. auretenra. This assemblage of free-living corals is likely related to a combination of abiotic factors consisting of wave exposure (swell), depths that waves can reach, a horizontal sea floor with little relief, an unconsolidated substratum, and low coral cover.

16. Transient overshoot extensional rheology of long chain branched polyethylenes: experimental and numerical comparisons between filament stretching and cross-slot flow

Abstract

This work analyses the high-strain extensional behaviour of long chain branched (LCB) polyethylenes, employing two novel extensional rheometer devices, the filament stretching rheometer and the cross-slot extensional rheometer. The filament stretching rheometer uses an active feedback loop to control the imposed strain rate on a filament, allowing Hencky strains of around 7 to be reached. The cross-slot extensional rheometer uses optical birefringence patterns to determine the steady state extensional viscosity from planar stagnation point flow. The two methods probe different strain-rate regimes and in this paper we demonstrate the agreement when the operating regimes overlap and explore the steady state extensional viscosity in the full strain rate regime that these two complimentary techniques offer. For long chain branched materials the cross-slot birefringence images show a double cusp pattern around the outflow centre line (named W-cusps). Using constitutive modelling of the observed transient overshoot in extension seen in the filament stretching rheometer and using finite element simulations we show that the overshoot explains the W-cusps seen in the cross-slot extensional rheometer, further confirming the agreement between the two experimental techniques.

21. Cross-slot extensional rheometry and the steady-state extensional response of long chain branched polymer melts

Abstract

Stress-optical measurements at a flow stagnation point in confined geometries such as the cross-slot provide an elegant way to perform extensional testing for polymer melts. This technique is especially useful for samples which have a steady-state that cannot be reached (easily) in standard elongational rheometry, for example, highly branched polymers which show a non-homogeneous deformation that occurs in stretching experiments for Hencky strains above 4. In contrast to filament stretching, the cross-slot provides one point at which steady-state extensional flow may be sustained indefinitely. In this study, a Cambridge multi-pass rheometer [Coventry, K. D., and M. R. Mackley, J. Rheol. 52, 401-415 (2008)] is used to generate planar elongational flow in a cross-slot geometry for different polyethylene melts. The experimental results are compared to finite element flow simulations using the multi-mode Pompom constitutive equations. The steady-state elongational viscosity at the stagnation point is computed from the flow-induced stress birefringence and the strain-rate determined from numerical calculations of the flow field. We apply this technique to a range of different branched high- and low-density polyethylene melts. This demonstrates both the effectiveness of this technique and shows how the stress distribution in a complex flow depends on molecular structure. Cross slot extensional rheometry therefore provides a very promising technique for parameterizing molecular constitutive equations for LCB melts. © 2011 The Society of Rheology, Inc.

Abstract

The higher relaxivity of gadobenate dimeglumine compared with gadodiamide is potentially advantageous for contrast-enhanced brain MR imaging. This study intraindividually compared 0.1-mmol/kg doses of these agents for qualitative and quantitative lesion enhancement. MATERIALS AND METHODS: Adult patients with suggested or known brain lesions underwent 2 identical MR imaging examinations at 1.5T, one with gadobenate dimeglumine and the other with gadodiamide. The agents were administered in randomized order separated by 3-14 days. Imaging sequences and postinjection acquisition timing were identical for the 2 examinations. Three blinded readers evaluated images qualitatively for diagnostic information (lesion extent, delineation, morphology, enhancement, and global preference) and quantitatively for contrast-to-noise ratio (CNR). RESULTS: One hundred thirteen of 138 enrolled patients successfully underwent both examinations. Final diagnoses were intra-axial tumor, metastasis, extra-axial tumor, or other (47, 27, 18, and 21 subjects, respectively). Readers 1, 2, and 3 demonstrated global preference for gadobenate dimeglumine in 63 (55.8%), 77 (68.1%), and 73 (64.6%) patients, respectively, compared with 3, 2, and 3 patients for gadodiamide (P < .0001, all readers). Highly significant (P < .0001, all readers) preference for gadobenate dimeglumine was demonstrated for all qualitative end points and for CNR (increases of 23.3%-34.7% and 42.4%-48.9% [spin-echo and gradient-refocused echo sequences, respectively] for gadobenate dimeglumine compared with gadodiamide). Inter-reader agreement was good for all evaluations (kappa = 0.47-0.69). Significant preference for gadobenate dimeglumine was demonstrated for all lesion subgroup analyses. CONCLUSION: Significantly greater diagnostic information and lesion enhancement are achieved on brain MR imaging with 0.1-mmol/kg gadobenate dimeglumine compared with gadodiamide at an equivalent dose.

Abstract

Four high resolution atmospheric general circulation models (GCMs) have been integrated with the standard forcings of the PRUDENCE experiment: IPCC-SRES A2 radiative forcing and Hadley Centre sea surface temperature and sea-ice extent. The response over Europe, calculated as the difference between the 2071-2100 and the 1961-1990 means is compared with the same diagnostic obtained with nine Regional Climate Models (RCM) all driven by the Hadley Centre atmospheric GCM. The seasonal mean response for 2m temperature and precipitation is investigated. For temperature, GCMs and RCMs behave similarly, except that GCMs exhibit a larger spread. However, during summer, the spread of the RCMs - in particular in terms of precipitation - is larger than that of the GCMs. This indicates that the European summer climate is strongly controlled by parameterized physics and/or high-resolution processes. The temperature response is larger than the systematic error. The situation is different for precipitation. The model bias is twice as large as the climate response. The confidence in PRUDENCE results comes from the fact that the models have a similar response to the IPCC-SRES A2 forcing, whereas their systematic errors are more spread. In addition, GCM precipitation response is slightly but significantly different from that of the RCMs.

Abstract

Demonstrating the effect that climate change is having on regional weather is a subject which occupies climate scientists, government policy makers and the media. After an extreme weather event occurs, the question is often posed, 'Was the event caused by anthropogenic climate change?' Recently, a new branch of climate science (known as attribution) has sought to quantify how much the risk of extreme events occurring has increased or decreased due to climate change. One method of attribution uses very large ensembles of climate models computed via volunteer distributed computing. A recent advancement is the ability to run both a global climate model and a higher resolution regional climate model on a volunteer's home computer. Such a set-up allows the simulation of weather on a scale that is of most use to studies of the attribution of extreme events. This article introduces a global climate model that has been developed to simulate the climatology of all major land regions with reasonable accuracy. This then provides the boundary conditions to a regional climate model (which uses the same formulation but at higher resolution) to ensure that it can produce realistic climate and weather over any region of choice. The development process is documented and a comparison to previous coupled climate models and atmosphere-only climate models is made. The system (known as weather@home) by which the global model is coupled to a regional climate model and run on volunteers' home computers is then detailed. Finally, a validation of the whole system is performed, with a particular emphasis on how accurately the distributions of daily mean temperature and daily mean precipitation are modelled in a particular application over Europe. Abstract

The patient described had a markedly enhanced response to warfarin when he was given erythromycin concomitantly. Before the initiation of erythromycin, his warfarin anticoagulation had been well controlled. After withdrawal of erythromycin, his response to warfarin returned to normal. These findings and previously reported clinical and laboratory data lead to the conclusion that, at least in some patients, a potentially serious interaction between erythromycin and warfarin can occur.

Abstract

Polymers of septin protein complexes play cytoskeletal roles in eukaryotic cells. The specific subunit composition within complexes controls functions and higher-order structural properties. All septins have globular GTPase domains. The other eukaryotic cytoskeletal NTPases strictly require assistance from molecular chaperones of the cytosol, particularly the cage-like chaperonins, to fold into oligomerization-competent conformations. We previously identified cytosolic chaperones that bind septins and influence the oligomerization ability of septins carrying mutations linked to human disease, but it was unknown to what extent wild-type septins require chaperone assistance for their native folding. Here we use a combination of in vivo and in vitro approaches to demonstrate chaperone requirements for de novo folding and complex assembly by budding yeast septins. Individually purified septins adopted nonnative conformations and formed nonnative homodimers. In chaperonin- or Hsp70-deficient cells, septins folded slower and were unable to assemble posttranslationally into native complexes. One septin, Cdc12, was so dependent on cotranslational chaperonin assistance that translation failed without it. Our findings point to distinct translation elongation rates for different septins as a possible mechanism to direct a stepwise, cotranslational assembly pathway in which general cytosolic chaperones act as key intermediaries.

Abstract

Key Points: Sickle cell disease (SCD) results in cardiopulmonary dysfunction, which may be exacerbated by prolonged exposure to environmental hypoxia. It is currently unknown whether exposure to mild and moderate altitude exacerbates SCD associated cardiopulmonary and systemic complications. Three months of exposure to mild (1609 m) and moderate (2438 m) altitude increased rates of haemolysis and right ventricular systolic pressures in mice with SCD compared to healthy wild-type cohorts and SCD mice at sea level. The haemodynamic changes in SCD mice that had lived at mild and moderate altitude were accompanied by changes in the balance between pulmonary vascular endothelial nitric oxide synthase and endothelin receptor expression and impaired exercise tolerance. These data demonstrate that chronic altitude exposure exacerbates the complications associated with SCD and provides pertinent information for the clinical counselling of SCD patients., Abstract: Exposure to high altitude worsens symptoms and crises in patients with sickle cell disease (SCD). However, it remains unclear whether prolonged exposure to low barometric pressures exacerbates SCD aetiologies or impairs quality of life. We tested the hypothesis that, relative to wild-type (WT) mice, Berkley sickle cell mice (BERK-SS) residing at sea level, mild (1609 m) and moderate (2438 m) altitude would have a higher rate of haemolysis, impaired cardiac function and reduced exercise tolerance, and that the level of altitude would worsen these decrements. Following 3 months of altitude exposure, right ventricular systolic pressure was measured (solid-state transducer). In addition, the adaptive balance between pulmonary vascular endothelial nitric oxide synthase and endothelin was assessed in lung tissue to determine differences in pulmonary vascular adaptation and the speed/duration relationship (critical speed) was used to evaluate treadmill exercise tolerance. At all altitudes, BERK-SS mice had a significantly lower percentage haemocrit and higher total bilirubin and free haemoglobin concentration (P < 0.05 for all). right ventricular systolic pressures in BERK-SS were higher than WT at moderate altitude and also compared to BERK-SS at sea level (P < 0.05, for both). Critical speed was significantly lower in BERK-SS at mild and moderate altitude (P < 0.05). BERK-SS demonstrated exacerbated SCD complications and reduced exercise capacity associated with an increase in altitude. These results suggest that exposure to mild and moderate altitude enhances the progression of SCD in BERK-SS mice compared to healthy WT cohorts and BERK-SS mice at sea level and provides crucial information for the clinical counselling of SCD patients., (© 2018 The Authors. 50. Wave-swept coralliths of Saba Bank, Dutch Caribbean.

Abstract

During a recent reef coral survey at the submarine Saba Bank (Eastern Caribbean), an uncommon and diverse assemblage of unattached scleractinian corals (coralliths) was encountered, which has not been reported from the Atlantic before. Four different types of these free-living (unattached) corals were distinguished. They were observed on a relatively flat seafloor (15-20 m deep) with poor coral cover and full exposure to oceanic swell. Much of the substratum was not consolidated and consisted mainly of sand and fragments of branching coralline algae. One of the four types is the (1) anthocyathus stage in the life history of the free-living species Manicina areolata and Meandrina danae . The other three are coralliths formed as ecophenotypic varieties: (2) spheroidal-amoeboidal (= globular and (sub)massive) in Porites astreoides , Siderastrea radians , S. siderea , and Stephanocoenia intersepta ; (3) tumbleweed-like (= globular and ramose) in Porites divaricata and P. furcata ; and (4) discoidal (flat and circular with short branches) in Madracis decactis and possibly in M. cf. auretenra . This assemblage of free-living corals is likely related to a combination of abiotic factors consisting of wave exposure (swell), depths that waves can reach, a horizontal sea floor with little relief, an unconsolidated substratum, and low coral cover.
